1.Effects of fuling shengmai yin on superoxide dismutase, malondialdehyde and lipofuscin level of rat hippocampal neurons in vitro
Chinese Journal of Tissue Engineering Research 2005;9(37):180-182
BACKGROUND: Deposition of lipofuscin (LF), decrease of superoxide dismutase (SOD) level and increase of malondialdehyde (MDA) level are important manifestations of senility. Some Chinese medicinal herbs have obvious advantage in delaying senility.OBJECTIVE: To explore the effects of fuling shengmai yin (FLSMY)serum on SOD, MDA and LF level of rat hippocampal neurons in vitro with vitamin E serum and blank serum as controls.DESIGN: Randomized controlled study.SETTING: Experimental Animal Department of Tongji Medical College,Huazhong Science and Technology University; Cell Culture Center of Anatomy Teaching and Research Section of Tongji Medical College,Huazhong Science and Technology University; Scientific Research Department of Wuhan Municipal Hospital of Traditional Chinese Medicine.MATERIALS: From October 2000 to February 2001, the experiment was conducted at the Experimental Animal Department of Tongji Medical College as well as Cell Culture Center of Anatomy Teaching and Research Section of Tongji Medical College, Huazhong Science and Technology University. Six Japanese male white rabbits with big ears, fed normally and weighting (2.5±0.1)kg, were divided into three groups with 2 in each group.Thirty newly born SD rats aged 1-2 days, of clean grade and either gender, were selected.METHODS: [1] Hippocampal neurons were inoculated to 24-well culture plates with the density of 2.5×105/L. Cells in each well were randomized into three groups: A. FLSMY group (fuling shengmaiyin composed of Poria,Radix Ginseng, Radix Ophiopogonis and Fructus Schisandrae; each milliliter contained 1.5 gram of the herbs); B. drug control group (serum containing vitamin E); C. blank control group (blank serum). Each group had 8 wells. After 24 hours, 1 mL serum containing FLSMY, 1ml serum containing vitamin E, or 1 mL blank serum was added into each plate well of corresponding groups. [2] On the 6th day of cell culture, XO-HPX reaction system was added to induce cell injury, in which senility model was established by producing hyperoxide anion free radicals. [3] In the process of cell culture different drug was added to the corresponding different group plate well during replacement of culture fluid each time. [4] The level of SOD, MDA, and LF was detected after 15 days of culture.MAIN OUTCOME MEASURES: Level of SOD, MDA and LF in rat hippocampal neurons.RESULTS: SOD level was higher in FLSMY group than in blank control group (P < 0.01), whereas MDA level in FLSMY group was lower than that in vitamin E group (P < 0.05); LF level was lower in FLSMY group than in blank group (P > 0.05).CONCLUSION: FLSMY as a drink is composed of traditional Chinese medicinal herbs: Poria, Radix Ginseng, Radix Ophiopogonis and Fructus Schisandrae; each has the properties of anti-oxidation and eliminating free radicals. FLSMY can strengthen SOD activity, lower MDA level, and inhibit LF formation. Therefore, FLSMY can delay senility through protecting neurons.

Objective ① To investigate the level of the vitamin D endocrine system in peripheral relationships with bone mineral density (BMD) and the disease activity respectively. Methods The level of the 25-hydroxylate vitamin D3 (25OHD3) and 1,25-dihydroxyvitamin D3 [1,25(OH)D3] in plasma from 43 SLE patients and 44 normal controls were detected by enzyme linked immunosorbent assay. Vitamin D receptor (VDR) gene expression was determinied by real-time PCR in peripheral blood. BMD measurements in the lumbar spine (L1-4) and left proximal femur (femoral neck) were performed using dual X-ray absorptiometry before treatment. The relationship between the vitamin D endocrine system and the bone mass were studied. We also discussed the relationship between the vitamin D endocrine system and the disease activity. Results The levels of 25OHD3 and 1,25 (OH)2D3 were lower in the initial SLE patients than normal controls (P<0.01, P<0.01). The expressions of VDR gene were significantly increased in initial SLE compared with normal controls (P<0.01). The initial SLE patients had significantly lower BMD values, and higher frequency of osteopenia (35%) at both sites of measurement compared with matched healthy controls (P<0.01). The initial SLE patients were divided into two groups by BMD, abnormal group and normal group. There were no differences in 25OHD3, 1,25 (OH)D3 and VDR gene expression (P0.05). There was no correlation between the vitamin D endocrine system and BMD in initial SLE patients. There was no correlation between the vitamin D endocrine system and the disease activity either. Conclusion Vitamin D endocrine system may play an important role in SLE, but the level of VDR gene is not correlated with BMD and disease activity.

