2.Polymorphism of ATP-binding cassette transporter A1 gene in sporadic Alzheimer's disease

Objective To investigate the association of the single nucleotide polymorphisms (SNPs)G→A (R219K) in the exon 6 of ATP-binding cassette transporter A1 (ABCA1) gene with sporadic Alzheimer's disease (SAD) in the Han Chinese population.Methods Genotypes were determined by PCR-restriction fragment length polymorphism (PCR-RFLP) in 168 SAD patients and 215 healthy controls. The association between the polymorphism and AD was estimated with the odds ratios (OR).Results The frequency of A allele in SAD cases was significantly lower than in controls (37.8 % vs 48.1%,x~2=8.204, P=0.004),and the frequency of AA genotype in SAD cases was also significantly lower than in control (14.3% vs 22.8%,x~2=8.230,P=0.016).Logistic regression analyses adjusted for age,gender and ApoEe4 status showed that the risk for AD was decreased in A allele (G/A+A/A genotypes,OR 0.57, 95% CI 0.36—0.91,P=0.019) or AA homozygote carriers (OR 0.40,95% CI 0.21—0.77,P= 0.006) compared with GG genotypes carriers.Conclusion Our data indicate a genetic association between polymorphism in exon 6 of ABCA1 gene and the risk of AD and reveal that AA genotype or A allele of ABCAI gene may have a protective effect on AD in the Han Chinese.
3.Analysis of Pathogens Distribution and Antibiotics Use in 93 Cases of Bronchiolitis Child

OBJECTIVE:To provide reference for antibiotics use. METHODS:A retrospective study was conducted to analyze the detection of pathogens and antibiotics use in 93 children with bronchiolitis in our hospital between Jan. 2013 to Dec. 2014. Pathogens detection mainly included respiratory virus,pneumonia mycoplasma IgM antibody and pneumonia chlamydia IgM anti-body and sputum culture. RESULTS:Among 93 cases,22 cases (23.66%) were positive in respiratory virus IgM antibody test, mainly for respiratory syncytial virus;17 cases(18.28%)were positive in sputum culture test,mainly for the Moraxella catarrha-lis,Haemophilus influenza and Streptococcus pneumonia;10 cases(10.75%)were positive in Atypical pathogens test,mainly for mycoplasma pneumonia. There were 47 cases(50.54%)of antibiotics;Among them,only 12.77%(6/47)had clear indications for using antibiotics;the children who were 1-3 months old had the highest percentage of antibiotics,accounting for 81.82% at the same age;the duration of disease was significantly prolonged in the cases who used antibiotics,with average duration of(8.27± 3.32) d. CONCLUSIONS:Bronchiolitis is still usually caused by virus infection in our hospital,the ratio of antibiotics use are high and most of them don't have bacterial infection indications.
4.Relaxin and cardiovascular fibrosis

Cardiovascular fibrosis is the common pathological manifestation of many heart diseases.Relaxin inhibits fibrosis through several mechanisms as follows relaxin inhibits fibroblast function,collagen synthesis/secretion and the influence of several profibrotic factors.Relaxin stimulates MMP expression.Relaxin rapidly inhibits cardiovascular fibrosis and has no effect on normal tissue.It is a potential and selective antifibrotic target of cardiovascular diseases.
5.Effect of irbesartan on activation of renal nuclear factor-kappa B in experimental type 2 diabetic rat

Objective To develop a rat model of type 2 diabetes, and to investigate the effect of AT1 receptor antagonist-irbesartan on activation of renal NF-?B in type 2 diabetic rat. Methods The rats of model groups were intraperitoneally given low-dose streptozotocin (STZ, 30 mg/kg) after having the sucrose-and fat-enriched diets(20% sucrose, 10% pig fat, 2. 5% cholesterol) for one month. Immuohistochemistry and computer image-pattern analysis system were used to analyze activation of NF-?B and expression of monocyte/macrophage (ED-1) in renal tissues. Results (1) Insulin resistance was induced by feeding diets enriched in sucrose and fat, and hyperglycemia was induced with a dose of STZ that did not cause diabetes in chow-fed rats. After 6 weeks, rats of model group presented itself early changes of diabetic nephropathy (DN). (2) Compared to normal group, the activation of NF-?B and expression of ED-1 increased in glomeruli of experimental type 2 diabetic rat. Irbesartan inhibited significantly the activation of NF-?B, ameliorated monocyte/macrophage infiltration, partly improved the renal function and matrix accumulation. Conclusions (1) A rat model of type 2 diabetes mellitus is developed successfully by combination of dietary-induced insulin resistance and low-dose STZ-induced hyperglycemia. (2) Renal NF-?B activation is greatly increased in experimental type 2 diabetic rat. The protection of irbesartan against kidney is associated, at least in part, with down-regulating NF-?B activation and monocyte/macrophage recruitment in renal tissue.
6.Difference in Pulmonary Function between COPD and Overlap Syndrome Patients

