1.Correlation analysis of methylation status of UNC5C gene promoter with colorectal cancer UNC5C

Objective To investigate the correlation between methylation status of UNC 5C gene promoter with colorectal cancer UNC5C .Methods 54 cases of sporadic colorectal cancer and related normal mucosal tissue ,as well as 6 colon cancer cell lines and fiber cell lines(FCL) in the oncology department of the Kailuan General Hospital from February 2010 to March 2013 were selected as the research objects and performed the mRNA and methylation analysis for exploring the correlation between the netrin‐1 receptor UNC5C defects with colorectal cancer disease .Results mRNA of UNC5A and UNC5B was expressed in the detected colorectal cancer cell lines .Except FCL for UNC5C ,all of the cancer cell lines had no mRNA expression .In colorectal cancer tissue , UNC5C methylation was significantly higher than that in the normal mucosa ,the difference was statistically significant (P<0 .05) . The methylation levels of UNC5C gene in the stage Ⅰ to Ⅱ ,and the stage Ⅲ to Ⅳwere significantly higher than non‐methylation levels ,the differences were statistically significant (P<0 .05) .The correlation analysis by the Spearman method showed that the UNC5C defects was positively correlated with colorectal cancer disease (r= 0 .856 ,P< 0 .05) .Conclusion Netrin‐1 receptor UNC5C defect has certain correlation with colorectal cancer disease .
2.Management of infertility in patients with non-obese polycystic ovary syndrome by micro-pulse infusion of GnRH

[Summary] Patients with infertility and non-obese polycystic ovary syndrome ( PCOS) were treated with continuous subcutaneous pulse infusion of GnRH. After the treatment, a 32-year old female had regular menstrual cramps. Dominant follicle occurred after 2 months of treatment. The patient was pregnant and now has a healthy baby boy. It shows that the pulse infusion of GnRH could induce spontaneous ovulation and natural fertilization of the patients with non-obese PCOS.
3.Protective effects of glutamine pretreatment on occludin protein in rats with intestinal ischemia-reperfusion injury

AIM:To determine the effects of glutamine ( Gln) pretreatment on occludin protein in the rats with intestinal ischemia-reperfusion ( I/R ) injury.METHODS: Male Wistar rats ( n =30 ) were randomly divided into 3 groups (n=10):sham group, I/R group and Gln pretreatment group.The rats in Gln pretreatment group were pretreated with Gln at dose of 1 g? kg-1? d-1 by orogastric route for 7 d, and those in the other 2 groups were pretreated with the same volume of normal saline .Intestinal I/R was induced by 30-min occlusion of the superior mesenteric artery followed by 24 h of reperfusion.After the operation, the levels of IL-10, IL-2, TNF-α, SOD and MDA were measured.The occludin protein was determined by the methods of immunohistochemistry and Western blotting .RESULTS: The occludin protein level in I/R group was significantly lower than that in sham group and Gln group (P<0.05).The levels of MDA and TNF-αin I/R group were significantly higher than those in sham group and Gln group ( P<0.05 ) .The levels of SOD , IL-10 and IL-2 in I/R group were significantly lower than those in sham group and Gln group ( P<0.05 ) .CONCLUSION:Glutamine has a protective effect on occludin protein in intestinal ischemia-reperfusion injury .The mechanism may be rela-ted to oxidative stress response and inflammatory inhibition .
4.Study of cytogenetic characteristics on myelodysplastic syndrome and acute myeloid leukemia

ObjectiveTo evaluate the cytogenetic characteristics of the patients with acute myelomonocytic leukemia (AML-M4), acute monocytic leukemia (AML-M5)and myelodysplastic syndrome (MDS).MethodsChromosomes of bone marrow cells were prepared by short-term culture.Karyotype analysis was performed in 100 AML-M4,46 AML-M5 and 115 MDS by R-banding technique.Results26 % (26/100) AML-M4 had clonal cytogenetic abnormalities which mainly include +8,t(8;21) and -7.26 % (12/46) AML-M5 had clonal cytogenetic abnormalities which mainly include +11. 39 %(45/115)had clonal cytogenetic abnormalities which mainly include +8, Hypodiploid and -7. ConclusionCytogenetic detection is very important for the diagnosis of myelodysplastic syndrome and acute myelocytic leukemia.
5.Study of inhibitory effect of bortezomib on leukemia cell line HEL

