1.Preliminary study of rabbit model with corneal neovascularization after thermal burn under the constant temperature

AlM:To explore the suitable conditions in rapid model of corneal neovascularization ( CNV ) after thermal burn under different constant temperature in rabbit.
METHODS: Total 45 New Zealand white rabbits were divided randomly into five groups ( A, B, C, D, E ) . A groups:100℃ ( n = 10 ) , B groups: 200℃ ( n = 10 ) , C groups:300℃ ( n=10 ) , D groups: 400℃ ( n=10 ) , and E groups:control group ( n=5 ) . All left eyes of rabbits in A,B,C,D groups were induced corneal neovascularization by constant temperature burning device. The growth of CNV was observed by slit lamp microscope and the area of CNV were recorded on 4 th , 7 th , 14 th , 30 th days postoperatively. SPSS 19. 0 statistical package was used for data analysis, and the data was recorded by mean ± standard deviation. Comparison by analysis of variance was made by repeated measures in the area of neovascularization at each time point in groups. Statistical tests were considered significantly when P values were less than 0. 05.
RESULTS:On postoperative 4 th , 7 th , 14 th , 30 th days: no neovascularization was found after corneal thermal burn in A group, but only a few nebula left (n=2);the area of CNV were (9.16±1.45)mm2, (37.73±5.49)mm2, (62.44± 7. 54 ) mm2 , ( 40. 28 ± 7. 39 ) mm2 in B group respectively;and (11.45±1.04)mm2, (44.51±4.64)mm2, (66.13±4.13)mm2, (43.04±2.33)mm2 in C group respectively; and (13.23± 0.86)mm2,(47.26±4.59)mm2,(67.57±4.56)mm2,(45.59± 4. 44 ) mm2 in D group respectively, and part corneal carbide ( n = 4 ) was observed as well as corneal perforation ( n= 6 ) were found on 3d in D group. No neovascularization was found in normal control group. Comparison of the areas of CNV at each time point between groups was statistically different, P < 0. 05. Statistical differences were found among B, C, D groups, P<0. 05.
COCLUSlON:ln 4 to 7d, the higher the temperature is, the more the neovascularization area of CNV are. lt has no significant difference in 14 to 30d. But corneal carbide and corneal perforation are often found in 400℃ group, so its modeling failure rate is high. lt is between 200℃ and 300℃ that repeatability and uniformity of the corneal neovascularization model of rabbit are superior.
3.Effect of high glucose environment on the NF-κB/Iκ B signal pathway in cultured rat glomerular mesangial cell and its mechanism

To explore the effect of high glucose on the NF-κB/IκB signal pathway in cultured rat glomerular mesangial cells. The results showed that high glucose increased the degradation of IκB-α and the translocation to nucleus of NF-κB. These changes could be reverted mostly by MG132, a proteasome inhibitor. It suggests that the activation of the NF-κB signal pathway by high glucose concentration may probably be via the ubiquitin-proteasome pathway.

6.Effect of tianma gouteng decoction on the endothelial function and the renal protein expression in spontaneously hypertensive rats.

OJECTIVETo observe the effect of tianma gouteng decoction (TGD) on the endothelial function and the renal protein expression of spontaneously hypertensive rats, and to analyze its possible mechanism.
METHODSTotally 18 6-week-old SHR were randomly divided into 3 groups according to randomized block design, the SHR control group, the TGD group, and the captopril group, 6 in each group. Meanwhile, Wistar Kyoto (WKY) rats of the same age were recruited as a WKY control group. Rats in the TGD group were administered with TGD at the daily dose of 10. 260 g/kg. Rats in the captopril group were administered with captopril at the daily dose of 3. 375 g/kg. 2 mL/100 g distilled water was administered to rats in the SHR control group and the WKY control group. All medication was performed by gastrogavage once per day till rats were 24 weeks old. Changes of blood pressure were measured once per two weeks. The relaxation of the thoracic aorta and the superior mesenteric artery was determined by vascular ring in vitro to reflect the endothelial function. The total renal protein was separated by two-dimensional electrophoresis (2-DE). The significantly deviated protein was verified by Western blot.
RESULTS(1) Compared with the SHR control group, blood pressure was significantly lowered in rats (10 - 24 weeks old) of the captopril group (P <0.01, P <0.05). The hypotensive effect of TGD was obvious at the beginning of hypertension (10 -12 weeks) (P <0. 01). But along with the progression of hypertension, its hypotensive effect was not obvious (P>0. 05). (2) Compared with the SHR control group, the relaxation of the superior mesenteric artery was obviously improved in the TGD group (P <0. 05); the relaxation of the thoracic aorta and the superior mesenteric artery was obviously superior in the WKY control group (P <0. 01, P <0. 05). But there was no statistical difference in each relaxation index between the captopril group and the SHR control group (P >0. 05).(3) RESULTS: of 2-DE found 16 significantly differential renal protein, mainly involved nitric oxide (NO) system, oxidative stress, and cytoskeleton-related proteins. Results of Western blot showed that TGD could significantly improve expressions of Cu-Zn superoxide dismutase (SOD), N(G, N(G)-dimethylarginine dimethylaminohydrolase 2 (DDAH2), and pterin-4-alpha-carbinolamine dehydratase 1 (PCBD1) (P <0. 05).
CONCLUSIONGTD could protect the endothelial function of the superior mesenteric artery in SHR, and its intervention mechanism of hypertension induced early renal injury might be relevant to regulating the NO system and antioxidative stress.

