In order to confirm the role that the 49th amino acid residue plays in enzymatic inactivity of Glutamine 49 phospholipase A2(Gln49-PLA2),site-directed mutagenesis of its 49th amino acid gene codon was conducted using PCR.Aspartic acid 49 phospholipase A2(Asp49-PLA2-Q49D-PLA2),the mutant of Gln49-PLA2 was expressed in E.coli with pET32a+ vector. The fusion protein, expressed as inclusion body, after being denatured, was on-column refolded and purified by immobilized metal affinity chromatography(IMAC),and then cleaved by Factor Xa.The mature Q49D-PLA2 mutant was obtained by Hitrap SP cation exchange and Superdex 75 gel filtration chromatography, with the recovery rate of 1.3%,and the specific activity of the mature Q49D-PLA2 mutant was 72 U/mg.It has been demonstrated that the 49th glutamine amino acid residue is the main reason in enzymatic inactivity of Gln49-PLA2 and the results are helpful for denatured protein refolding, especially in rich disulfide bonds conditions.

6.Silk fibroin/chitosan scaffolds for corneal tissue engineering: theoretical development and applications
Chinese Journal of Tissue Engineering Research 2018;22(6):952-957
BACKGROUND:Keratitis, chemical injury, trauma, and other diseases often lead to irreparable damage to the corneal tissue, which can cause scars and finally vision loss.Corneal transplantation is the only way for treatment, but is of limit use by the deficiency of donors. Moreover, biosynthetic implants are expensive and complex.Silk fibroin and chitosan are common materials for tissue engineering and have been widely used as bio-scaffold materials. However, the technology applied in corneal repair is still in infant stage.OBJECTIVE:To systemically analyze the literature regarding the application of silk fibroin/chitosan scaffold materials in corneal repair, and to offer new directions and theoretical support for further studies on corneal repair.METHODS:A computer-based search of CNKI,WanFang and PubMed was performed for relevant articles with the keywords of "silk fibroin, chitosan, cornea, keratoprosthesis, scaffold".Then,we traced the development progress of silk fibroin/chitosan scaffolds in corneal repair and summarized its current applications and prospects.RESULTS AND CONCLUSION:In total,178 articles were preliminarily retrieved, and finally,52 eligible articles were reviewed.(1) The silk fibroin/chitosan scaffold for corneal repair has low cytotoxicity and good biocompatibility.(2) This kind of scaffold supplies proper pericellular environmental for corneal cells that is good for hydroelectrolyte exchange.(3) The presence of ABCG2 protein implies that the scaffold offers possibilities of limbal stem cell proliferation.(4) Increasing animal model tests have shown that the scaffold can barely cause an inflammatory reaction of corneal cells, which is highy important to remain the vitreousness of corneal tissues.(5) Because of the deficiency of corneal donors, searching for new artificial materials for corneal repair is the developing direction of corneal transplantation.Silk fibroin chitosan scaffold has huge potential in this area, and it deserves more researches for clinical use in the future.

Abstract: Objective To explore the correlation between monocyte to high-density lipoprotein cholesterol ratio (MHR) and insulin resistance (IR) in male patients with type 2 diabetes mellitus (T2DM) combined with metabolic-related fatty liver disease (MAFLD). Methods A total of 454 male patients with T2DM combined with MAFLD in National Metabolic Management Center (MMC) of the Affiliated Hospital of Jiangsu University from May 2018 to July 2020 were enrolled. The general clinical data of subjects were collected, blood routine and biochemical indexes were tested, homeostasis model insulin resistance index (HOMA-IR) was calculated, visceral fat area (VFA) and subcutaneous fat area (SFA) were measured. Accordingtothe MHR quartile, patients were divided into group Q1 (MHR≤0.38), group Q2 (0.38

