1.The influence of Yupingfeng polysaccharide on T lymphocyte subgroup of adjuvant arthrits rat

Aim To explore the effects of Yupingfeng polysaccharide(YPF-P)on T cells of adjuvant arthritis(AA)in rats.Methods Freund's complete adjuvant was used to induce AA in rat.Secondary paw swelling of AA rats was measured with volumeter.Peripneral blood lymphocyte proliferation response induced by concanavalin A(ConA)was examined with MTT assay.The CD4+ and CD8+ T cell in peripheral blood were detected by flow cytometer.ELISA method was applied to determine the content of factor IFN-? and radio-immunity assay of factor IL-4.RT-PCR procedures were utilized to determine the expression of IFN-? mRNA.Results 80,160 mg?kg-1 YPF-P could singnificantly inhibit the secondary paw swelling of AA rats.The suppressesion of lymphocyte proliferation of peripheral blood T cell in AA rats was reversed.After treatment by YPF-P,the number of CD4+ cell and the ratio of CD4+/CD8+ markedly decreased.The level of IFN-? in YPF-P treating group was obviously decreased in comparison with that of model control group,while the level of IL-4 was increased.The mRNA expression of IFN-? was inhibited as well.Conclusion Yupingfeng polysaccharide can regulate the turbulence of periphery abnormal T cell subgroups of adjuvant arthritis rats and can recover Th1/Th2 balance to move towards Th1,which may be one of its mechanisms of inhibiting the secondary paw swelling.
2.Effect of neural cell adhesion moleculeon adhesion, migration and morphology of mouse BMSCs

Objective To explore the effect of neural cell adhesion molecule (NCAM) on adhesion,migration and morphology of mouse bone marrow-derived mesenchymal stem cells (BMSCs).Methods We isolated and cultured BMSCs from wild-type and NCAM gene knockout mice.The expression of NCAM was detected by Western blot and immunofluorescence.Wound healing and adhesion assays were used to detect cell migration and adhesion ability respectively.The morphological changes were observed and the expressings of protein β1 integrin,E-cadherin,β-catenin and N-cadherin were analysed by Western blot.Results The migration and adhesion of BMSCs were significantly reduced after NCAM gene knockout.Meanwhile,the expression of β1 integrin was lower than those in wild-type BMSCs (P<0.01).The morphology of NCAM gene knockout BMSCs changed from irregular to flattened,and expressed epithelial identification marker E-cadherin and β-catenin (P<0.05).However,the expression level of mesenchymal identification marker N-cadherin was decreased (P<0.01).Conclusions NCAM is involved in adhesion and migration of BMSCs via regulating the expression of β1 integrin.Furthermore,NCAMmay negatively regulate the mesenchymal-epithelial transitions of BMSCs.
3.Da Vinci robot surgical system versus traditional open surgery for old patients with upper abdominal surgical diseases: a case control study

Objective To compare the clinical application of Da Vinci robot surgical system (RSS) with traditional open surgery (TOS).Methods From Feb 2015 to Jul 2016,48 cases of upper abdominal surgical disease patients were divided into RSS group (23 cases) and TOS group (25 patients) randomly.Results The anesthesia time [(194 ±16)min vs.(181 ±11)min,t=3.262,P=0.002] and operation time [(167 ± 14) min vs.(158 ± 14) min,t =2.292,P =0.027] were much longer in the RSS,while the blood loss during operation significantly less than the TOS [(128 ± 62) ml vs.(190 ± 86) ml,t =-2.886,P =0.006].The RSS has obvious advantages in 24 h-drainage [(69 ± 27) ml vs.(114 ± 54) ml.t =-3.680,P =0.001],time to out-of-bed activity [(27.7 ± 8.0) h vs.(35.7 ± 9.9) h,t =-3.067,P =0.004],BPS,postoperative exhausting time[(27.2 ±5.9)h vs.(32.8 ±8.3)h,t =-2.690,P =0.01] and length of hospital stay [(10.4 ± 1.8) d vs.(11.8 ± 1.9) d,t =-2.600,P =0.013].But the total hospital cost was higher in RSS [(117 000 ± 10 000) yuan vs.(77 000 ± 8 000) yuan,t =15.087,P =0.000)].Conclusions The RSS is a much minimally invasive surgery,reducing blood loss and postoperative pain,promoting rapid recovery,shortening hospital stay.
4.Ultrasonographic application in predicting axillary lymph nodes metastasis in patients with breast cancer

