1.Application of flow-through hybrization and gene chip on human papillomavious subtype detection in clinical

Objectives To evaluate the application of flow-through hybrization and gene chip (HybriMax) on human papillomavious (HPV) subtype detection in clinical.Method 3 086 sexually active females from out-patient department were selected for HPV subtype analysis using HybriMax. Cervical tissue samples were taken under the colposcope from 463 females who had cervical lesion for pathological analysis. The predictive value of HybriMax in cervical abnormality was compared with pathological results, which were used as a golden standard. DNA sequence in HPV E6/E7 region was performed among 80 females with HPV 16 positive by HybriMax to determine the accuracy of HybriMax.Results All 21 different subtypes were found and total HPV positive rate was 63.1%(1 947/3 086). Among the 21 HPV subtypes, 5 of them had a infection rate over 5% and they were HPH16(15.9%, 490/3 086), 58(11.2%, 346/3 086), 52(8.5%、261/3 086)、33(6.3%、195/3 086)、53(6.2%、192/3 086)、6(5.6%、172/3 086), and CP8304(5.0%、155/3 086). The sensitivity for high degree squamous intraepithelial dysplasia(HSIL, CINⅡ+CINⅢ) by HybriMax was 95.49%(95%CI=95.44%~98.27%), while specificity was 34.85%(95% CI= 32.59%~37.57%)、Positive predictive value for HSIL was 37.13%(95%CI=30.79%~40.96%),while negative predictive vale was 95.04%(95%CI=89.24%~98.44%). Eighty sequence results in E6/E7 region completely matched to HybriMax results.Conclusion HybriMax has a high accurate rate in HPV subtype diction with good sensitivity and specificity for HSIL and above. It is an effective method to detect HPV subtype in clinical.

Objective To explore an efficient way to modulate the expression of estrogen receptor (ER) ? and ?, and to build up a model of endometrial cancer cell expressing predominantly one isoform of ER and to verify the roles of ER ? and ? in the tumorigenesis of endometrial cancer associated with estrogen and tamoxifen (TAM). Methods A series of oligodeoxyribonucleotides (ODN) against ? or ? regions of ER ? or ? were synthesized and tested in human endometrial cancer cell lines (Ishikawa) that express functional ER ? and ?. The expressions of two ER isoforms were detected by western blot using specific antibodies. Then we studied the change of Ishikawa proliferation in response to 17?-estradiol and TAM under the influence of antisense ODN. Results (1) Transfection with antisense ODN directed against the ER? and ER? could significantly inhibit target protein production. (2) 17?-estradiol could increase the proliferation of Ishikawa cells, but they lost the ability to proliferate in response to 17?-estradiol after transfected with ER? antisense ODN especially at hours 24, 48 and 72 ( P
4.Clinical analysis of pregnancy complicated with cervical epithelial neoplasia-18 cases

Objective To discuss the diagnosis,treatment and following-up of cervical intraepithelial neoplasia (CIN) during pregnancy.Methods Eighteen pregnant women with CIN,presented to the hospital from Jan.2004 to May.2008,were retrospectively reviewed.Pap smear,HPV,copolscopy and cervical biopsy were performed for diagnosis and all were followed up by Pap smear and copolscopy during pregnancy every 12 weeks,while 15 were followed up until 6-8 weeks postpartum.Results The incidence of CIN during pregnancy was 0.16%.All of the 18 women had abnormal Pap smear,followed by colopscopy and biopsy.Conservative management during the pregnancies every 3 months showed no progress in CIN.Among the 18 cases,17 delivered at term and only 1 preterm (36+4 weeks).Two out of the 18 women had normal delivery (11.1%),2 by forceps(11.1 %) and 14 by cesarean sections (77.8%).No neonatal apnea or neonatal respiratory distress syndrome had been observed.Pathological diagnosis was confirmed in all cases prenatally,including 4 CIN1,4 CIN2 and 10 CIN3.Four out of the 10 CIN3 remained after delivery,while 1 changed to CIN2.These 5 cases underwent leep electrical excision procedure and Pap smear results turned to normal at 6 to 11 months after delivery.One HSIL case were lost.The rest 4 CIN3 and all of the 8 CIN1 and CIN2 cases turned to normal or lesser degree.Fourteen of the 18 women received HPV test during pregnancy,and 12 (85.7%) were positive,among which 7 women were followed up at postpartum and 6 of them were HPV positive.Conclusions The three steps method for diagnosis of CIN during pregnancy is safe and effective.Conservative management of CIN during pregnancy is recommended.
5. Molecular signaling pathways in the occurrence and development of osteoarthritis

