1.Effects of continuous infusion of vascular endothelial growth factors with mini-osmotic pumps on survival rate of autogenous fat grafts in rats

BACKGROUND: Above curative dose, there is a dose-dependent correlation of the survival rate of fat transplantation with vascular endothelial growth factor (VEGF), the curative dose of VEGF determines the tissue volume of transplanted fat. OBJECTIVE: To investigate the changes of the blood supply rebuilding and the survival rate of the fat transplant in the microenvironment of continuous infusion of VEGF using osmotic pump. DESIGN, TIME AND SETTING: A cellular morphology observational experiment was performed from September 2007 to October 2008 in Department of Plastic Surgery at the Affiliated Hospital of Guangdong Medical College. MATERIALS: Ninety healthy SD rats were used to establish autologous fat transplantation models. Human VEGF was produced by American ONCOGEN Corporation. Type Alzet2002 mini-osmotic pump was produced by American Alzet Corporation. METHODS: Ninety rats were equally divided into 3 groups randomly. The operation control group was only processed into autologous fat transplantation; Experimental control group received autologous fat transplantation and continuous infusion of 2 000 ?L physiological saline using osmotic pump at a speed of 0.5 ?L/h for 14 days; Experimental group received autologous fat transplantation and continuous infusion of 2 000 ?L physiological saline containing 1 ?g VEGF using osmotic pump at a speed of 0.5 ?L/h for 14 days. MAIN OUTCOME MEASURES: At the 7th, 14th, 28th, 42nd, 64th days postoperation, the graft’s survival volumes were measured using mini-scale test tube; the cell morphology changes were observed with hematoxylin-eosin stain; The newly born microvessels on experimental group and control groups were researched with immunohistochemistry staining. RESULTS: At 14 days postoperation, the graft’s survival volumes of experimental group was higher than operation control group and experimental control group (P 0.05). At each time point, the vascular density of the fat transplant in the experimental group was higher than that in other groups (P 0.05). CONCLUSION: VEGF can speed up vasifaction and raise fat graft survival significantly.
2.Clinical application of retrograde medial and lateral gastrocnemius muscle flap for repairing the soft tissue defects of the middle and lower third of the leg

Objective To explore clinical application of retrograde medial and lateral gastrocnemius muscle flap for soft tissue defects of the middle and lower third of the leg. Methods From August 2008 to December 2009, in our hospital we adopted retrograde medial and lateral gastrocnemius muscle flap to renovate 5 cases of refractory soft tissue defects of the middle and lower third of the leg. Results Five cases of retrograde medial gastrocnemius muscle flap were survived, morphology and function of soft tissue defects were renovated well. Conclusion This operation is an effective and reliable technique for soft tissue defects of the middle and lower third of the leg, which is performed without sacrificing the major blood vessels, probing vascular pedicle and matching vascular anastomosis.
4.Relationship between Ulcerative Colitis and Lung Injuries.

OBJECTIVETo explore the relationship between ulcerative colitis (UC) and lung injuries by assessing their clinical manifestations and characteristics.
METHODSFrom July 2009 to April 2012, 91 UC patients presenting to Longhua Hospital who met the established inclusion and exclusion criteria were enrolled in this retrospective study. According to the scores of disease activity index, the patients were divided into the mild, moderate, and severe groups. Meanwhile, the records of pulmonary symptoms, chest X-ray image, and pulmonary function were reviewed.
RESULTSSixty-eight (74.7%) patients had at least 1 pulmonary symptom, such as cough (38.5%), shortness of breath (27.5%), and expectoration (17.6%). And 77 (84.6%) had at least 1 ventilation abnormality. Vital capacity value was significantly lower in the severe group than that in the mild group (91.82%±10.38% vs. 98.92%±12.12%, P<0.05).
CONCLUSIONSLung injury is a common extraintestinal complication of UC. According to the theory in Traditional Chinese Medicine that the lung and large intestine are related, both the lungs and large intestine should be treated simultaneously.

5.Recent advances and perspective in the study of the nano-reinforcing materials for molecular imprinting of proteins.