7.Recent advances in small-molecule inhibitors targeting influenza virus glycoproteins

Hemagglutinin and neuraminidase, two important glycoproteins on the surface of influenza virus, play a considerable role in the entry and release stage of the viral life cycle, respectively. With in-depth investigation of influenza virus glycoproteins and the continuous innovation of drug discovery strategies, a new generation of glycoproteins inhibitors have been continuously discovered. From the point of view of medicinal chemistry, this review summarizes the current advances in seeking small-molecule inhibitors targeting influenza virus glycoproteins, hoping to provide valuable guidance for future development of novel antiviral drugs.
8.Current status and influence factors of beta blocker prescription in Chinese patients with stable angina pectoris

Objective To analyze the status of beta blockers (BBs) prescription in Chinese patients with stable angina pectoris.To determine the gap between clinical practice and treatment guidelines and factors influencing the use of BBs in Chinese patients.Method A questionnaire survey was conducted in patients diagnosed with chronic stable angina pectoris in 2011.A total of 298 Chinese hospitals attended the survey.The questionnaire collected patient demographic information,clinical and medication information.In the analysis,patients were divided into two groups according to whether they were on BBs at the time of the interview.Hospital characteristics,patient demographic information,cardiovascular risk factors,diagnostic method of coronary heart disease,past history of coronary revascularization and use of secondary prevention medications were compared between the two groups.Binary logistic regression analysis was used to identify factors related to BBs use.Patients were followed-up for 3 months.Patients who were on BBs at baseline were divided into two groups according to whether they were still on BBs after 3 months.Comparison of the clinical information previously mentioned was made between the two groups,and binary logistic regression analysis was used to identify factors related to BBs adherence.Results Of the 5 011 questionnaires,2 859 cases were male,mean age was (67.6 ± 11.5)years old,3 060 (61.1%) patients were on BBs at baseline.Binary logistic regression analysis showed that when adjusted for other factors,old age (OR =0.895,95% CI 0.849-0.944,P <0.001) was related to no BBs use,while comorbidity with hypertension (OR =1.255,95% CI 1.103-1.427,P =0.001) or hyperlipidemia (OR =1.456,95% CI 1.287-1.648,P < 0.001),diagnosed with coronary disease by imaging (OR =1.250,95% CI 1.070-1.461,P =0.005) and past history of coronary revascularization (OR =1.490,95% CI 1.220-1.820,P < 0.001) were related to higher percentage of BBs use.A total of 2 533 cases (82.8%) were still taking BBs 3 months after baseline interview.Binary logistic regression analysis showed that,when adjusted for other factors,treated at traditional Chinese medical hospitals (OR =1.612,95% CI 1.154-2.251,P =0.005),low levels of education (OR =1.316,95 % CI 1.151-1.504,P < 0.001),hypertension (OR =1.345,95% CI 1.096-1.651,P =0.005),high baseline heart rate (OR =1.020,95% CI 1.010-1.030,P < 0.001) and diagnosed with coronary disease by imaging (OR =1.342,95% CI 1.095-1.644,P =0.005) were factors related to higher BBs adherence,while low baseline CCS class (OR =0.794,95% CI 0.704-0.896,P < 0.001) was related to lower BBs adherence.Conclusions In Chinese patients with clinically diagnosed chronic stable angina,BBs use was not sufficient.Old age,comorbidity with hypertension or hyperlipidemia,past history of coronary revascularization were independently related to higher percentage of BBs use.Treated at traditional Chinese medical hospitals,low levels of education,hypertension,high baseline heart rate and diagnosed with coronary disease by imaging were independent factors related to higher BBs adherence,while low baseline CCS class was related to lower adherence.
9.Rapid identification of crude and sweated dipsaci radix based on near-infrared spectroscopy combined with principal component analysis-mahalanobis distance.

In order to discriminate the crude and sweated Dipsaci Radix correctly and rapidly, the crude and sweated Dipsaci Radix were scanned by the NIR spectrometer, and an identifying model was developed by near infrared spectroscopy combined with principal component-Mahalanobis distance pattern recognition method. The pretreated spectra data of 129 crude samples and 86 sweated ones were analyzed through principal component analysis (PCA). The identifying model was developed by choosing the spectrum for 9 881.46-4 119.20 cm(-1) and "SNV + spectrum + S-G" to the original spectral preprocessing with 14 principal components, and then was verified by prediction set, identifying with 100% accuracy. The rapid identification model of the crude and sweated Dipsaci Radix by NIR is feasible and efficient, and could be used as an assistant means for identifying the crude and sweated Dipsaci Radix.
