To explore the feasibility of stable expression of Hantavirus H8205 strain G1 segment and human IL-2 fusion gene in Vero cells, and to examine the immune protection effects on mice vaccinated with this recombinant eukaryotic expression vector containing Hantavirus G1 gene and IL-2 gene. With the help of lipofectamine, the Vero cells were transfected with pcDNA3.1/HisB-IL-2-G1 and the positive cells were selected by G418. IFAT and SDS-PAGE electrophoresis were used to determine the stable transfection and expression of recombinant protein. Each mouse was inoculated with plasmids intramuscularly (i.m.) three times, 2 boosts were given at 2-week intervals, serum anti-hantavirus antibodies were detected by ELISA and neutralizing antibodies (NAb) were detected by Plaque Reduction Neutralization Test. The fusion protein expressed in Vero cells was 78 kD, corresponding to the estimated molecular size. The neutralizing antibody titers of mice with pcDNA3.1/HisB-IL-2-G1 were 1:20-1:80. IL-2/G1 fusion gene could be transferred in Vero cells and stably express the fusion protein. Specific humeral immune responses in mice can be induced with the recombinant eukaryotic expression vector containing the fusion gene, which lays the foundation for further development of therapeutic HTNV vaccine.

Coronary heart disease (CHD) is a common kind of cardiovascular disease which does serious harm to human health. It has become the first cause of people's death in many countries and regions. This article analyzed CHD-related animal experiments in recent ten years. It made a review on progress in the establishment of chronic and acute animal models of CHD from the animal selection and model establishment. It can provide guidance for the further research of CHD.
4.Interleukin-17 in apical exudates of periapical periodontitis treated with minocycline controlled-release formulation

BACKGROUND: Experimental proof for the efficacy, safety, and immunological assessment is needed when minocycline is used for root canal disinfection.OBJECTIVE: To investigate the effect of minocycline for root canal disinfection on levels of interleukin-17 in apical exudates of periapical periodontitis and periapical exudate volume.METHODS: Sixteen patients with acute periapical periodontitis (16 teeth) scheduled for root canal therapy were enrolled and randomly divided into calcium hydroxide and minocycline groups, respectively, followed by root canal disinfection.One week after disinfection, periapical index, periapical exudate volume and interleukin-17 level were detected prior to the root canal filling. Another 16 patients with normal pulp vitality (16 teeth) scheduled for single root canal filling were enrolled as control group, in which periapical index, periapical exudate volume and interleukin-17 level were detected.RESULTS AND CONCLUSION: The periapical exudate volume and interleukin-17 level in the calcium hydroxide and minocycline groups were significantly higher than those in the control group (P < 0.05). The periapical index and interleukin-17 level in the calcium hydroxide and minocycline groups were decreased significantly at 1 week after root canal disinfection (P < 0.05), while there was no difference between these two experimental groups in the periapical index, periapical exudate volume and interleukin-17 level. To conclude, the use of minocycline significantly reduces interleukin-17 level and periapical exudate volume, and thus achieves effective outcomes in periapical disease.

6.Neurolymphomatosis, a case report

Neurolymphomatosis(NL) is characterized by lymphomatous infiltration of the peripheral nervous system. We report a case of neurolymphomatosis(NL) which was confirmed by sural nerve biopsy. Sural nerve specimen from a 49-year-old female patient with weakness of limbs was examined with routine histochemical and immunohistochemistry staining, in which the first antibodies against CD3, CD20, CD45, CD45RO and CD68 were used. Numerous T-lymphoma cells invaded in the adipose tissue of epineurium of sural nerve. The nerve biopsy showed marked axonal degeneration of myelinated fibers. The clinical and histopathologic findings confirmed the diagnosis of neurolymphomatosis.
8.CT and MRI Manifestations of Lumbar Spinal Epidural Angiolipoma

Purpose To investigate the imaging features of lumbar spinal epidural angiolipoma, and to improve the imaging diagnostic capability of the disease. Materials and Methods Four patients with lumbar spinal epidural angiolipoma confirmed by pathology were recruited in the study. CT and MRI images were reviewed and the imaging characteristics including the shape, size, location, density, and signal intensity were analyzed. Results The lesions located at the right front of the spinal canal in two cases, at the left front in one case, and the residual one case located directly behind the spinal canal. The longitudinal axis of the lesions paralleled to the longitudinal axis of the spine. The dura mater spinalis was compressed and inward shifted. All of the four cases showed homogeneous iso- or hypo-density on CT without calcification or necrosis in the lesions. The maximum diameter was 3 to 5 cm. The boundary was clear and smooth. Three lesions showed dumbbell-shaped and crossed foramen, but the adjacent bone were not absorbed or destructed. One lesion showed scallop-like. On MRI, four cases displayed slightly hypointense on T1WI and hyperintense on T2WI. On STIR images, the lesions showed hyperintense with patchy low signal intensity in it. The boundary of the lesions was clear. After administration of contrast media, two lesions presented remarkably homogeneous enhancement, one lesion showed dual tail sign, and one lesion displayed pen-tip-like at the both ends. Conclusion MRI plays an important role in locating the lesion and distinguishing internal tissue components of spinal angiolipoma, which is the gold standard for the diagnosis of the tumor. CT provides excellent supplement. The Combination of CT and MRI will improve the diagnostic accuracy of the spinal angiolipoma.
9.Analysis of the clinical and laboratory features of 67 cases of scleroderma overlap syndrome

Objective To investigate the clinical and serologic features of scleroderma overlap syndromes.Methods Scleroderma overlap syndrome was diagnosed in 67 patients referred to our department from January 2003 to August 2013.Clinical and laboratory date were retrospectively analyzed.Comparisons between groups were tested by t test and chi-square test and Fisher exact method.Results ① Sixty-seven patients satisfied the criteria for scleroderma overlap syndrome,and 66 were female.The incidence of additional connective tissue diseases in the overlap syndrome group was systemic lupus erythematosus (SLE) in 27%,Sj(o)gren's syndrome (SS) in 24%,rheumatoid arthritis in 16%,dermatomyositis or polymyositis in 10%,two or more connective tissue diseases in 22% respectively.② Systemic sclerosis preceded the development of additional connective tissue diseases was frequent (58%).The median onset age of SSc in SSc-SLE was younger,in contrast,SSc-SS was older.③ Antinuclear antibody (ANA) was positive in 87% of patients,rheumatoid factor (RF) was in 42%.Anticentromere antibody (ACA) was more common in SSc-SS,RF and anti-cyclic citrullinated peptide (CCP) antibody were more frequent in SSc-RA,anti ds-DNA antibody was more common in SSc-SLE.Conclusion SSc-SLE is more common in scleroderma overlap syndrome in particular,and the median onset age of SSc in SSc-SLE is younger than others.There are distinct clinical and serological features that may predict the possibility of SSc patients with overlap syndrome.
