1.POLYMORPHISM OF DNA REGION ADJACENT TO THE 5' END OF INSULIN GENE OF HAN SUBJECTS IN CHINA

The polymorphic characteristics of DNA region adjacent to the 5' end of the insulin gene in 83 Han subjects in China including 26 normal people and 57 diabetics were analysed by restriction endonuclease Bgl 1 gene mapping of chromosomal DNA. DNA fragments obtained by Bgl 1 digestion mainly contained 2.8 ? 0.3 kb (5%), 3.5?0.3kb (10%), 4.2 ? 0.3kb (46%) and 5?0.4kb (36%). In contrast with western white and black people, the above region is homozy-gous in 86% of the Han subjects analysed. This difference may be associated with the racial characteristics. No definite correlation between such polymorphism and the type of diabetes has been found.
2.The applied value of abnormalities of fetus with oligohydramnios in mid-pregnancy by amnioinfusion

Objective To investigate the applied value of abnormalities of fetus with oligohydramnios in mid-pregnancy by anmioinfusion. Methods Fifty women with oligohydramnios diagnosed by ultrasonograph were analysed retrospectively. During the procedure 60-300 ml saline solution at 37 ℃ were infused into the amniotic cavity,made prenatal diagnosis at the same time necessarily. And examined the women to know whether the fetal anomaly existed or not after the amnioinfusion by the ultrasound. Results There were 15 cases with fetal anomaly, 8 cases with genitotirinary anomaly(2 cases associated with multiple organs anomaly), 1 case with single umbilical artery, 4 cases with chromosomal abnormality (2 cases associated with fetal growth restriction), 2 cases with pulmonary hypolasia. There was no significant difference in dilivery outcome between others and 35 cases with normal pregnant women (P >0.05). Conclusion Transabdominal amnioinfusion has double function on beth diagnosis and therapy for the oligohydramnios in the mid-pregnancy.
3.Analysis of the characteristics of multi-drug resistant acinetobacter baumannii infection in elderly patients with severe respiratory

Objective To investigate the clinical characteristics of multiple drug resistant Acinetobacter baumannii infection in respiratory elderly patients.Methods A total of 102 elderly patients infected with multidrug-resistant Acinetobacter baumannii were enrolled in our hospital from January 2014 to December 2015.At the same time,no multiple drug-resistant Acinetobacter baumannii infection elderly patients was selected as a control group.The gender,age,antimicrobial use,white blood cell count,mechanical ventilation time and other differences between the two groups of elderly patients were compared.Results There were significant differences in the days of hospitalization [(18.7±7.5) d vs.(10.0±2.7)d],the time of application of invasive ventilator[(24.6±10.3) d vs.(11.6±6.9)d] and the time of application of antimicrobial agents[(26.2±13.1) d vs.(8.0±2.6)d] (t=19.463、15.436、26.905,all P< 0.05).There was no significant difference in immunosuppressant use,white blood cell count,serum protein level and APACHEⅡ score (all P>0.05).Conclusions Reasonable scientific application of antimicrobial agents,reducing invasive mechanical ventilation time and improving patient immunity can help to reduce the risk of respiratory infection in elderly patients with multiple drug resistance to Acinetobacter baumannii.
4.Effects of BCL6 B on proliferation and migration of human colorectal car-cinoma LoVo cells and its potential mechanism

