1.Optimization of the Extraction Technology of Bushen Huoxue Capsules by Central Composite Design-re-sponse Surface Method

OBJECTIVE:To optimize the extraction technology of Bushen huoxue capsules. METHODS:Central composite de-sign-response surface method was used to optimize the extraction technology with the amount of water and boiling time as main fac-tors using normalized value of the contents of icariin and salvianolic acid B,the yield of dry extract as index. Validation test was al-so conducted. RESULTS:The optimal extraction technology was as follows as 15-fold water,reflux extracting for 75 min,extract-ing for 2 times. The deviation of measured value and predicted value was 0.000 3% in validation test. CONCLUSIONS:The cen-tral composite design-response surface method can be used to optimize the extraction technology of Bushen huoxue capsules with good prediction,and the technology is simple and stable.

3.Development and application of foldable lest tube rack

Objective To develop a new foldable test tube rack to solve the inconvenience and safety issues arising from transferring the specimens to test all together.Methods 482 urine specimens of 69 daily routines collected in the morning from various sections of the hospital were randomly divided into experimental group and control group.The experimental group was placed in the foldable test tube rack; however,the control group was managed with traditional methods.Results The index of stability in the experimental group was 98.3%,95.0% in the control group,and there was significant difference in both groups ( x2 =4.06,P < 0.05).Environmental pollution did not occur in the experimental group,while it happened six times in the experimental group,there was significant difference between both groups( x2 =4.06,P < 0.05 ).The time for transferring specimens was (18.72 ± 2.92) minutes in the experimental group,(27.28 ± 3.87 ) minutes for the control group,there was significant difference between them( t =3.04,P < 0.01 ).Conclusions Foldable test tube rack for transferring specimen is time-saving,as well as avoids re-sorting and places specimens,improves work efficiency; enhances the specimen placement stability,avoiding environmental pollution arising from dumping,overflowing,and sprinkling.
4.Effects of acupuncture on neuro-electrophysiological activities in hippocampal CA1 and CA3 areas of rats with post-traumatic stress disorder

Objective:To observe the effects of acupuncture on the characteristics of neuro-electrophysiological activity in hippocampal CA1 and CA3 areas of rats with post-traumatic stress disorder (PTSD).Methods:Fifty Sprague-Dawley (SD) rats were randomly divided into a blank group,a model group,a grasping group,a Western medicine group and an acupuncture group,with 10 rats in each group.Except for the blank group,rats in the other 4 groups all received the combined stress modeling method.Rats in the Western medicine group were intragastrically administrated with paroxetine hydrochloride,those in the acupuncture group received acupuncture intervention,those in the grasping group received grasping fixation,and those in the model group and the blank group did not receive any interventions.After 14 d of intervention,the interspike interval (ISI) and power spectral densities (PSD) were analyzed and mapped by in vivo multiple channels to record the neuron clusters discharge in the hippocampal CA1 and CA3 areas.Results:Compared with the blank group,ISI was prolonged in the CA1 and CA3 areas of the model group and the grasping group,and the concentrated PSD distribution area moved down (P<0.05 or P<0.01).Compared with the grasping group,the ISI of the CA1 and CA3 areas in the Western medicine group and the acupuncture group was shortened,and the concentrated PSD distribution area moved up (P<0.05 or P<0.01).The ISI and PSD distributions in the CA1 and CA3 areas of the acupuncture group were not statistically different from those in the Western medicine group (both P>0.05).Conclusion:Both acupuncture and paroxetine hydrochloride can significantly regulate the neuro-electrophysiology activity of hippocampal CA1 and CA3 areas in PTSD rats,which may be one of the mechanisms of acupuncture intervention to promote PTSD recovery.
5.The effects of gutter oil on liver function,renal function and serum lipid metabolism in mice

