1.Simultaneous Determination of 6 Antidepressants in Human Plasma by HPLC

OBJECTIVE:To establish a rapid and sensitive HPLC method for simultaneous determination of plasma concentrations of 6 antidepressants.METHODS:With diprozin as internal standard,the alkalized samples were extracted by liquid-liquid extraction and separated on a Phenomenex-C18 column using acetonitrile-0.05 mol?L-1 sodium dihydrogen phosphate(pH was adjusted to 2.5 by phosphoric acid) as mobile phase at a flow rate of 1.2 mL?min-1 by a gradient elution.The column temperature was set at 40℃.Venlafaxine was detected by fluorescence detection at an Ex wavelength of 276 nm and Em wavelength of 598 nm;doxepin,paroxetine,sertraline,fluoxetine and amitriptyline were detected at an UV detection wavelength of 200 nm.RESULTS:The linear ranges of venlafaxine,doxepin,paroxetine,sertraline,fluoxetine and amitriptyline were 5~1 000 ?g?L-1,40 ~200 ?g?L-1,20~800 ?g?L-1,40~1 000 ?g?L-1 and 10~400 ?g?L-1,respectively,with correlation coefficients ≥0.990 for all.Both the intra-day RSD and inter-day RSD were less than 15%;the extraction recovery rates were greater than 60% and methodological recovery were greater than 90% for all the samples.CONCLUSION:The method is simple,economical,rapid,accurate and sensitive,and it is applicable for the clinical monitoring of plasma drug concentrations as well as the analysis and pharmacokinetic study of toxic drugs.
2.Clinical trial of Kelisu combined nasal continuous positive airway pressure for neonatal respiratory ailure

Objective To evaluate the effect of calf pulmonary surfactant (Kelisu) combined nasal continuous positive airway pressure (NCPAP) in the treatment of neonatal respiratory failure (NRF).Methods Forty-eight NRF in our hospital were selected,and divided into the control group (n =24) and the treatment group (n =24) according to random number table.Control group were given NCPAP therapy,treatment group were treated with Kelisu 70-100 mg · kg-1 combined NCPAP.After using Kelisu 0,6,12,24 h,the results of arterial blood gas analysis,respiratory function indexes,clinical effects and incidence of complications in the two groups were compared.The adverse drugs reaction (ADR)were investigated.Results After 12 h treatment,the level of PaO2 in treatment group with (75.7 ± 1.6) mmHg was higher than the control group with (70.8 ± 1.8) mmHg.The level of PaCO2 in treatment group with (51.2 ± 1.7) mmHg was lower than the control group with (53.8 ± 0.7) mmHg.The time of the oxygen curein in treatment group with(180.65 ± 21.64)h was shorter than the control group with (217.52 ±19.58)h.The incidence of ADR in treatment group with 16.7% (4 cases/24 cases) was lower than the control group with 45.8% (11 cases/24 cases).Camparison between control group and the treatment group on the factors,the differences were statistically significant(all P < 0.05).Conclusion Endotracheal dripping of calf pulmonary surfactant combined with NCPAP is an effective treatment for NRF,and had good safety.
3.Expressions of Caveolin-1 and Transforming growth factor betal 1 in A549 cells exposed to hyperoxia and their relationship

Objective To observe the expressions of Caveolin-1 (CAV-1) and transforming growth factor bata 1 (TGF-β1) in human adenocarcinoma of lung A549 cells exposed to hyperoxia,and to explore the changes in CAV-1 and TGF-β1 expressions and the role in hyperoxic lung injury.Methods After the A549 human lung cancer cell lines were resuscitated,the cells were packaged and cultured in 50 mL/L CO2 culture chamber,when approaching to the condition of confluence in CO2 culture chamber(37 ℃,50 mL/L CO2),the cells were randomly divided into control group and hyperoxia group.Eighteen bottles of A549 cells in each group.The hyperoxia group received attacking factor that was mixed with oxygen (950 mL/L)and CO2 (50 mL/L) at a velocity of 3 L/min for 10 min;then the flasks were enclosed and cultured in culture chamber,while those in control group were still exposed to 5 mL/L CO2.After cells were cultured for 12 h,24 h,48 h,the changes in morphology were observed under the inverted microscope,the expressions CAV-1 and TGF-β1 were detected by double immunofluorescence staining technique,followed by laser scanning under the focus microscope.Results In the control group,there was high expression of CAV-1 associated with low expression of TGF-β1 (44.25 ± 3.23,10.18 ± 2.74).Compared with control group,the content of CAV-1 reduced continuously (12 h:35.25 ± 1.88,24 h:20.75 ± 1.68,48 h:9.86 ± 2.80) under prolonged hyperoxia exposure,but TGF-β1 increased gradually(12 h:17.32 ± 1.50,24 h:26.51 ± 1.82,48 h:41.94 ±4.47),and the expression of CAV-1 had highly negative relation to TGF-β1 at the same time (r =-0.91,-0.62,-0.53,-0.88,all P < O.05) in the control group and the hyperoxia group in 12 h,24 h,48 h.Conclusions The reduction of CAV-1 expression can attenuate the inhibitory effect of TGF-β1 and activate TGF-β1 signaling pathway,which may result in hyperoxic lung injury.
4.Protective effect of Shenmai injection on the behavior change in adult rats after neonatal maternal separation

