2.Effect of ginsenoside total saponinon on regulation of P450 of livers of rats after γ-ray irradiation.

Effect of ginsenoside total saponin (GTS) on the regulation of P450 of livers of rats after γ-ray irradiation was studied. Rats were irradiated by the ⁶⁰Coγ-ray for one-time dose of 5.5 Gy, dose rate of 117.1-119.2 cGy. The cocktail probe, qPCR and Western blot were used to detect expression of enzymatic activites, mRNA and protein of rats. Contrasted with blank group, expression of CYP1A2, 2B1, 2E1, 3A4 of irradiation group showed a up-regulated (P < 0.05). Contrasted with irradiation group, exprression of CYP1A2, 2B1, 2E1, 3A4 of GTS group showed a downward trend. GTS had negative agonistic action against expression of P450 of rats by irradiatied.

3.Effects of angiotensin Ⅱ and captopril on outward potassium channel currents in canine atrial myocytes

Objective To observe effects of angieminⅡ(AngⅡ)and captopril on outward potassium channel currents in canine atrial myoeytes,and to study mechanisnof Ang II and capupril on atrial arrhythmia.Method Ten healthy adult mongrel dogs(general class),weighing 15 to 20 kg,male and female informality,were provided bythe service centre of Tianjin Li-qun experimental animals.Single canine atrial myotcyte was acutely isolated and whole-cell configtmtion of the patch-clamp tchnique was used to detec trapidly activating delayed reefifier outward K+ current(Ikr),slowly activating delayed recti fier outward K+ current(Iks),ultra-rapidly aetivatin delayed rectifier outward K+ current(Ikur)and transient outward potassium current(Ito)before and after An II and captopril peffion.Software of pClamp 7.0 for windows and pClampfit 7.0 Was used to measure current and data were expressed as mean±standard deviation(x±s).SPSS 10.0 statistical was used for statistical analysis.The paired t test was useel for comparison betwn before and after treatment.P<0.05 was comidered as statistical significance.Results AngII(0.5/mol/L)increased Ikr and Iks,ilfibited Ito[(19.54±2.41)pA/pF vs.(24.83±2.52)pA/pF,P=0.001;(20.69±2.29)pA/pF Vfl.(25.59±3.42)pA/pF,P:0.0003;(6.34±1.93)pA/pF vs.(3.71±1.50)pA/pF,P=0.001)],and had no effect on k[(19.78±1.22 pA/pr Vs.(20.39±1.50)pA/pF,P=0.258)].Captopril(5tot/L)had no significant effect on Ikr.,b.k and[(19.11 4-4.91)pA/pF vs.(18.99 4-4.04)-∥pF,P=0.808;(20.76 4-2.89)pA/pF vs.(20.27 a-3.46)pA/pF,P=0.305;(18.50 4-3.78)pA/pF vs.(18.25 4-4.02)pA/pF,P=0.704;(7.31±1.99)pA/pF vs.(6.89±2.12)pA/pF,P=0.136)].Conclusioas AngⅡmay promote atrial electrical remocof atrial fibrillation through outward potassium currents.As angiotemin-eonverting enzy/ne inhibitor.captioruk can prevent atrial electrical rodding of atrial fibrillation by inhibiting renin-angiotensin-system.
4.Effects of the compatibility of Caragana sinica Radix and Astragali Radix on a rat model of diabetic kidney disease via PINK1/MFN2/Parkin pathway

AIM To investigate the impact of the combination use of Caragana sinica Radix and Astragali Radix on a rat model of diabetic kidney disease(DKD).METHODS The SD rats were randomly divided into the normal group,the model group,the Engelgin group,the Caragana sinica Radix group,the Astragali Radix group and the Caragana sinica Radix-Astragali Radix compatibility group,with 10 rats in each group.Following the successful establishment of a DKD model by unilateral amputate renal combined with intraperitoneal injection of streptozotocin(STZ),the corresponding gastric gavage of drugs were administered for 8 weeks.The rats had their 24 h urinary microalbumin(24 h U-mALB)detected at 0,4 and 8 weeks;their levels of Scr,BUN,CysC,MDA and SOD activity detected by ELISA;their renal ROS expression detected by fluorescence probe method;their renal pathological changes observed by HE,PAS,Masson and PASM-Masson staining;their renal expressions of NOX4,Drp1,MFN2 and P62 detected by immunohistochemistry;and their renal expressions of PINK1,MFN2,Parkin,LC3-Ⅱ/Ⅰ,P62 and p-Drp1 proteins detected by Western blot.RESULTS Compared with the model group,each treatment group displayed lower contents of 24 h U-mALB,BUN,Scr and CysC in the serum of rats(P<0.01);reduced pathological structure damage of the renal tissue;decreased MDA level in serum and kidney(P<0.01);increased SOD activity(P<0.01);increased renal protein expressions of PINK1,MFN2,Parkin and LC3-Ⅱ/Ⅰ(P<0.05,P<0.01);and decreased protein expressions of p-Drp1 and P62(P<0.01).And the Astragali Radix group and the Caragana sinica Radix-Astragali Radix compatibility group took the lead(P<0.05,P<0.01).CONCLUSION Upon the rat model of DKD,the compatibility of Caragana sinica Radix and Astragali Radix may alleviate their renal pathological damage and improve their renal function by activating the mitochondrial autophagy to improve mitochondrial dynamics and inhibit their oxidative stress via PINK1/MFN2/Parkin pathway.
6. Effects of polysaccharides from cryptoporus volvatus(peck) schear on leukotrienes generation in guinea pig lung

