1.Study on histone deacetylase inhibitor LBH589 reversal of multiple myeloma cells’ drug resistance mechanism in vitro

Objective To study on a new generation of histone deacetylase inhibitor LBH589 on multiple myeloma (MM) resistant cells associated with changes in gene expression,and the mechanism of LBH589 reversal of MM cells' drug resistance.Methods By Western blot analysis,LBH589 (0,20,50 nmol/L) and 50 nmol/L LBH589 combined respectively with dexamethasone of 5 μmol/L at 24 h,expression of histone H4 acetylation and bcl-X gene on MMIR cells were detected.By real-time fluorescence quantitative PCR analysis,LBH589 (0,20,50 nmol/L) and 50 nmol/L LBH589 combined respectively with dexamethasone of 5 μmol/L at 24 h and 48 h,expression of TOSO on MM1R cells were detected.Results Western blot analysis showed that single LBH589 and combined with dexamethasone showed at 24 h the up-regulation on expression of the histone H4 acetylation[(0.205±0.008) %,(0.346±0.009) %,(0.580±0.053) %,(0.986±0.012) %,F =992.957,P =0.032],while down-regulation on expression of the bcl-X in MM1R cells in a dose-dependent manner[(1.210±0.160) %,(0.930±0.036) %,(0.730±0.017) %,(0.488±0.029) %,F =56.432,P =0.028].However,the group of single LBH589 was less effective than the combined group (all P < 0.05).Real-time fluorescence quantitative PCR analysis showed single LBH589 and that combined with dexamethasone showed at 24 h and 48 h,the down-regulation on expression of the TOSO in MM1R cells in a dose-dependent manner,24 h specific numerical were (1.00±0.00) %,(0.55±0.01) %,(0.47±0.01) %,(0.38±0.01) % (F =1006.344,P =0.041),48 h specific numerical were (1.00±0.00) %,(0.39±0.04) %,(0.05±0.01) %,(0.03±0.00) % (F =2383.977,P =0.045),however,the group of single LBH589 was less effective than the combined group (all P < 0.05).Conclusion Histone deacetylase inhibitor LBH589 can recover dexamethasone sensitivity of MM1R through effect the gene expression of bcl-X and TOSO,promot histone acetylation,and inducing cell apoptosis to treat tumor.
2.Expression of enhancer of zeste homolog 2 in U266 cell line and bone marrow mononuclear cells of multiple myeloma patients

Objective To investigate the expression of DNA methylation related protein enhancer of zeste homolog 2 (EZH2) in multiple myeloma (MM) cell line U266 and enriched MM cells (MM-BMMNC) from 10 patients,and analyze the potential role of DNA methylation in the occurrence and development of MM.Methods Human MM cell line U266 and MM cells from 10 MM patients were studied.Enriched bone marrow mononuclear cells (N-BMMNC) from 10 healthy persons were used as the control.The expression of EZH2 were determined by Western blot using the ratio of target protein EZH2 and the internal reference β-actin expression as the final results.Results The preliminary in vitro results showed that EZH2 expression was higher in MM cell line U266 and patient MM-BMMNC (N-BMMNC 0.2277±0.1306,MM-BMMNC 0.9220±0.1301,U266 0.9730±0.1029),with statistically significant differences (P < 0.05).Conclusion EZH2 is enriched in U266 and MM-BMMNC.DNA methylation may have a close correlation with the occurrence and development of MM.
3.Origin and development of umbilical therapy in traditional Chinese medicine.

The origin and development of umbilical therapy in traditional Chinese medicine is explored from related literature in the history. As a result, the Shang period is regarded as initial period of umbilical therapy, while periods from Han Dynasty, Jin Dynasty and Southern-Northern Dynasties to Sui Dynasty and Tang Dynasty could be taken as stage of primary development. Time from Song Dynasty, Jin Dynasty and Yuan Dynasty to Ming and Qing Dynasties is believed as mature stage. Also the manipulation, application principle, indications and contraindications of umbilical therapy are explained. A brief overview of modern development of umbilical therapy is also described.