ObjectiveTo evaluate the implication of ultrasonographic features of primary breast cancer tumors and axillary lymph nodes in predicting axillary lymph nodes metastasis in patients with breast cancer.MethodsA total of 108 patients with breast cancer were underwent examination of primary breast tumors and axillary lymph nodes by high frequency linear-array probes of ultrasound.The ages of patients,locations of primary tumors,numbers of tumors,maximum diameters of tumors,the longitudinal transverse axis ratio of tumors,mass boundary,ultrasonic patterns,micro-calcification,classification of blood supply,color pixel density(CPD),peak systolic velocity,resistance index,the longitudinal transverse axis ratio of axillary lymph nodes and maximum cortical thickness of axillary lymph nodes were recorded.ResultsOut of 108 patients with breast cancer,the longitudinal transverse axis ratio of tumor were greater than 1 in 75 (69.4 % ),micro-calcification in 57(52.8 % ),classification of blood supply were Ⅱ - Ⅲ in 57 (52.8% ),CPD were greater than or equal to 10% in 48 (44.4%),maximum cortical thickness of axillary lymph node were greater than or equal to 3 mm in 51 (47.2%),and longitudinal transverse axis ratio of lymph nodes were less than 1.5 in 59 (54.6%).Univariate analysis revealed that these six parameters were correlated to the axillary lymph node metastasis in breast cancer ( P <0.05).However,ages of patients,location of tumor in the breast,numbers of tumors,maximum diameters of tumors,mass boundary,ultrasonic patterns,peak systolic velocity and resistance index were not related to the axillary lymph node metastasis( P >0.05).Multivariate logistic regression analyses showed that CPD (OR:16.337,95% CI:4.537- 58.826),longitudinal transverse axis ratio of lymph nodes (OR:3.754,95% CI:1.269- 11.108) and microcalcificationand (OR:3.033,95 % CI:1.040 - 8.840) were risk factors of axillary lymph nodes metastasis in patients with breast cancer.ConclusionsThe application of ultrasonography in patients with breast cancer is useful in predicting axillary lymph nodes metastasis.
5.Naringin protects human umbilical vein endothelial cells against injury in-duced by high glucose through PI3 K/AKT/eNOS pathway

[ ABSTRACT] AIM:To investigate the protective effect of naringin ( Nar) on the injury of human umbilical vein endothelial cells ( HUVECs) induced by 33 mmol/L high glucose ( HG) and to explore its possible mechanisms.METH-ODS:The injury model was established by treating HUVECs with HG medium for the indicated time (6, 12, 24, 48 and 72 h) , and then the levels of NO, eNOS and p-eNOS were detected, respectively.The effects of Nar on high glucose-in-duced endothelial cell injury were observed.HUVECs were treated with Nar at concentrations of 5, 10, 25, 50 and 100 mg/L for 6 h, 12 h, 24 h, 36 h and 48 h.The levels of NO in the supernatants were measured.The effects of Nar on HG-injured HUVECs were explored by treating the cells with 10 μmol/L of LY294002, a PI3K inhibitor, or 0.5 μmol/L of AKT inhibitorⅣ, an AKT inhibitor, and then the levels of NO, PI3K, AKT, eNOS and their phosphorylated proteins were determined by Western blot.RESULTS:Nar at concentration of 50 mg/L significantly attenuated the injury of endothelial cells induced by high glucose ( P<0.01) , and the protective effects of Nar were abolished by pretreating with the inhibitor of PI3K or AKT (P<0.01).CONCLUSION: Nar protects endothelial cells against the injury induced by high glucose through PI3K/AKT/eNOS pathway.
6.Effect of donor and recipient gender on the recovery of renal function after donation after cardiac death renal transplantation