AIM:To detect the endogenous expression of B-cell leukemia/lymphoma 6 member B (BCL6B) in FHC and LoVo cells, and to investigate the effects of BCL6B on proliferation and migration of LoVo cells for further explo-ring the underlying mechanism .METHODS:The endogenous expression of BCL 6B in the FHC and LoVo cells was detec-ted by RT-PCR and Western blot .The methods of MTT assay , colony formation assay , wound healing assay and Transwell chamber experiment were employed to examine the biological functions of BCL 6B in the LoVo cells.The mRNA and protein levels of BCL6B, cyclin D1 and matrix metalloproteinase-9 ( MMP-9) were determined by RT-PCR and Western blot , re-spectively.The level of phosphorylated protein kinase B (p-AKT) was detected by Western blot.RESULTS:BCL6B ex-pression was notably repressed in the LoVo cells as compared with the FHC cells , which were significantly increased by transfection with pcDNA3.1-BCL6B.The abilities of proliferation and migration of the LoVo cells at 72 h were inhibited by 28.33%(P<0.01) and 36.11%(P<0.05) in BCL6B group.The mRNA levels of cyclin D1 and MMP-9 in the cells of BCL6B group were decreased by 39.90%(P<0.01) and 77.36% (P <0.05), and the protein levels of cyclin D1, MMP-9 and p-AKT were reduced by 44.00%(P<0.05), 47.06%(P<0.01) and 32.88% (P<0.05), respectively. CONCLUSION:BCL6B inhibits proliferation and migration of the LoVo cells , and the PI3K/AKT signaling pathway is in-volved in this process .
5.Evaluation of the invisible bracketless appliance on maxillary molar distalization

Objective:To evaluate the clinical effects of invisible bracketless appliance on the upper molar distalization.Methods:15 patients with class Ⅱ malocclusion(at the average age of 25.3 years) were treated by invisible bracketless appliance,the pre and post three-dimensional digital models were superimposed and measured,the effects of molar distalization were analysed.Results:After treatment,the maxillary first molars were distalized by 2.58 mm on both sides.The left and fight second molars were distalized by 2.57 mm and 2.68 mm respectively.Bilateral central incisors were moved mesially by 0.34 mm.There was no significant difference in the horizontal movement of central incisor.But the left and fight first molars were moved buccally by 0.96 mm and 0.97 mm respectively,the left and fight second molars were moved buccally by 1.01 mm and 1.11 mm separately.Bilateral first molars were intruded by 0.26 mm,the left and right second molars were intruded by 0.37 mm and 0.36 mm,respectively.But the central incisors had no significant vertical movement.There was no significant difference in the buccally or palatally rotation of bilateral first and second molars.Conclusion:Invisible bracketless appliance is efficient for distalization of upper molar,but it may result in mild molar intrusion and anterior anchorage loss.
6.Effect of pediatric obstructive sleep spnea hypopnea syndrome with attention deficit hyperactivity disorder on sleep and quality of life

Objective Related factors of pediatric obstructive sleep apnea hypopnea syndrome (OSAHS) combined with attention deficit hyperactivity disorder (ADHD) were retrospectively analyzed in OSAHS children. Methods The research included 437 cases of children hospitalized for OSAHS from January, 2014 to December, 2014. All the cases were divided into OSAHS group and OSAHS combined with ADHD group, according to the presence or absence of ADHD. Patient's general conditions (gender, age, height, weight, allergic rhinitis history, tonsil grade and adenoid grade), OSA-18 quality of life score, intention-hyperactivity score, polysomnography parameter (apnea hypopnea index, lowest oxygen) were collected as variables. SPSS20.0 was used to perform statistical analyses.Results There were 437 cases in this research, 298 of them were males, and 139 of them were females. Of 437 patients, 147 had OSAHS combined with ADHD (33.64%). Higher apnea hypopnea index (P <0.01) and severer oxygen deflciency (P < 0.01) were found in OSAHS combined with ADHD group than those in OSAHA group. Five projects of OSA - 18 score of in OSAHS combined with ADHD group were all higher than those in OSAHS group (P <0.01).Conclusions The incidence of pediatric OSAHS combined with ADHD in this research was more than 30%. More OSAHS combined with ADHD were found in male children. The quality of life was lower in pediatric OSAHS combined with ADHD. Oxygen deflciency was suggested to be the most important risk factor of ADHD in pediatric OSAHS.
7.Clinical study on ambulatory labor analgesia used in latent phase of the first stage of labor

