Microbiology 1992;0(03):-
Baculovirus DNA polymerase gene belongs to an early gene of baculovirus. It is a necessary gene required for replication of virus in insect cells. It can encode DNA polymerase induced by virus. In the process of replication, DNA polymerase can bind to homologous regions and non-homologous regions, which are believed to act as the origins of virus DNA replication with other replication factors. In addition, DNA polymerase has advantages over occlusion protein and egt gene for resolving deep branching taxonomic relationships of baculovirus phylogenies.

Microbiology 1992;0(02):-
In order to mutate a carbofuran degrading strain CFDS-1,transposon of pSC123 was introduced into the genomic DNA of strain CFDS-1 by conjugation with E.coli DH5? (pSC123) as donor and strain CFDS-1 as recipient,6 mutants which lost carbofuran degrading ability were obtained with a kanamycin resistant gene in the middle of transposon and the disappearance of a red compound during the degrading of carbofuran as preliminary selecting marks, they were designated as CFDS-M1~CFDS-M6. UV scanning and GS assay results also proved their mutations. PCR was carried out respectively with primers designed according to the sequence of transposon and genomic DNA of 6 mutants as templates, restriction analysis of PCR products showed that the mutation of carbofuran degrading genes of these mutants was caused by transposon insertion.

OBJECTIVETo systematically assess the efficacy and safety of Rhodiola in treating chronic stable angina pectoris.
METHODSOur group searched the Cochrane library, PubMed, Embase, Chinese biomedical literature database (CBM), VIP database (VIP), Chinese Journal Full-text Database (CNKI) for the literature published in English and Chinese till April 2013. Randomized controlled trials (RCTs) were included on the therapeutic effect of Rhodiola or Rhodiola plus conventional Western medicine in comparison with the conventional Western medicine treatment on stable angina. Data were extracted according the data extraction form. The literature methodological quality was assessed by using the Cochrane handbook, and data analyzed by Rev-Man 5.2 Software for Meta-analysis. The effect indicators of outcomes was expressed by odds ratio (OR) and 95% CI.
RESULTSA total of 7 randomized controlled trials, 662 cases of stable angina pectoris patients met the inclusion criteria and all published in Chinese, without one scientific design and high quality literature. Compared with the conventional Western medicine treatment, combined with oral administration of Rhodiola could improve the efficiency of anti-angina (OR = 2.49, 95% CI: 1.02 - 6.09). Combined with intravenous infusion of Rhodiola could also improve the efficacy of angina pectoris (OR = 4.86, 95% CI: 2.4 - 9.82). Oral administration of Rhodiola couldn't improve ECG efficacy (OR = 1.25, 95% CI: 0.67 - 2.34). Intravenous infusion of Rhodiola could improve the clinical efficacy (OR = 2.94, 95% CI: 1.61 - 5.35). Combined with the conventional treatment, intravenous infusion of Rhodiola could improve the whole blood viscosity (low and high shear rates) and inverse variance (IV) (-1.36 and -0.99, 95% CI: -1.65 - 1.07 and -1.26 - 0.71), but could not reduce serum fibrinogen and D-dimer level. The incidence rate of adverse reactions was higher than that of the conventional treatment combined with Rhodiola (OR = 0.1, 95% CI: 0.02 - 0.51).
CONCLUSIONSOn the basis of routine treatment, Rhodiola could further improve patients' symptoms. Combined with intravenous medication, Rhodiola could increase the ECG improvement rate, and reduce adverse reactions. But the methodological quality of included studies was poor, the number of samples was small, and influence factors such as the intervention period was short. This conclusion needs scientific and rational design in a larger sample, multicenter clinical trial to verify.

