1.Effects of p38 Mitogen-Activated Protein Kinase on Apoptosis of Small Intestinal Epithelial Cells after Transplantation in Rats

Objective To explore the effects of p38 mitogen-activated protein kinase (MAPK) on apoptosis of small intestinal epithelial cells after transplantation in rats. Methods Small intestinal transplantation was performed in SD and Wistar rats. The recipients were divided into three groups: isograft group (Wistar→Wistar group), allograft group (SD→Wistar group) and allograft+cyclosporine A group (SD→Wistar+CsA group). The grafts were harvested on day 1, 3, 5 and 7 after operation. All graft samples were subjected to histological examination. The apoptosis of graft epithelial cells was detected by TUNEL method. p38 MAPK was measured by Western-blotting method and serum TNF-? was determined by ELISA. Results Mild, moderate and severe rejection reaction occurred in the SD→Wistar group, it was showed that the number of apoptotic cells increased with the severity of the rejection reaction by TUNEL. In SD→Wistar group, the numbers of apoptotic cells were significantly higher than those of the other two groups (P0.05). 3.A clinical study of XELOX chemotherapy combined with Chinese herbal medicines for obstructive colorectal cancer

Objective To evaluate the efficacy and safety of XELOX chemotherapy combined with Chinese herbal medicines in patients with obstructive colorectal cancer.MethodsA total of 70 patients with obstructive colorectal cancer were recruited and divided into a XELOX treatment group and a combined treatment group, 35 in each group. The XELOX treatment group received XELOX chemotherapy, while the combined treatment group were received XELOX chemotherapy combined with Chinese herbal medicines. All patients received a 3-week treatment as a cycle, and for 4 cycles. The outcomes were Karnofsky performance status (KPS) and the Zubrod-ECOG-WHO Scale (ZPS). Adverse effects were recorded.ResultsAfter the treatment, the clinical symptoms score (1.62 ± 0.21vs. 2.12 ± 0.31;t=7.900,P<0.01) and ZPS score (1.07 ± 0.33 vs. 2.88 ± 0.41;t=20.346,P<0.01) in the combined treatment group were significantly lower than those in the XELOX treatment group, but the KPS score was significantly higher (93.42 ± 5.56vs. 86.43 ± 5.93;t=5.087, P<0.01). The follow-up showed that the 3-year survival rates in the combined treatment group was significantly lower than those in the XELOX treatment group (80.00%vs. 57.14%;χ2=4.242,P=0.039). The incidence of leukopenia (11.43%vs. 34.29%;χ2=3.970,P<0.05), anemia (5.71%vs. 22.86%;χ2=4.200,P<0.05), nausea/vomiting (8.57%vs. 28.57%;χ2=4.629,P<0.05), and diarrhea (2.86%vs. 20.00%;χ2=5.081,P<0.05) in the combined treatment group were significantly lower than those in the XELOX treatment group. Conclusion XELOX chemotherapy combined with Chinese herbal medicines can improve clinical symptoms, performance status, and alleviate adverse effects in patients with obstructive colorectal cancer.

Objective:To investigate the relationship between hyper-homocysteinemia ( HHcy) and experiment autoimmune en-cephalomyelitis (EAE) mice,and to study the cause and effect of HHcy in multiple sclerosis (MS).Methods: Four groups were included in this study:healthy C57BL/6 mice and EAE mice fed with chow(HC and EAE group),healthy mice and EAE mice supple-mented with 4%(w/v) methionine in the drinking water (HC+Hcy and EAE+Hcy group).Clinical symptoms,the levels of Hcy and cytokines ( TNF-α,IFN-γand IL-17) were evaluated at different time points.Results:EAE group had no greater Hcy than HC group ( P>0.05 ).The clinical scores and weight loss of mice from EAE+Hcy group were more severe than those from EAE group during the course of EAE ( P<0.05 ).EAE+Hcy group had higher levels of TNF-αand IFN-γthan EAE group ( P<0.05 ) ,while IL-17 was about the same.Conclusion:Mice do not have gradually increased Hcy during the course of EAE.HHcy worsen the symptoms of EAE by pro-moting the production of TNF-αand IFN-γ,but not IL-17.
8.Intravenous administration of embryonic stem cells home into injured hearts and improve cardiac function post myocardial infarction in rats

Objective To clarify whether the intravenous administration of embryonic stem cells(ESCs)could home in on injured myocardium caused by acute myocardial infarction(MI)in rats and to improve the cardiac function afterwards,and to explore the possible effect of inflammatory factor TNF-?on it.Methods ESCs were cultured in vitro and transfected with green fluorescence protein(GFP).The model of heart failure post MI in rats was established by permanent ligation of left coronary anterior descend artery.After operation the rats were administered with 10~7 ESC via tail vein every other day for 6 days.The sham and MI without ESCs infusion groups served as control.Six weeks later,the hemodynamic measurement was employed to evaluate the cardiac function.Immunohistochemistry assay was used to detect the GFP expression in heart and other organs,and the cardiac specific protein Tropinin I(Tn I)expression in GFP positive spot of heart.Meanwhile,the co-culture in vitro of ESC and neonatal rat cardiomyocytes was used to observe the effect of overexpression of TNF-?in cardiomyocytes on ESC migration.Results Six weeks after MI,cardiac function was significantly improved in rats administered with ESC in comparison with those of MI group.Histological changes demonstrated that infused ESCs formed GFP-positive grafts in infracted myocardium.Alternatively,the positive immunostaining for Tn I was found in the area corresponding to GRP positive staining.In regard to other organs,only in spleen a few GFP positive cells were found.This would indicate that circulating ESC could translocate to infracted myocardium and to form cardiac tissue.Migration assay of ESC in vitro indicated that cultured cardiomyocytes with overexpression of TNF-aobviously enhanced the migration of ESCs in comparison with cardiomyocytes without transfection. Conclusion Intravenously infused ESC could home in on infarcted myocurdium and futher differentiate into cardiomyocytes which led to the improvement of cardiac function.In the setting of acute MI the homing mechanism Could be associated with locally released inflammatory factors such as TNF-?which may play as a chemotactic agent on circulating ESC.
10. Starvation-induced autophagy in glioma and changes in Beclin-1 expression

Objective: To investigate the starvation-induced autophagy in rat C6 glioma cells and the changes of autophagy-related protein Beclin-1. Methods: Rat C6 glioma cells of the logarithmic growth phase were cultured with amino acid-free EBSS medium instead of the DMEM medium. Cells were collected at 1 h, 3 h, 6 h, 12 h, and 18 h after EBSS culture. Western blotting analysis and immunofluorescence were used to detect the specific markers of autophagic microtubule-associated protein 1 light chain 3 (LC-3) and autophagy-related protein Beclin-1, and transmission electron microscope was used to observe the autophagic body. Results: LC-3 expression was found in C6 glioma cells 1 h after starvation; the expression increased with the prolongation of starvation, and reached a peak 12 h later and then began to decline. Autophagic bodies were observed under transmission electron microscopy, and autophagosomes were found in immunofluorescence. Beclin-1 expression began to increase in C6 glioma cells 1 h after starvation, reached a peak at 3 h and then decreased gradually. Increased expression of Beclin-1 was also observed by immunofluorescence. Conclusion Starvation can induce autophagy in C6 glioma cells, which is correlated with the expression of autophagy-related protein Beclin-1. This study lays a foundation for studying the autophagy mechanism in glioma cells.
