OBJECTIVE:To establish the quality standard for Cangbai qutong capsule. METHODS:TLC was conducted to iden-tify the Rhizoma atractylodis,Asarum sieboldii,Paeoniae Radix Rubra and Cortex phellodendri;RP-HPLC was conducted to deter-mine the content of berberine hydrochloride. The column was Waters Symmetry-C18 with mobile phase of acetonitrile-0.1% phos-phoric acid (gradient elution) at flow rate of 1.0 ml/min,the detection wavelength was 265 nm,column temperature was 30 ℃, and the injection volume was 10 μl. RESULTS:TLC showed R. atractylodis,A. sieboldii,Paeoniae Radix Rubra and C. phelloden-dri had clear spots and good separation. The linear range of berberine hydrochloride was 21.12-84.48 μg/ml(r=0.999 6);RSDs of pre-cision,stability and reproducibility tests were lower than 3%;recovery was 96.439%-100.950%(RSD=1.34%,n=9). CONCLU-SIONS:The method is simple and reproducible, and can be used for the quality standards of Cangbai qutong capsule.

Objective To compare the efficiency between mini-percutaneous nephrolithotomy (MPCNL) and extracorporeal shock wave lithotripsy (ESWL) for monotherapy of renal calculi in infants less than 3 years. Methods Forty-six infants were treated with either SWL (22 infants) or MPCNL monotherapy (24 infants). The mean age was (22.6 ± 8.7) months vs (23.5 ± 6.6) months and the stone size was (21.4 ± 3.5) mm vs (21.7 ± 1.7) mm,and there were no significant difference. Results For MPCNL,mean operating time was (76.2 ± 23.4) min and mean hospital stay was (14.13 ± 5.8) d.The stone-free rates were 84.0% (21/25) after first session and 96.0% (24/25) after second-look procedure.Postoperative fever happened in 4 (16.0%) cases.Hemoglobin drop was (8.5 ± 4.4) g/L and no one needed blood transfusion. For ESWL,the stone-free rate were 31.8% (7/22) after first session and 86.3% (19/22) after second session for 11 infants (50.0%).Mean hospital stay was (6.6 ± 2.3) d and 10 cases (45.5%)had complications.Hemoglobin drop was (10.6 ± 12.7) g/L.MPCNL was lower than ESWL in complications rate and re-treatment rate, and the stone-free rate was higher, but the hospital stay and operation time was longer (P < 0.05).GFR revealed improve or stable after operation in both groups. Conclusions For a higher success rate, lower complication rate and re-treatment rate,MPCNL was an effective option for the management of relatively larger stones in children (even in infants).

Objective To establish the extraction process of Zhechong Chuangyu Capsule. Methods The difference of analgesic effect of water extraction and alcohol extraction in mice was observed by body-torsion test to determine the extract solvent. With the rate of aqueous extraction, n-butyl alcohol extraction and asperosaponin Ⅵ as evaluating indicator, the influencing factors including solvent volume, time and times of extraction were investegated to evaluate extracting procedure by orthogonal experiment. Results There was no obvious difference in analgesic effect between water extraction and alcohol extraction. Given the requirement of produce, aqueous extraction was a better choice. The optimum extracting condition was extracted 3 times with 20 folds volume of solvent, and extraction time was 150 minutes. Conclusion The extraction process is feasible to be applied into production.

Objective To explore the clinical characteristics, therapy and prognosis of primary central nervous system lymphoma (PCNSL).Methods Forty-three PCNSL patients were retrospectively analyzed. Results The most frequent PCNSL clinical presentations were an increased intracranial pressure, focal neurologic deficits. The median survival time of patients was 18.0 months (14.2—21.8 months,95% CI).Cox regression model analysis revealed that ependymal and leptomeningeal involvement was the only independent factor for overall survival (P=0.002).Conclusions PCNSL is a aggressive disease with a poor prognosis and difficult to diagnose before operation. The optimal treatment modality for newly diagnosed PCNSL is still under investigation.

OBJECTIVETo control the quality of the product, quantitative fingerprint was used to evaluate the composition of the amino acids in the Xingnao Tongluo injection.
METHODThe method of the quantitative fingerprint to the amino acids composition was established through AccQ Tag precolumn derivatization. The quality was evaluated by the quantitative test of the amino acids and the similarity in ten batches.
RESULTThe Xingnao Tongluo injection contained 12 amino acids and the contents of these amino acids were stable. All the ten batches of the samples had similarity of more than 0.90.
CONCLUSIONThe method was accurate, feasible and could be a simple and effective way to evaluate the quality of the traditional Chinese medicine.

BACKGROUND:Until now, little has been reported on establishment of pancreatic stem cell strains and lines, and the purification of pancreatic stem cells is difficult since the cell line establish rate is low.OBJECTIVE:To explore a more rational and effective technique of in vitro separation and continuous passage of pancreatic stem cells, with the hope to establish cell strains and even cell lines and to lay the foundation for the follow-up study of pancreatic stem cells in the treatment of diabetes.METHODS:Firstly, Percoll discontinuous density gradient centrifugation method was applied to separate the mouse pancreatic endocrine portion from the exocrine portion, then to obtain cell strains with highly proliferative ability and low differentiation from pancreatic endocrine portion-the islet. We used mouse embryonic fibroblasts treated with mitomycin C as a feeder layer, for in vitro continuous culture of islet-derived pancreatic stem cells under feeder layer conditions until they were transferred to the 30th passage to establish cell lines. Then pancreatic stem cell line derived from pancreatic islet was detected and identified by a series of tests including growth characteristic test, morphological observation, related molecular marker identification and differentiation characteristic identification.RESULTS AND CONCLUSION:In the continuous process of passage, pancreatic stem cells showed active proliferative ability, and maintained the typical morphological characteristics of stem cells and expression of pancreatic stem cell marker-Nestin. After induction, pancreatic stem cells showed insulin gene expression, reflecting their differentiation potential. Therefore, under the condition of feeder layer, the pancreatic stem cell line derived from Kunming mice was successfully established and the related identification was completed, which lays the foundation for the following research.
