Objective:
To develop a headspace gas chromatography ( HS-GC ) assay for simultaneous determination of dichloroacetic acid ( DCA ) and trichloroacetic acid ( TCA ) in urine.
Methods:
Urine samples (5 mL) were transferred to a 22 mL headspace bottle, added with 0.5 mL 10% sodium acetate solution , immediately sealed, and shaken evenly. The bottle was placed in the HS-GC system, and equilibrated at 90 ℃ for 60 minutes. The mixture was separated with the HP-INNOWAX chromatographic column, and the DCA and TCA concentrations were detected with the hydrogen flame detector.
Results:
Under the optimal experimental conditions, the correlation coefficient of DCA and TAC was both > 0.999 0 within the range of 10-500.0 μg/L, and the lowest detection limits of DCA and TAC were 2.0 and 3.5 μg/L, with the spike recovery rate of 87.40% to 101.44%, and relative standard deviations of 1.89% to 3.25%. Of the 35 urine samples sampled from occupational populations, DCA and TCA were not detected.
Conclusions
The establishment of the HS-GAS assay through addition of sodium acetate and optimization of the headspace conditions, has high recovery and precision, which is effective to meet the requirements for daily determination of DCA and TCA in urine samples.

OBJECTIVE:To preliminarily study the stability of 3 pieces of Chinese medicinal formula(CMF)after decoction, and provide reference for guaranteeing storage quality of decocted liquid and improving safety of drug use. METHODS:3 represen-tative formulas of Gegen Huangqin Huanglian decoction(A formula),Wuling powder(B formula)and Didang decoction(C for-mula)from Shanghan Zabing Lun were selected, the decocted liquid were stored under ambient temperature(25 ℃)and refrigerat-ed temperature (4 ℃) after decocting by automatic boiling-machine and packing. The microorganism, precipitation,pH and con-tents of total flavonoids, alkaloid, polysaccharide, total protein after 1,7,14,21,28 d were detected. RESULTS:Compared with the first day, contents of total flavonoids, polysaccharide in formula A at ambient temperature group were significantly in-creased on the 28th(P<0.05),content of polysaccharide in refrigerated temperature group was significantly increased(P<0.05). Content of polysaccharide in formula B at ambient temperature group was significantly decreased(P<0.05). The pH and content of total flavonoids in formula C at ambient temperature group and refrigerated temperature group were significantly increased (P<0.05 or P<0.01). Other indexes showed no obvious changes during the trial period. CONCLUSIONS:Under ambient temperature and refrigerated temperature, liquid ingredients of above decocted CMF will change when storing for 4 weeks. It indicates that the storage time of decocted CMF should not be more than 3 weeks.

OBJECTIVE:To investigate the effect of polyamide on the separation on phenolic acids components by nanofiltra-tion process. METHODS:Phenolic acids components were selected as targets, extracted from the Salvia miltiorrhiza,Lonicera ja-ponica or intermediate preparations (tea polyphenols) with water, and filtered with membranes with different pore sizes. The con-tent changes of components were analyzed by HPLC. RESULTS:Danshensu, protocatechuic aldehyde and caffeic acid could better transmit nanofiltration membranes with 3 different pore sizes;with the decrease of pore size, the rejection rates of neochlorogenic acid, chlorogenic acid, rosmarinci acid,ECG and EGCG grew in different levels. But salvianolic acid B was almost retained by the membrane with low molecular weight except 600-800 Da. CONCLUSIONS:Molecular weight importantly influenced nanofiltra-tion separation on phenolic acid components;nanofiltration has a good prospect in enrichment of phenolic acids components.

