1.Effects of Nitric Oxide on Growth and Metastasis of Tumor

Objective To study the effects of nitric oxide (NO) on the growth and metastasis of tumor.Methods The literatures of recent years were reviewed.Results NO had double effects on the growth and metastasis of tumor. NO promoted the growth and metastasis by regulating the expression of tumor proliferation gene and inducing tumor angiogenesis. On the other hand, NO had antitumor effects by interfering with the metabolism of tumor cells, inducing the damage of DNA, forming high toxic free radical, inducing apoptosis of tumor cells and mediating the antitumor action of endothelial cells and macrophages.Conclusion Selective blockage or induction of synthesis of NO may be a new way for tumor therapy.
2.Comparison of Drug Release in vitro of Diclofenac Sodium Sustained-release Capsules and Sustained-release Tablets from Different Manufacturers

Objective: To establish an HPLC method for the determination of diclofenac sodium sustained-release preparations, and compare the in vitro dissolution of diclofenac sodium sustained-release capsules and tablets from different manufacturers to provide reference for the clinical reasonable medication. Methods: According to the first dissolution method described in Chinese Pharmaco-poeia (2010 edition),the dissolution of each sample was determined by HPLC and the cumulative release percentage was calculated. The dissolution parameters were fitted by Weibull-equation and the results were analyzed. Results:The HPLC method for the determi-nation of diclofenac sodium release was accurate with good reproducibility, and the dissolution parameters of different manufacturers had remarkable difference. Conclusion:The in vitro dissolution of diclofenac sodium sustained-release capsules and tablets from different manufacturers is various, which should be paid attention in the clinical use.
3.The biological effect of exogenetic GM-CSF to dentritic cells in spesis

Objective To investigate the biological effect of GM-CSF on splenic dentritic cells (DCs)when used to treat severe sepsis and the influence on the prognosis.Methods All 160 male Kunming mice were randomly(random number)divided into four groups:control group(n =50),the mice didnt receive special treatment; CLP group(n =42),the mice underwent cecal ligation and puncture;conventional treatment group(n =34),the mice received intraperitoneal injection of 5 mg/kg ceftriaxone at 12,24,36,48,60,72 and 84 h after surgery; GM-CSF treatment group(n =34),the mice received hypodermic injection of 20 μg/kg GM-CSF besides ceftriaxone at 12,36,60,and 84 h after surgery.The mice were sacrificed at 0,12,24,48 and 72 h after CLP by cervical dislocation.The amount of splenic DCs and apoptosis rate were measured by flow cytometry,and the serum concentrations of IL-12 were detected by ELISA in each group.Meanwhile the survival prognosis was observed at 96 h.Results At every time point,the apoptosis of splenic DCs increased and the amount markedly reduced in severe sepsis(P <0.05),the serum concentration of IL-12 increased on an one-way curve type(P <0.05).The treatment of cephalosporin exacerbated the loss of DCs(P < 0.05),while hadnt any effect on IL-12(P > 0.05).GMCSF treatment increased the amount of DCs(P < 0.05),and decreased IL-12 concentrations(P < 0.05).The OR of GM-CSF was 0.079 computed by Logist regression analysis.Conclusions GM-CSF treatment severe sepsis can increase the amount of splenic DCs,decrease serum levels of IL-12,and improve prognosis.

The application of MNIS have played an important role in confined the nursing profession, disease management and pharmaceutical management, more users discover problem from different professional perspective, thus the MNIS should be improved for all-round care. This article is consist of the domestic and care information systems development, application and effect, as well as its application in the clinical factors of its research progress in clinical application were reviewed to provide a basis for the development of leadership and decision-making about MNIS, and thus, enhance the mobile application of the effectiveness of care information systems and promoting their improvement.