Objective To determine the effects of gutter oil on liver function,renal function and serum lipid metabolism in mice.Methods A total of 72 mice were averagely divided into 6 groups.Mice were gavaged with different dosages (5 μL/g,1 0 μL/g and 20 μL/g)of gutter oil or cooking oil every other day for continuing 8 weeks.The indexes of liver, kidney and serum lipid parameters were assayed.Results The moisture,acid value and peroxide value of the gutter oil exceeded the national standard.After continuing 8 weeks'intake of oils,alanine aminotransferase (ALT)of gutter oil group with a dosage of 5 μL/g,as well as aspartate aminotransferase (AST)of gutter oil group with a dosage of 1 0 μL/g significantly increased (P <0.05),compared with the cooking oil group.Serum creatinine (Scr)of the gutter oil groups with 3 dosages significantly increased compared with the control groups (P <0.05 ).Meanwhile,blood urea nitrogen (BUN)of gutter oil groups with a dosage of 5 μL/g and 1 0 μL/g were also significantly increased respectively(P <0.05). However,no significant differences were observed in triglyceride (TG)and total cholesterol (TC)among the groups. Conclusion Eight weeks'intake of gutter oil could cause the damage to liver and renal functions.
6.Detection of interleukin-8, matrix metalloproteinase-9 and interferon gamma levels in the cerebrospinal fluid of patients with tuberculous meningitis and viral meningitis

Objective To investigate the diagnostic values of interleukin-8 (IL-8), matrix metalloproteinase-9 (MMP-9) and interferon gamma (INF-γ) levels in patients with tuberculous meningitis and viral meningitis by detecting the contents of these biomarkers in the cerebrospinal fluid (CSF). Methods Enzyme-linked immunosorbent assay (ELISA) was used to detect the levels of IL-8,MMP-9 and INF-γ in the CSF of patients with tuberculous meningitis (n=20),viral meningitis (n=15) and noninfectious neurologic diseases (n=20) who admitted to our hospital from August 2010 to November 2011. Results The IL-8,MMP-9 and INF-γlevels in the samples from the tuberculous meningitis patients were significantly higher than those from either viral meningitis or noninfectious neurologic diseases (P<0.05).The contents of IL-8 and MMP-9 in patients with viral meningitis were significantly higher than those in patients with noninfectious neurologic diseases (P<0.05),while that of INF-γ showed no significant difference between the 2 groups (P>0.05). Conclusion Detection of IL-8,MMP-9 and INF-γ in CSF is useful in the diagnosis of tuberculous meningitis; IL-8 and MMP-9 participate in the progress of viral meningitis; the detective value of high concentration of INF-γ in CSF is more significant in diagnosis of tuberculous meningitis than that of IL-8 and MMP-9.
7.Preparation of amylopectin modified dipyridamole liposome and its tissue distribution in mice.

AIMTo prepare amylopectin anchored dipyridamole (DIP) liposome and to study its tissue distribution in mice.
METHODSThe regular DIP liposomes were prepared by film-scatter method. The amphiphilic O-palmitoyl amylopectin was synthesized and added to modify the surface of liposome. The entrapping efficiency, zeta potential, mean diameter, span of modified and regular liposomes were assayed. The RP-HPLC was used for the determination of DIP concentration in mice tissue.
RESULTSAfter modification, the entrapping efficiency depressed, zeta potential was raised, mean diameter and span had no obvious change. The level of DIP in lung, liver and spleen for regular liposomes were higher than that of injections. Compared with regular liposomes, the modified liposomes increased the DIP level in lung, and decreased the DIP level in liver, spleen, moreover, lengthened the retention time of DIP in lung.
CONCLUSIONThe distribution of modified liposome in mice was markedly changed as compared with regular liposomes and injections. The modified liposomes had obvious lung targeting property.

