4.Expression of Endothelin in Kidneys of Rats with Unilateral Ureteral Obstruction

Objective To determine the expression of endothelin in kidneys of rats with unilateral ureteral obstruction (UUO). Methods Rats were evenly divided into two groups. Some were subjected to complete ligation of left ureter and the others with sham operation were used as control group.Four weeks later, we examined the mRNA expressions of ET-1 in kidneys of all these rats using reverse transcription-polymerase chain reaction (RT-PCR) method.The immunohistochemistry analysis was carried out to investigate the protein level of ET-1. The urinary concentration of ET-1 was determined by radioimmunoassay.Results In UUO group, the urinary concentration of ET-1 increased substantially;both gene expression and protein level of ET-1 were much higher than those in control group. At the same time, tubulointerstitial fibrosis was present in the kidneys of rats with UUO. Conclusion ET-1 is involved in the pathogenesis of the injury of UUO.

ObjectiveTo explore the renal functions of vasopressin-deficient Brattleboro(BB) rats by using different concentrated Gadolinium-diethylenetetramine pentaacetic acid(Gd-DTPA) dynamic enhanced magnetic resonance imaging(MRI).MethodsThe study included 14 BB rats(male rats of 3 month-old) and 14 normal male Wistar rats used as control group.Dynamic MRI was performed by using either a low dosage(0.05 mmol/kg) or a high dosage of Gd-DTPA(0.5 mmol/kg).Data of 0-60 min renal cortex,medulla and pelvic were obtained after using contrast medium.MRI of kidneys at different time was analyzed and the mean relative signal intensity(RSI) was measured.Then the RSI curves of different groups were marked.Data of each group were caculated separately by SPSS 11.0 software.ResultsThe findings demonstrated that RSI curves of the vasopressin-deficient kidneys showed different patterns as compared with those of the control group(P
6.Six to Ten Years Follow-Up Survey of Triancinolone Acetonide Therapy in Children with Nephrotic Syndrome

Objective To observe the long term effects of triancinolone acetonide(T-A)as a substitution for prednisone,a maintenance therapy in treating children with nephrotic syndrome(NS).Methods Twenty-five children with idiopathic NS(10 children with simple type NS and 15 children with nephritic type NS).Prednisone treatment for 4-8 weeks would bring remission,and immunosupression was added if remission failed.Then oral prednisone was reduced to 2 mg/kg per alternate day for 2 weeks,followed by a small dose of T-A(0.5-1.0 mg/kg)intramuscular injection,once a year per month for the first year,and every other month for the second year,as for the 8 frequently relapse cases,it would be every 3 months for the third year,other 17 cases stopped T-A intramuscular injection in the third year.Two weeks after T-A treatment stopped using prednisone.The relapse and the side-effects was observed through the observation of 25 NS children's clinical manifestation,routine urine test,and biochemistry variation before and during the T-A treatment,along with a 6 to 10 years follow-up survey after T-A therapy stopped.Statistical analysis was performed by using SPSS 10.0 software.Results 1.Median dose of T-A was 0.97 mg/kg per time,total doses of T-A was 18.1 mg/kg(13.5-24.4 mg/kg).2.There were 16 relapse cases by prednisone treatment,while 7 relapse cases by T-A treatment for 2 years,there existed a significant comparison(P0.05).4.There were only 4 relapsed cases in the 6-10 years of follow-up observation,the difference was sharp compared with the 16 relapsed cases before T-A treatment(P
7.Protective Effects of Bosentan on Kidney in Rats with Unilateral Ureteral Obstruction

Objective To research the effects of bosentan in on unilateral ureteral obstruction (UUO) rats. Methods Wistar rats were randomly divided into three groups. UUO group was subjected to complete ligation of left ureter, sham operation group with sham operation was used as control. Bosenten group was subjected to complete ligation of left ureter and to stomach with bosentan suspension. Three groups were killed at 5,10,15,20 d. We examined renal pathological changes and the expressions of transforming growth factor-?1 (TGF-?1) in kidney of all rats using immunohistochemistry analisis method. The plasma and ureteral obstructive kidney urinary concentration of endothelin - 1 (ET - 1) was determined by radioimmunoassay. Results In Bos and UUO groups, the urinary concentration of ET - 1 increased substantially and the expression of TGF - ?1 were much higher than those in the S group. At the same time.tubulointerstitial fibrosis was present in the kidney of rats in Bos and UUO group. But the expression of TGF- ?1 and tubu-lointerstitial fibrosis in Bos group were lower than those of UUO group. Conclusion Bosentan can effectively amelirate tubulointerstitial fibrosis in rats with UUO by ET and TGF - ?1 machanism.

10.Application Investigation of Xiyanping Injections in Bone Surgery Department Inpatients in a Certain Hos-pital

Objective:To investigate the application of Xiyanping injections ( XYP) in the bone surgery department inpatients in a certain hospital to provide reference for the clinical rational medication. Methods:A retrospective investigation was adopted to randomly survey 180 hospitalized medical recorders with XYP from a certain hospital in 2013. The usage, dosage and medication purpose of XYP were analyzed. Results:The reasonable rate of usage and dosage of XYP was 97. 2%. Among the surgical patients, 82. 8% were giv-en XYP, and among them, 97. 3% was accordance with the medication purpose. Among the nonsurgical patients, 17. 2% were given XYP, and among them, 71. 0% was accordance with the medication purpose. The alone-use rate was 60. 0% and the rate of combina-tion with antibacterials was 40. 0%. The longest treatment course was 20d, the shortest one was 1 d, and the mean value was (7. 7 ± 5. 3) d. Conclusion:Irrational use of XYP exists in clinics, and the use should be strengthened and regulated.
