1.Effect and mechanism of intravenous bone marrow stromal cells transplantation against chronic ethanol-induced cognitive dysfunction in rats

Objective To investigate the effect and mechanism of intravenous bone marrow stromal cells (BMSCs)transplantation against chronic ethanol-induced cognitive dysfunction in rats.Methods BMSCs from rats were isolated,cultured and identified in vitro.Male Sprague-Dawley rats(n=28)were randomly divided into control group(n=7),chronic ethanol group(n=7),BMSCs group(n=7)and PBS group(n=7).BMSCs(1×10~7 cells/animal)or PBS were intravenously injected to the rats of BMSCs or PBS group at the 7th week after ethanol gavage,respectively.All rats were sacrificed at the end of the 8th week.The learning and memory behavior was assessed using Moms water maze task.Immunofluorescence was used to examine cell proliferation maker(BrdU)in hippocampal dentate gyrus.Immunohistochemistry was used to evaluate NGF and BDNF expression in hippocampal region.Results In Morris water maze,escape latency at each time point in BMSCs group((33.83±3.26)s,(13.14±2.64)s,(6.53±0.98)s,(4.45±0.85)s)were significantly shorter than those in chronic ethanol group((53.78±4.29)s,(36.53±2.39)s,(18.28±2.13)s,(8.67±0.46)s,all P<0.01))and time percentage of platform quadrant in BMSCs group((73.99±1.49)%) was dramatically longer than that in chronic ethanol group((51.25±3.69)%),but no significant difference with contro1.The number of BrdU-positive cells in the dentate gyrus was significantly higher in BMSCs group than that in chronic ethanol group(P<0.01).Comparing to chronic ethanol gavage,BMSCs transplantation significantly increased the expression of NGF alld BDNF (P<0.01)in hippocampus.Conclusion Intravenous BMSCs transplantation is able to result in enormous attennation of the learning and memory abnormalities in chonic alcoholism rats,and which might be involved to promote hippocampal endogenous cellular proliferation by secretary neurotrophic factors.
2.Fall risk assessment and relative interventions in the elderly: A latest advances

Fall is one of major causes of disability, reduced mobility, shortness of life span and increased medical costs in the elderly.There are multiple risk factors for falls in the elderly, but they can be preventable and controlled.This article reviews the evidences regarding the clinical effectiveness, practicalness and security of these fall risk assessment methods and measures of interventions by evidence-based medicine in the elderly and analyzes their research directions.
5.Effect of ginsenoside total saponinon on regulation of P450 of livers of rats after γ-ray irradiation.

Effect of ginsenoside total saponin (GTS) on the regulation of P450 of livers of rats after γ-ray irradiation was studied. Rats were irradiated by the ⁶⁰Coγ-ray for one-time dose of 5.5 Gy, dose rate of 117.1-119.2 cGy. The cocktail probe, qPCR and Western blot were used to detect expression of enzymatic activites, mRNA and protein of rats. Contrasted with blank group, expression of CYP1A2, 2B1, 2E1, 3A4 of irradiation group showed a up-regulated (P < 0.05). Contrasted with irradiation group, exprression of CYP1A2, 2B1, 2E1, 3A4 of GTS group showed a downward trend. GTS had negative agonistic action against expression of P450 of rats by irradiatied.