8.Fluorescence labeling for human bone marrow mesenchymal stem cells with PKH26

Objective To establish a method of labeling human mesenchymal stem cells (MSCs) with PKH26 in vitro.Methods MSCs were cultured and labeled with PKH26 according to the manufacturer's instruction.The growth,fluorescence intensity and serial subcuhivation of labeled MSCs were analyzed with the confocal laser microscope and the flow cytometry.The biological characteristics of labeled MSCs were investigated by RT-PCR.Results The labeled MSCs appeared red fluorescence and the labeling rate was 100 percent.During serial subcuhivation of labeled MSC from passage 1 to passage 7,the fluorescence intensity and the labeling rate of MSCs were gradually decreased.The biological features such as morphology,growth,expression level of nucleostemin and GAPDH gene and capability of differentiation into osteoblast in vitro were not affected by labeling.Conclusion Labeling the human MSCs with PKH26 is an effective and practical method,which can be used as an important tool in the study on the homing, plasticity and transplantation of MSCs.
9.The efficacy and safety of autologous platelet-rich plasma in the treatment of moderate to severe dry eye disease

Objective:To evaluate the efficacy and safety of using platelet-rich plasma (PRP) eyedrops for the treatment of moderate to severe dry eye disease (DED).Methods:A total of 395 patients (790 eyes) with moderate to severe DED diagnosed and treated in the Armed Police Liaoning Corps Hospital and the PLA 967 Hospital from March 2018 to October 2019 were collected. Random number table method was used to divide into autologous PRP treatment group (196 cases, 392 eyes) treated with autologous PRP and control group (199 cases, 398 eyes) treated with artificial tears. The changes of subjective symptoms of DED, Schirmer test (ST), corneal fluorescein staining (CFS) and Ocular Surface Disease Index (OSDI) before and after treatment were observed in both groups.Results:After 1 course of treatment, the ST values of both groups increased, which was statistically significant compared with before treatment ( P<0.05). After treatment, the OSDI and CFS scores of the two groups were reduced. The difference in OSDI and CFS scores of the PRP treatment group before and after treatment was statistically significant ( P<0.05), and there was no statistically significant difference in the control group before and after treatment ( P>0.05). After treatment, the OSDI and CFS scores of the PRP treatment group were lower than those of the control group [(16.8 ± 18.7) scores vs. (43.2 ± 14.5) scores, (0.21 ± 0.53) scores vs. (1.62 ± 0.69) scores], the differences were statistically significant ( P<0.05). After one course of treatment, the total effective rate of the autologous PRP treatment group was higher than that of the control group [80.1% (157/196) vs. 51.76% (103/196)], the difference was statistically significant ( P<0.05). Conclusions:Autologous PRP can treat patients with moderate-to-severe autologous PRP treatment group, which can greatly improve patients' eye discomfort and other symptoms.
10.Efficacy of intravitreal Conbercept injection for macular edema secondary to macular branch retinal vein occlusion

AIM:To evaluate the efficacy and safety of intravitreous injection of Conbercept for macular edema secondary to macular branch retinal vein occlusion(MBRVO).METHODS:Clinical records of 19 patients(19 eyes) who were diagnosed MBRVO with macular edema (ME) in our hospital,from July 2015 to September 2016 were retrospectively analysed.All patients were treated with an intravitreal dose of 0.5mg or 0.05mL conbercept ophthalmic injection by the 3+ pro re nata (PRN) project.All patients were evaluated by best corrected visual acuity (BCVA),central retinal thickness(CRT),the number of the injection,and the complications.RESULTS:During the 1,2,3 and 6mo after treatment the mean BCVA all improved significantly(P<0.01),and the mean CRT reduced statistical significantly(P<0.01);3 eyes had refractory ME,and micro-aneurysm leakage were identified by FFA,and the ME was suppressed effectively after local laser photocoagulation.No complications,such as secondary vitreous haemorrhage,retinal detachment,persistent high intraocular pressure and endophthalmitis were observed during subsequent follow-up.CONCLUSION:Intravitreous injection of conbercept for macular edema secondary to MBRVO may reduce macular edema and improve visual acuity effectively and safely in the short term.
