2.Clinical value of human papillomavirus genotype distribution in HPV infection

Objective To analyze the distribution of human papillomavirus (HPV) genotype in the diseases associated with HPV infection and explore the role of HPV genotype in these diseases. Methods Retrospective analysis was carried out to compare HPV genotypes of 353 outpatients suffering the diseases associated with HPV infection including cervical caner,cervical intraepithelial neoplasia (CIN),genital duc condyloma,and subclinical cervical human papillomavirus infection (SPI) in our department from January to October 2008. HybriMax were used to detect HPV genotype. The distribution data of HPV genotype were statistically analyzed. Results Totally 176 HPV positive females were found with a positive rate of 49.86%. HPV positive rate in cervical cancer was 100%,and all of them were high risk HPV. Sleep disorders are commonly occurred among patients with Parkinson's disease,such as difficulties in the initiation of sleep,fragmented sleep,sleep behavior disorder and excessive daytime sleepiness.The mechanism of sleep disorders associated with Parkinson's disease is not clear,which may be associated with the injury of brain stem,nuclei of median raphe,nuclei fasciculi solitarii,thalamencephalon and the changes of neurotransmitters as dopamine,hypocretin(orexin) and melatonin.This article gives an overview of the mechanism of sleep disorders associated with Parkinson's disease.
4.Nuclear magnetic resonance-based metabonomics features before and after 800 meters race

BACKGROUND:Metabolic characteristics of the 800 meters race is not very clear, which creates a lot of obstacles for the exercise training of 800 meters race. OBJECTIVE: To investigate the characteristics of material and energy metabolism of 800 meters race, to improve the scientific training of the 800 meters race and to verify the feasibility of nuclear magnetic resonance-based urine metabonomics in sports training monitoring. METHODS: Seven middle distance athletes were subject to an 800 meters race at ultimate intensity, and their urine metabonomics analysis was conducted in combination with the Human Metabolism Database and other databases before and after exercise. RESULTS AND CONCLUSION:OPLS-DA model showed that the metabolic samples before and after movement presented the cluster-like distribution. The change was mainly caused by the folowing metabolites, including leucine, lactic acid, creatinine, succinic acid, hypoxanthine, and adenosine. Through the analysis we could infer that the 800 meters race is a sport event mainly dependent on glycolysis for energy supply, working together with raw phosphate and aerobic oxidation for energy supply. The amino acid metabolism and methane metabolism of the athletes were more vigorous after 800 meters running, and moreover, there were certain oxidative stress during the 800 meters race. These findings indicate that nuclear magnetic resonance-based urine metabonomics takes the advantages of invasive, high sensitivity, less sample requirement, simple processing.
5.Application effect of surgical simulator to improve the microsurgical skills in junior ophthalmologist

AlM: To discuss the application effect of surgical simulator to improve the microsurgical skills in junior ophthalmologist. METHODS: Lecture teaching, training in surgical simulator and operation in animal eyes were received in all these ophthalmologists. Results of the ability of operation in cataract surgery after this training were analyzed. RESULTS: After taught theory, students completed cataract surgical procedures on simulator and the mean test score was 75. 91 ± 6. 53 points. After trained on simulator repeatedly, the mean test score was 85. 57±4. 64 points. There was statistically significant difference ( P<0. 01) . During the third stage of practicing on animal eyes, the score was 89. 77 ± 7. 61 points, there was statistically significant difference compared with former two stages (P<0. 05).CONCLUSlON: Comprehensive training can improve microsurgical skills of junior ophthalmologist, but the long effect need to be observed.
6.Effects of electroacupuncture at "Fenglong" (ST 40) on formation of macrophage-derived foam cell and efflux of cholesterol in hyperlipidemia rats.

OBJECTIVETo explore action mechanism of electroacupuncture (EA) at "Fenglong" (ST 40) for treatment of hyperlipidemia.
METHODSForty SPF-grade SD rats were randomly divided into a normal group (group A), a model group (group B), a model diet-control group (group C), a model + EA group (group D) and model diet-control + EA group (group E), 8 cases in each one. The rats in group A were fed with normal diet continuously while those in the rest 4 groups were fed with high-fat diet to establish hyperlipidemia model. Afterwards, the rats in group C and group E were fed with normal diet, while EA at "Fenglong" (ST 40) was applied in group D and group E, 30 min per time, once a day. After 28 days of treatment, macrophage was collected in each group. Oil red O-staining was applied to detect the formation of foam cells, and enzyme-linked immunosorbent assay (ELISE) was adopted to measure the contents of total cholesterol (TC) in macrophage and analyze the rate of cholesterol efflux.
RESULTSThe counts of positive cells of oil red O-staining and the contents of TC in the group B, group C and group D were significantly increased compared with those in the group A (all P < 0.01). The counts of positive cells and contents of TC in the group C, group D and group E were significantly clecreased compared with those in the group B (all P < 0.01), and the decline in group D was more obvious than that in the group C (all P < 0.01). Compared with group C and group D, the counts of positive cells and contents of TC in the group E was obviously decreased (all P < 0.01), which was not statistically different from group A (P > 0.05). In the meantime, compared with group C, the rate of cholesterol efflux in group D and group E was significantly increased (both P < 0.01), and the rise in group E was more obvious than that in the group D.
CONCLUSIONThe electroacupuncture at "Fenglong" (ST 40) could significantly prohibit the transformation of macrophage into foam cell and increase rate of cholesterol efflux in macrophage, which could prevent and reverse the formation of foam cell and play an essential role in treating hyperlipidemia and stopping it from developing into a further level.

