1.Effect of High Iodine Level on Proliferation of Cultured Fibroblast

Objective To study the effects of iodine on the proliferation activity of fibroblasts at different doses. Methods The cultured fibroblasts were treated with different dose iodine(100 ?g/L, 500 ?g/L, 1 000 ?g/L, 3 000 ?g/L, 5 000 ?g/L, 7 000 ?g/L, 9 000 ?g/L, 11 000 ?g/L)for 24 h and observe the morphology of fibroblast. MTT colorimetry was used to detect the fibroblast proliferation activity. Results Iodine could significantly increase the fibroblast proliferation activity at certain range of concentration (7 000 ?g/L). The proliferation activity was highest from 5 000 ?g/L to 7 000 ?g/L, the differences were significant compared with the iodine free control. Conclusion Iodine, the exposure level is 7 000 ?g/L.
2.Effect of Fluoride on Proliferation of Human Blood Vessel Endothelial Cell in Cultured

Objective To observe the effects of fluoride of different dose on the proliferation of the cultured human umbilical vein endothelial cells(HUVEC). Methods The cells were treated with NaF at different doses. Cell counting, MTT colorimetry, flow cytometry (FCM) and immunohistochemistry were used to detect the endothelial cells activities. Results Compared with the fluoride free control, the positive rate of PCNA and Ki-67 were higher as NaF concentration was at 120-240 ?mol/L and were highest at 240 ?mol/L, the same was seen in the values of A and PI, as NaF concentration was at 600-960 ?mol/L,these indexes decreased. Conclusion In vitro, the low concentration of fluoride can promote the proliferative activity of HUVEC, whereas the high concentration can inhibit it.
3.Early assessment of left ventricular dysfunction in mice with doxorubicin-induced cardiotoxicity by high-frequency echocardiography

Objective To evaluate the diagnostic value of high frequency echocardiography on mice with doxorubicin-induced cardiotoxicity and find out the most valuable monitoring indexes for early doxorubicin-induced myocardial damage.Methods Forty mice were divided into doxorubicin-injection group (ADM group) and control group randomly.Twenty mice in each group were injected with doxorubicin and saline at the same dose for 5 weeks continuously.The parameters of cardic function of ADM group and contol group were observed by high-frequency echocardiography after infection.Brain natriuretic peptide (BNP) tested by euzymelinked immunosorbent assay(ELISA) and pathological findings were used as the golden standard.Results ①The left ventricular long axis view and the apical four chamber view could be displayed clearly by transthoracic high-frequency echocardiography.②After 5 weeks intervention,weight,heart rate,left ventricular ejection fraction (LVEF),left ventricular fractional shortening (LVFS),E/A,E wave deceleration time(EDT) and other parameters showed marked difference between ADM group and control group(P <0.05).③The decrease of diastolic function was earlier than systolic function in ADM group.④LVEF and E/A presented good negative linear correlation to BNP(r =-0.622,P <0.05; r =-0.593,P <0.01).EDT showed positive linear correlation to BNP(r =0.501,P <0.05).Conclusions Cardic function of mice with doxorubicin-induced myocardial damage could be early evaluated accurately by high-frequency echocardiography,which contained good relativity with BNP.
4.Assessment of Bax gene transfer influence on survival of heterotopic cardiac allograft in rats by high-frequency echocardiography

Objective To evaluate the value of high-frequency echocardiography in assessing Bax gene transfer influence on survival of heterotopic cardiac allograft in rats.Methods Thirty rat models of heterotopic heart transplantation were established.Group A received heart transplantation only; Group B received cyclosporin (CsA) after operatiom Group C received ultrasound targeted microbubble destruction (UTMD) combined with Bax-shRNA.All rats were tested by high-frequency echocardiography at day 1,3,6 after transplantation.The ultrasound parameters included left ventricular internal dimension diastole (LVIDd),left ventricular internal dimension systole(LVIDs),left ventricular ejection fraction(LVEF),left ventricular thickness (LVT),left ventricular thickening (LVTR) and so on.The pathologic examinations were carried out on five rats at day 6 after echocardiography exam.The other rats in each group were observed for the survival time of cardiac allograft.Results ①The left ventricular long axis view and the apical four chamber view could be displayed clearly by high-frequency echocardiography.②The survival time of allografts in group C was (16.21 ± 5.01)d,which was longer than that in group B [(11.14 ± 1.72)d,P < 0.05] and group A [(7.26 ± 1.50)d,P <0.01].③LVT index got higher after transfection.At day 6,LVEF of group A was lower than group B and C markedly(P <0.05) while no significant difference was found between group B and C(P >0.05).At day 3 and 6,LVT of group A and B were higher than group C (P <0.05) while LVTR was lower(P <0.05).Conclusions The Bax gene transfer influence on survival of heterotopic cardiac allograft in rats could be evaluated accurately by high-frequency echocardiography which could assess cardiac structure and function.LVT and LVTR can be considered as early evaluation indexes for its high sensitivity over LVEF.
5.Protective effect of bicyclol on liver function in patients after liver resection: a randomized control trial