Objective To evaluate the clinical effect of ambulatory labor analgesia used in latent phase of the first stage of labor, which include labor progress, Apgar score after ambulatory labor analgesia begun to use when the cervix was different size dilatated. Methods Seventy-five parturient primiparas who had no complication were randomly divided into three groups: group Ⅰ: ambulatory labor analgesia was begun to use when the cervix was 1.0 cm dilated, group Ⅱ: ambulatory labor analgesia was begun to use when the cervix was 2-3 cm dilated, group Ⅲ: control group without use of ambulatory labor analgesia. Analgesic effects were observed, changes of uterine contraction were recorded by fetal monitor. Meanwhile, total stage of labor ,outcome of delivery and Apgar score were recorded. Results Duration of total stage of labor had no significant difference between group Ⅰ and group Ⅲ.The duration of the first labor stage was significantly longer in group Ⅰ than that in group Ⅱ(P
8.Clinical features of eosinophilia in peripheral blood in children with parasitic diseases

Objective:To summarize the characteristics of eosinophilia in peripheral blood in children with parasitic diseases.Methods:All clinical data of children with parasitic diseases who were diagnosed in Shenzhen Children′s Hospital from June 2002 to June 2020 were collected to analyze the characteristics of eosinophil changes in peripheral blood after parasitic infection.Results:(1)A total of 37 cases of parasitic diseases were diagnosed in children, with 25 males and 12 females, aged from 10 months to 12 years and 7 months, and the median age was 5 years and 3 months.(2)Among 37 cases, the absolute number of eosinophils in peripheral blood was increased in 32 cases (86.49%), with a range of (0.55-43.80)×10 9/L: 6 cases (18.75%) were slightly increased, 8 cases (25.00%) were moderately increased, and 18 cases (56.25%) were severely increased.(3)As for eosinophilia in peripheral blood of different insect species, there were 16 cases (50.00%) of fasciola infection, 4 cases (12.50%) of fasciola infection, 3 cases (9.38%) of mixed infection, 2 cases (6.25%) of schistosomiasis, plerocercoid, hydatid and cysticercosis infection, and 1 case (3.13%) of ascaris infection.The degrees of the increase of eosinophils in peripheral blood infected by different species of parasites were different ( Fisher′ s value 17.97, P=0.01), with statistically significant differences.(4)Eosinophilia in peripheral blood was involved in different systems, with respiratory system in 18 cases (48.65%), digestive system in 13 cases (35.14%), skin in 7 cases (18.92%), central nervous system in 5 cases(13.51%), circulatory system in 3 cases (8.11%), and others in 2 cases (5.41%). (5)There were 5 cases of parasitic infection without eosinophilia, including 2 cases of Blastocystis hominis, and 1 case of mixed infection with plerocercoid and cysticercosis, 1 case of cysticercosis and 1 case of trichinella spiralis.The increase of eosinophils in peri-pheral blood after different systems was different, without statistically significant differences ( Fisher′ s value 7.37, P=0.06). Conclusions:Eosinophil in peripheral blood is a common phenomenon of parasitic infection in children.The increased severity is related to parasite species, instead of the site of systemic involvement.
9.MIRAGE syndrome caused by SAMD9 gene variation in a Chinese child and literature review

The clinical data of a case diagnosed with MIRAGE syndrome in the Respiratory Department of Shenzhen Children′s Hospital in June 2020 were analyzed retrospectively.The 11-month-old boy was admitted to the hospital because of " intermittent fever for 1.5 months and blood oxygen decline for half a day" . Whole exome detection was carried out by using second-generation sequencing technology.The results showed spontaneous, heterozygous, missense variation in SAMD9 gene (NM_017654) and the mutation site was c. 2471G>A.Review of the literature found that all of the children (47 cases) were born prematurely and their parents were not intermarriage.Besides, they had overall growth retardation, and some suffered from myelodysplasia, recurrent infection, adrenal insufficiency, genital phenotypes and enteropathy.Among SAMD9 gene variation, mutations c. 1376G>A and c. 2471G>A are most frequent.Attention should be paid to the MIRAGE syndrome in children with premature birth and full development lag after birth.
