1. Determination of six flavonoids in Schizonepeta tenuifolia from different areas by HPLC

Objective: To detect the contents of six kinds of flavonoids in Schizonepeta tenuifolia from different areas, so as to provide a basis for their quality control. Methods: Ultra performance liquid chromatography (UPLC) was used to establish the detection scheme of flavonoids content, and clustering analysis was conducted by the results of content detection. Results: The contents of six kinds of flavonoids in S. tenuifolia from different areas had large differences. The linear ranges were: 0.030-0.148 μg for cynaroside, 0.222-1.108 μg for quercetin, 0.357-1.784 μg for hesperidin, 0.058-0.292 μg for luteolin, 0.054-0.269 μg for apigenin, and 0.050-0.247 μg for diosmetin, respectively. The average recovery rate was between 96.32% and 99.97%, RSD < 2.20%; Among this, the Schizonepeta Briq. herb from Anhui was as one class, the herbs from Henan 1 and Guangdong were as one class, and the herbs from Henan 2, Hebei, Yunnan, Sichuan, Hubei, and Gansu were as the other class. Conclusion: The method used for quality control of the multi-index level in this experiment is simple and sensitive; on the other hand, the herbs from Henan 1, Henan 2, Hebei, Yunnan, Sichuan, Hubei, Guangdong and Gansu have a close relationship; They can be used as the same medicinal source.
2.Combined application of a self-expanding neurovascular stent and endovascular coils in the management of wide-necked intracranial aneurysms

Objective To assess the technical feasibility and efficacy of the combined application of a flexible,self-expanding neurovascular stent(Neuroform)and Gugliebni detachable coils(GDC)in the management of wide-necked intracranial aneurysms in humans.Methods Sixty-five wide-necked aneurysms which underwent 65 endovascular procedures were performed by using intracranial stent and GDC.There was a total of 30 aneurysms at basilar artery including 16 at the basilar tip,9 at the basilar trunk and 5 at the beginning of the basilar artery.And there were 30 aneurysms located at the posterior communicating artery, and 5 aneurysms located at the vertebral artery.The Neuroform stents were deployed to cover the neck of aneurysms.Another microcatheter was introduced into the aneurysm sac through the stent interstices and then detachable coils were released to embolize the aneurysms.Results The combined procedures were successful in all of the 65 patients with wide-necked aneurysms.The stent could pass smoothly through the intracranial artery and got released.Complete occlusion was achieved in 60 patients and incomplete occlusion in 5 patients.In-stent thrombosis occurred in 2 patients.All patients recovered well.Forty-two patients had followe-up angiography at 3 to 6 months after the procedure.Among them,no filling was found for the 39 aneurysms which were densely packed,and 3 aneurysms had neck remnant.Conclusion The implantation of Neuroform stent as a complimentary device to GDC coiling is easy and safe for embolization of wide-necked intracranial aneurysms.It has great advantage for treatment of wide-necked intracranial aneurysms.

4.Service Middleware of Medical Information Integration and Exchange Based on HL7 and DICOM.

Medical information exchange and integration is the effective method to solve the interoperability and medical information island, and is the basis of medical information sharing. In this paper, we take medical texts and medical images as the basic integrated objects, DICOM, HL7 messages and datasets as the integrated units, efficient DI-COM, HL7 message construction and parsing methods as basis, design and realize a universal medical information integration and exchange service middleware. Experimental results show that the prototype system could perform medical information integration and exchange among relational database, HL7 and DICOM message, provide a feasible scheme to solve the medical information island and lay a good foundation for establishing the unified medical information integration and sharing platform. The middleware has been applied in the project named "development and demonstration of opened medical information integration system".

5.Clinicopathological features, treatment and prognosis of soft tissue sarcoma and sarcomatoid carcinoma of urinary and male reproductive system in adults