BACKGROUND: Osteoarthritis is a refractory degenerative disease that seriously affects patients’ quality of life, and brings a heavy financial burden to their families. Nowadays, treatments for osteoarthritis can only alleviate symptoms, improve joint function in a short period of time, but cannot prevent or reverse the pathological process of osteoarthritis. To date, there are no prophylactic or regenerative therapies for osteoarthritis. The signaling pathway can explain the occurrence and development of disease at the molecular level. Therefore, the regulation of signaling pathway can be used as a potential treatment for osteoarthritis. OBJECTIVE: To review the occurrence and development of osteoarthritis, aiming at providing new ideas for clinical diagnosis, treatment and deeper understanding of diseases in the future. METHODS: CNKI, Wanfang, VlP, CBMdisc, PubMed and MEDLINE databases were retrieved respectively with the keywords of “osteoarthritis; signaling pathway; EGFR; Hedgehog; NF-kB; Notch” in Chinese and English, and included literature reports regarding the signaling pathways related to osteoarthritis published in 2008-2019 were retrieved. RESULTS AND CONCLUSION: Normal articular cartilage function depends on the regulation of multiple signaling pathways, and the occurrence and development of osteoarthritis are also regulated by corresponding signaling pathways, providing deep understanding of osteoarthritis at the molecular level and new ideas for clinical diagnosis and treatment. Regulation of signaling pathways through drug action and gene modification is a potential treatment for osteoarthritis. However, there is a common problem in related research: most of the research on signaling pathway is isolated, and the occurrence and development of osteoarthritis may involve the joint action of multiple signal pathways. Therefore, further research is needed on the synergy of signaling pathways.
6.Radiofrequency ablation of cardiac fat pad at right pulmonary vein-atria junction attenuates Bezold-Jarisch reflex in canine

Objective:To observe the influence of radiofrequency catheter ablation of the fat pad at the right pulmonary vein- atria junction(RPV fat pad)on Bezold-Jarisch reflex(BJR)induced by veratridine in canine.Methods:Ten mongrel dogs were anesthetized and their chest were opened through a right thoracotomy to expose the RPV fat pad.Veratridine was injected into the left ventricle at bolus doses of 10?g/kg through a catheter to induce BJR.The sinus cycle length(SCL),systolic arterial pressure(SAP),diastolic arterial pressure(DAP),mean arterial pressure(MAP),left ventricle systolic pressure(LVSP), left ventricle diastolic pressure(LVDP)and mean left ventricle pressure(MLVP)were measured under the baseline and after ablation of the RPV fat pad;the changes of the above parameters(?SCL,?SAP,?DAP,?MAP,?LVSP,?LVDP,and?MLVP)were calculated.Then the same dose of veratridine was injected under the pad after ablation and the changes of the a- bove parameters were observed.Results:BJR was successfully induced in the 8 animals;the animals has prolonged sinus cycle length(SCL)and decreased MAP and MLVP.After ablation,the prolongation of SCL after injection of veratridine was markedly reduced([229.2?92.3]ms vs[39.3?14.1]ms,P

Objective To review the clinical characteristics of paraganglioma of the urinary bladder with 5 cases reprt.Methods The clinical data of 5 patients (2 males and 3 females,age 23-68 years)with paraganglioma of the urinary bladder were reported.Two cases were diagnosed by ultrasonography during health examination,1 case was found by hematuria,1 had difficulty of voiding,and 1 presented with palpitation,chest discomfort while urination.Two cases were clinical diagnosed as bladder paraganglioma,1case urachal carcinoma,and 2 cases bladder tumor.Cystoscopy showed a protruding tumor within the bladder or bladder had compressed changes.One case of tumor located in the triangle,1 in the posterior wall,1in the top,2 in the anterior wall.Three cases of biopsy were negative.Three cases of preoperative endocrine examinations showed norepinephrine significantly higher.Results Laparoscopic partial cystectomy was performed in 3 cases,open partial cystectomy in 1 case,and transurethral resection of bladder tumor (TURBT) in 1 case.One case had the complication of stress cardiomyopathy during TURBT,3 cases found intraoperative hypertensive crisis.The bleeding volume was 20-800 ml (average 126 ml),and I case received blood transfusion.During the follow-up period for 3-48 months,the blood pressure was normal,and no recurrence was found.Conclusions Bladder paraganglioma is uncommon and easily misdiagnosed.For the patients with bladder tumor,accompanied by changes in blood pressure,palpitations during urination should be highly suspicious of bladder paraganglioma.Partial cystectomy is the main treatment method.
8.Effects of curcumin preconditioning on activity of xanthine oxidase during intestinal ischemia-reperfusion in rats