Objective This study investigates the protective effect of bicyclol on liver function in patients after liver resection.Methods One hundred and twenty patients undergoing liver resection with Pringle's maneuver were randomly divided into groups A,B,and C,and given bicyclol (50 mg),diammonium glycyrrhizinate (150 mg),and silybum marianum (77 mg),respectively.The medication was orally given preoperatively for 5 days and postoperatively for days 3 to 7.The fasting serum ALT,AST,TB,ALP,and PAB levels were determined before operation and on days 1,3,5,and 7 after operation.Results ALT levels in the A group were significantly lower than those in the B and C group on post operative days 1,3,5,and 7 (P<0.01).On postoperative day 7,the ratio of serum ALT returned to normal was significantly higher than the B and C group ratios (P<0.05).Conclusion Therefore,oral bicyclol given before and after liver resection can significantly inhibit the rapid increase and promote the normalization of serum ALT levels.
6.The effects of exercise therapy in children with congenital heart disease:a systematic review

Objective To evaluate the effects of exercise therapy in children with congenital heart disease. Methods To search databases such as PubMed, Medline, Science Direct, Embase, Cochrane Library, China National Knowledge Infrastructure, WanFang, VIP, China Biology Medicine for all the randomized controlled trials. After evaluating the quality of each article, the software of RevMan 5.3 were used to analyze. Results A total of 10 randomized controlled trials were included. Compared with the routine nursing, meta-analysis showed that exercise therapy can improve the peak oxygen uptake(MD=4.82,95%CI 2.25-7.39, P=0.0002) and the postoperative compliance (RR=2.84, 95%CI 1.75-4.63, P<0.01), shorten the postoperative hospital time(MD=- 4.41,95% CI - 6.15-- 2.68, P <0.01). Conclusions Exercise therapy can improve the pulmonary function and quality of life on children with congenital heart disease, shorten the postoperative hospital time, increasing the postoperative compliance. However, there were few research on present, so we need a large sample randomized controlled trials of long time to confirm the effects of exercise therapy.
7.Surgical Treatment for 1102 Cases of Primary Liver Cancer

The purpose of this study was to retrospectively analyse the results of 1102 primary liver cancer (PLC) patients underwent liver resection in the past thirty years and to search some effective approaches for improving the longterm effect or PLC treatment. 95% were with hepatocellular carcinoma (HCC), 85.2% with cirrhosis of hepatitis and 25.6% with tumor equal to or smaller than 5 cm in diameter. The mortality rate (MR) within 1 month after operation was 1.8%, the operative MR was 8.8% before 1977 and only 0.4% after that. The total 5-year survival rate (SR) was 28.2% while in the group of small tumor (≤5cm), it was 75.0%. Our experience was as follows: (1) Early diagnosis and early resection of PLC is the key point for improving the operative result of longterm survival. In 282 cases of small cancer, tumor resection rate was 90.0%. Of 48 cases of tumor equal to or smaller than 3 cm in diameter, the 5-year SR was 83.3%. (2) .Rehepatectomy for recurrent liver cancer is an important approach for improving the surgical result. In our series, recurrent rate within 5 years postoperation was 72.3% in larger tumor group and 34.5% in small tumors. There were 78 cases undergoing reoperation in a total number of 170 times of rehepatectomy with 54.7% of 5-year SR after the 1st operation and 34.6% after the 2nd one. (3) For unresectable large tumors, two-stage operation is an important development in liver surgery. We had 26 cases of such patients with 60.0% of 5-year SR. (4) Improvement of operating techniques plays an important role in reducing postoperative complications, lowering operative mor- tality and obtaining better operative result. (5) Postoperative comprehensive treatments are also important for solidating operative effect and preventing tumor recurrence.
8.Study on the Quality Standard of Xianrengu Oral Liquid

