1.Influence of Different Extraction Conditions on Naringin Content in Drynaria Extract

Objective To study the different extraction condition on naringin content in Drynaria extract.Methods The Naringin content in Drynaria extract was determined by HPLC.The e ffect of extraction solutions,drying temperature(60℃,100℃and 120℃)and drying time (0,0.5,1.0,1.5,2.0,5.0,10.0,25.0,50.0hours respectively)on naringin content in Drynaria extract was investigated.Results The optimal extraction solution for Drynaria extract was water,and the loss of naringin enhan ced with the increase of drying temperature and the prolongation of dryin g time.Con-clusion The result can provide references fo r the extraction technology,preparation selection and quality control of the compound including Rhizoma Drynariae.
2.Determination of Heavy Metals Content in Five Kinds of Chinese Medicinal Material by ICP-MS

Objective To establish an ICP-MS method for the determination of heavy metals,including copper(Cu),arsenium(As),cadmium(Cd) and plumbum(Pb),in five kinds of Chinese medicinal material.Methods The samples were digested by closed-vessel microwave.The four heavy metals were directly analyzed by ICP-MS,with elements 72Ge,115In,and 209Bi as the internal standards.Results For all of the analyzed heavy metals,the correlative coefficient of the calibration curves was in the range of 0.999 5~ 0.999 7.The recovery rates were in the range of 92.3 % ~ 108.0 %,and RSD were in the range of 2.4 % ~ 5.8 %.Conclusion The method is convenient,rapid,accurate and can be applied to determine four heavy metals,including Cu,As,Cd and Pb,in Chinese medicinal material.
4.Effects of Total Saponins of Chaenomeles speciosa on Degranulation Ratio of RBL-2H3 Mast Cells

Objective To study the effects of total saponins of Chaenomeles speciosa on release of β-hexosaminidase from rat basophilic leukemia-2H3 ( RBL-2H3 ) mast cells. Methods After rat RBL-2H3 mast cells were prepared, total saponins of Chaenomeles speciosa and the RBL-2H3 mast cells were co-cultured.The toxic effects of total saponins of Chaenomeles speciosa on mast cells were detected by MTT method, β-hexosaminidase release rate was measured by fluorescence quantitative spectrophotometric method, and cell supernatants of tumor necrosis factor-α( TNF-α) release were detected by ELISA. Results After total saponins of Chaenomeles speciosa were co-cultured with RBL-2H3 mast cells with different antigen stimulation, β-hexosaminidase release rates and the levels of TNF-α of mast cells significantly decreased compared with the control group. Conclusion Total saponins of Chaenomeles speciosa inhibit the degranulation of RBL-2H3 mast cells in a dose dependent manner, which provids basis for studying mechanism of inhibiting allergic reactions.
5.Optimized formulation of Xiao'er Magan Orally Disintegrating Tablet by central composite design-response surface methodology

AIM:To optimize formulation of Xiao'er Magan Orally Disintegrating Tablet by central composite design-response surface methodology. METHODS:By two factors and five levels central composite design,independent variables were the proportion of optimized microcrystalline cellulose 50 and mannitol and the pressure,while disintegrating time and tensile strength were dependent variables. Binary linear and quadratic models were used to estimate the relationship between the dependent and the indepedent variables,and to delineate response surface method and overlay contour plots in order to select the optimal formulations. RESULTS:The binomial regression equation were TS =0. 096 88X1 + 0. 027 6X1X2(r = 0. 759),DT = 9. 845 4X1 -63. 196X2 + 40. 092 9X22 (r = 0. 838) which indicated the quantitative relationship between the factors and the evaluation indexes was characterized. The optimal formulation were the ratio of microcrystalline cellulose 50 and mannitol in the range of 0. 35 and 0. 6,and the pressure was in the range of 6 and 8 KN. CONCLUSION:The optimized formulation validation is close to the estimated value,indicating that the method has a good predictability and reliability.
6.Correlation study of interleukin-28B rs12978960 genetic polymorphism and the retreatment efficacy of peginterferon/ribavirin in chronic hepatitis C genotype 1 b relapsers