Molecular imprinting technique (MIT) involves the synthesis of polymer in the presence of a template to produce complementary binding sites in terms of its size, shape and functional group orientation. Such kind of polymer possesses specific recognition ability towards its template molecule. Despite the rapid development of MIT over the years, the majority of the template molecules that have been studied are small molecules, while molecular imprinting of proteins remains a significant yet challenging task due to their large size, structural flexibility and complex conformation. This review, we summarized the research findings over the past years, and discussed the nano-reinforcing materials used to prepare molecular imprinting of proteins and the perspective of these nano-reinforcing materials.

6.Screening and Metabolic Flux Analysis of Glutathione-high-yielding Strain from Saccharomyces cerevisiae

The zinc chloride and cystine resistant strain of S.cerevisiae YZM-14(ZnCl2r,Cysr) was screened with the mutant processing of the protoplast of S.cerevisiae by combinative mutagens of ultraviolet and nitrite.The glutathione(GSH) production(84.72 mg/L),dry cell weight(7.63 g/L) and the intracellular GSH content(11.10 mg/g) of YZM-14 were 2.79,1.63 and 1.71 times compared with that of the initial strain.The biosynthetic process of GSH was divided into three phases according to the time course of the specific cell growth rate and GSH yielding coefficient.In the second phase,the metabolic flux of the pentose phosphate pathway and the GSH precursors biosynthetic pathway of the mutant strain increased by 8.1 mmol/(g?h),compared with that of the initial strain.Furthermore,the metabolic flux of the organic acids secretion of the mutant strain decreased.Through these mechanisms,the utilization efficiency of the carbon sources was enhanced and high production of GSH was obtained.
7.The experimental study on effect of the spinal neuron flow with the nerve repair time

Objective To measure Ihe effect on rats spinal neuron flow according nerve roots repair time.Methods We adopted the experimental rats on the root avulsion and extravertebral foramen nerve root divison of C_(5~7).We divided them into four groupsin each which there were 16 ratsaccording the type of nerves root injury and repair timeGroup AC:the avulsed roots were reimplanted into the spinal cord and the transeeted roots were sutured to the proximal stump immediately.Group B,Dthe avulsed roots and the transected roots were reimplanted into the spinal cord or were sutured to the proximal stump in delayed 3 weeks each with 16 rats.At the different time point(3 weeks3 months6 months)through pathological examina- tion and immunohistological lechniques and nerve tracing techniqueswe examined the spinal cord and distal nerve trunk in order to observe the pathologic changes and axonal regeneration.Results Group A、C were much better than group B、D in the numberthe conformation and the degree of abatement of spinal motoneu- rons and nissl body.It is the same on the number and the development level of regenerating nerve fiber. Conclusion It had the advantage of neuronal protection and nerve regeneration that reparing the injured nerve roots earlv after nerve roots injury.
8.Treatment of plantar cutaneous deficiency with retrograde medial pedicled skin flap of the leg

Objective To introduce clinical application of retrograde medial pedicled skin flap of the leg to repair the plantar cutaneous deficiency. Methods From January 2002 to May 2008, in our hospital we adopted retrograde medial pedicled skin flap to renovate 12 cases of plantar cutaneous deficiency. The size of the skin flaps ranged from 10 cm× 7 cm to 13 cm × 12 cm. Results Ten cases of retrograde medial pedicled skin flap were survived, the rest had partial necrosis because of distal blood flow obstacle. One was cured by changing dressings; the other was repaired by secondary operation. Morphology and function of soft tissue defects were renovated well with 6-18 months follow-up. Conclusion This operation is an effective and reliable technique for plantar cutaneous deficiency.
9.Clinical trial of Danshen dripping pill in the treatment of diabetic coronary heart disease

