Objective To study the clinical, image and pathological features of cerebral gliomatosis. Methods 2 cases with cerebral gliomatosis underwent routine MRI scan, contrast enhance MRI scan and pathologic examination of the lesions. Their clinical manifestations were observed during the past 2 years. Results Main clinic features of the 2 patients were headache, dizziness, nausea, vomit, diplopia, hemiplegia, hemianesthesia and ataxia. Cranial MRI scan showed long T 2 signals in the bitemporal lobes, biparietal lobes, corpus collosum, thalamus, caudate nucleus and putamen. Brain stem and cerebellum were also involved in 1 patient. The borders of the lesions were unclear and no mass-effect phenomenons were found. Contrast enhancement occurred only in 1 patient after Gd-DTPA injection. The biopsies in the 2 patients showed diffuse infiltrative growth of most astroglioma cells. The shape of nucleus was round or ellipse and the staining of nucleus was comparatively deep. Cleavage of nucleus was seldom. The 2 patients died in 4 to 6 months after the onset of the disease. Conclusions MRI scan and pathologic examination are essential diagnostic methods for cerebral gliomatosis. The prognosis of cerebral gliomatosis is poor.

OBJECTIVE To investigate the applicability of 3DMIA software to upper airway modeling in children with obstructive sleep apnea hypopnea syndrome (OSAHS). METHODS A total of 30 children diagnosed with OSAHS by polysomnography were included in this study. Data regarding upper airway structure were collected via spiral CT while sleeping and awake, from which a three-dimensional model of the upper respiratory tract from the nasopharynx to the supraglottic region using 3DMIA software was constructed. The upper airway volume and airway minimum cross-sectional area were measured employing software algorithms. RESULTS The upper airway volume and airway minimum cross-sectional area of the 30 children during sleep were significantly less than those while awake (P<0.01). CONCLUSION 3DMIA software modeling and software algorithm measurement are more objective than traditional radiology (e.g. Fujioka) with respect to evaluation of the extent of the upper airway narrowing in OSAHS children, and has good applicability to study upper airway morphology and function in children with OSAHS.

Objective:To investigate the balance of Th1/Th2 and Tc1/Tc2 in peripheral whole blood from SLE patients and find out the relationship between disease activity and the balance of type 1 and type 2 cytokines. Methods:Peripheral blood cells from SLE patients(active,n=15,inactive,n=20) were stimulated and normal subjects(n=20) with phorbol myristate acetate and ionomycin(PMA/I) for 4 h,then IFN-? and IL-4 in CD3~+CD8~-T cells(Th) and CD3~+CD8~+T cells(Tc) were detected by recently developed intracytoplasmic cytokine-staining techniques,anti-dsDNA antibodies in serum was detected with enzyme linked immunosorbant assay(ELISA) and immunoglobulin in serum and protein in urine were analyzed with rate nephelometry. Results:In SLE patients with active disease, the percent of IFN-?-secreting Th cells significantly increased compared with SLE patients with inactive disease and normal after in vitro PMA/I stimulation, the percent of IFN-?-secreting Tc cels significantly increased compared with normal, but IL-4-secreting Th cells and Tc cells did not vary significantly. In SLE patients with inactive diseases, only the percent of IFN-?-secreting Tc cells significantly increased compared with control. The percent of IFN-?-secreting Th and Tc cells in patients with anti-dsDNA antibody(+) in serum decreased significantly compared with patients with anti-dsDNA antibody(-).Although the percent of IL-4-secreting Th and Tc cells in patients with aberrantly high level of immunoglobulin had no change, but the mean fluorescent intensify(MIF) of IL-4-secreting Th and Tc cells increased significantly compared with patients with normal level of immunoglobulin. There was no correlation between the percent of IFN-? or IL-4 in Th or Tc cells and the content of protein in patients' urine. Conclusion:The balance between type 1 and type 2 cytokine-secreting T cells is disrupted in SLE patients, especially IFN-?-secreting T cells, which is significantly in favour of Th2 cells. The level of IFN-? or IL-4 in Th or Tc cells varied partly with disease activity, which deserved further study.

China Oncology 2017;27(3):161-168
Background and purpose: MORC2 (microrchidia family CW-type zinc finger 2, MORC2) is a newly identified chromatin remodeling protein that plays key roles in DNA-based biological processes including gene transcription and DNA damage repair. However, its functional role in breast cancer development and progression re-mains unknown. ALDH1A3 (aldehyde dehydrogenase 1 family member A3), a member of the aldehyde dehydrogenases (ALDH) superfamily, is a putative breast cancer stem cell marker, but its regulatory mechanism in breast cancer is poor-ly characterized. This study aimed to investigate the effects of knockdown of endogenous MORC2 on the expression levels of ALDH1A3 and the breast cancer stem-like phenotype in MCF-7 cells. Methods: Human breast cancer MCF-7 cells were infected with negative control short hairpin RNAs (shNC) and specific shRNAs targeting human MORC2 (shMORC2), followed by selection with puromycin to generate stable MORC2 gene knockdown cell lines. Western blot and real-time fluorescent quantitative polymerase chain reaction (RTFQ-PCR) were used to examine the protein and mRNA levels of ALDH1A3 in MCF-7 cells stably expressing shNC and shMORC2. Microsphere formation and fluo-rescence-activated cell sorting (FACS) assays were used to analyze the effects of knockdown of MORC2 on the breast cancer stem-like phenotype. Results: Western blot and RTFQ-PCR analyses revealed that the protein and mRNA levels of ALDH1A3 were significantly down-regulated in shMORC2 expressing cells as compared with shNC -transfected control cells. Moreover, mammosphere formation assay showed that knockdown of endogenous MORC2 in MCF-7 cells significantly reduced the ability of cells to form microspheres. Consistently, FACS assays demonstrated that shMORC2-transfected cells had a lower proportion of ALDH-positive stem cells as compared with shNC expressing cells. In contrast, knockdown of MORC2 did not significantly affect the CD44+CD24- stem cell population. Conclusion:MORC2 promotes a breast cancer stem-like phenotype through, at least in part, regulating ALDH1A3 expression.