Objective:Clinicopathological features, treatment and prognosis of urinary and male reproductive system soft tissue sarcoma (STS) and sarcomatoid carcinoma in adults were compared.Methods:A retrospective analysis was performed on the clinical data of 73 patients with STS and 15 patients with sarcomatoid carcinoma in adult urinary and male reproductive system in the First Affiliated Hospital of Zhengzhou University. There were 59 males and 14 females in STS group, with a median age of 41 (18-78)years old. The maximum tumor diameter ranged from 0.5 to 19.0 cm. The primary tumors were located in testis and peritesticular (23 cases), kidney (23 cases), prostate (15 cases), bladder (8 cases), ureter(3 cases), other parts(1 case). There were 18 cases of lymph node metastasis and 8 cases of distant metastasis. Among 73 patients with STS, 66 patients underwent surgical resection, of which 31 patients underwent radical resection. Among the 66 patients who underwent surgery, 3 patients received neoadjuvant chemotherapy; 22 patients received adjuvant chemotherapy; 5 patients were treated with adjuvant radiotherapy. Among 7 patients with STS did not receive surgical treatment, 2 patients received radiotherapy combined with chemotherapy, 2 patients received chemotherapy alone, and 3 patients received symptomatic support treatment.There were 11 males and 4 females in sarcomatoid carcinoma group, with a median age of 65 (23 - 84)years old. The measurable tumor diameter ranged from 0.4 to 16.9 cm. The primary tumors were located in kidney (6 cases), bladder (5 cases), ureter(2 cases) and prostate(2 cases). There were 2 patients of lymph node metastasis and 4 patients of distant metastasis. Of the 15 patients with sarcomatoid carcinoma, 12 patients underwent surgical resection, of which 5 patients underwent radical resection. 2 patients were treated with adjuvant therapy after operation. Among the 12 patients who received surgical treatment, 2 patients had distant metastasis before operation, all of which originated from the kidney. Among the 3 patients without surgical treatment, 1 patients received systemic chemotherapy and 2 patients received symptomatic supportive treatment. There was no significant difference in gender, tumor maximum diameter, distant metastasis and operation, chemotherapy, radiotherapy and operation combined with chemotherapy ( P>0.05) and there were significant differences in age, tumor primary location and lymph node metastasis ( P<0.05) between STS and sarcomatoid carcinoma patients.The categorical variables of the two groups were compared by χ2.With Kaplan-Meier method for univariate survival analysis, the Cox was used for multivariate analysis. Results:The median follow-up time was 18.3(0.3-90.4) months.In STS group, there were 14 patients of synovial sarcoma, 11 patients of liposarcoma, 15 patients of rhabdomyosarcoma, 16 patients of leiomyosarcoma, 10 patients of other types, and 7 patients of spindle cell sarcoma without specific classification. Among 66 patients with STS, 8 patients recurred, 14 patients metastasized after operation, 4 patients recurred and metastasized after operation. The 7 patients without surgical treatment all progressed. Among the 10 patients of sarcomatoid carcinoma without distant metastasis before operation, 3 patients recurred and 3 patients metastasized after operation. Two patients of renal sarcomatoid carcinoma with distant metastasis were treated with nephrectomy and chemotherapy. One of them had overall survival (OS) up to 2 years, and one recurred 2 months after operation. The 3 patients without surgical treatment all progressed without remission. The median OS of STS patients were 59.3 (95% CI 24.1-94.5) months and that of sarcomatoid carcinoma patients were 8.7 (95% CI 6.1-11.2) months. The OS of STS patients were better than those of sarcomatoid carcinoma patients ( HR=2.874, 95% CI 1.118-7.386, P=0.022). Conclusions:The onset age of STS in adult urinary and male reproductive system was lower than that in sarcomatoid carcinoma. The primary lesions of STS were mainly in testis, peritesticular and kidney. The primary lesions of sarcomatoid carcinoma were mainly in kidney. Among STS, leiomyosarcoma was the most common type.STS and sarcomatoid carcinoma should be diagnosed and treated with surgery quickly, and systemic therapy should be performed for patients who cannot be treated with surgery.
6.Study on the Fingerprints of Different Parts of Rhubarb from Genuine Medicinal Materials in Gansu Province

OBJECTIVE:To investigate the differences of the active components of different parts of rhubarb(the head,the body and the end part of the root)from genuine medicinal materials in Gansu province so as to provide theoretical basis for its quality control.METHODS:A comparative study of the fingerprints of different parts of rhubarb extract were conducted by HPLC,in which,the C18 was used as the chromatographic column under room temperature,the mobile phase consisted of menthol-0.1%H3PO4(70∶30)with a flow rate of 1.0ml/min,the column's temperature of room temperature and detection wavel_ ength of 280nm.RESULTS:Significant differences were noted in the HPLC fingerprints of different parts of rhubarb,there were significant differences in the contents of the active components such as emodin,chrysophanol,etc.,the contents decreased progressively from the head of root,the body part to the end part.CONCLUSION:In order to ensure the medical value and the quality of rhubarb medicinal material,different parts of which should be differentiated in the quality control.
7.Research advances on the chemical components and pharmacological activities of Zanthoxyli Radix

Zanthoxyli Radix is a traditional Chinese medicine. It can be used for the treatment of wind-cold-dampness arthralgia, muscle and bone pain, fall fracture, hernia, sore throat, toothache and other diseases. Due to possessing many excellent and mild pharmacological properties, there are lots of reports about Zanthoxyli Radix worldwide. At present, more than 100 bioactive components have been extracted and purified from Zanthoxyli Radix. Nitidine chloride (NC), one of the most important alkaloids in Zanthoxyli Radix, has the activities of anti-tumor, anti-inflammation, anti-bacteria, etc. In this review, we summarize the chemical components of Zanthoxyli Radix, pharmacological activity and mechanism of action of NC to provide references for further research and utilization of Zanthoxyli Radix.
8.Effects of sex on chronic pancreatitis induced by L-arginine in Kunming mice