Objective To study the protective effect of Shenmai injection (SI)on the behavior change in adult rats after neonatal maternal separation.Methods Neonate-mother separation (NMS) animal model was replicated.The rats in maternal separation group were separated for 3 h per day on postnatal days 2 to 21.Neonatal rats were divided randomly into three groups (n =12):control group,model group (NMS),experimental group (SI).Each group had twelve rats.After two months,the behavior changes of adult rats were tested.The expression of Bcl-2 in hippocampus was detected by immunohistochemical method.Results The total distances in experimental group and model group were (15.17 ± 5.32),(12.32 ± 2.74) m with significant difference (P < 0.05).The average speeds in experimental group and model group were (0.09 ± 0.03),(0.07 ± 0.04) m · s-1 with significant increase (P < 0.05),But not to the same as that of control group.The expression of hippocampus Bcl-2 in model group,control group and experimental group were 110.34 ± 37.32,92.62 ± 52.13,152.17 ± 29.65 with significantly increased in model group compared with control group (P <0.05);while decreased in experimental group compared with model group (P <0.05).But not to the same as that of control group.Conclusion Shenmai injection possibly can improve the cognitive impairment,which may be related to the upregulation of the expression level of apoptosis gene Bcl-2.
5.Serum homocysteine and metabolic syndrome in middle-aged and elderly men.

OBJECTIVETo investigate the relationship among serum reproductive hormone levels, serum homocysteine (Hcy) levels, metabolic syndrome (MS), and the components of MS in middle-aged and elderly males.
METHODSUsing the cluster and stratified sampling methods and a unified structured questionnaire, we conducted a survey among 948 men aged 40 - 80 years in the rural community, measured their basic physical parameters, and obtained their reproductive hormone levels, serum Hcy concentrations, and metabolism-related indicators. We collected 868 valid questionnaires along with their serum samples, divided the subjects into an MS and a non-MS control group in a 1:1 ratio, and measured their serum Hcy concentrations.
RESULTSAmong the subjects included, 132 were diagnosed with MS. Nonparametric tests showed statistically significant differences between the MS and non-MS groups in the waist circumference (WC), waist-hip ratio (WHR), body mass index (BMI), systolic blood pressure (SBP), and diastolic blood pressure (DBP) (P < 0.05), but not in age (P > 0.05). Significant differences were also observed between the two groups in the levels of serum tT, SHBG, LH, and FTI (P < 0.05) , but not in the concentrations of serum Hcy (P > 0.05). The concentration of serum Hcy exhibited no correlation with BMI, SBP, DBP, FBG, TG, and HDL-C (P > 0.05) and had no influence on MS.
CONCLUSIONThe concentration of serum Hcy is not significantly correlated with MS, nor with its components. The levels of male serum reproductive hormones are associated both with MS and with its components.

6.Improvement of matrine on the cardiac function and left ventricular remodeling in cardiac hypertrophy of rats