OBJECTIVETo investigate the features and causes of complications of unilateral pedicle screw fixation combined with contralateral percutaneous translaminar facet screw fixation and lumbar interbody fusion in treating lower lumbar diseases.
METHODSThe clinical data of 166 patients with lower lumbar diseases who underwent unilateral pedicle screw fixation combined with contralateral percutaneous translaminar facet screw fixation and lumbar interbody fusion with intervertebral cages from January 2008 to December 2013 were retrospectively analyzed. There were 64 males and 102 females, aged from 24 to 74 years with a mean of 51.9 years old, suffered from lower lumbar lesions for 47.5 months on average (ranged, 8 months to 30 years). Among these patients, lumbar intervertebral disc degeneration was found in 49 patients, recurred lumbar intervertebral disc protrusion in 17 patients, massive lumbar intervertebral disc protrusion in 23 patients, lumbar intervertebral disc protrusion accompany with spinal canal stenosis in 27 patients, lumbar degenerative spondylolisthesis with degree I (Meyerding grade) in 21 patients, far lateral lumbar intervertebral disc protrusion in 5 patients. Single segmental diseases occurred in 124 patients and two segmental diseases in 42 patients. The diseases occurred at L(3,4) segment in 6 patients, at L(4,5) segment in 97 patients, at L5S1 segment in 21 patients, at L(2,3), and L(3,4) segments in 1 patient, at L(3,4) and L4,5) segments in 26 patients, and at L(4,5), and L5S1 segments in 15 patients.
RESULTSThere was no abnormal bleeding in the patients and no patient received blood transfusion. During the surgery, spinal dura mater injury with cerebrospinal fluid leakage complicated in 1 patient, a fracture of vertebral pedicle in 4 patients, and end plate injury in 2 patients. No postoperative cerebrospinal fluid, incision infection and skin necrosis were found after operation. Nerve root injury was found in 1 patient. According to the position of pedicles crew, 371 screws of 163 patients were in degree I and 3 screws of 3 patients were in degree II; position of translaminar facet screw, 199 screws of 157 patients were type I, 8 screws of 8 patients were type II, 1 screw of 1 patient was III. Translaminar facet screw was slightly short in 2 patients. Five patients were lost to follow-up, two patients were died. The remaining patients were followed up for 35.4 months on average (ranged, 12 to 60 months). During the follow-up period , end plate was cut off and intervertebral cages were embedded in 14 segments of 14 patients. Abnormal pain of both lower extremities was found in 1 patient. With the exception of 11 unidentified segments in 11 patients, 189 segments of 148 patients obtained intervertebral fusion. No loosening, displacement, breakage of pedicle screw or translaminar facet screw, displacement of intervertebral cages or obvious degeneration of adjacent segments were found. The coronal and sagittal planes balance of lumbar vertebra were obviously improved. Postoperative JOA score was significantly increased than that of preoperative.
CONCLUSIONUnilateral pedicle screw fixation combined with contralateral percutaneous translaminar facet screw fixation and lumbar interbody fusion with intervertebral cages is a good choice for the treatment of lower lumbar diseases, but it has a risk of complications. Abundant surgeon's surgical experience, careful operation, and rational use of imaging technique can effectively reduce the incidence of complications.

Objective To investigate the immunological rejection in the brain of cloning goats received neural stem cell transplantation. Methods Eight cloning goats of CL series were chosen at random and divided into 2 groups. Neural stem cells and saline at the same dosages were transplanted into the fixed site by surgical intervention in the brain cortex of each group, respectively. The levels of IL-2 and IL-10 in the blood of each group were detected at different times (1 w before, and 0, 1 and 3 w,and 3 months after the cell transplantation) to reflect the systemic immune rejection of the goats after the transplatation. The CD3+ cells in the cell transplantation areas in each group were also detected by the method of immunohistochemistry to reflect the local immune rejection after the transplatation. Results The level of IL-2 was obviously higher and the level of IL-10 was obviously lower in the neural stem cell transplantation group than those in the control group 1 and 3 w, and 3 months after the cell transplantation (P<0.05). The quantity of CD3+ cells in the neural stem cell transplantation group was much larger than that of control groups at the acute period (1 w after cell transplantation) and chronic period (3 months alter cell transplantation, P<0.05). Conclusion Systemic and local immunological rejections at acute or chronic periods will appear at the brain of cloning goats with neural stem cells transplantation.

OBJECTIVETo investigate the effect of Kangquan Recipe (KQR) on apoptosis regulatory genes bax and bcl-2 mRNA in prostate of rats.
METHODSBenign prostatic hyperplasia model rat was established by injecting testosterone after castration. The model rats were killed and prostate glands were removed for examination after being treated with administration of KQR by gastrogavage for 30 days. The wet weight of prostate was measured and the mRNA expressions of bax and bcl-2 in rats' tissue of abdominal lobe of prostate were determined by RT-PCR.
RESULTSCompared with the model group, wet weight of prostate was lower significantly in the groups treated with different dosages of KQR (P < 0.05 or P < 0.01), and that in the high dose KQR treated group was similar to that in the normal group (P > 0.05). Compared with the model group, the expressions of bax mRNA and ratios of bax/bcl-2 were significantly higher and the expressions of bcl-2 mRNA significantly lower in the KQR treated groups (P < 0.01), and these indexes in the high dose KQR treated group were insignificantly different from those in the normal group (P > 0.05).
CONCLUSIONKQR shows an obvious treatment effect on rats with benign prostatic hyperplasia, the mechanism might be through effectively regulating the expressions of bax mRNA and bcl-2 mRNA in prostatic tissue to accelerate the cell apoptosis of prostate in obvious dose-effect manner.

Chronic osteomyelitis is one of the most common disorder in clinic. In recent years due to diabetes, peripheral vascular disease and trauma induced disease increased, the prevalence rate increased. With the development of magnetic resonance imaging and CT imaging technology, it greatly improved the accuracy of clinical diagnosis of chronic osteomyclitis and ability to describe the infection characteristics, and provide a reliable basis for clinical treatment. The current research on chronic osteomyelitis mainly concentrated on the aspects of imaging applications and ways of using antibiotic optimization control inflammation, defect restoration and reconstruction of blood supply and treatment. But the best time to the antibiotic therapy and the use of program is still uncertain, for after debridement, bone grafting time and defect repair function of fast recovery still need further research.