AIM:To explore the effect of sex on the formation of chronic pancreatitis(CP)by comparing the differences in L-arginine-induced CP model between male and female mice.METHODS:Male(n=42)and female(n=42)Kunming mice were randomly divided into 4 groups:female control group, female CP group, male control group and male CP group(n=18 in each control group, n =6 at each time point; n=24 in each CP group, n=8 at each time point).The mice in CP groups were intraperitoneally injected with 20%L-arginine(3 g/kg,twice/d,1 d/week).After modeling for 2 weeks,4 weeks and 6 weeks,the mice were anesthetized and sacrificed.The morphological changes and the degree of fibrosis in the pancreas were observed by HE and Masson staining.The positive expression rate of F4/80 in the pancreatic tissues was observed by the method of immunohistochemistry.The mRNA expression of interleukin-6(IL-6), α-smooth muscle actin(α-SMA)and fibronectin(FN)in the pancreas were detected by real-time PCR.The protein of pancreas was extracted to detect the expression of α-SMA and FN by Western blot.RESULTS:At 2 weeks,4 weeks and 6 weeks after intraperitoneal injection of L-arginine, the pancreatic tissues were obviously injured and exhibited different degrees of fibrosis in female and male CP groups.At the same time,there were significant differences in the degree of pan-creatic injury between male and female mice,and the degree of pancreatic lesion in the male mice was significantly more severe.The positive rate of F4/80 in the pancreas of male CP mice was significantly higher than that in female CP group at the same time point after modeling.At every time point, the mRNA expression of IL-6 in the pancreas was increased in both female and male CP groups,but that in male CP group was higher(P<0.05).The fibrosis indexes,α-SMA and FN, were both highly expressed at mRNA and protein levels after modeling, but compared with the female group, the time of positive expression in male mice was earlier and the expression level was higher(P<0.05).CONCLUSION: The CP model is successfully established by intraperitoneal injection of 20%L-arginine,and a difference in the degree of pathologic alteration in pancreas between male and female mice exists.CP is more effectively induced by L-arginine in male mice,and the degree of fibrosis is more pronounced.The reason may be related to the sensitivity of male mice to L-arginine,causing more serious inflammatory response.Therefore,male mice are more suitable for establishing CP animal model.
9.Quantitation and detection of deletion in tumor mitochondrial DNA by microarray technique.

OBJECTIVETo develop a method to rapidly quantitate and detect deletion of mitochondrial DNA (mtDNA) by microarray technique as a tool to study its relationship to tumorigenesis.
METHODSA modified PCR was used to amplify full length mtDNA sequence in two samples of normal human blood leukocytes and five samples of gastric cancerous tissues, which were simultaneously labeled with fluorescin. The amplified products were verified by polyacrylamide gel electrophoresis (PAGE) and silver staining. Then, 17 pairs of overlapping primers of mtDNA were designed and their PCR products were used as mitochondrial probes. They were spotted onto amino-slides as microarray and hybridized. Hybridization image was scanned with GeneTAC laser, mtDNA copy number was counted by ScanAnalyzer software.
RESULTSPAGE analysis showed that the designed probes were quite reasonable and strongly specific. The modified PCR method was efficient to amplify the whole mitochondrial genome with high-yield specific bands. The hybridizing spots were distinct, and background was clear. The signals of negative probes were close to those of background, and there was no significant difference between them (P > 0.05). The results were identical to those in the designed experiment. There were no significant differences between the results when the same sample of blood leucocytes or cancer tissues repeatedly examined with the same positive probes (P > 0.05), while there were significant differences when different types of samples were examined (P < 0.01). The hybridizing signals were stable and most of the data distributed in the range of mean +/- 2xSD.
CONCLUSIONThe method here reported can rapidly, correctly and massively determine whether there exist special deletion and/or quantitative changes of mtDNA in patients with tumors. It will be helpful for the study of the relationship between mtDNA alteration and tumor development.

10.Correlation analysis on the malocclusion and articulation of skeletal angle III malocclusion in mixed dentition.

OBJECTIVETo investigate the effects of skeletal Class III malocclusion in mixed dentition on speech articulation and to look for which factors lead to the speech errors.
METHODSThirty-eight children with skeletal Angle Ill malocclusion in mixed dentition were selected as a sample group and 40 children with normal occlusion in mixed dentition as a control group. Two phoneticians evaluated their articulations and wrote down error phonemes respectively. The correlation analysis was undertaken between the number of errors and the measurements of patients' cephalometry.
RESULTSThe number of errors were correlated significantly with overbite, UI-LI, OBJ (OB+OJ) and TD-PW.
CONCLUSIONThere is articulatory malfunction in the majority of skeletal Angle III malocclusion patients in mixed dentition. Articulatory malfunction is related to the position of incisors and the tongue.