OBJECTIVE:To establish the quality standard for Xianrengu oral liquid. METHODS:Thin layer chromatography (TLC) was used for qualitative identification of Epimedii Folium,Astragali Radix,lycium barbarum,Atractylodes macrocephala and Panax ginseng in Xianrengu oral liquid;RP-HPLC was preformed on the column of Dikma Diamonsil C18 with the mobile phase of acetonitrile-0.1% phosphoric acid (30∶70,V/V)at the flow rate of 1.0 ml/min,the detection wavelength was 270 nm,the temperature was 30℃and the volume was 10μl. RESULTS:TLC identification had good separation,clear spots and high specifica-tion. The linear range of icariin was 7.97-510.00 μg/ml(r=0.999 3);RSDs of precision,stability and reproducibility tests were no more than 0.83%;and the average recovery was 97.69%(RSD=1.30%,n=6). CONCLUSIONS:This method is simple,accu-rate and simple,which can be used for the quality control of Xianrengu oral liquid.
9.Ultrasound-targeted microbubble destruction combined with polyethylenimine mediated Bax gene silence in H9c2 cells

Objective To silence Bax gene in H9c2 cells by ultrasound-targeted microbubble destruction(UTMD) combined with polyethylenimine.Methods The shRNA expression plasmid targeting Bax gene was co-treated with microbubbles and polyethylenimine.Then the mixture added to H9c2 cells were irradiated by ultrasound.Grouping:① plasmid group;② Lipofectamine2000 + plasmid group; ③ UTMD + plasmid group;④ PEI + UTMD + plasmid group;⑤ PEI + SonoVue + plasmid group (without ultrasound irradiation).The transfection efficiency was assessed by fluorescence microscope and FCM.The expression of Bax mRNA and protein were detected by RT-PCR and Western Blot.Results Bax shRNA wasn't destroyed by ultrasound exposure.The transfection efficiency of PEI + UTMD + plasmid group was higher than any other groups obviously (P <0.05).The result of RT-PCR and Western Blot showed inhibition efficiency of Bax mRNA and protein expression in PEI + UTMD + plasmid group were (69.41 ± 4.91) % and (64.09 ± 2.38)% separately,which were higher than any other groups significantly (P <0.05).Conclusions UTMD combined with polyethylenimine can inhibit the expression of Bax gene in H9c2 cells effectively.
10.Effects of grain-sized moxibustion on expression of Abeta(1-42) in prefrontal cortex and hippocampus in double-transgenic AD mice.

OBJECTIVETo explore the therapeutic effect of grain-sized moxibustion at "Xinshu" (BL 15) and "Shenshu" (BL 23) on early-stage Alzheimer's disease (AD) in transgenosis AD mice.
METHODSThe genotyping of amyloid precursor protein/presenilin 1(APP/PS1I) double-transgenic AD mice were detected by PCR method. Seventeen 1.5-month female transgenic (Tg 6799) mice were randomly divided into a model group (9 cases) and a treatment group (8 cases). Nine female C57BL/6J wild-type mice with identical age and background were selected into a normal group. The treatment group was treated with grain-sized moxibustion at bilateral "Xinshu" (BI. 15) and "Shenshu" (BL 23), once a day, ten treatments were considered as one course, and total 9 courses were given. The model group and normal group were treated with stimulus such as grabbing, immobilization and non-ignited moxa cone. Morris water maze (escape latency, crossing times and dwell time in the target quadrant) was applied to evaluate the learning and memory ability. Hematoxylin-Eosin (HE) staining was used to observe morphology changes in the brains of AD mice. beta-amyloid protein 1-42 (Abeta(1-42)) in the area of prefrontal cortex and hippocampus was detected by immunohistochemical method.
RESULTSAfter the treatment of grain-sized moxibustion, learning and memory ability in the treatment group was increased; compared with the model group, the escape latency was shorten, crossing times was increased, and dwell time in the target quadrant was prolonged (all P<0. 05). The crossing times and dwell time in the target quadrant in the treatment group were not significantly different from those in the normal group (both P>0.05). Compared with the normal group, the positive area and integral optical density of Abeta(1-42) in prefrontal cortex and hippocampus in the model group were increased (all P<0.01). Compared with the model group, the positive area and integral optical density of Abeta(1-42) in prefrontal cortex and hippocampus in the treatment group were reduced (P<0.01, P<0.05).
CONCLUSIONThe grain-sized moxibustion at "Xinshu" (BL 15) and "Shenshu" (BL 23) could significantly improve the learning and memory ability in APP/PS1 double- transgenic AD mice, and inhibit the over expression and accumulation of Abeta(1-42).
