Objective To compare the specificity and sensitivity of antibodies against cell membrane associated DNA (cmDNA)、 nucleosome (AnuA)、 double stranded DNA (dsDNA)、 deoxyribonucleoprotein (DNP) and antinuclear antibodies (ANA) in systemic lupus erythematosus (SLE). To analyze the relationship between these auto-antibodies in SLE. Methods One hundred and twenty five SLE patients and 118 other rheumatic diseases patients were studied. The latter included primary Sj?觟gren syndrome, ploymyositis, dermatomyositis, rheumatoid arthritis, osteoarthritis and systemic sclerosis. Indirect immunofluorescence was used to measure the anti-cmDNA antibodies and ANA. The enzyme-linked immunosorbent assay (ELISA) was used to measure the AnuA. Anti-DNP antibodies were detected by latex agglutination text and anti-dsDNA antibodies were detected by colloidal gold-technique. Results The seropositive rates of AnuA、 anti-cmDNA、 DNP、 dsDNA antibodies and ANA were 68%、 38.4%、 51.2%、 49.6% and 95.2% respectively in SLE patients. Objective To detect the mutation of A379,we develop a TaqMan fluorogenic probe based amplification refractory mutation system (TaqMan-ARMS) and investigate whether the A379V variant and activity of Lp-PLA2 are the risk factors for CAD.Methods According to the amplification refractory mutation system(ARMS-PCR)combined with a TaqMan fluorogenic probe,we established and evaluated the assay teehnique of TaqMan-ARMS for measuring the genotype of variant.At the same time,we tested the aetivity of Lp-PLA2 in 395 patients with coronary heartdisease and 396 controls,whose clinical information [ages,CHO,GLU,TG,HDL,LDL,Hs-CRP,Lp (a)] were collected.Date was analyzed by using Independent-samples t test,Chi-square test,One-Way ANOVA,Binary Logistic Regression.Results CAD group had significantly higher Lp-PLA2 activity than the controls(31.51 nmol · ml-1 · min-1 ＞21.31 nmol ·ml-1 · min-1,F =16.40,P ＜ 0.001).Comparing the highest quartile of Lp-PLA2 activitv to the bottom quartile,OR was 7.50 (95％ CI:2.34-24.05) after adjustment for various traditional cardiovascular risk factors,including ages,sex,CHO,TG,Hs-CRP,Lp (a) and GLU; the genotype VV of A379V was associated with higher risk of CAD (OR =2.95; 95％ CI:1.22-7.15,P ＜ 0.05).Conclusions The TaqMan-ARMS real time PCR technique is established to analyze A379V genotype.Lp-PLA2 activity is significantly higher in CAD group and is a risk factor for CAD; the genotype VV of A379V is also a risk factor for CAD.

Objective To investigate the change and significance of regulatory T lymphocytes in peripheral blood of rats model of chronic abacterial prostatitis.Methods Twelve Wistar rats with weight of approximate 390 g were randomly divided into two groups,model group and control group.Rats in the model group was injected subcutaneously 17β-estradiol(0.25 mg/day,for 30 days) after castration to establish rat model of chronic abacterial prostatitis.Flow cytometry was applied to detect the frequency of CD+4 CD+25 cells and CD+8CD-28 cells in peripheral blood of rats after model establishment.Results Compared with control group (11.63±1.36)％,the proportion of CD+4 CD+25T lymphocytes in model group (7.90±1.74)％ significanlty decreased (P＜0.01).Compared with control group (24.64±4.76)％,the proportion of CD+8CD-28T lymphocytes in model group (17.18±2.83) ％ also significantly decreased (P＜0.01).Conclusions The changes of the ratio of CD+4 CD+25T lymphocytes and CD+8CD-28T lymphocytes in peripheral blood of rats model of chronic abacterial prostatitis provided evidences for pathogenic mechanism of regulatory T lymphocytes participating in the development of chronic abacterial prostatitis.

Objective To detect the sealing ability of iRoot as root end retrograde filling material in vitro experiment,and to compare it with mineral trioxide aggregate(MTA) in order to provide reference basis for clinical use of iRoot.Methods The dental crown was removed in 67 single root canal anterior teeth.The routine root canal preparation and filling were performed.The root end was resected by 3 mm for conducting root end retrograde preparation.Sixty-three teeth in the samples were randomly divided into 4 experimental groups.Then the different materials were used for root end filling:iRoot BP (group A,n=17),iRoot BP plus (group B,n=17),MTA (group C,n =17),and group D(iRoot BP + gutta percha,n =12),the residual 4 samples served as the control group.The apical microleakage of 10 teeth in each experimental group was detected by the fluid dynamics.The microleakage situation at the time points of 1,7,14,28 d was observed.In addition,2 teeth selected from each group were performed the filling material ultramicrostructure observation by SEM.Each 5 teeth were remained in the group A,B and C and performed the Micro CT scanning.The porosity rate of filling material was detected.Results No obvious edge gap was found between the filling materials and dentin surface in the group A,B and D by SEM,but obvious gap could be seen in various samples of group C.Micro CT scanning found that the porosity rates in the group A,B and C were 0.020 ±0.023,0.045 ±0.035 and 0.031 ± 0.011 respectively,but showing no statistical difference(P＞0.05).The fluid dynamic experiment showed that the microleakage detection value in each group had no statistical difference(P＞0.05).Conclusion The sealing ability of iRoot and MTA used in root end retrograde sealing has no obvious difference,But iRoot operability is superior to MTA.

