1.Correlation Study on Changes in Chemical Composition and Anti-inflammatory Effects of Fried Paeoniae Radix Alba-Glycyrrhizae Radix et Rhizoma Praeparata cum Melle

Objective To investigated the correlation between the effects of the compatibility of fried Paeoniae Radix Alba-Glycyrrhizae Radix et Rhizoma Praeparata cum Melle on chemical composition and anti-inflammatory effects.Methods An HPLC-DAD method was employed for the simultaneous determination of various components of fried Paeoniae Radix Alba-Glycyrrhizae Radix et Rhizoma Praeparata cum Melle,including paeoniflorin,albiflorin,benzoylpaeoniflorin,oxypaeoniflorin,glycyrrhizic acid and glycyrrhizin.An adjuvant-induced arthritis rat model was established.Administration groups were given fried Paeoniae Radix Alba,Glycyrrhizae Radix et Rhizoma Praeparata cum Melle,and fried Paeoniae Radix Alba-Glycyrrhizae Radix et Rhizoma Praeparata cum Melle liquid by gavage for 2 weeks,respectively.Enzyme-linked immunosorbent assay was conducted to detect the levels of inflammatory factors IL-1β,TNF-α and PGE2 in serum of each group.Factorial design,bivariate correlation analysis,and multiple linear regression were employed to explore the effect of compatibility on chemical composition and its correlation with anti-inflammatory efficacy.Results The established HPLC-DAD analysis method revealed a significant increase in the content of oxypaeoniflorin,albiflorin and benzoylpaeoniflorin following the compatibility of fried Paeoniae Radix Alba-Glycyrrhizae Radix et Rhizoma Praeparata cum Melle(P<0.05).Compared with the model group,the contents of IL-1β,TNF-α and PGE2 in serum of each administration group decreased(P<0.01),and the contents of IL-1β,TNF-α and PGE2 in serum of compatibility group were lower than fried Paeoniae Radix Alba group and Glycyrrhizae Radix et Rhizoma Praeparata cum Melle group(P<0.05,P<0.01).The results from the factorial design indicated the presence of an interaction effect between fried Paeoniae Radix Alba-Glycyrrhizae Radix et Rhizoma Praeparata cum Melle.Bivariate correlation analysis results showed negative correlations between oxypaeoniflorin,albiflorin,paeoniflorin,glycyrrhizin,benzoylpaeoniflorin and glycyrrhizic acid with the contents of IL-1β,TNF-α and PGE2,respectively.Multiple linear regression results revealed that glycyrrhizin and glycyrrhizic acid were negatively correlated with the contents of IL-1β,TNF-α and PGE2.Conclusion The established method for analyzing multiple constituents in the compatibility of fried Paeoniae Radix Alba-Glycyrrhizae Radix et Rhizoma Praeparata cum Melle is straightforward,rapid,highly specific,accurate,and reliable,suitable for quality evaluation and control.The compatibility of fried Paeoniae Radix Alba-Glycyrrhizae Radix et Rhizoma Praeparata cum Melle can inhibit the expressions of IIL-1β,TNF-α and PGE2,and the content of chemical composition is correlated with anti-inflammatory effects.
2.Relationship between gH genotyping and clinical characteristics of children with congenital cytomegalovirus infection

Objective To study the relationship between human cytomegalovirus (HCMV) envelope glycoprotein gene H and clinical features of children with congenital cytomegalovirus infection. Methods A cohort study was conducted. Newborns diagnosed with congenital cytomegalovirus infection, hospitalized in the Department of Neonatology and Neonatal Intensive Care Unit (NICU) of the Children′s Hospital, Zhejiang University School of Medicine, were included from July 2013 to December 2015. HCMV‐DNA gH typing in urine, sputum or blood was conducted. Patients then were divided into gH1 group and gH2 group according to gH genotypes. Patients′data during hospitalization in newborn and 3-5 years of follow‐up were collected. The relationships between gH genotype and clinical manifestations, laboratory examinations, hearing loss and neurological prognosis were analyzed by chi‐square test, t test and non‐parametric test. Results A total of 21 cases were enrolled as congenital HCMV infection and followed‐up for 3-5 years. Among them, 14 (67%) were gH1 type and 7 (33%) were gH2 type. No mixed infection was found. In the two groups, there were no significant differences in the ratio of males (9/14 vs. 3/7, P=0.397), or birth weight ((2 609±686) vs. (3 021±451) g, t=-1.436, P=0.167). Gestational age of gH1 group was younger than that of gH2 group (38 (29-40) vs. 39(38-40) weeks, Z=-2.18, P=0.029). Moderate to severe hearing loss detected by neonatal auditory brainstem response were found in 40 ears (20 cases). It was higher in gH1 group than that in gH2 group (4/22 vs. 0/18, χ2=5.145, P=0.023). In the imaging examination of the nervous system, the Alarcon score of gH1 group was lower than that of gH2 group (0.4±0.3 vs. 1.3±1.1, t=-2.459,P=0.024). No significant statistical difference was found in the probability of motor or language development lag in gH2 group and gH1 group (4/7 vs. 4/14, P=0.346). Conclusions Compared with gH2 infection, gH1 infection in children has a younger gestational age. The major type of hearing loss in neonatal period is gH1 infection. Children with gH2 congenital infections are more likely to suffer from nervous systems damage.
3.Biomechanical Analysis of Lower Limbs during Walking under Simulated Reduced Gravity

Objective To investigate the influence of gravity levels on lower limb motions during human walking. Methods A suspended microgravity simulation system was designed for the experiment. Kinetic parameters from lower limb joints of twelve volunteers during walking were measured by the motion capture system and 3D force plate under simulated Mars gravity (1/3 G), lunar gravity (1/6 G) and earth gravity (1 G). Results Under simulated Mars and lunar gravity, the ranges of motion (ROMs) of hip and knee joints in sagittal plane significantly decreased (P<0.01) while the ROMs of ankle joints obviously increased (P<0.05). The ROMs of hip, knee and ankle joints in sagittal plane under earth gravity were 45.2°, 67.7°, 32.5°, respectively, while the ROMs of hip, knee and ankle joints under lunar gravity were 25.1°, 50.8°, 42.4°, respectively. In addition, the force and torque of lower limb joints in sagittal plane under lower gravity also decreased significantly (P<0.01). Conclusions The results obtained by this study were almost the same as the research findings obtained by using treadmill. Astronauts can use the treadmill and microgravity simulation system for walking exercises under lunar gravity on the earth.
4.Clinical and imaging analysis of COVID-19-related osmotic demyelination syndrome

Objective:To analyze the clinical and imaging features of patients with COVID-19-related osmotic demyelination syndrome (ODS).Methods:COVID-19-related ODS cases diagnosed in the Department of Neurology, Peking Union Medical College Hospital from January 2020 to September 2023 were retrospectively reviewed. And their past medical history, possible triggers, clinical manifestations, imaging manifestations, treatment and prognosis were summarized.Results:A total of 5 patients with COVID-19-related ODS were included. Electrolyte disturbances acted as an inducement of ODS in all patients (5/5),4 of whom with hyponatremia. Four of 5 patients first presented with disturbance of consciousness, followed by predominant dystonia. Imaging of all patients (5/5) showed isolated extrapontine myelinolysis (EPM). With the prolongation of the course of disease, such signal intensity could return to normal, and lesions showed atrophic changes in some patients. The patients′ clinical symptoms were partly relieved within a few days to a few months after treatment.Conclusions:COVID-19-related ODS is mostly associated with hyponatremia, and EPM is more common. COVID-19 should be considered as a risk factor for ODS.