Objective To investigate the effect of matrine on the cardiac function and left ventricular remodeling in rats with isoproterenol (ISO) -induced cardiac hypertrophy.Methods Seventy-two adult male SD rats were randomly divided into six groups as follows:normal group,model group,control group (matrine 200 mg · kg-1 · d-1),large-,medium-and small-dose experimental groups(matrine 200,100,50 rng · kg-1 · d-1),and there were 12 rats in each group.The 5 mg · kg-1 · d-1 isoproterenol (ISO) was subcutaneously injected to establish the model of chronic pathological left ventricular hypertrophy in model group and experimental groups.The 0.9% NaCl was administrated to rats in normal group and control group.The aforementioned medications were offered once daily to 12 rats of each group for 7 successive days.On day 8,cardiac function was evaluated by the biological function experiment system.The hematoxylin-eosin staining was used to observe pathological morphology changes of the left ventricular tissues.Insulin-like growth factor-1 (IGF-1) and transforming growth factor-β1(TGF-β1)levels in serum were determined by ELISA.Expression levels of IGF-1and TGF-β1 genes in myocardial tissues were detected by real time fluorescence quantitative PCR method.Results After drug-concerned treatment,serum IGF-1 levels in normal group,model group,control group,large-,medium-and small-dose experimental groups were separately (2.04 ± 0.52),(3.66 ± 0.97),(2.08±0.95),(1.98 ± 0.98),(1.93 ±0.78) and (1.59 ± 0.48)ng · mL-1;moreover,serum TGF-β1 levels in those groups were (94.11 ±10.63),(139.98 ± 37.64),(90.36 ± 30.02),(84.55 ± 45.70),(74.74 ± 42.18) and (72.84 ± 21.43) pg · mL-1.In contrast with model group,the serum IGF-1 or TGF-β1 levels were lowered in other groups (P < 0.05).Similarly,expression levels of cardiac IGF-1 genes in normal group,model group,control group,large-,medium-and small-dose experimental groups were sequentially 3.34 ± 3.27,10.91 ± 7.44,3.09 ± 1.86,0.62 ± 0.44,0.64 ± 0.26,1.00 ± 0.35;furthermore,expression levels of cardiac TGF-β1 genes were successively 0.70 ± 0.67,10.97 ± 9.86,0.63 ± 0.32,1.25 ± 0.98,1.76 ± 2.40,0.86 ± 0.46;and hence the TGF-β1 gene levels in model group were much higher than those of other groups (P < 0.01).Conclusion Matrine significantly improved the cardiac function in rats with ISO-induced left ventricular hypertrophy and simultaneously regulated the cardiac hypertrophy-causing cytokines.
7.Study on the ARIMA model application to predict echinococcosis cases in China

Objective To predict the monthly reported echinococcosis cases in China with the autoregressive integrated mov-ing average(ARIMA)model,so as to provide a reference for prevention and control of echinococcosis. Methods SPSS 24.0 software was used to construct the ARIMA models based on the monthly reported echinococcosis cases of time series from 2007 to 2015 and 2007 to 2014,respectively,and the accuracies of the two ARIMA models were compared. Results The model based on the data of the monthly reported cases of echinococcosis in China from 2007 to 2015 was ARIMA(1,0,0)(1,1, 0)12,the relative error among reported cases and predicted cases was-13.97%,AR(1)=0.367(t=3.816,P<0.001),SAR (1)=-0.328(t=-3.361,P=0.001),and Ljung-Box Q=14.119(df=16,P=0.590).The model based on the data of the monthly reported cases of echinococcosis in China from 2007 to 2014 was ARIMA(1,0,0)(1,0,1)12,the relative error among reported cases and predicted cases was 0.56%,AR(1)=0.413(t=4.244,P<0.001),SAR(1)=0.809(t=9.584, P<0.001),SMA(1)=0.356(t=2.278,P=0.025),and Ljung-Box Q=18.924(df=15,P=0.217).Conclusions The different time series may have different ARIMA models as for the same infectious diseases.It is needed to be further verified that the more data are accumulated,the shorter time of predication is,and the smaller the average of the relative error is.The estab-lishment and prediction of an ARIMA model is a dynamic process that needs to be adjusted and optimized continuously accord-ing to the accumulated data,meantime,we should give full consideration to the intensity of the work related to infectious diseas-es reported(such as disease census and special investigation).