Objective To evaluate the effects of curcumin preconditioning on the activity of xanthine oxidase (XOD) during intestinal ischemia-reperfusion (I/R) in rats.Methods Thirty female Sprague-Dawley rats,aged 180-220 g,were randomly divided into 3 groups (n =10 each) using a random number table:sham operation group (S group),intestinal I/R group (I/R group),and curcumin preconditioning group (Cur group).Intestinal I/R was induced by clamping the superior mesenteric artery for 75 min followed by reperfusion.Curcumin 200 mg/kg was given everyday for 5 days before intestinal I/R in Cur group and the equal volume of normal saline was given instead of curcumin in S and I/R groups.The rats were sacrificed at 4 h of reperfusion and the intestinal specimens were obtained for microscopic examination of pathologic changes which were graded using Chiu scoring system and for determination of XOD activity,content of malondialdehyde (MDA),and superoxide dismutase (SOD) activity.Results Compared with S group,the Chiu score,activity of XOD and content of MDA were significantly increased,while the activity of SOD was decreased in I/R and Cur groups (P < 0.05).Compared with I/R group,the Chiu score,activity of XOD and content of MDA were significantly decreased,and the activity of SOD was increased in Cur group (P < 0.05).Conclusion Curcumin preconditioning can attenuate intestinal I/R injury in rats,which may be due to inhibition of XOD activity and decreased oxidative stress in intestinal tissues.
9.Intratumor Angiogenesis and Expression of Angiogenic Factors in Human Pancreatic Cancer

Objective:This study was designed to investigate microvessel density (MVD) and expression of vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF) in pancreatic cancer, and the relevance of these three indexes to the biological behavior of the cancer. Methods:Intratumor MVD and the expression of VEGF and bFGF were examined by immunohistochemical staining in 47 pancreatic cancer tissues (polyclonal antibody for FⅧ and VEGF, monoclonal antibody for bFGF), and the relationship between either MVD or the expression of these two angiogenic factors and the clinicopathological features, including survival were analyzed. Results:MVD in cancer tissues was higher (10.32± 3.32) than that in normal tissues (6.72± 1.73) (P<0.01). In 47 cases, the positive rate of expression of VEGF, bFGF protein in pancreatic cancer was significantly higher than that in normal tissues(P< 0.05), 57.44% of 47 pancreatic cancer were positive for VEGF protein in cancer cells,and 63.82% showed strong bFGF staining in cancer and infilbrating cells. The overexpression of VEGF and bFGF protein correlated significantly with high MVD (P< 0.05). Advanced pathological grade of the disease was significantly more frequent in the patients with high MVD, positive staining of VEGF and bFGF, (P< 0.01). The higher MVD and the overexpression of bFGF correlated with poor survival (P< 0.05). Conclusions: Intratumor MVD is strongly related to the progress, invasiveness and metastasis of pancreatic cancer, and may serve as a meaningful prognostic factor. The overexpression of VEGF and bFGF may play an important role in the process of tumor-associated neovascularization and suggest that VEGF or bFGF system may be promising target for antiangiongenic strategy of pancreatic cancer.
10.Treatment of thoracic vertebrae tumor with posterior total vertebral resection and internal fixation.

OBJECTIVETo evaluate the clinical outcome of posterior total vertebral resection in treating thoracic vertebrae tumor in order to provide a safe and effective method in rebuilding spine stability.
METHODSFrom 2002.1 to 2007.12, 18 patients with thoracic spine tumor underwent posterior total vertebral resection and internal fixation. Among the patients, 10 patients were male and 8 patients were female, ranging in age from 45 to 78 years, with an average of 56 years. The course of the diseases ranged from 2 to 13 months. After the operation, the tumor reccurence was monitored by X-ray, and the tumor markers were detected.
RESULTSAll the patients were followed up for a period ranging from 12 to 60 months, averaged 29 months. All the patients showed a postoperative neurologic improvement, as well as showed radiographic evidence of solid fusion in the follow-up examinations during 3 to 9 months, with an average of (8 +/- 1.4) months.
CONCLUSIONPosterior total vertebral resection for the treatment of thoracic spine tumor is safe and effective.