Background and Purpose:Lung cancer is the most malignant tumour in the world. Its incidence is growing and NSCLC is predominent(80%) in lung cancer. Most patients with lung cancer were diagnosed in late stages. The tumour could be shrunk by neoadjuvant chemotherapy when the case with stage Ⅲ NSCLC was considered not possible for radical operated neoadjuvant chemotherapy may lead to the following, operation could be improved, micrometastasis could be annihilated and survival could be extended. Objective of this paper was to analyse the prognostic factors for survival in patients treated by surgery and chemotherapy for NSCLC.Methods:98 cases of neoadjuvant chemotherapy combined with surgery for NSCLC,stageⅠ~Ⅲ,were collected retrospectively in our hospital from 1995 to 1997.35 cases were stageⅠ.21 cases were stage Ⅱ.42 cases were stage Ⅲ.83 cases had 1 cycle of chemotherapy pre-operatively.15 cases had 2 cycles chemotherapy pre-operatively.Regimes of chemotherapy were MVP,MOP and MAP et al.Response rate(RR) of chemotherapy was:45 cases had partial response(PR) and 53 cases were stable disease(SD).Operative mode was lobectomy and pneumectomy with lymph nodes dissection.Pathologic type was squamous,adeno,adeno-squamous and others. All the patients were treated by chemotherapy for two or three cycles after surgery except for the patients in stageⅠin 1996~1997.After being followed-up for more than 5 years, data were examined using life table,KaplanMeier method,Log Rank statistic and Cox-mantel test. The possible factors affecting survival were tested with univariate and multivariate analysis. Results:The median followed-up time of 98 cases for NSCLC was 41.2 months.36 cases were alive.62 cases were dead. The 1-,3-,5-year survival rate of 98 cases for NSCLC was 88.78%、49.63% and 18.46%.The 5-year survival rates of stageⅠ、ⅡandⅢ were 33.23%、20.26% and 5.52% respectively(P=0.0002).The 5-year survival rates of N_(0)、N_(1)、N_(2) were 35.49%、19.08% and 4.90% respectively(P=0.0004).In the 98 cases of NSCLC,better prognosis was correlated with earlier stage. The prognosis was better if the period from last chemotherapy before operation to operation was no more than 1 month. The prognosis of lobectomy, lung hila activity, thorax lymph nodes negativity and squamous cancer was better. The prognosis was poorer if the tumor had invaded big vessels, viscera, chest wall, pericardium and quantity bleeding during≥400ml.The prognosis was better if the tumor was fibrotic. The prognosis of 2 cycles of chemotherapy pre-operatively might be better than 1 cycle. The prognosis of tumor necrosis was poorer and the prognosis of chemotherapy post-operatively was better. Conclusions:The main prognostic factors affecting survival in patients treated by surgery and chemotherapy for NSCLC was stage, the period from last chemotherapy before operation to operation, operation mode, lung hila activity, thorax lymph nodes, site of tumor invasion, bleeding quantity, pathologic type, tumor fibrosis and necrotis, cycles chemotherapy pro-operation and chemotherapy post-operation.

Considering that photodynamic therapy (PDT)-induced oxygen consumption and microvascular damage could exacerbate hypoxia to drive more glycolysis and angiogenesis, a novel approach to potentiate PDT and overcome the resistances of hypoxia is avidly needed. Herein, morpholine-modified PEGylated bilirubin was proposed to co-deliver chlorin e6, a photosensitizer, and diclofenac (Dc). In acidic milieu, the presence of morpholine could enable the nanocarriers to selectively accumulate in tumor cells, while PDT-generated reactive oxidative species (ROS) resulted in the collapse of bilirubin nanoparticles and rapid release of Dc. Combining with Dc showed a higher rate of apoptosis over PDT alone and simultaneously triggered a domino effect, including blocking the activity and expression of lactate dehydrogenase A (LDHA), interfering with lactate secretion, suppressing the activation of various angiogenic factors and thus obviating hypoxia-induced resistance-glycolysis and angiogenesis. In addition, inhibition of hypoxia-inducible factor-1α (HIF-1α) by Dc alleviated hypoxia-induced resistance. This study offered a sequentially responsive platform to achieve sufficient tumor enrichment, on-demand drug release and superior anti-tumor outcomes in vitro and in vivo.
