1.Effect of transmembrane protein 45A on extracellular matrix synthesis by keloid-derived fibroblasts

Objective:To determine the expression of transmembrane protein 45A (TMEM45A) in keloid tissues and fibroblasts, and to evaluate its effect on extracellular matrix (ECM) synthesis by keloid-derived fibroblasts (KFs) .Methods:Samples of surgically excised keloid and normal foreskin tissues were collected from the Department of Dermatology and Department of Urology of Yanbian University Hospital from January 2019 to December 2020, and TMEM45A protein expression was determined in keloid tissues and KFs by Western blot analysis. KFs were divided into TMEM45A-specific small interfering RNA (siRNA) group and control siRNA group to be transfected with the TMEM45A-specific siRNA and control siRNA respectively. Then, Western blot analysis was performed to evaluate the effects of down-regulation of the TMEM45A gene on the expression of myofibroblast marker protein (α-smooth muscle actin) and ECM-related proteins.Results:Compared with normal skin tissues (1.00 ± 0.11) and fibroblasts (1.00 ± 0.20), TMEM45A expression levels significantly decreased in keloid tissues (0.26 ± 0.05) and KFs (0.41 ± 0.09), respectively ( t = 10.76, 4.75, P < 0.001, = 0.009, respectively). The expression levels of α-smooth muscle actin, ECM-related type Ⅰ collagen, type Ⅲ collagen, and fibronectin were significantly higher in the TMEM45A-specific siRNA group than in the control siRNA group ( t = -5.98, -4.57, -4.90, -7.19, P = 0.004, 0.010, 0.008, 0.002, respectively) . Conclusion:Lowly expressed TMEM45A in keloids may play an important role in the pathogenesis of keloids by promoting ECM synthesis.
2. Relationship between noise-induced hearing loss,hypertension and abnormal electrocardiogram in noise-exposed workers

OBJECTIVE: To explore the relationship between noise-induced hearing loss, hypertension and abnormal electrocardiogram(ECG) in noise-exposed workers. METHODS: A judgment sampling method was adopted to select 555 male workers with hearing loss as study group and 555 male workers with normal hearing as control group in the similar environment in an automobile manufacturing enterprise in Guangzhou. Pure tone audiometry,blood pressure measurement and ECG examination were performed in both groups to analyze the relationship between hearing loss and hypertension and abnormal ECG. RESULTS: The prevalence rate of hypertension and abnormal ECG was higher in the hearing loss group than the control group(P < 0. 05). The multiple logistic regression analysis showed that after adjusting confounding factors such as age,seniority,body mass index,drinking et al,the risk of hypertension in hearing loss group was higher than the control group(P < 0. 05) and the odds ratio(OR) was 2. 255 [95% confidence interval(CI) : 1. 093-4. 655 ],while adjusting the confounding factor of drinking,the risk of ECG abnormalities in hearing loss group was higher than the control group(P < 0. 05) and the OR was 1. 408(95% CI: 1. 027-1. 930). CONCLUSION: Workers exposed to noise with hearing loss increase the risk of hypertension and abnormal ECG.
3.Analysis of 66 children with atypical hemolytic uremic syndrome

Objective To summarize the clinical data of atypical hemolytic uremic syndrome (aHUS) and analyze the treatment and prognosis.Methods A prospective cohort study was conducted on 66 cases in Beijing Children's Hospital affiliated to Capital Medical University from January 2011 to December 2017.The children were divided into positive and negative auto-antibody groups according to the results of anti-factor H autoantibody test.The clinical characteristics,treatment plan and prognosis of the two groups were compared.Results Among the 66 children who met the inclusion criteria,there were 43 cases (65.2%) in the positive group,with an average onset age of (8.0±2.9) years.There were 23 eases (34.8%) in the negative group,with an average onset age of (3.0± 2.6) years.On the basis of plasma treatment,in the positive group,the usage rate of hormone was 83.3% (35/42) and the usage rate of immunosuppressive agents was 42.9%(18/42),while in the negative group,the rates were 63.6%(14/22) and 13.6%(3/22) respectively.The average follow-up time was 19.3 months.One child in each group was lost to follow-up.In the positive group,8 cases recurred (19.0%)and the average recurrence interval time was 16.1 months.In the negative group,7 cases recurred (31.8%) and the average recurrence interval time was 9.3 months.And the recurrent interval time in the positive group was more longer than the negative group (P < 0.05).A total of 85.9%(55/64) children had complete hemolysis control and complete recovery of renal function,in which the positive group was 85.7%(36/42) and negative group was 86.4%(19/22).However,7.8%(5/64) children had abnormal renal function,in which the positive group was 9.5%(4/42) and the negative group was 4.5%(1/22).And 4.7%(3/64) children died,in which the positive group was 2.4%(1/42) and the negative group was 9.1% (2/22).The one left (1.6%) showed dialysis dependence,which was positive for the auto-antibody.Multifactor Cox regression analysis showed that the age of less than 3 years old was the risk factor of poor prognosis (HR=4.651,95%CI 0.988-21.898,P=0.047).Conclusions The positive proportion of anti-factor H autoantibody in children with aHUS is high.The age of these children is older.Individualized therapy based on anti-factor H autoantibody and immunosuppressive therapy is of great significance for disease remission,preventing recurrence and improving the prognosis.Age less than 3 years old is the risk factor for poor prognosis.
4. Effect of cumulative noise exposure on hypertension in automotive manufacturing workers

OBJECTIVE: To analyze the relationship between the cumulative noise exposure( CNE) and prevalence of hypertension in automotive manufacturing workers. METHODS: A total of 1 535 noise-exposed male workers with length of service ≥ 1. 0 year from automobile manufacturing industries in Guangzhou City was chosen as study subjects by convenience sampling method. The noise intensity in the workplace was measured, the CNE calculated, and the occupational health examination conducted. RESULTS: The prevalence of hypertension of the study subjects was 8. 1%( 125/1 535). The prevalence of hypertension showed an upward trend with the increase of CNE( P < 0. 01). The multivariate logistic regression analysis showed that the prevalence risk of hypertension increased with the increase of CNE after adjusting for confounding factors such as body mass index,smoking and wearing personal protective equipment in automobile manufacturing workers( P < 0. 01). CONCLUSION: There is a dose-effect relationship between prevalence of hypertension and CNE in automobile manufacturing workers who exposed to noise.
5. Analysis of factors affecting the noise-induced high frequency hearing loss of male workers in a vehicle manufacturing enterprise

Objective:
To analyze the relationships between the exposure of occupational noise and the occurrence of noise-induced high frequency hearing loss in automobile manufacturing industry.
Methods:
From June to November 2017, 1579 male workers were selected for exposure to noise hazards in the automobile manufacturing industry in Guangzhou for more than one year (including one year) using the convenient sampling method. According to the hearing test results, 276 people in the high frequency hearing loss group and 1303 in the normal hearing group were surveyed, occupational health tests were conducted, and the noise intensity in the workplace was measured. The cumulative noise exposure (CNE) was calculated. The relationship between shift work and high frequency hearing loss of noise work is analyzed.
Results:
Compared with the normal hearing group, the noise exposure intensity, age, length of service, marital status, working system, and physical exercise of the high frequency hearing loss group were statistically significant (
