1.Fingerprint and Multi-components Determination of Saponins in Sanjie-zhentong Capsule

This study was aimed to establish a HPLC fingerprint of saponins in Sanjie-zhentong Capsule in order to make a quantitative analysis of the quality of Sanjie-zhentong Capsule. The Waters Symmetry ShieldTM RP18 (4.6 mmí 250 mm, 5 μm) column was used with a mobile phase of acetonitrile-water gradient elution. The flow rate was 1.2 mL/min. The column temperature was 30℃. The detection wavelength was 203 nm. The results showed that the fingerprint chromatography included 9 mutual peaks. The similarity among batches was more than 0.95. Compared with reference substance, five characteristic components were recognized. The five components are notoginsenoside R1, ginsenoside Rg1, ginsenoside Re, ginsenoside Rb1 and ginsenoside Rd. It was concluded that this method was rapid, simple and accurate and can be used as one of the effective methods for the quality control of Sanjie-zhen-tong Capsule.
2.The Analysis Methodology for Multiple Elements in Four Herbs ofSanjie Zhentong Capsules

To develop a method for the determination of 18 elements such as Pb, Cu, As, Hg, Mn, Ni, & Tl in Panax notoginseng,Bulbus fritillariae thunbergii,Coix seed, Resina draconis, and to control the contents of heavy metal elements in Sanjie Zhentong Capsule, the samples were digested by microwaves and then analyzed by appropriate determination parameters through ICP-MS, with the internal standard method to improve the matrix effect and interference. The correlation coefficientR2≥ 0.999 2. The lowest limits of quantification were from 0.002 8 to 0.54 μg·L-1. The experiments had better repeatability, while the recovery values ranged from 73.01% to 109.13%. The method is simple, accurate and high sensitive, and it can be used for the determination of rapid monitoring the multi-elements inPanax notoginseng, Bulbus fritillariae thunbergii, Coix seed,and Resina draconis.
3.Ilizarov technique in combination with bone grafting and ankle traction for treatment of periarticular nonunion at the distal tibia

Objective To evaluate the clinical efficacy of a combination of Ilizarov technique, bone grafting and ankle traction for treatment of nonunion of the distal tibia within 2 cm around the ankle joint. Methods A retrospective study was performed on the patients who had been treated at Department of Orthopaedics, Sir Run Run Shaw Hospital from September 2014 to January 2018 for nonunion of the distal tibia within 2 cm around the ankle joint. They were 8 men and 4 women, aged from 32 to 56 years ( average, 41 years). Their injury was all unilateral, involving 5 left and 7 right ankles. They had all received internal fixation at other hospitals for 6 to 18 months. After we treated them simultaneously with Ilizarov technique, bone grafting and ankle traction, they could walk with crutches as soon as 3 days after surgery in some cases. Their affected limbs were then subjected to gradual weight-bearing walking until they could walk normally without any aid 3 to 6 months after surgery. Their postoperative ankle functions were evaluated according to the ankle-hindfoot scores of the American Orthopedic Foot and Ankle Society ( AOFAS) system. Results The 12 patients were effec-tively followed up for 12 to 24 months ( average, 19 months). All their incisions healed well with no incision or pin-tract infection. All their bone nonunions healed after circular external fixation for 12 to 36 weeks. Their AOFAS scores ranged from 75 to 96 points, showing good functional recovery of the ankle joint. Conclusion A combination of Ilizarov technique, bone grafting and ankle traction is an effective treatment for nonunion of the distal tibia within 2 cm around the ankle joint, leading to preservation of the ankle function and prevention of posttraumatic ankle osteoarthritis.
4.Rapid preimplantation genetic diagnosis of -thalassemia SEA deletion with blastocyst cell whole genome amplification and short fragment Gap-PCR method.

OBJECTIVETo develop a rapid preimplantation genetic diagnosis method for -thalassemia SEA deletion based on blastocyst cell whole genome amplification (WGA) combined with short fragment Gap-PCR.
METHODSUsing multiple displacement amplification (MDA) WGA technique, we established a double-fluorescent PCR system of the housekeeping genes GAPDH and β-actin for WGA quality testing, and a genotyping PCR system of mutant and normal short sequences for α-thalassemia SEA deletion. The sensitivity and accuracy of this method for diagnosis of -thalassemia SEA deletion were evaluated by detecting lymphocyte samples containing different cell numbers from carriers of SEA deletion. The applicability of this method was evaluated by testing of 12 blastocyst biopsy samples.
RESULTSDetection of lymphocyte samples with different cell numbers using the method developed in this study revealed no ADO in 3-cell samples, and the product quantity of WGA became stable for 4-cell samples. Genotyping of the 10 blastocyst biopsy samples with successful WGA showed a genotype of --/ in 5 samples and / in the other 5 samples, which were consistent with the verification results.
CONCLUSIONSThe method developed in this study is a complete testing process for 4-6 blastocyst biopsy cells to allow rapid, accurate, and cost-effective PGD genotyping of -thalassemia SEA deletion using short fragment gap-PCR.
