1.Biological characteristics of silicon rubber and polyurethane in medical materials

OBJECTIVE: Polyurethane (PU) and silicon rubber have good biocompatibility, excellent mechanical properties, easy processing and controlled properties, etc. Therefore, their researches exhibit an important significance in the designs of the medical appliance and body apparatus.DATA SOURCES: The computer respectively indexed the database articles of abroad El, SCI, CA, Science Direct (Elsevier) and Chinese periodical full text from January 1995 to May 2006, the indexed words were "Silicon rubber, polyurethane, medical materials", the article language category was limited respectively in English and Chinese.STUDY SELECTION: The relevant articles (full text) that meet the study were selected by the first trial. Inclusion criteria: ① Related to silicon rubber and polyurethane. ② The medical rubber apparatus and body organ design. Exclusion criteria: The repetitive studies.DATA EXTRACTION: Collect totally 122 relevant articles that use the medicinal rubber materials, and expel the repetitive or similar research,therefore 16 articles meeting the request.DATA SYNTHESIS: ① Medicinal gum, hose and other products: The medical stopper used for sealing is requested to have the flexibility,resistant to cold, water, oil and disinfectant, and pinprick can still keep the original closeness and gastightness, etc. ② The abroad body organs: The material of repairing fracture, skull, meninges, bridge, auricle, mandible,windpipe, tear bolt, lung membrane, heart valve, stomach, intestine,peritoneum, penis, knuckle, muscle, knee membrane, skin and breast. ③ The research and expectation of the medicinal silicon rubber and polyurethane offer more medicinal apparatus and body organ.CONCLUSION: The study of the medicinal silicon rubber and polyurethane make people design and produce more cheap artificial organ and medicinal apparatus. More materials were applied to the clinic, so that satisfy the demand of many patients.
2.Efficient transient expression to analyze miRNA targets in rice protoplasts.

Compared with the transgenic approach, transient assays provide a convenient alternative to analyze gene expression. To analyze the relationship between miRNAs and their target genes, a rice protoplast system to detect target gene activity was established. The MIRNA and GFP-fused target sequence (or GFP-fused mutated sequence as a non-target control) were constructed into the same plasmid, and then delivered into rice protoplasts. The GFP expression level decreased significantly when the protoplasts were transfected with the plasmid containing GFP-fused target compared to that of the plasmid with non-target sequence either by fluorescence microscopy or qRT-PCR method. Two microRNA genes, osaMIR156 and osaMIR397, and their target sequences were used to prove the feasibility of the rice protoplast transient assay system. This method will facilitate large-scale screening of rice miRNA target in vivo, and may be suitable for functional analysis of miRNAs of other monocot plants that might share the evolutionarily conserved small RNA processing system with rice.

3.Comprehensive assessment of hepatic functional reserve before operation in liver cancer patients

Liver resection is the most efficiency measure to treat the liver cancer at present. About 80% patients with liver cancer encounter inordinately hepatocirrhosis. The regeneration of liver tissue in the patients with hepatocirrhosis is poor, liver functional reserve reduces, those patients with liver cancer combined with hepotocirrhosis will face greater risks when they receive hepatoectomy. Liver failure is the leading cause of death after hepatic resection. So we must attach importance to the assessment of liver functional reserve before operation. Here this article mostly summarizes the comprehensive assessment of hepatic functional reserve before operation in liver cancer patients.

5.Severe acute pancreatitis model by antegrade puncture injection of the sodium taurocholate in rat

Objective To improve the establishment of severe acute pancreatitis model in rat. Methods 36 Sprague-Dawley rats were randomly divided into experiment or shame operation group(n=18,each). Severe acute pancreatitis was induced by injection of 4% sodium taurocholate through puncturing common bile duct with a fine needle, puncture hole was closed by medical sealant glue. Ascites volume and blood amylase were measured at 3, 6 and 12 hours after injection. Gross and histological changes of pancreas were evaluated by a scoring system. Results Pancreatic changes in the experiment group was hemorrhagic and necrosis.The ascites volume(8.52±1.05)ml,serum amylase activity(5247.17±547.07)u/L, gross and histological scores(13.6±3.92) in experiment group was significantly higher than ascites volume(1.21±0.32)ml,serum amylase activity(1289.5±176.67)u/L, gross and histological scores((0.83±0.58) in shame operation group at every time point(P<0.05). Conclusions Combined injection through common bile duct and medical sealant glue in the experiment can minimize the trauma and simplize the procedure. This method produces a reliable model with high success rate and it is an ideal severe acute pancreatitis animal model.
6.Protective Effect of Electroacupuncture at Neiguan (PC 6) on Neurotransmitters Catecholamines in Rats with Myocardial Hypertrophy