BACKGROUND: Donation after cardiac death (DCD) is becoming the main source of organs for transplantation. Donor and recipient gender may play an important role in preoperative evaluation and recipient selection of transplantation. OBJECTIVE: To analyze the effect of donor and recipient gender on the recovery of renal function after DCD renal transplantation, and to further guide the selection of recipients. METHODS: The clinical data of recipients and donors of DCD kidney transplantation performed at the First Affiliated Hospital of Zhengzhou University from July 2012 to March 2015 were analyzed retrospectively. According to donor and recipient gender, the donors and recipients were divided into four groups: group A (male donor, male recipient), group B (male donor, female recipient), group C (female donor, male recipient), group D (female donor, female recipient). The renal function of the recipients was recorded at 1 and 2 weeks, 1, 3, 6, and 12 months postoperatively to compare the effect of donor and recipient gender on the recovery of renal function. RESULTS AND CONCLUSION: The serum creatinine level in the groups A and B was lower than that in the groups C and D at postoperative different time points, and there was a significant difference at 3, 6 and 12 months postoperatively (P < 0.05). The estimated glomerular filtration rate in the groups A and B was significantly higher than that in the groups C and D at postoperative different time points (P < 0.05). The serum creatinine level in the group C was significantly higher than that in the other three groups at postoperative different time points (P < 0.05). The serum creatinine level in the group A was significantly higher than that in the group B at 3, 6 and 12 months postoperatively (P < 0.05). The glomerular filtration rate in the group A was significantly higher than that in the group C at postoperative different time points (P < 0.05). The glomerular filtration rate in the group B was significantly higher than that in the group D at 12 months postoperatively (P < 0.05). These results suggest that the male donors show superior renal function to the female donors. The postoperative recovery of renal function is relatively slow when the male recipients receive a kidney of female donors. The female recipients with lower body surface area who receive the kidney of elderly male donors with poor renal function can achieve favorable clinical effects. Therefore, the gender should be used as a reference index for selecting potential recipients.
7.The application value of water flea Daphnia pulex for hypoxia model.

Hypoxia-inducible factor (HIF) is an important transcription factor under hypoxic condition in many organisms, and plays a key role in the induction of hypoxia tolerance. It is necessary to establish a hypoxia model for HIF and to perform further hypoxia tolerance research. To investigate the value of Daphnia as a model organism in hypoxia precondition, we developed a preconditioning protocol with a model organism, Daphnia pulex. We found that two episodes of exposure to hypoxic solution resulted in enhanced hypoxia tolerance which is dependent on HIF. Comparative genomic analysis was also made to highlight the homology of HIF-related genes among Daphnia, fruitfly and human. We found that Daphnia is suitable for the study of human hypoxic injury as a model organism.

9.Relationship between urinary metals and lung function in college students

Objective:
To explore the association between urinary metals and lung function among college students, and to provide a theoretical basis for related research on metal exposure and lung function injury.
Methods:
A total of 45 healthy college students were recruited from North China University of Science and Technology in Caofeidian between 2017-2018. During the four seasons, information was obtained from questionnaires and physical examinations, lung function parameters were assessed, including FVC, FEV1, PEF, FEV1/FVC and FEF 25-75 , and morning urine samples were collected simultaneously. The urinary levels of 15 metals were measured by inductively coupled plasma mass spectrometry (ICP/MS); a Kruskal Wallis H test was used to compare differences in urinary metals during the four seasons; and a mixed effect model was used to assess correlations between urinary metals and lung function.
Results:
There were significant differences in the levels of urinary chromium, iron, nickel, copper, zinc, arsenic, selenium, selenium, molybdenum, cadmium, antimony and lead from 15 metals over the four seasons ( H =9.79- 20.61 , P <0.05). The differences observed in five lung function parameters over the four seasons were statistically significant ( F =61.72, 45.30, 47.61, 25.47, 35.13, P <0.05). The linear mixed effect model analysis showed that urinary concentrations of vanadium, manganese, iron, cobalt, nickel and antimony were negatively correlated with FEV1( B =0.202, 0.192, 0.181, 0.154, 0.131 , 0.283); urinary concentrations of aluminum, vanadium, manganese, iron, cobalt, nickel, zinc, cadmium, and antimony were negatively correlated with FVC ( B =0.252, 0.290, 0.292, 0.271, 0.201, 0.180, 0.171, 0.163, 0.381); urinary concentrations of manganese and antimony were negatively correlated with PEF ( B =0.291, 0.354)( P <0.05).
Conclusion
The increase of multiple metal concentrations among college students was related to lung function decline, the long term metal exposure might lead to lung function damage. So environmental metal pollution should be controlled.
10.Expression of FOS protein in glioma and its effect on the growth of human glioma cells.

OBJECTIVETo study the expression of FOS protein in human glioma tissues and its effect on tumor growth.
METHODSFOS protein expression in glioma tissues was determined with immunohistochemical (IHC) staining. Subsequently, 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-diphenytetrazoliumromide (MTT) assay, flow cytometry, transwell invasion and Western blotting were used to assay U87 and U251 cells with reduced FOS expression.
RESULTSThe expression of FOS in glioma was increased and strongly correlated with its pathological grade. Abrogating expression of FOS has suppressed proliferation and invasion, and delayed cell cycle at G1 phrase for both U87 and U251 cells.
CONCLUSIONThe expression of FOS protein in human glioma was strong. FOS protein probably plays a critical role in the progression of gliomas.
