1.Expressions of focal adhesion kinase and matrix metalloproteinase-9 in pituitary adenomas

OBJECTIVE To explore the expression and its significance of focal adhesion kinase(FAK)and matrix metalloproteinase 9(MMP-9)in the pituitary adenomas. METHODS The expressions of FAK and MMP-9 were determined by immunohistochemical technique in 50 pituitary adenoma tissues samples obtained during operation, including 27 invasive pituitary adenomas and 23 non-invasive pituitary adenomas. The relationship of FAK and MMP-9 expression with tumor invasiveness were analyzed. 2.Optimization of Preparation Formulation of Tetrandrine Chitosa Microspheres by Central Composite Design

OBJECTIVE:The preparation of microspheres was optimized by central composite design in order to improve the lung targeting of tetrandrine chitosa microspheres. METHODS:Microspheres were prepared using an emulsion-chemical crosslink technique.Effects of three independent variables i.e. weight percent of tetrandrine and christon, volume percent of water phase and organic phase and christon concentration in aqueous phase were investigated on four response variables.Response variables selected in the research were yield,drug loading, envelop efficiency, mean diameter and span of dispersity.Second-order polynomial and linear equations were fitted to the data,and the resulting equations were used to produce three dimensional response surface graphs,by which optimal experimental conditions were selected. RESULTS:Five response variables were found to be dependent on three independent variables. According to optimal experimental conditions.An optimized formulation contained weight percent of tetrandrine and christon was 61.97%, volume percent of water phase and organic phase was 13.51% and christon concentration in aqueous phase was 2.37%. CONCLUSION:The central composite design can be used to optimazation of preparation formulation,diameter of the microspheres optimized by it can meet the demands of lung-targeting.
4.Resistance Analysis of Pseudomonas aeruginosa

OBJECTIVE To investigate the distribution and drug resistance of Pseudomonas aeruginosa.METHODS All specimens isolated and cultured from patients in our hospital were identified by using the automatic microorganism analyzer WalkAway-40,Dade Behring made in America,and bacteria′s drug susceptibility test and identifications were performed on strains using NC21 Microscan Panel.RESULTS From sixty-two strains of P.aeruginosa 36 strains were isolated of sputum.The resistance rate to the third generation of cephalosporins cefotaxime and ceftriaxone was 55.6% and 47.5%,respectively.The resistance rate to the other ?-lactamases antibiotics such as ceftazidime was 7.5%,piperacillin/tazobactam 11.0%,cefepime 14.8% and penicillin was 30.0%,the ratio of resistance for imipenem was 20.0%,the lowest one was amikacin(4.6%).CONCLUSIONS P.aeruginosa is one of the main bacteria in the lower respiratory tract infection.The drug-resistant mechanism of P.aeruginosa is very complex,including multidrug resistance characteristics,and it is originally resistant to several antibiotics.To avoid being produced ?-lactamases and result in resisting drug widely,the antibiotics should be selected according to low drug-resistant rate and taking into account sufficiently its drug resistance mechanism in the treatment of P.aeruginosa infection.

6.Establishment of gene expression profile and regulation of genes expression during plasma cell differentiation

Objective To study the genes modulated during plasma cell development and their regulating network. Methods The changes of gene expression during differentiation from mature B cells to plasma cells were studied using cDNA microarray and RT PCR. Results The gene expression profile during plasma cell differentiation was established. During mature B cell plasma cell differentiation, numerous genes were modulated, including genes involving immunity, cell growth, survival, and signal transduction. The expressions of several molecules in the B cell antigen receptor (BCR) signaling pathways, such as Ig?, Ig?, Lyn, Syk, BLNK, SWAP 70, and SHP 1 were down regulated. The decreased expression of some of these molecules might be associated with the down regulation of the transcription factor BSAP. Conclusion During plasma cell differentiation, molecules involving BCR signaling pathways are down regulated.
7.Correlation between plasma mitochondrial coupling factor-6 level in coronary circulation and coronary stenotic degree in CHD patients

