Journal of Third Military Medical University 2017;39(17):1734-1738
Objective To determine the effects of down-regulating serine/threonine/tyrosine kinase 1 (STYK1) on the proliferation, apoptosis and invasion and on MEK/ERK and PI3K/AKT signaling pathways in human colorectal cancer cell line HCT-15.Methods The expression of STYK1 protein was downregulated by RNA interference technique in HCT-15 cells. Then, the STYK1 protein expression was detected by Western blotting.CCK-8 assay was used to measure the effect of STYK1 down-regulation on the proliferation of the cells, flow cytometry was employed to detect the cell apoptosis, and Transwell chamber assay was adopted for cell invasion ability. The expression levels of p-ERK1/2 and p-AKT were detected by Western blotting. Results Compared with the siRNA NC group,STYK1 down-regulation significantly inhibited the proliferation (P < 0.01) and invasion (P < 0.01),induced cell apoptosis (P < 0.01),and suppressed the expression levels of p-ERK1/2 and p-AKT in the HCT cells transfected with STYK1 siRNA (P <0.01).Conclusion STYK1 down-regulation inhibits the proliferation and invasion and induces the apoptosis in colorectal cancer cells, which may be related to its inhibition on the MEK/ERK and PI3K/AKT signaling pathways.

ObjectiveTo design a weight-bearing pressure casting system for transtibial prostheses to make the total surface bearing socket and to control the stump and bearing during the casting process.MethodsThe weight-bearing pressure casting system was designed. 6 cases weared the socket by this method and traditional patellar tendon bearing socket by hand casting. Socket interface pressure was compared, and the effect of casting was assessed.ResultsCompared with patellar tendon bearing socket, the socket pressure in the socked by this method was distributed more equally throughout the transtibial residual limb during the casting process, and completed the stumps compression and stump-end pressure control.ConclusionWeight-bearing pressure casting system can make the total surface bearing socket, and improve the fit of the socket.

This study was aimed to optimize the extraction process conditions of lycopene in tomato. Ketchup was used as raw material. Method verification, single factor experiment and orthogonal method were used in the study on extraction process of lycopene. The results showed that the best optimization process conditions of lycopene extraction with ethyl acetate: extraction temperature at 50℃, extraction time for 40 min, ethyl acetate concentration of 80%, solid-to-liquid ratio of 1?2 (g·mL-1). Under these conditions, the extraction rate of lycopene reached 15.564 mg·100g-1. It was concluded that the extraction process of lycopene can provide experimental basis for further development and utilization of lycopene.

Objective To explore the atypical ultrasonic appearances of breast fibroadenoma and analyze the cause of mis diagnosis. Methods A total of 493 lesions in 485 patients of breast fibroadenoma detected by ultrasound examination andconfirmed by pathology were retrospectively analyzed.Atypical appearances were analyzed and misdiagnosis rate were cal-culated.Statistical methods were taken to compare the misdiagnosis rates in lesions with various atypical appearances andthose with none or single atypical appearance.Misdiagnosis rates in lesions with different sizes and pathological types wereanalyzed statically. Then misdiagnosing causes were analyzed. Results A total of 404 lesions were diagnosed correctly,in-cluding 99 lesions with atypical appearances, and the other 89 lesions were misdiagnosed, which showed atypical appear-ances.The misdiagnosis rate of lesions with ≥2 atypical features was higher than that with none or single atypical feature(x2 =256.40,P< 0.05).Large lesions (maximum diameter> 3 am) showed higher misdiagnosis rates than small ones(maximum diameter ≤3 cm,x2=6.73,P<0.05),and complex fibroadenoma lesions also showed higher misdiagnosisrate than simple ones (x2 =188.01,P<0.05).Conclusion The lesions with various atypical appearances, large size andcomplex fibroadenoma in pathology are easy to be misdiagnosed.

OBJECTIVE:To explore the method and approach for traditional Chinese pharmacists to play an active role in case of emergent epidemic situation. METHODS:Taking the opportunity of H1N1 flu treatment, the traditional Chinese pharmacists carried out pharmaceutical services such as drug counseling, establishing medication history for typical cases, offering guidance and advice on medication, retrospectively analyzing prescription and consulting records. RESULTS & CONCLUSION:In the differential treatment, pharmacists of traditional Chinese medicine should take full account of patients' individual differences, actively participate in the design of individualized dose scheme and offer medication guidance so as to enhance patients' medication compliance meanwhile enhancing the status of pharmacists of traditional Chinese medicine.

Objective To probe the characteristic ultrasonic appearances of hepatic adenoma. Methods The appearances of color Doppler flow imaging(CDFI)and contrast-enhanced ultrasound(CEUS)in 10 cases of hepatic adenoma were analyzed. Results Rich blood flow signals were found in all the hepatic adenoma lesions on CDFI.Large blood flow signals were detected in the periphery of all tumors. On CEUS,peritumoral feeding vessels displayed in early arterial phase and homogeneous enhancement in arterial phase was observed in all the 4 cases. The tumor capsule was enhanced continuously in all cases. One case with isoechoic enhancement in delayed phase was found the contrast agent wash out in portal phase. The other 3 cases showed the‘fast-in and slow-out'appearance.which was iso-echoic enhancement in portal phase and slightly hyperechoic enhancement in delayed phase. Conclusions The rich blood flow signals, early displaying of peritumoral feeding vessels, homogeneous enhancement in the arterial phase and continuously enhanced tumor capsule on CEUS are the distinctive appearances of hepatic adenoma.

An innovative approach to quantitatively analyze the histamine and its precursor histidine simultaneously in biological matrices was established for the first time based on double adsorption combined with high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS).The internal standard was 2-dihydroxybenzoic acid (DHB).The plasma and brain tissue homogenate was protein precipitated with 3-fold acetonitrile, and the supernatant was then sampled for injection analysis. The chromatographic separation of the target components was achieved on an amino chromatography column (ODS-SPXBridge? Amide).Gradient elution was carried out with the mobile phase consisting of solvent A (0.1% formic acid and 1mmol/L ammonium formate in water) and solvent B (acetonitrile).Mass spectrometry was employed for quantitative analysis with ESI ion source in multiple reaction monitoring (MRM) mode.In order to improve the specificity and accuracy, activated carbon and calcite were used for the double adsorption of biological matrices for the first time. The adsorbed matrix was then used for methodology validation. The results showed that histamine and histidine were linear in the quantitative range (correlation coefficient r ≥ 0.999).Accuracy, precision, extraction recovery, matrix effect and stability all met the requirements of biological sample analysis. All results suggested that the present method could not only be efficiently and reliably used for simultaneous quantitative analysis of histamine and histidine in biological samples, but also provide reference for the detection of other endogenous substances.