Objective To report clinical manifestations, electroencephalogram (EEG), and the genotypes of two siblings with type Ⅲ Gaucher disease. Methods Two patients with different features were siblings. Their clinical data, signs, peripheral leukocytes acid β-glucosidase activity, andGBA gene were analyzed. Results (1) The proband was a boy. He visited us at the age of nine years old because of hepatosplenomegaly, thrombocytopenia and growth retardation without any neurologic symp-toms. He had normal intelligence but abnormal EEG ifndings. The activity of acid β-glucosidase in his leucocytes decreased to 1.5 nmol h-1·mg-1 Pr (normal range 6.0-16.7 nmol h-1·mg-1 Pr), supporting the diagnosis of type Ⅲ Gaucher disease. (2) The elder sister of the proband was 12 years old. She had tonic-clonic seizure and myoclonus seizure from the age of seven years old. Mild hepatomegaly, abnormal EEG, poor effect for antiepileptics, and progressive deterioration of psychomotor abilities were found. Her blood leucocytes acid β-glucosidase activity decreased to 1.8 nmol h-1·mg-1 Pr (normal range 6.0-16.7 nmol h-1·mg-1 Pr). Two heterozygous missense mutations, c.680A>G, (p.N188S) and c.1342G>C (p.D409H) were detected from the two siblings, respec-tively.Conclusions Patients with type Ⅲ Gaucher disease usually have the onset in childhood with typical features of Gaucher disease without neurologic involvement. Abnormal EEG may be helpful to the differential diagnosis of type I or type Ⅲ. On the other hand, neurologic manifestations could be presented as the ifrst symptom in some patients without viscera enlargement. The patients of type Ⅲ Gaucher disease with the same genotype could have different phenotypes, even between the siblings.

There were 11 males and 10 females with an average age of 65.81±6.45 years (range 42-76 years). All of them were unilateral. There were 11 cases on the left and 10 cases on the right. There were 11 cases of osteoarthritis secondary to hip dysplasia, 4 cases of hip osteoarthritis, 4 cases of aseptic necrosis of femoral head and 2 cases of femoral neck fracture. Different wire tension belt ventral compression wiring technologies were used for each fracture type. Harris hip function score, Parker activity score, and visual analogue scale (VAS) score of hip pain were evaluated during follow-up. X-ray films were taken to evaluate the fracture healing, prosthesis position, loosening and dislocation. Results:Three new fracture types were proposed: A transverse fracture from the greater trochanter tip to the base (4 cases); B oblique fracture from the greater trochanter tip to the base (according to the fracture line direction, type B was further divided into types B1 (4 cases) and B2 (6 cases); and C fracture line from the greater trochanter to subtrochanteric plane (7 cases). Among the 21 patients, one died at an early stage, two were lost during follow-up, and 18 were followed up for an average of 30.7±7.6 months. In 18 patients, the mean operation time was 110.0±20.0 min, and the mean intraoperative blood loss was 356.9±115.7 ml. The patients' Harris score was 35.26±5.52 at the preoperative, 65.7±6.42 at the 3 months after operation, and 87.75±6.21 at the final follow-up. The difference was statistically significant ( F=377.23, P<0.001). The patients' Parker score was 2.17±0.98 at the preoperative, 5.94±1.11 at the 3 months after operation, and 8.01±0.77 at the final follow-up. The difference was statistically significant ( F=170.96, P<0.001). The patients' VAS score was 6.22±1.11 at the preoperative, 2.61±0.92 at the 3 months after operation, and 1.28±0.67 at the final follow-up. The difference was statistically significant ( F=139.71, P<0.001). Deep vein embolism, heterotopic ossification was noted in one and another patient, respectively. The patient with non-union refused reoperation and had a broken steel wire, lower-limb limp, and no notable pain at the 12-month follow-up examination. Radiographs of 17 patients showed good location of the femoral prosthesis and no chronic pain. Conclusion:Different types of greater trochanter fractures in total hip arthroplasty were proposed, using different wire tension belt ventral compression wiring technologies for the various types of femoral greater trochanter fractures during total hip replacement can improve clinical outcomes.
