1.Efficacy and safety of Wuling capsule on patients with post-stroke depression

Objective To explore the effect on efficacy and safety of Wuling capsule on patients with post-stroke depression.Methods From July 2012 to September 2014, 90 post-stroke depression patients were selected in our hospital.The patients were divided into treatment group (n=45) and control group (n=45) by random number table method.The post-stroke depression patients in control group were treated with fluoxetine as clinical drug program.The post-stroke depression patients in treatment group were treated with Wuling capsule as clinical drug program, six weeks for a course of treatment, both group were treated after two courses,the clinical indicators improve effect, clinical treatment, changes of cytokines, incidence of adverse reactions of the patients between two groups were compared and analyzed.Results Hamilton depression scale (HAMD) scores and Barther index (BI) scores of patients in treatment group post-treatment were better than control group (P<0.05) .Compared with control group, the ratio of clinical efficiency and total efficiency significant in treatment group were increased significantly (P<0.05).The patients in treatment group post-treatment, the levels of cytokine of interleukin-6 (IL-6) and tumor necrosis factor-α(TNF-α) decreased significantly (P<0.05) .Compared with control group, the incidences of adverse effect of patients in treatment group were decreased (P<0.05).Conclusions Wuling capsule has certain therapeutic advantages for improving clinical efficacy and reducing adverse reactions for patients with post-stroke depression.It could as the therapy of choice for post-stroke depression patients.
3.Single use of transarticular screws in atlantoaxial instability

Objective To research the biomechanical characteristics and clinical applicant of posterior transarticular screws (Magerl technique) with bene graft in atlantoaxial instability.Methods 30 fresh cadaver human cervical spine specimens were obtained and randomized subdivided into 5 groups:intact group,atlantoaxial destabilized group,atlantoaxial destabilized and fixed with pedicle screws group,fixed with Magerl screws group and fixed with Maged screws combined with Brooks way group.ROM(range of motion)and NZ(neutral zone)of extend,flex,right and left lateral bend,right and left rotation of each specimen in 5 groups were measured.Transarticular screws were placed in 23 patients with atlantoaxial instability from 2000 Feb.to 2005 Jan.,with 16 males and 7 females,age from 17 to 62 years.20 of them were traumatic dislocation and 3 were congenital instable.Results Biomechanical study indicated that stability of all 6 degrees were Magerl+Brooks>Magerl screws>Pedicle screws>Intact>Destabilized.23 patient were fixed with 46 Maged screws in total,none were suffered from vertebral artery injury and neurological deterioration.All patients were followed-up from 4 to 36 months.2 of them complained that difficult in rotation of head.Conclusion Single use of Magerl screw can provide enough biomechanical stable for adantoaxial instability,and is an effective,low-risky way combined with intra-spinal-process bone graft.
4.CT Analysis of Lung Carcinosarcoma(A Report of 11 Cases)

Objective To evaluate the diagnosis value of CT through analyzing 11 cases with the rare disease-lung carcinosarcoma.Methods 11 cases of lung carcinosarcoma proved by operation and pathology from 1994-2000 were analyzed.Results Lung carcinosarcoma had the characteristics of both lung cancer and sarcoma. The border of the carcinosarcoma was irregular, it had lobulation or spiculation, just like primary lung cancer. When IV contrast medium was used, it had some sarcoma’s characteristics such as lower density in center of mass. Conclusion CT is a good method to show the characteristics of lung carcinosarcoma. It is important to recognize the CT appearances of lung carcinosarcoma in order to improve diagnostic ability.
5.The change of plasma endothelin and atrial natriuretic peptide in heart failure infants and the effect of applying milrinone

Objective To determine the change and effect of milrinone on the concentration of plasma endothelin(ET)and atrial natriuretic peptide(ANP)in congestive heare failure(CHF)infants.Methods Forty-one CHF infants' plasma ET and ANP concentration were determined before and after having received milrinone intravenously,meanwhile compared with that of 40 healthy infants as control.Results The concentration of plasma ET and ANP in CHF infants were obviously increased,which has greatly decreased after intravenous dripping of milrinone.The difference has statistic significance.Conclusion The concentration of plasma ET and ANP in CHF infants are greatly increased and could be lowered by treating with milrinone.