OBJECTIVETo evaluate the efficacy and safety of long-term on-demand use of vardenafil in the treatment of erectile dysfunction (ED).
METHODSWe conducted a questionnaire investigation among 891 ED patients treated by on-demand use of oral vardenafil at 20 mg every 3 days from March 2007 to January 2010, covering the general information of the patients, their need for and attitudes towards the treatment, clinical efficacy and adverse events of the drug, and satisfaction of the patients and their partners after 12 weeks of treatment.
RESULTSTreatment and follow-up were completed in 700 patients, of whom 504 (72%) achieved sufficient hardness and duration of penile erection and overall sexual satisfaction, 84 (12%) admitted to improvement of erectile hardness and duration but not adequate satisfaction, and the other 112 (16%) experienced no significant changes. Significant differences were found in IIEF-5 scores, Rigiscan test results and partners TSS scores before and after the treatment (P < 0.05). Most frequent adverse events included flushing (15%), dizziness and headache (10%), dyspepsia (3%), and nasal congestion (1%).
CONCLUSIONLong-term on-demand use of oral vardenafil, in addition to its safety and good tolerance, can effectively improve the erectile function of ED patients, their success rate of sexual intercourse, and overall quality of sexual life.

Chinese Journal of Hepatology 2008;16(8):590-593
OBJECTIVETo study liver plasma membrane (PM) proteins affected by hepatitis B virus (HBV), and to study the molecular mechanism of HBV infection through plasma membrane proteome analysis of transgene mice livers.
METHODSPlasma membrane was purified using second antibody superparamagnetic beads that combine subcellular fractionation and immunoisolation strategies. Western blot was used to verify the purification of plasma membrane and the expression of HBV envelope protein. The proteins from plasma membrane were extracted, quantified and separated by two-dimensional electrophoresis. The gels were stained by silver nitrate or Coomassie Blue and analyzed by Imagemaster software. The different expressed proteins were identified by matrix-assisted laser desorption/ionization time-of-flight/time-of-flight mass spectrometry (MALDI-TOF-TOF MS).
RESULTSThe plasma membrane of mice livers was enriched 3-fold and the contamination from mitochondria was reduced 2-fold in comparison with the density gradient centrifugation method. HBV envelope protein was found only in HBV transgene mice livers. More than 500 protein spots were separated in Coomassie Blue stained gels. Twenty-six proteins with two-fold differences were found through Imagemaster software analysis. Seven proteins were identified.
CONCLUSIONAn optimized plasma membrane purification method was developed; protein profile of liver plasma membrane changed in the transgene mice livers; some proteins related to HBV infection were found, and this work may be helpful in the research of the molecular mechanism of HBV infection.

Journal of Third Military Medical University 2001;23(5):591-592
Objective To investigate the prognosis in risk of ventricular arrhythmia in coronary heart disease with heart rate variability (H RV), left ventricular ejection fraction (LVEF) and other clinical background dat a. Methods A total of 81 patients were divided into ventricular premature beats (VPBs)≥30/h group and VPBs<30/h group. Their LVEF, HRV and cli nical data were studied and analyzed. Results The age and blood pressure between 2 groups had no significant difference. LVEF, standard deviati on of all normal RR intervals (SDNN), SD of the average of NN interval (SDANN) a nd HRV triangular index (HRVI) were significant less in VPBs≥30/h group than in VPBs<30/h group (43.29±15.38 vs 67.33±11.47,P<0.01;90.05±22.2 9 vs 117.90±30.32,P<0.05;77.43±17.78 vs 105.69±28.79,P<0.05 ;24.54±8.70 vs 32.70±10.87,P<0.05, respectively). Incidence of myo cardial infarction (MI) was larger in VPBs≥30/h group than VPBs<30/h group. LVE F was the independent predictable factor in risk of ventricular arrhythmia with multinomial regression logistic analysis(B=0.119, P=0.032). Co nclusion Our findings indicate that LVEF is an independent predictable factor i n risk of ventricular arrhythmia in coronary heart disease. Although HRV and MI history can not be used to predict VPB, significant difference is found between 2 groups. High-risk patients could be selected successfully when these data are considered in combination.