Objective To explore the difference in pulmonary function between chronic obstructive pulmonary disease (COPD) and overlap syndrome patients.Methods 50 hospitalized COPD patients who were randomly recruited were divided into two groups, which contained 33 COPD patients and 17 overlap syndrome patients respectively. All the 50 patients’pulmonary function was evaluated and the results were analyzed statistically.Results The pulmonary function test showed that only PEF and V75 significantly decreased in overlap syndrome patients compared with COPD patients(P=0 032 and 0 048 respectively),But pulmonary volume, small airway function and pulmonary diffusion function in 5 COPD patients with severe sleep apnea syndrom (SAS) were significantly reduced compared with COPD patients.Conclusions Upper airway obstruction in SAS patients aggravates the pulmonary ventilation function in overlap syndrome. And obesity deteriorates both of the pulmonary function and hypoxemia in patients suffered from COPD.

12.Meanwhile 10 healthy elderly subjects of similar age and gender ratio were matched as controls.There was no significant difference of age,gender,education,onset duration and MMSE.Attention task was carded out with silent color-word task:all participants pressed the left or right button to select the answer of congruence or incongruence when color nouns in red,green,blue or yellow ink appearing on the left part of screen and a target color appearing on the right part of the screen by.During the tasks,reaction time,false ratio,missing ratio and fMRI image acquisition was respectively recorded,which were later analyzed on SPSS 11.0 statistical parametric mapping software 2 (SPM2) respectively.Results The reaction time in dAD,AD and control group were (2214.4?1071),(2020.6?558.3) and (840.0?254.5) ms respectively.The false ratio were 8.3%,6.9% and 0.7% respectively,and the omit ratio were 3.6%, 2.9% and 0 respectively.Either AD with or without depression was slower than the controls in reaction time (P

Objective Hepatic stimulator substance(HSS)is newly defined as a growth mitogen to hepatocytes.This sutdy is aiming to investigate the effect of recombinant human hepatic stimulator substance(rhHSS) on growth of liver oval cells(OVCs). Methods OVCs were prepared from intoxicated rats with 3'-methyl-dimethylaminoazobenzene.After confirmation of OVC morphologically and histochemically,the cell cultures of OVC were exposed to various dosages of rhHSS for 12 and 24?h,respectively.The cellular proliferation was analyzed by MTT and flow cytometry. Results Administration of rhHSS(160-400 mg/L)inhibited the proliferation of OVC,as indicated by MTT and cell cycle analysis,the effect appeared non-dose dependent pattern.The peak of inhibition occurred at 240?mg/L.After incubation with 240?mg/L of HSS for 12 and 24?h,the percentages of S-phase were reduced 47.8% and 35.8% to those of the untreated cells.respectively.Conclusion rhHSS exhibits the inhibitory effect on growth of OVC.

10.Early-phase changes of parameters related to myocardial fibrosis in acute myocardial infarction

AIM:To investigate the changes in parameters related to myocardial fibrosis of H2 relaxin (H2RLX) and procollagen type I C-terminal peptide (PICP),transforming growth factor-?1 (TGF-?1) level in the plasma after acute anterior myocardial infarction. METHODS:Forty-one patients with anterior myocardial infarction (MI) and 10 controls with normal coronary vessels were selected. Plasma H2RLX,PICP,and TGF-?1 levels were determined by enzyme linked immunosorbent assay (ELISA) in 10 control subjects and 41 post-MI patients at day 3 and 7 post-MI. RESULTS:(1) H2RLX level at day 3 post-infarct had no difference between the patients and the controls. No marked change in H2RLX at day 3 versus day 7 post-infarct was observed. (2)The level of PICP at day 3 post-infarct was markedly increased followed by a fall at day 7 post-infarct,but was still higher than that in control. (3) TGF-?1 level,which was similar at day 3 and 7 post-infarct,was higher than that in control. (4) At day 7 post-infarct,the level of H2RLX was positively correlated with the fasting blood glucose in MI group. CONCLUSION:In early period of acute MI,profibrotic factors such as TGF-?1 expression and collagen expression increase. However,antifibrotic relaxin has no change.