Objective To investigate the virological response in hepatitis C virus (HCV)genotype 1b relapsers after 48 weeks of peginterferon/ribavirin (peg-IFN/RBV)combination retreatment,and to explore the predictive value of interleukin (IL )-28B rs12978960 genetic polymorphismon virological response.Methods From 2012 to 2014,genotype 1b chronic hepatitis C (CHC)relapsers in He′nan Provincial People′s Hospital were retreated with combined peg-IFN/RBV for 48 weeks and followed up for 24 weeks off-treatment.Host IL-28B genetic polymorphism was detected.Predictive factors associated with virological response and sustained virological response (SVR)were analyzed.Independent-samples t test was conducted in continuous variables,whileχ2 test or Fisher exact probability test was conducted in counts data.Results A total of 61 patients finished 48 weeks of peg-IFN/RBV combination therapy and were further followed up for 24 weeks off-treatment.Mean age was (46.7 ±12.4)years.Thirty-seven patients (60.7%)were male and 49 were rs12978960 CC genotype.After 48 weeks of retreatment with peg-IFN/RBV and 24 weeks of off-treatment follow-up,40 patients (65 .6%)achieved SVR.Rapid virological response (RVR)and SVR of younger patients were both significantly higher than those of older patients (100.0% vs 67.4% and 85 .0% vs 47.6%,respectively;both P =0.006).IL-28B rs12978960 genotype was predictive to RVR and SVR.Patients with RVR and SVR had higher carriage rates of IL-28B rs12978960 CC genotype compared with those without RVR and SVR (both P <0.05 ).Patients with CC genotype had higher rates of RVR (34.1 % vs 0;χ2 = 10.625 ,P =0.006 ),end-of-treatment virological response (84.1 % vs 70.6%;χ2 =5 .563,P =0.039 )and SVR (77.3% vs 35 .3%;χ2 =9.572,P =0.007)than those with CT/TT genotype.However,there were no statistical differences of extended RVR (34.1 % vs 29.4%;χ2 =0.122,P =0.809)and early virological response (79.5 % vs 82.3%;χ2 =0.612,P =0.964).Conclusions Retreatment with antiviral therapy is necessary in CHC patients with genotype 1b. IL-28B rs12978960 genetic polymorphism is predictive to the SVR of retreatment,especially for patients without RVR,which will provide individualized treatment and optimize the treatment strategy.
7.Electrophysiologic study of f-wave amplitude in chronic atrial fibrillation associated with rheumatic heart disease

To investigate the electrophysiologic characteristics of f-wave amplitude and to evaluate its role in development and persistence of chronic atrial fibrillation (AF) associated with rheumatic heart disease (RHD). Methods: Epicardial mapping was performed in 44 patients with chronic AF of RHD who underwent heart valve surgery. Ten patients with supraventricular tachycardia served as the control group. Results:The f-wave amplitude of left atrium (LA) and middle and low LA posterior regions were significantly lower than those of the control group. The f-wave amplitudes of the upper, middle and low sections in LA posterior region were significantly lower than those in right atrium (RA) (P<0.05). The f-wave amplitudes were compared before and after electrocardioversion in 14 patients with chronic AF. The mean atrial electrogram amplitude during sinus rhythm was significantly higher than that during AF (P<0.01).The f-wave amplitude in left appendage was higher than that in LA posterior region (the upper,middle and the lower part),P<0.05.The f-wave amplitude in the upper section of LA was significantly higher than that in the middle section. The f-wave amplitude in AF group was not correlated to the diameter or volume of both atria. Conclusion: There are amplitudes differences between the upper, middle and lower LA,suggesting that the middle and lower sections of LA posterior wall may be the region producing anisotropy and reentrant circle.
8.Chin morphology in subjects with different vertical skeletal craniofacial pattern.

