2.Preventive effect of No. Ⅰ recipe for osteoarthritis on chondrocyte injury by hydrocortisone in rabbits

BACKGROUND:Kidney-nourishing and blood activating Chinese herbs in the No. Ⅰ recipe for ostooarthritis have the effects of treating osteoarthritis and preventing the side effect of glucocorticoid.OBJECTIVE: To build up rabbit chondrocyte culture system in vitro to investigate the effect of preventive effect of No. Ⅰ recipe for osteoarthritis on chondrocyte injury by hydrocortisone.DESIGN:A complete randomized design and controlled trial.SETTING: Department of Rheumatology and Immunology, Second Hospital of Fujian ProvinceMATERIALS:The experiment was conducted from November 2003 to March 2004 at the Molecular Laboratory of Department of Orthopedics &Grammatology, Fujian College of Traditional Chinese Medicine. No. Ⅰrecipe for osteoarthritis consisted of Herba Epimedii, Radix Morindae Officinalis, Rhizoma Ligustic Chuanxiong, Radix Notoginseng and Daphne geraldii Nitsche. Articular cartilage was digested with collagenase Ⅱ ,chondrocyte culture system in vitro was built up, then in the culture system the different concentrations of the mixture of No. Ⅰ recipe for osteoarthritis and hydrocortisone were added, according to the concentrations they were at random divided into two groups. One group was added 1.25 g/L hydrocortisone and/or 150 g/L No. Ⅰ recipe for osteoarthritis, the other group was added 2.50 g/L hydrocortisone and/or 300 g/L No. Ⅰ recipe for osteoarthritis. Each group was again subdivided as control, hydrocortisone, hydrocortisone + No. Ⅰ recipe for osteoarthritis and No. Ⅰ recipe for osteoarthritis groups, with 5 wells in each subgroup.teoarthritis group: In each well 0.1 mL mixture of 1.25 g/L hydrocortihydrocortisone group: In each well 0.1 mL mixture of phosphate buffer teoarthritis group: In each well No. 1 recipe for osteoarthritis 0.1mL was In each well 0.1 mL mixture of 2.50 g/L hydrocortisone and 300 g/L No.each well 0.1mL mixture of phosphate buffer and 2.50 g/L hydrocortisone after medicines were added, the morphological change of chondrocyte in all (A) value in each well was assayed with tetrazoline colorimetry as reflection of proliferation of the chondrocyte. MAIN OUTCOME MEASURES:Influence of No. Ⅰ recipe for osteoarthritis combined with hydrocortisone on chondrocyte proliferation in rabbits.croscope that the chondrocytes in the control group were mostly in multiangles or shuttle-like, and adhesive growth; those cultured for 24 hours with 1.25 and 2.50 g/L hydrocortisone were in pyknosis or flotation state.;those in the various concentrations of hydrocortisone +No. Ⅰ recipe for osteoarthritis, and No. Ⅰ recipe for osteoarthritis groups were mostly in multiangles or shuttle-like, and adhesive growth, without difference with those in 1.25 g/L hydrocortisone group than in the control group(0.007 6±0.0018,0.015 2±0.002 6,t=5.374 0,P<0.01);it was obviously higher in the 1.25 g/L hydrocortisone +150 g/L .No. Ⅰ recipe for osteoarthritis and 150 g/L No. Ⅰ recipe for osteoarthritis groups than in the 1.25 g/L hydrocortisone group (0.065 6±0.016 9, 0.086 6±0.019 0, t=7.630 9, 9.255 9,P<0.01); it was obviously lower in the 2.50g/L hydrocortisone group than in the control group 2(0.005 3±0.001 4,0.015 3±0.002 3 ,t=8.304 5,P<0.01) ;it was obviously higher in the 2.50 g/L hydrocortisone +300 g/L No. Ⅰ recipe for osteoarthritis and 300 g/L No. Ⅰ recipe for osteoarthritis groups than in the 2.50 g/L hydrocortisone group (0.085 8±0.015 0,0.092 6 ±0.016 8, t=11.948 3,11.579 4, P < 0.01).CONCLUSION :Hydrocortisone in higher concentration could cause serious injury or death of chondrocytes, however the chondrocyte grew well cultured in the mixture of hydrocortisone and traditional Chinese drug, No.Ⅰ recipe for osteoarthritis had the effect of preventing injury of chondrocyte from hydrocortisone.