8.Optimism and suicide risk:self-esteem mediation role and life events adjustment role

Objective To explore the mediating role of self-esteem in optimism and suicidal risks,and the moderate effect of life events in optimism,suicidal risk and self-esteem.Methods 1 750 college students were surveyed with the life orientation test-revised (LOT-R),the suicidal behaviors questionnaire-revised (SBQ-R),rosenberg self-esteem scale(SES) and adolescent self-rating life events check list (ASLEC).Results The total scores of SBQ-R,LOT-R,SES and ASLEC of college students were (4.78±2.10),(23.17±3.22),(29.58±4.43)and (36.54±21.73).Optimism scores were negatively correlated with suicidal risk scores(r=-0.23,P<0.01),optimism scores were positively correlated with self-esteem (r=0.50,P<0.01).Scores of self-esteem scales were negatively correlated with suicidal risk scores(r=-0.35,P<0.01).A structure equation model consists of optimism,self-esteem,suicidal risk and it had a better degree of fitting (the path form optimism to suicidal risk were r =-0.05,P<0.05,RMSEA =0.05,GFI =0.90,NFI =0.92,CFI =0.93).The relationship between optimism and suicide risk were mediated by self-esteem(P<0.01),which accounted for 76%of the total effects.The life events' moderating effects between self-esteem and suicide risk were significant(P<0.01).Conclusion Self-esteem has a significant correlation with optimism and suicidal behaviors,self-esteem plays an intermediary role between optimism and suicidal behaviors,and life events play an regulation role between self-esteem and suicidal behaviors.
9.Analysis of risk factors for susceptibility of subcutaneous tophi in people with gout

Objective To discuss the risk factors for susceptibility of subcutaneous tophi with an aim to provide clinical evidence for the prevention and treatment of tophi.Methods A total of 5 321 cases of gout patients whose course of disease was less than 10 years were selected and divided into two groups according to whether a subcutaneous tophus was present. The clinical information was collected and relevant biochemical indices were detected.Results There were significant differences in the ratios of regular exercise, involvement of upper limb joints, combining renal insufficiency, kidney stone and coronary heart disease between the tophus group and the non-tophus group(P<0.05 or P<0.01). There were significant statistical differences in age, disease duration, body mass index, systolic blood pressure, diastolic blood pressure, serum uric acid, serum creatinine, creatinine clearance rate, the number of joints involved between the two groups[(51.80±14.58 vs 48.45±14.45)year,(5.61±3.20 vs 3.28±3.06)year,(26.50±3.39 vs 27.05±3.45)kg/m2,(138±20 vs 133±18)mmHg, 1 mmHg=0.133 kPa,(89±13 vs 87±12)mmHg,(486.66±121.96 vs 460.02±119.10) μmol/L,(112.11±62.97 vs 104.43±46.28) μmol/L, (86.46±37.99 vs 93.00±40.11 )ml/min,(2.89±1.86 vs 1.76±1.22) joints, all P<0.01]. There were no statistical differences in family history, gender, history of smoking and drinking, the means of waistline, fasting blood glucose, triglyceride, and total cholesterol between the two groups(all P>0.05). Further logistic regression analysis indicated that disease duration, a large number of joints involved, the involvement of upper limb joints, combining kidney stones, higher serum uric acid level, and higher diastolic pressure were risk factors of the early onset of subcutaneous tophi, while regular exercise and body mass index were protective factors.Conclusion Patients with long duration, high serum level of uric acid, a large number of joints involved, upper limb joint involved, kidney stones, and hypertension were more likely to develop subcutaneous tophi. These risk factors should be intervened actively in clinic.
10.Expression and significance of erythropoietin and its receptors in rats with traumatic brain injury

Objective To study the expressions of erythmpoietin(EPO)and its receptors(EPOR)in the injured brain tissue ofthe rats with traumatic brain injury(TBI).Methods A total of78 SD rats were randomly divided into three groups including control group(six rats),sham group(36rats) and fluid percussion injury group(36 rats).The rats were sacrificed at 6,24 hours,3,5,7 and 14days after TBI in the sham group and the fluid percussion injury group(six rats at each time point).Then,the injured brain tissues were removed for observation of the mRNA and protein expressions of EPO and EPOR by meaDiB of real-time PCR and Western blot. Results The expression of EPO was increased at 24 hours and reached the peak at day 3 after TBI.The hish expression level of EPO could maintain for two days or so.began to decrease at day 7 and recovered to normal at day 14 after Till.While the expression of EPOR reached the peak at 24 hours after TBI and maintained hish level at day14. Conclusions The expressions of EPO and EPOR show increase within 24 hours after TBI.In fact,the expressions of both factors are not in consistency,with more transient expression of EPO.