Objective To explore the effects of proteasome inhibitor bortezomib alone and in combination with chemotherapeutic agents in acute leukemia cell line HEL. Methods Tumor cell growth inhibition and apoptosis was measured by MTT assay and FACS analysis, respectively. Apoptosis- and cell cycle-associatd protein expression levels were measured by Western blotting assay. Results Bortezomib induced the suppression of tumor cell growth and apoptosis in a dose-dependent manner with an inhibitor concentration IC50 values 7.15 nmol/L in acute leukemia cell line HEL. Further combination treatment with doxorubicin or cytarabine increased tumor cell growth suppression in acute leukemia cell line, as compared to single drug treatment alone. Bortezomib caused an increase in G2/M in a time-dependent manner. Bortezomib alone and in combination with chemotherapeutic agents treatment increased the bax and p27 expression and decreased the bcl-2 expression even without influencing p53 expression levels. Conclusion Bortezomib may be an effective therapeutic drug for patients with acute leukemia. Combining bortezomib with doxorubicin might be more effective for displaying tumor cell growth inhibitor effects in acute leukemia cells through regulation of bcl-2, bax and p27 proteins and independent of p53 status.
6.Effects of different disinfection methods on antibacterial activity of TiO2 nanotubes

Objective To study the influence of different kinds of disinfection methods on antibacterial activity of TiO2 nanotubes. Methods TiO2 nanotubes were fabricated with 0.5%wt HF on pure titanium to acid-etching combined with an? odization as experimental group. The pure Ti was control group. They were disinfected by three kinds of disinfection meth?ods, which were alcohol immersion, autoclaving and UV irradiation. The bacterial adhesion ability was evaluated by observ?ing the morphology of samples, measuring the roughness and contracting angle. The film contact method was used to evaluate the antibacterial activity of samples with Staphylococcus aureus (Sa) after different infection treatment. The bacterial morphol?ogy was observed by field emission scanning electron microscope (FE-SEM). Results The treatment group has higher sur?face roughness. The bacterial adhesion ability of experimental group was stronger compared with that of control group. The contracting angle of samples exposed to UV irradiation was smaller than that of alcohol immersion and autoclaving treatment group. The viable count and FE-SEM showed that there were more bacteria and more complete morphology on the sample surface after alcohol immersion. The viable count was less and bacterial morphology suffered some damage on the sample sur?face after high pressure steam sterilization. The least viable count and the most complete destruction of bacterial morphology were found after ultraviolet (UV) germicidal irradiation. Conclusion UV irradiation shows the best antibacterial activity, the second is autoclaving and the worst is alcohol immersion.
7.Cyclic fatigue performance of different types of stainless steel root canal files

BACKGROUND: Root canal files fracture is a kind of common oral clinical complications. Therefore, it is of clinical importance to study the cyclic fatigue of different types of stainless steel root canal files.OBJECTIVE: To explore the effect of the bending angle and cross-sectional area of root canal instruments on cyclic fatigue of stainless steel files.METHODS: Thirty 25# stainless steel K files (25 mm in length) were selected, the same to 30# and 35#. And these three kinds of files were respectively randomized into three groups (n=10 per subgroup). In the nine subgroups, the files were placed into self-made root canals at the bending angle of 45°, 60°, 90°, and driven by a 16:1 reducer (350 r/min).Fracture time in each subgroup was recorded and converted into the number of rotations. The microstructure of the fracture end of the files was observed under scanning electron microscope. Fractured files were collected and the crack tip length was measured by an electronic caliper.RESULTS AND CONCLUSION: At the same bending angle, the number of rotations resulting in file fracture was significantly reduced with the increasing of the cross-sectional area of the files (P < 0.05). When the type and cross-sectional area were same, the number of rotations resulting in file fracture was significantly reduced with the increasing of the bending angle of the root canals (P < 0.05), indicating a reduction in the cyclic fatigue performance of the files. Toughness fracture occurred in all the files, and circular or oval roughness nests with different sizes and microcavities formed on the fracture surface. In addition, brittle surfaces could be detective between the roughness nests. The diameter of roughness nests was gradually increased with the increasing of the bending angle of the root canals. For 30# and 35# files, the number of brittle surfaces was gradually increased with the increasing of the bending angle of the root canals. For 25# files, the crack tip length was gradually decreased with the increasing of the bending angle of the root canals, but there was no significant difference (P > 0.05). For 30# and 35# files, the crack tip length was significantly shortened with the increasing of the bending angle of the root canals (P < 0.05). But there were no significant changes in the 35#-60° and 35#-90° groups (P=0.095). At the same bending angle, there were no regular changes in the crack tip length in the different types of files. In conclusion, with the increasing of the cross-sectional area, the cyclic fatigue performance of the files with the same length and taper is reduced and the files become easy to be fractured; with the increasing of the bending angle of the root canals, the files with the same types are apt to be fractured near the root tip, indicating the shorter length of the crack tip indicates the more difficulty in the removal of the broken files.
8.Establishment of Total Quality Management of Hospital Pharmacy New Model Based on the Function Transformation of Drug Control Room