4.Clinical analysis of vitreous resection combined with vitrectomy combined with vitrectomy for proliferative diabetic retinopathy

Objective To investigate the clinical effect of vitrectomy with single slice vitrectomy (PDR) and preoperative injection of monoclonal antibody (mAb) for treatment of proliferative diabetic retinopathy. Methods According to clinical treatment were randomly 64 cases of PDR patients in our hospital in February 2016-February 2017 were divided into observation group and control group, 32 cases (37 eyes) single vitreous body resection were included in the control group, the remaining 32 cases (41 eyes) from preoperative intravitreal injection 0.5mg Lei Zhu monoclonal antibody drug treatment after resection of vitreous body were included in the observation group. The operation time, the best corrected visual acuity before and after the operation were recorded and compared between the two groups. The incidence of iatrogenic hiatus, frequency use, vitreous volume and hyphema were recorded in the two groups. Results The operation time of the observation group was (96.47±12.67) min, which was significantly shorter than that of the control group (128.56±13.78) min (P<0.05). The best corrected visual acuity of the two groups was basically at the same baseline level, and the corrected visual acuity of the 30d observation group was obviously improved and better than that of the control group (P<0.05). Compared with the control group, the incidence of iatrogenic hiatus, electrocoagulation, vitreous volume and hyphema was lower in the observation group (P<0.05). Conclusion Intravitreal injection of ranibizumab therapy after resection of vitreous body regimen in the treatment of PDR is safe and effective, not only helps to shorten the operation time, and lower incidence of postoperative complications, postoperative recovery of visual acuity is good, it is worthy of clinical popularization and wide application of bed.

7.Combined administration of pilocarpine and timolol eye drops for the treatment of glaucoma

Objective To analyze the therapeutic effect of combined administration of pilocarpine and timolol eye drops in the treatment of glaucoma, and to summarize the rational administration of the two drugs.Methods The study was conducted by animal experiment.The study was conducted from January 2016 to February 2016.Fifty New Zealand white rabbits were randomly divided into 5 groups: pilocarpine group, timolol group, conventional administration group, pilocarpine+timolol group, timolol group and pilocarpine group.each group had 10 New Zealand white rabbits, according to different modes of administration,,the changes in intraocular pressure after 10, 20, 30, 40, 60, 90, 120, 150, 180 and 240 min after the first administration were compared.Results Intraocular pressure changes with time points after administration of different administration methods, There was no significant difference of IOP changes in pilocarpine group and timolol group single drug.pilocarpine+timolol timolol group changes with time continuing to reduce intraocular pressure of rabbits, compared with the conventional dose group, pilocarpine+timolol group and timolol pilocarpine+group alkali group of intraocular pressure, intraocular pressure and good control effect,the difference was statistically significant (P<0.05).There was no significant difference between in the adverse reactions between pilocarpine group, timolol group, routine administration group, pilocarpine+timolol group, timolol group and pilocarpine group.ConclusionThe treatment of glaucoma can be treated with pilocarpine and timolol eye drops, pilocarpine+timolol compared with pilocarpine, timolol simple treatment, conventional administration of several modes of administration, from the continuous reduction of intraocular pressure time Point of view, lowering the intraocular pressure longer, fewer adverse reactions, with high therapeutic safety, worthy of clinical promotion and application.
8.Progress and Prospects on Next Generation Sequencing-Based Full Resolution STR Genotyping

Next generation sequencing (NGS),also known as massively parallel sequencing (MPS),with significant advantages such as high throughput,rapidness,integration and low cost,has been widely used in research and clinical diagnosis fields and promises bright future for forensic applications.Compared with traditional DNA sequencing methods,NGS technologies allow the simultaneous detection of various forensic genetic markers.The classical CE-based STR genotyping method only can differentiate alleles with length polymorphism.However,there are occasions like DNA base substitution and repeat sequence variations,which give different alleles with the same size of amplicons.These alleles are heritable from parents to off-springs,and could be of great importance for solving criminal cases.NGS-based STR genotyping can be compatible with current STR data output mode,and allows for the full resolution of STR loci by providing additional sequencing polymorphism information.In this article,the authors focused on the forensic STR genetic markers and next generation sequencing,and reviewed recent progresses on NGS-based STR genotyping.Perspectives of NGS-based full resolution STR genotyping were presented and possible challenges were discussed in the hope of providing a reference for related studies and applications.