5.Effect of aspheric multifocal toric IOL implantation on visual acuity and contrast sensitivity in patients with cataract

AIM: To observe the effect of intraocular lens ( IOL) implantation on visual acuity and contrast sensitivity in patients with cataract.
METHODS: Fifty - eight cases ( 72 eyes ) cataract patients with regular cornel astigmatism, in our hospital from May 2014 to May 2015 were randomly divided into two groups to undergo phacoemulsification and IOL implantation: the observation group: 29 cases (36 eyes) received multifocal toric IOL implantation; the control group: 29 cases (36 eyes) received monofocal toric IOL implantation. Uncorrected distance visual acuity ( UCDVA), uncorrected near visual acuity ( UCNVA), best corrected distance visual acuity (BCDVA), the best corrected near visual acuity ( BCNVA ), total eye astigmatism, and the dark contrast sensitivity were observed for these patients at 1 and 6mo after cataract surgery.
RESULTS: There were no statistical significant difference between the two groups at postoperative 1, 6mo on UCDVA, BCNVA, BCDVA and total eye astigmatism(P>0. 05). UCNVA of observation group at 1 and 6mo were better than those of control group ( P <0. 05); there were statistically significant difference in high frequency comparison at the sixth postoperative months (P<0. 05).
CONCLUSION: Both monofocal toric IOL implantation, and aspheric multifocal toric IOL implantation for cataract with regular corneal astigmatism are effective to improve visual acuity. But the latter treatment would contribute to the improvement of uncorrected near visual acuity and the dark contrast sensitivity.
6.Expression of COX-2 and p38MAPK of peripheral blood mononuclear cells in trauma patients with multiple organ dysfunction syndrome

Objective To explore the expression of COX-2 and p38MAPK in patients with trauma MODS. Methods Forty MODS patients were evaluated. The levels of peripheral blood mononuclear cells COX-2 and p38MAPK in MODS patients and 40 normal controls was detected by enzyme linked immunosor-bent assay (ELISA). RT-PCR was used to measure the COX-2 mRNA and p38MAPK mRNA expression of in PBMCs. ANOV and correlation analysis were used in statistical analysis. Results The levels of COX-2 and p38MAPK of PBMCs and the mRNA expression in MODS group were higher than in control group (all P <0.05). The levels of COX-2 and p38MAPK of PBMCs and the mRNA expression in dead group were higher than in survival group( all P <0.05). The levels of COX-2 and p38MAPK of PBMCs were positively correlated, (r =0.6 147, P<0.01). The expression of COX-2 mRNA, p38MAPK mRNA of PBMCs and APACHE I scoring were positively correlated (r1 =0.5 009, P1 <0.05,r2 =0. 5 316, P2 <0. 05). Conclusions COX-2 and p38MAPK of PBMCs take part in the onset of MODS, and may service as index to judge the prognosis of MODS.
7.Clinical Application for Detection of Helicobacter Pylori Infection in Children with Helicobacter Pylori Stool Antigen Test

Objective To evaluate the accuracy of the Helicobacter pylori stool antigen (HpSA) test for detection of Helicobacter pylori (Hp) infection before and after treatment in children.Methods stool specimens were collected from 62 consecutive patients (4-17 years) who received 13 C-urea breath test( 13C-UBT) and/or gastroscopy for gastrointestinal symptom and 31 patients after radical cure of Hp who received 13 C-UBT within 2 days.The evaluation of Hp status was defined as positive when 13 C-UBT were positive and gastroscopic biopsy were positive.Results The sensitivity of HpSA test for the diagnosis of Hp using a cutoff value of 0.121 was 92.30% and its specificity and accuracy was 91.30% and 91.94%,respectively.sensitivity was 83.33% and its specificity was 88.00% and accuracy was 87.10% after radical cure of Hp.Conclusions The precision ratio of HpSA test is higher to diagnose Hp infection before the radical cure in children.Sensitivity and specificity after the radical cure are lower than before.
8.Effect of the silver needle injection therapy on rat with Sports muscle injury

Objective To evaluate the effect of silver needle injection therapy on rat with sports muscle injury. Methods Twenty-one healthy male Wistar rats were randomly divided into the injury group (n = 3),the silver needle group (n=12) and the control group (n=3). The expressions of bFGF and GDNF in gastrocnemius muscle tendon junction were detected on 7 d ,14 d and 28 d post-injury. Results No significant difference in the appearance of the injured tissue was found in both two groups on 7 d post-injury. The appearance of the injured tissue was better in the silver needle group than that in the control group on 14 d and 28 d post-injury. The tissue was almost normal in the therapy group on 28 d post-injury; The expression of bFGF in the therapy group was higher than that in the injury control group on 7 d and 14 d post-injury (P < 0.01). The expression of bFGF markedly decreased in the therapy group compared with the control group (P < 0.01) on 28 d post-injury. The expression of GDNF in the therapy group was higher than that in the injury control group on 7 d ,14 d and 28 d post-injury (P<0.01). Conclusion The silver needle injection therapy has the therapeutic effect on sports muscle injury reparation, which can increase the expression of bFGF and GDNF efficiently.