Objective: To study correlation between plasma mitochondrial coupling factor-6 (MCF-6) level in coronary circulation and coronary stenotic degree in CHD patients. Methods: A total of 64 CHD patients were selected as CHD group, and they were further divided into single-vessel group, double-vessel group and multi-vessel coronary disease group; another 20 cases with normal coronary artery were enrolled as normal control group. Radioimmunoassay method was used to measure plasma MCF-6 concentration in peripheral, coronary sinus and aortic root in each group. Multivariant linear gradual regressive analysis was used to analyze above-mentioned correlation. Results: Compared with normal control group, there was significant increase in plasma MCF-6 concentration in peripheral, aortic root and coronary sinus in CHD patients (P<0.05 all) ;compared with aortic root and peripheral plasma, there was significant increase in MCF-6 concentration [(348±48) pg/ml vs. (340±51) pg/ml vs. (402±56) pg/ml, P<0.01 all] in coronary sinus of CHD group;Compared with single-vessel group, double-vessel group, there were increase in MCF-6 concentration in multi-vessel coronary disease group (P<0.05~0.01); Multivariant linear gradual regressive analysis indicated that Gensini score was significant positively correlated with plasma MCF-6 concentrations in coronary sinus and aortic root (r=0.650, P<0.01; r=0.711, P<0.01). Conclusion: Mitochondrial coupling factor-6 participates in the pathophysiological process of coronary heart disease, which may be an important vasoactive substance during development of coronary heart disease.
8.Effects of benazepril on inflammatory cytokines in patients with primary hypertension

Objective To investigate the effects of Benazepril on inflammatory cytokines in patients with primary hypertension and its efficacy analysis.Methods 72 patients with mild to moderate primary hypertension were randomly divided into benazepril group(37 cases)and control group(35 cases).The control group patients were given felodipine sustained release tablets Stag,1 times daily.Benazepril group were given benazepril tablets 10mg,1 times daily.Both groups were treated for 8 weeks.Changes in blood pressure and heart rate,plasma inflammatory cytokine levels and the adverse reactions were observed.Results After 8 weeks of treatment,the patients' blood pressures were significantly decreased (t =2.87,2.43,2.31,2.23,P < 0.05 or P < 0.01) than before in both groups.And the decrease in the benazepril group was more obvious than that in the control group (t =2.21,2.13,all P < 0.05),while the patients' heart rates in the two groups showed no significant change (P > 0.05) ; Patients' plasma IL-4 and IL-8 levels decreased significantly,plasma IL-10 levels increased significantly (t =2.99,2.87,3.21,2.13,2.19,2.23,all P<0.05 or P<0.01),and the magnitude of decrease or increase in the benazepril group were more obvious compared with the control group (t =2.21,2.13,2.34,all P < 0.05).Conclusion Benazepril in the treatment of primary hypertension has good antihypertensive effect and security which can significantly reduce the plasma levels of proinflammatory cytokines IL-4 and IL-8 levels,improve the anti-inflammatory cytokine IL-10 levels,and has the role of suppressing the inflammatory response of the vessel wall.

10. Toxicity of Danshen injection in Beagle's dogs by repeated iv injection

Objective: To re-evaluate the potential toxicity of Danshen Injection (DI) in Beagle's dogs by repeated iv injection. Methods: DI was iv given to the dogs at the doses of 0, 1.6, 5.4, and 16.0 g/(kg·d) (4 per sex per group) for 13 weeks. During the test period, the clinical signs, mortality, body weights, food consumption, rectal temperature, ophthalmoscopy, electrocardiography, urinalysis, hematology, serum biochemistry, organ weights, gross findings, and histopathology were examined. Results: Dogs iv given with DI at the doses of 0, 1.6, 5.4, and 16.0 g/(kg·d) for 13 weeks had no drug-related changes in mortality, body weight, food consumption, temperature, electrocardiography, ophthalmoscopy, urinalysis parameters, and organ weights. The hematological parameter data showed a significant decrease in red blood cells and hemoglobin concentration in the high-dose group and a significant increase in activated partial thromboplastin time suggesting an effect on haemopoiesis. For biochemical parameters, a significant decrease in glucose and a significant increase in total bilirubin were observed in the high-dose group, and the latter was considered to be toxicologically insignificant as lack of histopathological correlate. However, the histopathological examinations of the injection site showed that DI could cause dose-dependent focal inflammation. Conclusion: That the iv injection with DI into dogs at 16 g/(kg·d) for 13 weeks could cause the decreases in red blood cell parameters and glucose, as well as the lesions of the injection site. The no observed adverse effect level is 5.4 g/(kg·d), which suggests that safe clinical dosing be possible. © 2013 Tianjin Press of Chinese Herbal Medicines.
