3. Abnormal methylation of RASSF1A and BLU genes in renal carcinoma

Objective: To investigate the methylation status of the promoters of RASSF1A and BLU genes in the renal carcinoma tissues and to assess their roles in the tumorigenesis of renal carcinoma. Methods: Methylation specific PCR (MSP) method was used to examine the methylation status of the promoters of RASSF1A and BLU genes in the renal carcinoma tissues of 26 patients with renal carcinoma and their corresponding adjacent normal tissues; and the results were analyzed. Results: We found that 17 (65.4%) of the renal carcinoma tissues had abnormal methylation of RASSF1A gene, and there was no abnormal methylation of RASSF1A in the adjacent normal tissues. Eleven(42.3%) of 26 renal cancer tissues had hypermethylation of BLU gene, and there was no hypermethylation of BLU gene in the corresponding adjacent tissues. Conclusion: Hypermethylation of RASSF1A and BLU genes is present in the renal carcinoma tissues, suggesting the hypermethylation of RASSF1A and BLU might be associated with the tumorigenesis of renal carcinoma.
5.Laparoscopic total mesorectal excision for low rectal cancer of 126 cases

Objective To study the feasibility of laparoscopic total mesorectal excision(TME) for low rectal cancer.Methods Laparoscopic total mesorectal excision was performed in 126 patients with low rectal cancer according to TME principle.Results The operation time was 95 - 180 min,with an average time of ( 117 ± 21 ) min,the amount of bleeding 50 - 200 ml,with an average amount of (90 ± 27 ) ml.2 - 3 days after surgery,gastrointestinal function was restored.Hospital stay was 6 - 14 days,with an average of (8 ± 2)days.Four cases converted to open surgery,the conversion rate for laparotomy was 3.2%.The proportion of sphincter-preserving operation was 95.24% (120/126).No instant or delayed injury of ureters,large bleeding in front of sacrum and other operation-related severe complications happened intra-and after operation.Conclusion Laparoscopic surgery for low rectal cancer is safe and feasible.
6.Observation of the therapeutic efficacy of nasal septoplasty of nasal septal spur, ridge and/or elevated deviation in 134 cases under intranasal endoscopy

Objective To observe the therapeutic efficacy of different operative methods for nasal septal spur/ridge elevated deviation. Methods A total of 134 patients with nasal septal spur/ridge elevated deviation were randomly divided into endoscopic surgery group and control group. Patients in the endoscopic group were treated with nasal septoplasty under intranasal endoscopy, while those in the control group were treated with traditional submucous correction of nasal septum. 8.The Antagonistic Effect of PGE_1 on TGF?_1 mRNA Expression and Rat Renal Interstitial Fibrosis Induced by Unilateral Ureleral Obstruction

Objective To explore the possible role of PGE 1 in tubulointerstitial fibrosis formation. Methods Rat model of unilateral ureteral obstruction(UUO) was established.In situ hybridation(ISH) technique, morphological observation and biochemical analysis were applied to determine TGF? 1 mRNA expression, changes of tubulointerstitium and rencortical hydroxyproline content after used PGE 1. Results 3 days after UUO,the rats renal interstitial edema, a number of mononuclear cells recruitment. On day 14,tubule atrophy, interstitial proliferative fibrosis and area enlarged,hydroxyproline content,TGF? 1 mRNA expression increased, the peak value of the latter occurred early than that of hydroxyproline peak value and interstitial area enlargement.PGE 1 could down -regulate these changes.Conclusions The antagenistic effect of PGE 1 on renal interstitial fibrosis may be partly associated with down-regulating overexpression of TGF? 1 mRNA and inhibiting mononuclear cells recruitment.
9.Role of histamine on the expression of tight junctions gene in airway epithelia

OBJECTIVE To investigate and elucidate the role of histamine on the regulation of tight junctions(TJs) gene expression of the airway epithelial cells. METHODS We cultured primary human bronchial epithelial cells from healthy and asthmatic individuals in the presence of histamine for 96 hours in air-liquid interface (ALI) culture system. The TJs expression on the level of mRNA and protein was determined by RT-qPCR and immunofluorescence (confocal microscopy), respectively. RESULTS A RT-qPCR analysis revealed that the mRNA expression of several TJs were up- or down-regulated by histamine. However, examination by confocal microscope revealed no detectable alterations in the morphology of the tight junctions after stimulating with histamine. CONCLUSION Our findings that histamine could regulate the mRNA expression of TJs of bronchial epithelial cells. That might contribute to the pathogenesis of chronic airway inflammatory diseases.