6.The Study of the Feasibility of Improved Laparoscopic Common Bile Duct Exploration

Objective To explore the clinical value and feasibility of the open bile duct operative instruments in laparoscopic common bile duct exploration.Methods 43 operations of laparoscopic common bile duct exploration were accomplished by using open bile duct operation instruments for exploring common bile duct and removing stones without choledochofiberscopy.Results 43 operations of laparoscopic common bile duct exploration were achieved successfully by exploring common bile duct and removing stones with open bile duct operative instruments with no postoperative remanent stones of the common bile duct and no open operation was needed.The follow-up in 38 patients for 6 months showed that there was no postoperative complication.Conclusions This method has the advantages of convenient manipulation,safety and better effect,low cost.It was deserved to be extended.

Objective To study the effects of motor imagery therapy on functional performance of upper ex- tremity in stroke patients.Methods Sixty cases of stroke were divided into a treatment group(n=30) and a con- trol group(n=30).The control group was treated with routine rehabilitation training,while the treatment group was treated with motor imagery therapy in addition to the routine rehabilitation training.The Fugl-Meyer Assessment (FMA),upper extremity function test(UEFT)and simple test for evaluating hand function(STEF)were used to as- sess the upper extremity function before and after six months of treatment.Results After treatment,patients in both groups scored significantly better with the FMA scales,UEFT scales and STEF scales when compared to their scores obtained before the treatment(P
8.Inducible Nitric Oxide Synthase and p53 Expression in Hepatocellular Carcinoma and Their Relationship with Angiogenesis

Objective To investigate the expression of inducible nitric oxide synthase (iNOS) and p53 protein in hepatocellular carcinoma (HCC) and their relationship with angiogenesis. Methods Immunohistochemical method and image analysis technique were used to detect the expression of iNOS and p53 protein in tumor tissue sections of 59 HCC patients. Microvessel density (MVD) was counted by immunohistochemical staining with anti CD34 antibody.Results ①The expression rates of iNOS and p53 were 81.4%(48/59), 64.4%(38/59) in HCC patients, respectively. 9.shRNAs Aiming at Glycosyltransferase Inhibit Invasive and Proliferative Ability of LoVo Cell Line in vitro

To construct expression vectors of small hairpin RNA aimed at N-acetylglucosaminyltransferase Ⅴ(GnT-Ⅴ) gene, and to investigate effects of GnT-Ⅴ shRNA on proliferation, adhesion, migration and invasion of LoVo cell line. siRNAs were designed according to the coding sequence of GnT-Ⅴ gene, shRNA expression vectors were constructed and transfected into LoVo cell line, cell lines which stably expressed low level of GnT-Ⅴ were established by G418 screening. The mRNA and protein expression of GnT-Ⅴ were measured by semi- quantitative reverse transcription polymerase chain reaction(RT-PCR) and Western blot analysis, respectively. The effects of pGPU6/GFP/Neo GnT-Ⅴ shRNA vectors on proliferation, adhesion, migration and invasion of LoVo cell line were evaluated by CCK-8 assay, heterogenous adhesion, wound closure assay, chemotactic migration and cell invasive experiment, respectively. GnT- Ⅴ shRNA expression plasmid was constructed successfully and pGPU6/GFP/Neo GnT-Ⅴ shRNA down-regulated expression of GnT-Ⅴ dramatically in LoVo cell. Expression of LoVo GnT-Ⅴ/1564 and LoVo GnT-Ⅴ/2224 dereased by 82%, 71.5% respectively at mRNA level, and 68%, 56% respectively at protein level. The more effective interfered cell line, LoVo GnT-Ⅴ/1564, was chosen to do further experiment. mRNA display provides a new powerful tool for in vitro selecting of peptides and proteins.In the selecting process,peptides are covalently attached to their own mRNA to form mRNA-protein fusions.These mRNA-protein fusions enable in vitro selection of peptide and protein libraries of more than 1013 different sequences.The experiment conditions and protocols have been optimized in recent years.The application of mRNA display technology is mainly in the discovery of ligands for many kinds of target molecules and the analysis and mechanism elucidation on interaction between proteins.With its great potential,there will be a wide application foreground in the application of mRNA display.