7.Effect of morphine on P-glycoprotein expression in mouse brain microvascular endothelial cells

Objective To investigate the effect of morphine on P-glycoprotein (P-gp) expression in mouse brain microvascular endothelial cells. Methods The mouse brain microvascular endothelial cell line b. End3 was purchased from ATCC company (USA) and cultured at 37 ℃ in high glucose serum-free medium RPMI 1640 in 10 cm petri dishes and assigned to one of 3 groups(n = 9 each): Ⅰ control group (group C); Ⅱ morphine group (group M) and Ⅲ PDTC + morphine group (group P + M). In group M the cells were exposed to morphine 1 μg/ml while in group P + M the cells were pre-incubated with PDTC (NF-κB inhibitor) 5 μmol/L for 1 h before treatment with morphine. In group M and P+ M after being treated with morphine 1 μg/ml for 24 h, the cells were collected for determination of P-gp expression and NF-κB p65-abcb1b protein-DNA binding analysis. Results P-gP expression and NF-κB p65-abcb1b protein-DNA binding were up-regulated in group M as compared with group C. The up-regulation was negated by pre-incubation with PDTC in group P + M. Conclusion Morphine can induce up-regulation of the expression of endogenous P-gp in mouse brain microvascular endothelial cells by NF-κB mediated-abcb1 b gene activation.
8.Clinical evaluation of ¹⁸F-FDG-SPECT/CT imaging in diagnosis and staging of patients with suspected lung cancer.

BACKGROUNDCorrect diagnosis is very important to patients with suspected lung cancer. According to the higher ¹⁸F-FDG intake of tumor tissues much more than normal tissues, this study evaluates the clinical value of ¹⁸F-FDG dual-head coincidence SPECT/CT in diagnosis of lung cancer.
METHODSSixty-one patients with lung mass underwent ¹⁸F-FDG dual-head coincidence SPECT/CT imaging and thoracic CT imaging before operation. These imageological results were compared with histological examination of lung mass.
RESULTSThe sensitivity, specificity and accuracy of ¹⁸F-FDG-SPECT/CT in differentiating malignant and benign lesions were 82%, 87%, 84%, respectively. Compared with thoracic CT imaging, diagnostic rate of mediastinal lymphatic metastasis was significantly higher by ¹⁸F-FDG-SPECT/CT (52% vs 81%, P < 0.05) .
CONCLUSIONSCombined functional-anatomical image of ¹⁸F-FDG-SPECT/CT has potential to improve staging and localizing procedures of lung cancer. It is an excellent modality in diagnosis and differential diagnosis of lung cancer.
9.Laboratory study on reproducibility of no-albumin human plasma by rivanol precipitation Ⅰ.Purification or isolation of protein C and protein S

Purpose The aim is to isolate and purify Protein C(PC) and Protein S(PS) from no-albumin human plasma by rivanol precipitation. Methods The isolated and purified steps included adsorption onto and elution from barium, salting-out, ion-exchange chromatography, affinity chromatography,preparative isoelectric focusing and so on. Results The molecular weights of the obtained PC and PS were (61±0.9)kD and (83±0.8)kD, respectively, the isoelectric point, 4.70±0.03 and 5.20±0.03,and the yield, 28.3% and 12.6%. The purified PC and PS were shown to be highly homogeneous by capillary zone-electrophoresis(CZE), and rich in Glu, Leu and Gly or Asp, Glu and Leu respectively. Conclusion The methods could be used for large-scale isolation and purification of PC and PS from no-albumin human plasma.
10.Vascular smooth muscle cell senescence induced by reactive oxygen species and intervention effect of dehydroepiandrosterone

Objective To observe the onset of vascular smooth muscle cell (VSMC) senescence induced by tert-butyl hydroperoxide (t-BHP) and the intervention effect of dehydroepiandrosterone (DHEA). Methods The VSMCs were divided into four groups: blank control group, t-BHP group (incubated with 80 μmol/L t-BHP for 72 h), 10 nmol/L DHEA intervention group (pretreated with 10 nmol/L DHEA 30 min before t-BHP) and 100 nmol/L DHEA intervention group (pretreated with 100nmol/L DHEA 30 min before t-BHP). Two ageing markers of ageing associated β-galactosidase activity and cell proliferation activity were adopted as main indexes. β-galactosidase activity was measured with immunocytochemical method and cell proliferation activity was measured with flowcytometry. Results After continuous treatment with 80 mmol/L t-BHP for 72 h, the ratios of G0/G1 phase cells and SA-β-galactosidase staining positive cells increased as compared with blank controlgroup [(89.4±3.4)% vs. (49.5±5.5)%, (3.5±1.2)% vs. (75.3±4.3)%], which indicated that VSMCs senescence were successfully induced by t-BHP. While the above changes were smaller in 100 nmol/L DHEA intervention group than in t-BHP group. Conclusions With ageing,accumulation of damage produced by reactive oxygen species may be an important mechanism causing the onset of VSMCs senescence. DHEA may be able to retard the progression of VSMCs senescence through antioxidant effect.