Objective To evaluate the correlation of the ion selectivity electrode assay and arsenazo Ⅲ assay for serum calcium determination and compare the differences between two methods.Methods 142 serum samples were collected from patients and detected by ion selectivity electrode assay and arsenazo Ⅲ assay,the differences of two methods were compared,person correlation and Bland -Altman was used to discuss the correlation.Results The serum calcium concentration detected by ion selectivity electrode assay was (2.23 ±0.10)mmol/L and detected by arsenazo Ⅲ assay was (2.24 ±0.12)mmol/L,the difference between the two methods was not statistically significant (t =-1.09,P >0.05).The person correlation analysis demonstrated that the two methods had a positive correlation (r =0.393,P <0.01).The Bland -Altman consistency analysis showed that ion selectivity electrode assay was agreed with arsenazo Ⅲ assay.Conclusion The two methods for serum calcium concentration determination,the ion selectivity electrode assay was coincidence with the arsenazo Ⅲ assay.

In our gastroenterology clinic from Jun. 2008 to Jan. 2009, 20. 9% (277/1320) patients were diagnosed with functional dyspepsia (FD) and 14. 8% (41/277) of them with psychological problems according to Minnesota Multiphasic Personality Inventory (MMPI). In those with psychological disorders,92.7% were found with somatization, 60. 9% with depression and anxiety, 97.6% with abdominal pain,75.6% with bloating, 63.4% with early fullness and 36. 6% with nausea. More than 80% patients with anxiety and depression complained irritability, worry, fatigue, poor concentration, memory loss and insomni.a. After 4 weeks of psychological consultation and anti-depression treatment, 5 patients had significant improvement, 31 had improvement and 3 had no response. The overall response rate was 87. 8%.In summary,there is a high prevalence of depression and anxiety in population with FD. Hypochondria and somatization are common among these patients. Psychological consultation with anxiolytic drugs may have good therapeutic effects.

Objective To evaluate the effect of non-linear blending function for dual-energy CT on image of pulmonary angiogra-phy.Methods 27 patients underwent dual energy CT pulmonary angiography(CTPA).Data obtained with 100 kVp,140kVp,and non-linear blending were divided into group A,B,and C respectively.CT value of emboli were measured.Corresponding signal to noise ratio(SNR )and contrast to noise ratio(CNR)were calculated.One-way Anova analysis and Friedman test were used to ana-lyze statistical significance among these values.Results On quantitative analysis of 27 patients,there was no statistical difference for CT value of CTPA among the three groups(P >0.05).For the noise,CNR and SNR,there were statistically significant among the three groups(P <0.05).Conclusion The non-linear blending function has certain advantages in improved image signal-to-noise ratio and it can be used in CT pulmonary angiography examination for patients suspected pulmonary embolism clinically .

Objective To design and screen small interefere RNA (siRNA) targeting of HOXA7,and to investigate the effect of the siRNA on human lung cancer LETP-a-2 cells proliferation and apoptosis in vitro.Methods Three pairs of siRNA targeting of HOXA7 and one pair of siRNA for negative control were transfected respectively into LETP-a-2 cells through cationic liposome.The mRNA and proteion expression levels of HOXA7 were observed by reverse transcription polymerase chain reaction (RT-PCR) and Western blot analysis.The effect of HOXA7 siRNA on growth and apoptosis of LETP-a-2 cells were measured by MTT and flow cytometry.Results All the three pairs of siRNA could inhibit HOXA7 expression effectively,among which siRNA2 got the best effects,the silence rates were (57.344±4.743) ％ on mRNA level and (52.219±0.550) ％ on protein leval.The proliferation was inhibited and the apoptosis was promoted by the siRNA targeting HOXA7 in LETP-a-2 cells,among which siRNA2 got the favourite results,the inhibitory rate was (48.144±4.992) ％ and the apoptosis rate was (26.613±0.612) ％.Conclusion The siRNA2 targeting of HOXA7 enrolls in promoting apoptosis and inhibiting grows of LETP-a-2 cells,indicating that manipulation of HOXA7 expression may be a potential therapeutic strategy for cancer.