AIM: To study the effects of polysaccharides of ferment cultured cryptoporus volvatus (CVPS) on release of leukotrienes from guinea pig lung and Schultz - Dale reaction. METHODS: The bioassay method and reversed phase high-performance liquid chromatography (RP-HPLC) were used to analyze SRS-A or LTD4 from sensitized or normal guinea pig, after challenged with antigen or A23187 respectively. The antiallergic effects of CVPS were evaluated with Schultz-Dale reaction. RESULTS: The release of SRS-A or LTD4 from sensitized or normal guinea pigs were greatly reduced by CVPS after antigen or A23187 challenge. The CVPS could also inhibit the Schultz-Dale reaction of sensitized guinea pig (IC50 = 0.49 g·L-1). CONCLUSION: The effects of CVPS involved in inhibiting release of leukotrienes from lung and antiallergy.
7.Intratumor Angiogenesis and Expression of Angiogenic Factors in Human Pancreatic Cancer

Objective:This study was designed to investigate microvessel density (MVD) and expression of vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF) in pancreatic cancer, and the relevance of these three indexes to the biological behavior of the cancer. Methods:Intratumor MVD and the expression of VEGF and bFGF were examined by immunohistochemical staining in 47 pancreatic cancer tissues (polyclonal antibody for FⅧ and VEGF, monoclonal antibody for bFGF), and the relationship between either MVD or the expression of these two angiogenic factors and the clinicopathological features, including survival were analyzed. Results:MVD in cancer tissues was higher (10.32± 3.32) than that in normal tissues (6.72± 1.73) (P<0.01). In 47 cases, the positive rate of expression of VEGF, bFGF protein in pancreatic cancer was significantly higher than that in normal tissues(P< 0.05), 57.44% of 47 pancreatic cancer were positive for VEGF protein in cancer cells,and 63.82% showed strong bFGF staining in cancer and infilbrating cells. The overexpression of VEGF and bFGF protein correlated significantly with high MVD (P< 0.05). Advanced pathological grade of the disease was significantly more frequent in the patients with high MVD, positive staining of VEGF and bFGF, (P< 0.01). The higher MVD and the overexpression of bFGF correlated with poor survival (P< 0.05). Conclusions: Intratumor MVD is strongly related to the progress, invasiveness and metastasis of pancreatic cancer, and may serve as a meaningful prognostic factor. The overexpression of VEGF and bFGF may play an important role in the process of tumor-associated neovascularization and suggest that VEGF or bFGF system may be promising target for antiangiongenic strategy of pancreatic cancer.

9.Effect of combined therapy with hypha Cordyceps and ginkgo leaf tablet on micro-inflammation in patients undergoing maintenance hemodialysis.

OBJECTIVETo investigate the status of the micro-inflammation in patients undergoing maintenance hemodialysis and the effect of combined therapy with hypha cordyceps and ginkgo leaf tablet for its prevention and treatment.
METHODSSixty-five patients undergoing long-term maintenance hemodialysis were randomly assigned to the treated group (33 patients) and the control group (32 patients). Patients in both groups underwent hemodialysis regularly, meanwhile, 3 capsules of Jinshuibao (each capsule containing hypha cordyceps 0.33 g) and 1 tablet of ginkgo leaf tablet (0.23 g/tablet) were additiolally given to the patients in the treated group orally, 3 times per day, and 3 months as 1 treatment course. Their serum inflammatory factors, such as high sensitive C-reactive protein (hs-CRP), interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-alpha), which could reveal the state of microinflammation, were determined before and after treatment. And the same index was also determined in 30 healthy persons allocated in a normal control group.
RESULTSCompared with the normal control group, the serum levels of hs-CRP (4.94 +/- 2.21 mg/L), IL-6 (0.472 +/- 0.220 microg/L) and TNF-alpha (1.748 +/- 0.724 microg/L) in the treated group were higher before treatment, these indexes decreased markedly after treatment (3.28 +/- 1.28 mg/L, 0.379 +/- 0.163 microg/L, and 1.330 +/- 0.458 microg/L, respectively, all P < 0.01), but without significant change in the control group.
CONCLUSIONMicro-inflammation reaction exists popularly in patients undergoing maintenance hemodialysis, and the combined therapy with hypha cordyceps and ginkgo leaf tablet could effectively improve this kind of micro-inflammation reaction.

10.Studies on preparation of sinomenine hydrochloride-loaded bovine serum albumin microspheres.

OBJECTIVETo prepare Sinomenine hydrochloride-loaded bovine serum albumin microspheres (SM-BSA-MS).
METHODSM-BSA-MS was prepared by spray drying technique. The morphology, drug-loading and release in vitro of SM-BSA-MS was studied.
RESULTThe diameters of SM-BSA-MS were in the range of 1-3 m. The drug loading of microspheres, formulated with different drug/albumin ratios as 1, 2, 1:1, 2:1, were 31.6%, 47.7% and 67.9% , respectively. And the drug entrapment efficiencies of different drug/albumin ratios were higher than 94%. The results of in vitro release experiments showed that the drug loaded microspheres have the properties of sustained-release compared with the Sinomenine hydrochloride injection. Different release characteristics could be obtained by adjusting the prescription composition and the thermal denaturation condition.
CONCLUSIONSpray drying technique is a simple and feasible method for preparing SM-BSA-MS. The drug loaded microspheres had high drug-loading and sustained-release effect.