8.Analysis of prognostic factors for severe multiple trauma patients in ICU

Objective To explore the risk factors in the prognosis of patients with severe multiple trauma related factors.Methods Ninety-three patients with severe multiple trauma(ISS score≥16 points) were selected as our subjects,who all hospitalized in the second hospital affiliated to Dalian medical University.Among of which,38 cases' ISS score was 16-25,and 55 cases' ISS score was higher and equal to 25.Clinical data and general information were recorded.ISS score,APACHE Ⅱ score and 6 h lactate clearance rate score were measured.Results Twenty-seven dead among these 93 cases and the mortality was 29.03%.There were significant differences in terms of ISS score,APACHE Ⅱ score,lactate level when entering ICU,6 h lactate level and 6 h lactate clearance rate between death patients (27 cases) and survival patients (66 cases) (t =9.846,9.812,7.112,7.012,9.831 ; P < 0.001).In multiple organ dysfunction syndrome (MODS) patients,the mortality of ISS score:16-25 group was different from that of ISS score≥25 (15.78% (6/38) vs.35.18% (21/93),x2 =21.52,P <0.001).Among the 93 cases of severe multiple trauma,35 cases with MODS(37.6% (35/93)),and 20 cases of which were dead(57.1% (20/35)).There was statistic difference between MODS group(35 cases) and non-MODS(58 cases) in term of mortality(37.6% (35/93) vs.12.1% (7/58) ;x2 =21.52,P < 0.001).Multivariate logistic regression analysis showed that the ISS score,APACHE Ⅱ score,6 h lactate clearance rate and MODS were independently prognostic correlation factors (P < 0.05).Operating characteristic curve (ROC) showed that AUC of ISS score was 0.735 (95 % CI 0.629-0.842),of APACHE Ⅱ score was 0.888 (95% CI 0.821 =0.956),of 6 h lactate clearance rate score was 0.951 (95% CI 0.906 -0.997).Conclusion The factors of ISS score,APPCHE Ⅱ assessment,6 h lactate clearance rate,the occurrence of MODS are independent prognostic factors.
9.EFFECT OF CHICKPEA PROTEIN HYDROLYSATE ON ANTIOXIDANT ABILITIES IN AGED MICE INDUCED BY D-GALATOSE

Objective To study the effects of chickpea protein hydrolysate(CPH) in different doses on of antioxidant abilities in serum,liver and heart in aged mice induced by D-galactose .Method The subacute aged model mice were made by neck back subcutaneous injection of D-galactose continually.The content of malondialdehyde(MDA) and activity of superoxide dismutase(SOD) and glutathione peroxidase(GSH-PX) in the serum,liver and heart of the mice were tested.Results The SOD and GSH-Px activities in aged model group were significantly decreased and MDA significantly increased compared with normal mice;CPH in different doses evidently increased the SOD and GSH-Px activities and reduced the MDA level in serum,liver and heart in mice.Conclusion CPH can obviously increase the antioxidant ability of mice and has certain anti-aging effects,especially the high dose .
10.Median effective target effect-site concentration of sufentanil inhibiting responses to insertion of laryngeal mask airway when combined with propofol in elderly patients

Objective To determine the median effective target effect-site concentration (EC50) of sufentanil inhibiting responses to insertion of laryngeal mask airway (LMA) when combined with propofol in the elderly patients.Methods ASA physical status Ⅰ or Ⅱ patients,aged 66-75 yr,with body mass index of 19-30 kg/m2,scheduled for elective transurethral resection of prostate under general anesthesia,were included in this study.Anesthesia was induced with propofol given by target-controlled infusion.The initial target effect-site concentration (Ce) of propofol was 1 μg/ml,and the Ce was increased by 0.5 μg/ml every 1 min until it was increased to 2 μg/ml and maintained at this level for 5 min.Sufentanil was simultaneously administered by target-controlled infusion.The target Ce of sufentanil was 0.3 ng/ml in the first patient.The concentration of sufentanil was determined using the modified Dixon's up-and-down method.After the patients lost consciousness,LMA was inserted after the target effect-site and plasma concentrations were balanced.The insertion response was defined as positive when MAP and HR increased by 20% of the baseline value within 5 min after insertion of LMA.The Ce of sufentanil was increased/decreased in the next patient if the insertion response was positive or negative.The ratio between the two successive concentrations was 1.2.The EC50 and 95 % confidence interval of sufentanil blunting responses to insertion of LMA was calculated using Probit analysis.Results When combined with propofol,the EC50 (95% confidence interval) of sufentanil blunting responses to insertion of LMA was 0.160 (0.130-0.188) ng/ml.Conclusion The EC50 of sufentanil inhibiting responses to insertion of LMA is 0.160 ng/ml when combined with propofol in the elderly patients.
