Objective To determine the value of vascular indication of sexual excitation response (VISER) in the diagnosis of erectile dysfunction (ED). Methods Fifty three cases of ED underwent PGE1 intracavernous injection,VISER and color duplex ultrasonography.The comparison of the results was performed. Results Forty three cases were in Porst Grade 3, whose average cavernous pressure and carvenous pressure index were 144.29 mmHg and 1.24,respectively.Of them,42 cases were of normal penile vascular function,the remaining one was of venous ED;10 cases were in Porst Grade 2, whose average cavernous pressure and carvenous pressure index were 87.76 mmHg and 0.81,respectively.There were significant differences in the average cavernous pressure and carvenous pressure index between the two groups ( P

To explore the related risk factors and outcome in pregnancy- induced hypertension patients with Ⅲ phase of retinopathy.
●METHODS: A total of 105 pregnancy - induced hypertension patients with Ⅲ phase of retinopathy in our hospital from Januany 2012 to December 2013 were enrolled. Clinical date of them were collected to analyze.
●RESULTS: The occurrence of pregnancy - induced hypertension patients with Ⅲ phase of retinopathy were positively correlated with the course of the disease, blood pressure, proteinuria, and it was higher occurred in cold winter and spring, timely termination of pregnancy and appropriate hormone therapy can promote the recovery of vision, and improve outcomes of pregnancy.
●CONCLUSlON: The occurrence of pregnancy - induced hypertension patients with Ⅲ phase of retinopathy associated with season and disease severity. Timely treatment can restore normal vision, improve maternal and neonatal prognosis. Routine examination of fundus examination should be used as the pregnancy induced hypertension syndrome.
7.To Explore the Core Composition and Compatibility Features of Tibetan Medicine Ershiwuwei Songshi Pills in the Treatment of Liver Disease

Objective: To explore the core composition and compatibility features of Ershiwuwei Songshi pills in the treatment of liver disease, in order to provide ideas and methods for the optimization of Tibetan medicine prescriptions and the creation of new medicine. Methods: In this study, I checked common prescriptions for the treatment of liver diseases from the Tibetan Medical Classics, such as Four Medical Classics, Xin Xiu Jing Zhu Ben Cao, etc., summarizing the flavor, nature and efficacy of the 25 herbs in Ershiwuwei Songshi pill, and analyzing the compatibility features of flavor and of nature and efficacy of the 25 herbs in Ershiwuwei Songshi pill. And I also sorted out 21 common prescriptions for the treatment of liver diseases in the Tibetan Medical formula, counting and analyzing the using frequency of the ingredient medicine of Ershiwuwei Songshi pill in the 21 Tibetan medical formula for the treatment of liver and the compatibility frequency. Conclusion: The results show that Ershiwuwei Songshi pills are composed of a heat clearing formula, mainly for the treatment of heat liver diseases.
8.Palindromic sequence-mediated de novo chromosome translocation in sperm samples derived from normal and oligospermic males.

OBJECTIVETo systematically explore the occurrence of a novel type of chromosome translocation in human sperm samples.
METHODSSpecific translocation junction fragments were quantified using nested and/or multi-nested PCR in sperm DNA derived from 28 oligospermic patients and 32 normal controls.
RESULTSt(11;22) was detected in 49 samples. At least 4 samples were found to have t(1;22) (p21.2;q11.2), t(17;22) (q11;q11) or t(X;22) (q27;q11). The mutation rate seemed to be associated not with age or semen volume, but with sperm concentration (r = -0.389, P < 0.05) and motility (r = -0.397, P < 0.05). Correlation was not found between homology of palindromic sequences and mutation rate.
CONCLUSIONPalindromic sequence mediated chromosome translocation is common in human sperm, and associated with sperm concentration and motility. Measurement of such mutations may provide a molecular-level reference for assessing sperm quality.

The src homology 2 (SH2)-domain containing inositol-5-phosphatase (SHIP) is another recently identified lipid phosphatase after phosphatase and tensin homology deleted on chromosome ten gene (PTEN). It plays an important role in negatively regulating the proliferation of hematopoietic cells. The relationship between SHIP and the inhibition of tumor proliferation is rarely reported. The purpose of this study is to evaluate the apoptosis induced by SHIP gene in K562 cell line and to explore the involved signaling pathway. The K562 cells were transfected with human SHIP gene by using the lentiviral vector containing SHIP, and the transfection was verified by fluorescent quantitative PCR (FQ-PCR) and Western blot. Then the effects of SHIP protein expression on cell growth and apoptosis were measured. The levels of p-Akt, bcl-2 family, caspase and the activity of NFkappaB were assayed by Western blot and ELISA, respectively. The results are as follows: (1) Human leukemia cell line K562 was SHIP-negative; (2) Transfection with SHIP gene led to the re-expression of SHIP mRNA and protein in K562, as shown by FQ-PCR and Western blot; (3) The expression of SHIP protein inhibited cell growth and significantly increased apoptosis in K562 cells; (4) Compared to that in control group, the expression level of p-Akt-308 and p-Akt-473 in SHIP-expressing cell group decreased significantly (P<0.01); SHIP activated caspase-9, caspase-3, up-regulated protein levels of bad, p27, down-regulated expression of bcl-xL, while it had no effect on the expression of bcl-2 and bax. Furthermore, the inhibition of NF-kappaB was achieved along with the inactivation of Akt. These data suggest that SHIP gene has potential abilities to inhibit K562 leukemic cell proliferation and induce its apoptosis via inactivating PI3K/Akt pathway. The loss of SHIP might be the explanation of aberrant high-level p-Akt in human leukemia. It may be at least one of the mechanisms by which the loss of SHIP expression contributes to leukemia progression.

10.Effects of SHIP gene mutation on cell cycle related proteins and phosphorylated Akt in K562 cells.

OBJECTIVETo investigate the effect of SHIP gene mutation on the cell cycle and its related gene expression in K562 cells.
METHODSThe recombined green fluorescent protein (GFP) containing FIV-SHIP gene was transfected into K562 cells. The transfection efficiency and cell cycle of K562/SHIP were assessed by flow cytometry (FCM). The proliferation of K562 cells was detected by MTT assay, the mRNA levels of SHIP by real-time fluorescent relative-quantification reverse transcriptional PCR (FQ-PCR), and the protein levels of SHIP, Cyclin D1, p21(WAF1/CIPI) and p27(KIP1) by Western blot.
RESULTSWild type SHIP inhibited K562 cell proliferation and caused a G(0)/G(1) arrest \[(34.2 +/- 7.8)% vs (0.7 +/- 8.3)% (P < 0.01)\]; while the point mutation of SHIP gene did not show such effect. Western blot results showed that the Akt phosphorylation and cyclin D1 expression was significantly decreased (P < 0.01), and the expression of p27(KIP1) and p21(WAF1/CIPI) increased. Site-directed mutation of SHIP gene SH2 domain (TTC-->CTC, Phe-->Leu) did not influence the Akt phosphorylation and cyclins (P > 0.05).
CONCLUSION(1) wtSHIP gene can down-regulate Akt phosphorylation and result in inhibition of cyclin D1 expression, up-regulating p27(KIP1) and p21(WAF1/CIPI) expression, finally leading to the reduction of K562 cell proliferation, and inducing G(0)/G(1) phase arrest. (2) SHIP gene suppresses the proliferation of K562, being dependent on its intact structure and function.