ObjectiveTo investigate the clinical features of systemic lupus erythematosus (SLE) patients with fever and find out the related factors.MethodsData was collected by the same methods in the past ten years in fifteen hospitals in Jiangsu province and then the data wereretrospectively analyzed.The potentially possible risk factors of fever in SLE were selected and then analyzed by chi-square test,Wilcoxon rank sum test and Logistic regression analysis.ResultsAll 1762 patients were investigated.Seven hundred and twenty-nine had active fever.Age at hospitalization,initially treated patients,photosensitivity,serositis,nervous system involvement,generalized lymphadenopathy/hepatosplenomegaly,white blood cell count (WBC),haemoglobin (HB),erythrocyte sedimentation rate (ESR),C-reaction protein (CRP),alanine aminotransferase(ALT),albumin(ALB),serum creatinine (Scr),complement C3,anti-dsDNA antibodies positive rate,anti-Sm antibodies positive rate,SLEDAI score and past therapies were factors associatedwith SLE fever.Logistic regression analysis showed that abnormal WBC count (OR=1.396,95%CI 1.114-1.711,P=0.004),CRP(OR=1.005,95%CI 1.002-1.009,P=0.002),ALT(OR=1.003,95%CI 1.001-1.005,P=0.005),Scr (OR=0.997,95%CI0.995-0.999,P=0.007),HB (OR=0.986,95%CI 0.981-0.992,P=0.000),age (OR =0.984,95% CI 0.974-0.993,P=0.001 ) and past usage of cyclophosphamide (CTX) (OR =0.557,95%CI 0.382-0.813,P=0.002) were correlated with SLE fever.ConclusionFever is one of the most common clinical manifestations of SLE patients.Leucopenia,elevated CRP levels,liver function abnormalities,anemia,younger age are risk factors for SLE fever,while renal impairment and past usage of CTX are protective factors.
9.Status of the poor vision among primary and secondary school students in a division of Xinjiang Production and Construction Corps and analysis of the influencing factors

AIM: To understand the status of poor vision and analyze the main influencing factors among primary and secondary school students in a division of the Xinjiang Production and Construction Corps, and provide a scientific basis for prevention and control of poor vision.METHODS: A total of 12 primary and secondary schools in a divisional urban district and regiment field of the Xinjiang Production and Construction Corps were chosen using stratified cluster random sampling from August to October 2020. A total of 2 982 primary and secondary school students were selected for visual inspections and questionnaires. Logistic regression analysis was used to explore the influencing factors of poor vision.RESULTS: The rate of poor vision was 65.66% among 2 982 primary and secondary school students in the division of the Xinjiang Production and Construction Corps, with the majority severe poor vision. Furthermore, the rates of poor vision were higher in girls(70.17%)than that in boys(61.47%)(χ2=4.993, P<0.001)and the urban area(70.03%)were higher than the rural area(58.96%)(χ2=38.680, P<0.001)and the Han nationality students(66.83%)were higher than Uyghur students(52.82%)(χ2=19.772, P<0.001). The risk of poor vision increased with age(χ2trend =300.144, P<0.001), and by the age group ≥18 years old, the rate of poor vision reached 80.47%. The results of the multivariate analysis showed that the women, the age≥13 years old, writing and reading duration every day ≥2h, cram school on weekends and presence of myopia in parents increases the risk of poor vision, increase the outdoor exercise time, sleep time ≥8h, and do eye exercises every day can reduce the occurrence of poor vision.CONCLUSION: Poor vision rate of primary and secondary school students in the division of the Xinjiang Production and Construction Corps is higher than that of many regions in the country. Girls with the age(≥13 years old)whose parents are myopic should receive special attention. Increasing outdoor exercise time, doing eye exercises every day, and keeping your eyes more than one foot away from books when reading and writing is effective in preventing poor vision.
10.Nomogram for predicting overall survival in children with neuroblastoma based on SEER database

Purpose:
This study was performed to establish and validate a nomogram for predicting the overall survival in children with neuroblastoma.
Methods:
The latest clinical data of neuroblastoma in Surveillance, Epidemiology, and End Results (SEER) database was extracted from 2000 to 2016. The cases included were randomly divided into training and validation cohorts. The survival curves were drawn with a Kaplan-Meier estimator to investigate the influences of certain single factors on overall survival. Also, least absolute shrinkage and selection operator regression was applied to further select the prognostic variables for neuroblastoma. Additionally, receiver operating characteristic (ROC) curves and calibration curves were used to evaluate the accuracy of the nomogram.
Results:
In total, 1,262 patients were collected and 8 independent prognostic factors were achieved, including patients’ age, sex, race, tumor grade, radiotherapy, chemotherapy, tumor site, and tumor size. Then we constructed a nomogram by using the data of the training cohort with 886 cases. Subsequently, the nomogram was validated internally and externally with 886 and 376 cases, respectively. The internal validation revealed that the area under the curves (AUC) of ROC curves of 1-, 3-, and 5-year overall survival were 0.69, 0.78, and 0.81, respectively. Accordingly, the external validation also showed that the AUC of 1-, 3-, and 5-year overall survival were all ≥0.69. Both methods of validation demonstrated that the predictive calibration curves were consistent with standard curves.
Conclusion
The nomogram possess the potential to be a new tool in predicting the survival rate of neuroblastoma patients.
