1.Study on the construction and expression of anti-IL2R? genetically engineering an-tibody Fab

Abstract Objective: To construct and express anti-IL-2R? genetically engineering antibody Fab and identify its biological activity.Methods: PCR were used to construct cloning vector 5G1-pA22 of anti-IL-2R? antibody Fab,by digestion of vector 5G1-pA22, anti-IL-2R? an-tibody Fab gene fragment was obtained and then this fragment was coupled with digesting product of pET28b plasmid by EcoRI and HindIII toget a recombinant expression vector 5G1-pET28b, it was highly expressed in the E. coli BL21(DE3) in the form of inclusion bodies, compoing20% of the total protein. After the renaturation of antibody Fab, its binding activity was identified by ELISA and antibody competitive inhibitionassay in vitro. Results: Anti-IL-2R? antibody Fab was successfully constructed and highly expressed in the form of inclusion bodies, its yield wasup to 1 mg/ml.When applied to ELISA, antibody Fab exhibited good spectfic reactivity with rIL-2R, antibody competitive inhibition assay invitro indicated that antibody Fab can compete with IL-2 for binding IL-2R in activated T cell surface.As a result,the activation and proliferationof T cells were inhibited and inhibition percentage exceeded 90%. Conclusion: Had successfully constructed and expressed anti-IL-2R? geneti-cally engineering antibody. Fab,it had good biological ativity.
2.Development of mobile monitor for risk factor control on chronic disease.

In order to improve the control rate of risk factors of chronic disease, it is developed a status control method of risk factor and its mobile monitor. The monitor uses 32 bit RISC microprocessor of S3C2410X as a controller based on ARM920T core, and MC35i cellular engine GSM/GPRS supported by SIEMENS as the communication unit. The proving tests show that the physical activity, diet, smoking and alcohol of the controlled people can be controlled using the status control method, and the monitor plays a key role in the method. The conclusion is that status control method and mobile monitor can become an alternation method and technology for the risk factor control of chronic disease.

3.Clinical analysis of whole body gamma knife conformal radiotherapy for 169 patients with non-small cell lung cancer

Objective To evaluate the therapeutic effects and complications of whole body gamma knife conformal radiotherapy for patients with non-small cell lung cancer(NSCLC).Methods 169 patients with NSCLC received whole body gamma knife conformal radiotherapy.Total dose,fractions,and radiation time were determined based on disease course,tumor location,tumor features and tumor volume,etc.Small target volume(tumor size less than 3cm in diameter) received 5-10Gy each time,40-50Gy in 4-10 fractions;moderate target volume(tumor size 3cm-5cm in diameter) received 4-8Gy each time,48-56Gy in 4-16 fractions;and big target volume(tumor size more than 5cm in diameter) received 3-4Gy per time,40Gy in 10-14 fractions and followed by 10-30Gy using reduced fields.Results Of 269 tumors in 169 patients,complete response(CR) rate,partial response(PR) rate,and rate of no change(NC) were 41.6%(112/269),44.6%(120/269) and 13.8%(37/269),respectively.Overall response rate(CR+PR) was 86.2%(232/269),with 1-year local control rate of 92.3%.1-year and 2-year survival rate was 75.1% and 46.2%,respectively,with median survival time of 19 months.The incidence of Grade 1,2 and Grade 3 radiation pneumonia was 15.4% and 3.1%,respectively.Conclusions With minor adverse effects,whole body gamma knife conformal radiotherapy may have short-term therapeutic effects for NSCLC patients with amelioration of disease symptoms,obvious elevation of tumor local control rate,a raise of the survival rate,and improvement of life quality.

Objective To study the pathologic characteristics of eutopic endometrium in patients with endometriosis.Methods Pathologic characteristics of eutopic endometrium were studied in 176 patients with endometriosis in Peking Union Medical College Hospital from January 2007 to December 2008 retrospectively.Results About 72.2%(127/176)of eutopic endometrium were in proliferative phase,19.9%(35/176)of were observed as endometrial polyp,including 32 cases with simple endometrial polyp and 3 cases with abnormal hyperplasia combined with endometrial polyp.And 4.0%(7/176)showed abnormal hyperplasia.The incidence of pathologic changes in eutopic endometrium was 22.2%(39/176).Among 53 endometriosis patients combined with infertility,the incidence of pathologic changes of eutopic endometrium was 35.9%(19/53),which was significantly higher than 16.3% in non-infertile patients (x2 =8.24,P =0.004).Among 65 cases with irregular menstruation,the incidence of endometrial polypus and endometrial hyperplasia were 20.0%(13/65)and 10.8%(7/65),which were significantly higher than 17.1%(19/111)and 0 in normal menstruation patients(x2 =13.839,P =0.003).Conclusions The eutopic endometrium of endometriosis were in proliferative phase state.The pathologic changes of eutopic endometrium were more in patients combined with infertility and irregular menstruation.
6.Qualitative study on psychological state of male nursing students