OBJECTIVE:To establish and carry out total quality management and evaluation system in hospital pharmacy. METHODS: The background and the implementation steps of the total quality management in hospital pharmacy were analyzed and summarized. RESULTS: Seven steps were included in total quality management of hospital pharmacy, i.e. role shifting of drug control room, transpositional thinking, orientation defining, network construction of quality management, improvement of quality management regimen, fusion of quality management with the construction of department culture. CONCLUSIONS: The practice of the new model of the total quality management in hospital pharmacy has contributed to the preliminary establishment of the quality evaluation standard for dispensing as well as the forming of the total quality control system in hospital pharmacy and the upgrading of the scientific management in hospital pharmacy.
9.Relationship between urinary albumin excretion rate and cardiac structure and function in type 2 diabetes mellitus

Objective To study the relationship between urinary albumin excretion rate(UAER)and cardiac structure and function in type 2 diabetes mellitus(T2DM).Methods Five hundred and twenty patients with T2DM were divided into three groups by 8 h UAER:normoalbuminuria group(424 cases),microalbuminuria group(75 cases)and macroalbuminuria group(21 cases).At the same time,50 healthy people were selected as control group.The cardiac structure and function in four groups were evaluated by echocardiographic measurements.Results Left atrial dimensio[(34.39±3.46)mm],left ventricular posterior wall thickness[(10.34±1.05)mm],relative wall thickness(0.46±0.05),interventricular septal thickness[(10.49±1.07)mm],left ventricular mass[(167.37±32.12)g],left ventricular mass index[(87.62±12.16)g/m2]in macroalbuminuria group were significantly higher than those in control group[(32.59±2.71)mm,(9.30±0.77)mm,0.42±0.04,(9.75±0.81)mm,(147.33±27.23)g,(80.14±13.81)g/m2](P<0.05),mean while the ratio of E/A(0.82±0.19)in macroalbuminuria group was significantly lower than that in control group(1.21±0.16)(P<0.05).Conclusions In T2DM patients with albuminuria,the cardiac structure and function have changed.With the increase of UAER,the impairments are more prominent.And it may predict diabetes cardiomyopathy.
10.Treatment of metacarpal and phalangeal articular fracture dislocation: external fixation device or mini plate

Objective To compare the clinical outcome of external fixation device with Kirschner wire or fragment fixation pin with mini plate and screw fixation in treatment of metacarpal and phalangeal articular fracture dislocation. Methods From October 2002 to March 2008, 106 patients with metacarpal and phalangeal articular fracture dislocation were randomly divided into A and B group. The 53 patients in A group were treated with external fixation device with Kirschner wire or fragment fixation pin. There were thumb injury in 24 cases, fracture-dislocation of proximal interphalangeal joint(PIP) in 36 cases, fracture-dislocation of metacarpophalangeal joint (MP) in 17 cases. The 53 patients in B group were treated with mini plate and screw fixation. There were thumb injury in 22 cases, fracture-dislocation of PIP in 30 cases, fracture-dislocation of MP in 23 cases. Duncan rating criteria were used to compare finger range of motion. Results The mean follow-up of 16.8 and 17.5 months in A group and B group. According to Duncan rating criteria, there were excellent in 33 cases, good in 16, fair in 3, and poor in 1 case. The excellent and good rate was 92.5% in A group. There were excellent in 30 cases, good in 17, fair in 5, and poor in 1 case. The excellent and good rate was 88.7% in B group. In A group, 1 case of wound infection was found. The average arc of motion of thumb joint was 134°±21° while the average arc of motion of other fingers was 248°±19°. In B group, no wound infection occurred. The average arc of motion of thumb joint was 122°±18° while the average arc of motion of other fingers was 225°±17°. Conclusion External fixation device with Kirschner wire or fragment fixation pin was better than the application of mini plate and screw fixation in treatment of metacarpal and phalangeal articular fracture-dislocation.