OBJECTIVETo evaluate whether there is difference with regard to chin morphology in subjects with different vertical skeletal craniofacial pattern and the relationship among them.
METHODSThe sample was composed of 80 adolescents who denied orthodontic treatment history and presented Class I skeletal pattern, aged (12.69+/-0.70) years. They were divided into three groups according to mandibular plane angle: High angle group (21 cases, FH/MP> or = 32 degrees), average angle group(43 cases, 22 degrees RESULTSSNA, SNB and ANB presented no significant difference among three groups. Compared with the low angle group, high angle subjects exhibited increased chin height, depth/height ratio, concavity and decreased chin angle. Therefore the chin looked lathier and less protrusive in high angle group. Positive correlation was found between mandibular plane angle and chin height, concavity, curvature (P<0.05), negative correlation between mandibular plane angle and chin minimal depth, depth/height ratio, angle. CONCLUSIONDifference emerged with regard to chin morphology in subjects with different vertical craniofacial skeletal pattern, which deserves to be taken into consideration in orthodontic treatment planning.

9.The correlation analysis of MMP-13 and AGG and Col-Ⅱin the chondrocytes caused by nutritional deficiencies in rab-bits

Objective To explore the relationship among the expression level of MMP?13, AGG and Col?II in the chon?drocytes caused by nutritional deficiencies in rabbits. Methods 30 New Zealand white rabbits were randomly divided into autolo?gous chondrocyte transplantation group (control group, n=10), nutrition block group (surgery group, n=10), and peripheral nutrition block group (sham surgery group, n=10). 4 weeks after treatment, the rabbits were sacrificed for undergoing the observations on general and histological level;real?time PCR assay was employed for testing the expression level of MMP?13, AGG and Col?II;cel?lular apoptosis percentage was observed through TUNEL stain. The relationship among the apoptosis level, cartilage cells histologi?cal Mankin score as well as the expression level of MMP?13, AGG and Col?II were analyzed. Results Based on the Mankin score, there was a statistic difference between surgery group and control group. On the other side, there were no statistic differenc?es between sham surgery group and control group. 4 weeks after treatment, surgery group presented a higher apoptotic percentage compared with control group;this value between sham surgery group and control showed no significant differences. There was an increased mRNA expression level of MMP?13 and a decreased mRNA expression level of AGG and Col?II in surgery group com?pared with control group;no statistic differences of all these values was found between sham surgery group and control group. His?tological Mankin score and apoptotic percentage presented positive correlation (r=0.922, P<0.001), the regression equation:Y=-0.548+0.404X, R2=0.844 (F=157.735, P<0.001); the mRNA expression level of MMP?13 and apoptotic percentage presented positive correlation (r=0.942, P<0.001), the regression equation:Y=0.951+0.116X, R2=0.883 (F=219.054, P<0.001). There was a nega?tive correlation between the mRNA expression level of MMP?13 and the mRNA expression level of AGG as well as Col?II (r=-0.956,-0.945, P<0.001). Conclusion Damage of cartilage cells causes the up?regulation of the MMP?13 expression which could ex?acerbate the degeneration of cells. It could induce the down?regulation of AGG and Col?II mRNA expression, which will cause the extracellular matrix synthesis disorder.
10.Study on the establishment of data management system of emergency response in fighting against flooding in China.

OBJECTIVETo make the emergency response to the diseases control and prevention with flooding more scientific ally-based and efficient.
METHODSHubei and Jiangxi province were chosen as the study fields and to establish a data management system of emergency response on flooding using remote sensing (RS) and geographical information system (GIS).
RESULTSSpatial and basic data together with data on flooding were linked to the code of location, and established on the platform of GIS. Through this system, information on flooding was able to be more rapidly and visually analysed.
CONCLUSIONUsing this system, the extent of flooding and the possible epidemic of some communicable diseases during flooding could be ascertained and predicted scientifically and the resource and capacity of emergency response to the disease control and prevention in flooding areas could also be evaluated.