Objective To observe the clinical efficacy and safety of Danshen dripping pill combined with atorvastatin calcium tablets and metformin hydrochloride tablets in the treatmem of diabetic coronary heart disease.Methods Ninety-four patients with diabetic coronary heart diseases were randomly divided into control group (n =47 cases) and treatment group (n =47 cases).Control group was treated with atorvastatin calcium 20 mg qd + metformin hydrochloride 0.5 g tid.Treatment group was given Danshen dripping pill 270 mg per time,tid,on the basis of the control group.Two groups were treated for 3 months.The clinical efficacy,the levels of serum total cholesterol (TC),triglycerides (TG),low-density lipoprotein cholesterol (LDL-C),high density lipoprotein cholesterol(HDL-C),hemoglobinA1 c (HbA1 c),fasting blood glucose (FBG) and adverse drug reactions were compared between two groups,and the homeostasis model assessment insulin resistance (HOMA-IR) were calculated.Results After treatment,the total effective rates in treatment and control groups were 91.49% (43/47 cases) and 74.47% (35/47 cases) with significant difference(P < 0.05).After treatment,the main indexes in treatment and control groups were compared:TC were (3.05 ±0.21),(3.55 ±0.16)mmol · L-1;TG were(1.14 ± 0.16),(1.45 ± 0.14) mmol · L-1;LDL-C were(1.42 ±0.19),(1.74±0.19) mmol · L-1;HDL-C were(1.11 ±0.12),(1.40 ±0.15) mmol · L-1;HbA1c were (4.09 ± 0.50) %,(5.06 ± 0.71) %;FBG were (4.68 ± 0.61),(5.42 ± 0.93) mmol · L-1;HOMA-IR were (2.63 ±0.60),(3.66 ±0.59),the differences were statistically significant(P <0.05).The adverse drug reactions in two groups were based on increasing transaminase,flatulence,constipation,acute myocardial infarction,and so on.The incidences of treatment and control groups were 31.94% and 34.04% without significant difference (P > 0.05).Conclusion Danshen dripping pill combined with atorvastatin calcium tablets and mefformin hydrochloride tablets has a definitive clinical efficacy in the treatment of diabetic coronary heart disease,which can improve the levels of blood lipid and function of islet cells,decrease blood glucose,without increasing the incidence of adverse drug reactions.
10.Development and Standardization of a New Cognitive Assessment Test Battery for Chinese Aphasic Patients: A Preliminary Study

Background: Nonlinguistic cognitive impairment has become an important issue for aphasic patients, but currently there are few neuropsychological cognitive assessment tests for it. To get more information on cognitive impairment of aphasic patients, this study aimed to develop a new cognitive assessment test battery for aphasic patients, the Non?language?based Cognitive Assessment (NLCA), and evaluate its utility in Chinese?speaking patients with aphasia. Methods: The NLCA consists of five nonverbal tests, which could assess five nonlinguistic cognitive domains such as visuospatial functions, attention test, memory, reasoning, and executive functions of aphasic patients. All tests are modified from the nonverbal items of the current existed tests with some changes to the characteristics of Chinese culture. The NLCA was tested in 157 participants (including 57 aphasic patients, 50 mild cognitive impairment (MCI) patients, and 50 normal controls), and was compared with other well?established relative neuropsychological tests on the reliability, validity, and utility. Results: The NLCA was fully applicable in the MCI patients and the normal controls, almost working in the aphasic patients (57/62 patients, 91.9%). The NLCA scores were 66.70 ± 6.30, 48.67 ± 15.04, and 77.58 ± 2.56 for the MCI group, the aphasic group, and the control group, respectively , and a significant difference was found among three groups (F = 118.446, P < 0.001). The Cronbach's alpha of the NLCA as an index of internal consistency was 0.805, and the test?retest and interrater reliability was adequate (r=0.977 and r= 0.970, respectively). The correlations of the cognitive subtests and their validation instruments were between 0.540 and 0.670 (all P < 0.05). Spearman's correlation analysis indicated that the coefficient of internal consistency of each subtest itself was higher than other subtests.When choosing the Montreal Cognitive Assessment score of <26 as the diagnostic criteria of cognitive impairment, the area under the curve for all participants in the control and MCI groups was 0.942 (95% confidence interval: 0.895–0.989), and an optimal cutoff point of 75.00 seemed to provide the best balance between sensitivity and specificity. Age (r = ?0.406, P < 0.001) was the main influence factor for the NLCA. Conclusions: The NLCA could efficiently differentiate the cognitive impairment patients from the normal controls and is a reliable and valid cognitive assessment test battery to specially find nonlinguistic cognitive function for aphasic patients.