Objective To analyze the psychological state of male nursing students in clinical practice.Method The method of qualitative research was used to interview 20 male nursing students to analyze data and come out with a theme.Results The psychological state of the male nursing students included the following three aspects:intense psychological pressure,unstable mood, low self-esteem and anxiety and depression.Conclusions Establishing nursing communication course using situational simulation and role play and inviting psychological consultants to have face-to-face communication with them for the improvement of mental conditions are good for them to complete their courses and find a nursing position in the future market?
7.Schistosomiasis status and control strategy in Hubei Province from 2008 to 2012

Objective To evaluate the effects of the schistosomiasis control mode(“Hubei mode”)that includes“compre-hensive measures in a whole endemic county”,“co-action of Health Ministry and Province”and“replacing cattle with machine”in Hubei Province. Methods The data of schistosomiasis control in Hubei Province from 2008 to 2012 were collected and a data-base including the annual schistosome infections of human and cattle,acute schistosome infection,outbreak of schistosomiasis en-demic,schistosome infected Oncomelania hupensis snails,and other prevention indicators was established and analyzed by using SPSS. Results Compared with 2008,in 2012,the infection rates declined by 64.91%and 88.63%in human and cattle respec-tively. The area with snails decreased from 5 423.85 hm2 in 2008 to zero. There were no acute schistosomiasis patients and out-break of schistosomiasis endemic. Conclusion The schistosomiasis control mode(“Hubei mode”)is effective significantly.
8.The association between serum albumin levels and prognosis in children with hand-foot-mouth disease

Objective To observe levels of the serum albumin and their association with prognosis in children with hand-foot-and-mouth disease (HFMD) . Methods A total of 122 HFMD children admitted from 2011 to 2013 were selected and divided into three groups:common group (n=57), severe group (n=52) and critical group (n=13). Serum albumin levels, blood glucose levels, peripheral blood white cell counts of the children in the three groups were compared within 24 hours of admis-sion. Moreover, the incidence of hypoalbuminemia and mortality of the children were analyzed in each group. Results In critical group, serum albumin levels were signiifcantly decreased while blood glucose levels and peripheral blood white cell counts were signiifcantly increased than those in the severe group and common group, and the differences were all statistically signiifcant (all P<0.05). There were no signiifcant differences in serum albumin levels, blood glucose levels and peripheral blood white cell counts between severe group and common group (all P>0.05). There was no hypoalbuminemia and death in severe group and common group. The percentage of hypoalbuminemia and mortality of children in critical group were higher than those in the other two groups (all P<0.01). Conclusions The mortality was extremely high in children with critical HFMD. The decrease of serum albumin levels was a high risk factor for death in children with critical HFMD. Early and dynamic monitoring serum albu-min levels may be helpful in evaluation of disease condition and prognosis of HFMD.

10.The effect of spinal interleutkin-33 on radicular pain after non-compressive lumbar disc herniation

Objective:To explore the effect of spinal interleutkin-33 (IL-33) on radicular pain in rats modeling non-compressive lumbar disc herniation.Methods:A total of 80 male Sprague-Dawley rats were randomly divided into a sham operation group, a model group, a lentivirus negative control group, a low-dose IL-33 recombinant lentivirus group and a high-dose IL-33 group, each of 16. Non-compressive lumbar disc herniation was successfully induced in all except the rats in the sham operation group. Two days later, the model group was injected intrathecally with 10μl of enhanced infection solution. The lentivirus control group received 10μl of negative lentivirus, the low-dose IL-33 recombinant lentivirus group received 5μl of IL-33 recombinant lentivirus and the high-dose IL-33 recombinant lentivirus group received 10μl of IL-33 recombinant lentivirus. The 50% paw withdrawal threshold (50% PWT) was measured one day before the modeling and on the 1 st, 3 rd, 5 th, 7 th, 9 th, 11 th, 13 th, 15 th, 17 th, 19 th, and 21 st day afterward. On the 12 th day the expressions of IL-33 protein and mRNA were evaluated. Results:The average expression of IL-33 protein and mRNA in the model and the lentivirus negative control group increased significantly after the modelling compared with the sham group, while expression in the low- and high-dose IL-33 recombinant lentivirus groups was significantly lower than in the lentivirus negative control group. Compared with one day before the modelling, average 50% PWTs on the affected side decreased significantly in all of the modelling groups. From the 9 th to the 21 st day significantly increased 50% PWTs were observed on the affected side in the low-dose and high-dose IL-33 recombinant lentivirus groups compared with the other two modelling groups. Immunostaining showed significant increase in the expression of IL-33 in the dorsal horn of the spinal cord in the model group, compared with the sham operation group. Significant decrease in the average expression of IL-33 in the spinal dorsal horns was observed in the low-dose and high-dose IL-33 recombinant lentivirus groups. Conclusions:Intervertebral disk herniation may increase the expression of IL-33 in the spinal cord, and may cause radicular pain.
