Temporal and spatial expressions of ?-catenin investigated in the hair follicle and epidermis of the hoof periphery in bovine embryonic development. IHC (immunohistochemical method) was applied to qualitatively detect the temporal and spatial expressions of ?-catenin. ?-catenin was detected in suprabasal, epidermal basal layer, placode, hair bud in early phase(E68~93),and expressed strongly in epidermal basal layer, placode, and hair bud, in suprabasal expressed less strongly; in metaphase(E94~184), ?-catenin was detected in epidermis, hair peg, and in suprabasal, epidermal basal layer, hair follicle bulge, inner root sheath, outer root sheath, follicular infundibulum expressed less strongly; in late phase(E184~225), ?-catenin expressed weakly in epidermal basal layers, while expressed strongly in epidermal keratinocytes. The result suggested that ?-catenin plays an important role in hair follicle morphogenesis in the periphery of bovine hoof in bovine embryos.

Objective To study the biomechanical mechanism of Essex-Lopresti injury, and provide biomechanical basis for diagnosis and treatment of Essex-Lopresti injury. Methods Twelve fresh frozen adult upper limbs were addressed. Firstly, 12 samples ("complete state group") were loaded 100 N of a compressive force lasting 30 seconds in pronation, supination and neutral position on the mechanical testing machine. Secondly, 12 specimens were randomly divided into 2 groups. In the group named resection of radial head, the radial head was removed and interosseous membrane (IOM)was intact. In the group named the section of interosseous membrane, IOM was cut off. Finally, the radial head were removed and IOM was cut off in all specimens. The group was named as resection of radial head and IOM. Each sample was tested according to the method as described. Results The forearm rotation or single excision of the IOM had no effect on radial longitudinal displacement. Simple radial head excision or resection of the IOM and the radial head increased the vertical displacement of the radius. The radial stiffness had a gradual decline in forearm supination, neutral position and pronation. Simple excision IOM has no effect on the radial stiffness. The radial stiffness had decreased under the condition of excision of radial head or resection of the IOM and the radial head. Conclusion These in vitro measurements validate that the radial head fracture with IOM injury may be important reason for complications of the Essex-Lopresti injury. Radial head fracture play a key role for Essex-Lopresti injury and the injury of IOM is secondary cause. IOM is responsible for maintaining the vertical stability of the forearm after radial head resection.

Objective:To investigate the whole genome characteristics of coxsackievirus A4 (CVA4) circulating in Qingdao city. Methods:Four CVA4 isolates circulating in Qingdao city during 2013 to 2015 were selected. Whole genome sequences of these strains were amplified by one-step reverse transcription-polymerase chain reaction (RT-PCR). Sequence alignment and phylogenetic analysis were performed using MEGA7.0 software package. Genetic recombination analysis was performed using similarity plots 3.5.1 software package. Results:Phylogenetic analysis showed that based on the sequences of the whole genome and P1, P2 and P3 regions, HS312/QD/CHN/2013 and HS605/QD/CHN/2014 strains together with the early domestic isolates belonged to the same clade, while FY218/QD/CHN/2015 strain and CV-A4/P1033/2013/China strain collected in Wenzhou in 2013 formed another clade in each phylogenetic tree. HS144/QD/CHN/2014 strain belonged to the same clade as HS312/QD/CHN/2014, HS605/QD/CHN/2014 and the early domestic CVA4 isolates in the phylogenetic tree based on the P1 region, but formed a separate clade in the phylogenetic trees based on the whole genome, P2 region and P3 region. Genetic recombination analysis revealed that there was genetic recombination between HS144/QD/CHN/2014 strain and the CVA2 strain of CV-A2/P373/2013/China isolated in mainland China in 2013 in the region of 2C-3D (5 081-7 301); FY218/QD/CHN/2015 and CV-A4/P1033/2013/China strains were highly homologous and recombination signal sequences were detected in the region of 2A-2B (3 821-4 161) between the two strains and the CVA2 strain of CV-A2/P373/2013/China.Conclusions:The CVA4 isolates circulating in Qingdao city presented obvious genetic diversity at the genome-wide level.

OBJECTIVETo determine the dynamic expression of survivin gene in hepatocarcinogenesis of rats induced by aflatoxin B1 (AFB1).
METHODS78 Sprague-Dawley rats were used in this study. Hepatocellular carcinoma was induced in the rats by aflatoxin B1. Liver and HCC tissues were examined by immunohistochemistry and reverse transcriptase-polymerase chain reaction (RT-PCR).
RESULTSThe earliest hepatocellular carcinoma occurred at 46th week after AFB1 treatment. The HCC incidence was 54.9% (28/51) at 46th week and 64.9% (24/37) at 58th week. The positive rates of survivin protein expression in 24 HCC, para-cancerous liver tissues of experimental group were 41.7% and 54.2%, respectively, with no significant difference between them (P > 0.05). No survivin expression was detected in the experimental group before 46th week, neither in the rats without HCC occurrence nor the normal controls. The level of survivin mRNA expression in HCC at 58th week was significantly higher than that in pre-HCC, no-HCC and normal liver tissues in the control group (P < 0.01). The level of survivin mRNA expression in para-carcinoma tissues was also significantly higher than that in no-HCC and normal liver tissues of the control (P < 0.01). The level of survivin mRNA in pre-HCC at 12th, 20th, 36th, 46th weeks were significantly higher than those in normal liver tissues taken from control group during the same periods (P < 0.01).
CONCLUSIONThe over-expression of survivin gene is related to the occurrence of HCC and may play an important role in the carcinogenesis of HCC.