Objective To discuss whether acupuncture at Neiguan (PC 6) inhibits myocardial hypertrophy via monoamine neurotransmitters.Method Sixty female Sprague-Dawley rats were randomized into a control group, a model group, a sham electroacupuncture (EA) group, and an EA group, 15 cases in each group. The rat models of myocardial hypertrophy were developed by subcutaneous injection of Isoprenaline hydrochloride (ISO) for 14 d. The EA group was intervened by EA at Neiguan (PC 6), and the sham EA group was by EA at a sham acupoint [5 mm away lateral to Neiguan (PC 6). The Electrocardiogram (ECG) of each group was recorded, electron microscope was used to observe the microstructure of the rats’ myocardial tissues, and high performance liquid chromatography (HPLC) was adopted to detect the contents of catecholamines in myocardial tissues.Result The heart rate, R-R interval, ST-elevation, and the contents of catecholamines in rats’ myocardial tissues (adrenaline, noradrenaline, and dopamine) in the model group and sham EA group were significantly different from that in the control group (P<0.05). The heart rate, R-R interval, ST-elevation, and the contents of catecholamines in rats’ myocardial tissues (adrenaline, noradrenaline, and dopamine) of the EA group were significantly different from that of the model group (P<0.05).Conclusion EA at Neiguan (PC 6) can improve myocardial hypertrophy, which is related to the regulation of the topical neurotransmitters; it has provided laboratory evidences for the relationship between Neiguan (PC 6) and heart, i.e. the relationship between acupoints and internal organs.
7.Effects of glutamine on level of cytokines in serum of rats with severe acute pancreatitis

Objective To explore the effects of glutamine (Gln) on tumor necrosis factor-alpha (TNF-α),interleukin-6(IL-6),and interleukin-10 (IL-10) in rats with severe acute pancreatitis (SAP).Methods 54 Sprague-Dawley rats were randomly divided into 3 groups ( n =18 for each group):Gln treatment group ( Gln),SAP model group (SAP),and negative control group (NC).SAP was induced by injection of 4% sodium taurocholate through common bile duct with a fine needle and the puncture hole in the bile duct was closed by medical sealant glue.Sham operation was performed in the rats of NC group.Rats in Gln group received Gln injection and rats in SAP group and NC group received normal saline instead of Gln.Ascites volume and blood amylase were measured at 3,6 and 12 hours after injection and plasma cytokines were detected by enzyme-linked immunosorbent assay (ELISA).Gross and histological changes of pancreas were evaluated by a scoring system.Results The ascites volume,serum amylase level,gross and histological scores,TNF-α level,and IL-6 level were significantly higher in SAP group than in NC group at each time point(P<0.05).IL-10 level was significantly higher in SAP group than in NC group at 3 hour point(P<0.05).The ascites volume,serum amylase level,and gross and histological scores were significantly lower in Gln group than in SAP group at the time point of 3 hour and 6 hour after injection( P < 0.05 ).Level of TNF-α and IL-6 was significantly lower in Gln group than in SAP group at each time point( P <0.05 ).IL-10 level was lower in Gln group than in SAP group at 3 hour point( P <0.05 ).Conclusion Gln is effective in SAP treatment by decreasing the serum level of proinflammatory cytokine and ameliorating the pathological damage of pancreatic tissues in rats.
8.Application of mixed reality technology and situational awareness in clinical teaching of cardiac macrovascular surgery

Mixed reality technology can achieve a 3D reconstruction of the patient's imaging data and organically combine the real scene. This allows to simulate a real medical operation in clinical teaching. Combined with large data analysis, mixed reality technology allows to build situational awareness data models, thus providing data support for teaching interaction and disease simulations. In the present work, the application of mixed reality technology and situational awareness in clinical teaching of cardiac macrovascular surgery is explored innovatively. These technologies open a new avenue for teaching, solving the issue that stymies traditional teaching approaches and improving the teaching efficiency. It is thus important to explore and promote the applications of this innovative teaching methodology particularly in clinical teaching.

10.Determination of MIL50, a humanized antibody, in different tissues of rats by a sandwich enzyme-linked immunosorbent assay

Objective To develop a sandwich enzyme-linked immunosorbent assay ( ELISA) for the determination of a humanized antibody MIL50.Methods RiVax, a mutant of RTA (a chain of ricin) expressed in E.coli, was used as the coating protein.Horseradish peroxidase (HRP) labeled IgG of goats against humans was used to determine MIL 50 captured by RiVax coated on the plate.Results Compared with ricin, RiVax could bind well with MIL50,indicating it could be used as a coating protein to determine MIL50 instead of holotoxin.Using this method, the detective limit of MIL50 in PBST (PBS containing 0.1%Tween 20) could be as low as 0.030 mg/L.The absorbance value of ELISA for MIL50 in the ser-um and homogenate of the liver , spleen, lung, kidney, muscle of rats was lower than that in PBST .Conclusion The sandwich ELISA is a sensitive method for the analysis of distribution of MIL 50 in rat tissues.
