1.Case review of Wilson′s disease and correlation analysis of factors except heredity

Objective To retrospectively analyze the clinical classification ,laboratory and imageological examinations of Wil‐son′s disease for strengthening the cognition on this disease and increasing the diagnosis rate .Methods The clinical data of the pa‐tients with WD in our hospital from January 2011 to September 2014 ,were collected and performed the statistical analysis .Results Among different ages ,nationalities and genders of WD patients ,first onset age ,clinical classification and laboratory examination had some difference ,the average age of first onset in male WD patients was earlier than that in female WD patients ,the low density shadow change of nervous type WD existed in MRI .Conclusion Heredity is not a unique factor affecting WD ,gender ,age and na‐tionality may be one of factors affecting the first onset age and clinical symptoms in the WD patients .
2.Effects of prenatal pulsed electromagnetic fields on neural stem cell proliferation and nestin protein expression in the hippocampus of rat offspring

BACKGROUND:Electromagnetic fields can cause changes of the body,especially the nervous system.Effect of pulsed electromagnetic fields(PEMFs)on neural stem cells has been detected.OBJECTIVE:To investigate the effect of prenatal pulsed electromagnetic fields(PPEMFs)on neural stem cell proliferation and nestin protein expression in the hippocampus of rat offspdng.METHODS:Sprague Dawley female rats weighing 240-260 g were included and randomly divided into two groups:control and PPEMFs.Rats from the control group were given no interventions.Rats from the PPEMFs group were given PEMFs stress at gestational days 14-20.Each stress was given three times daily for 10 minutes.The male and female offspring rats were sacrificed at 1 month of age and their brains were sectioned to determine the expression of nestin protein and Brdu-positive cells in the hippocampus by immunohistochemistry.RESULTS:The expression of nestin-and Brdu-positive cells in the hippocampus of female and male PEMFS offspring were significantly higher compared with the control group(P<0.001),and there was a significant difference between female and male offspring(P<0.001).The nestin-and Brdu-positive cells in female offspring outnumbered those in male offspring(P<0.001);however,there was no significant difference between female and male offspring in the control group(P>0.05).CONCLUSION:PPEMFs can increase the number and proliferative capability of the neural stem cells in offspring.It may be a pdmary stage of the cascade reaction of the body to the brain damage caused by PPEMFs stress.
3.Expression and role of connective tissue growth factor in the peripheral nerve after chronic compression injury

Objective To investigate the effects of connective tissue growth factor (CTGF) on the chronic peripheral nerve compression injury and explore the function of CTGF in peripheral nerve compression injury and repair. Methods From July 2010 to September 2010, fifty aduh male SD rats were randomly divided into group A and B: group A (sham-operated group): only exposed the sciatic nerve; group B (compression group): undergone sciatic nerve entrapment operation on the right hind leg according to the method which Mackinnon adopted when he established the model of chronic sciatic nerve compression.Electron microscopy,immunohistochemistry,RT-PCR and Western-blot were performed to observe the morphological changes of the compressed nerve tissue and to determine the level of CTGF,collagen- Ⅰ,Ⅲ (COL- Ⅰ,Ⅲ),2,4,6,8,10 weeks after the surgery,respectively. Results After sciatic nerve compression,the collagen in nerve increased ; The expression of CTGF and COL- Ⅰ, Ⅲ in sciatic nerve of compressed group increased, which was statistically different compared with the sham-operation group (P < 0.05); In the meanwhile,the contents of CTGF and COL- Ⅰ,Ⅲ were positively correlated in a certain period. Conclusion Peripheral nerve fibrosis can be caused by chronic nerve compression.The expression of COL- Ⅰ,Ⅲ in sciatic nerve increased and CTGF get involved in the pathophysiological process, which suggests that CTGF plays an important role in the process of neural injury and fibrosis.
4.Role of iatrogenic transforming growth factor-β1 antibody in peripheral neural fibrosis after chronic entrapment

ObjectiveTo investigate the effect of transforming growth factor-β1 (TGF-β1) antibody on peripheral neural fibrosis after chronic entrapment.MethodsA total of 75 rats were randomly divided into three groups, ie, Group A (sham operation, only the sciatic nerve exposed), Group B (compression only, treated with sciatic nerve entrapment) and Group C (compression plus antibody injection).Electron microscopy, immunohistoehemistry, RT-PCR and Western blot were performed to observe the morphological changes of the compressed nerve tissue and to determine the level of TGF-β1 , collagen Ⅰ and collagen Ⅲ at 2, 4, 6, 8, 10 weeks after sciatic nerve compression, respectively.Results The levels of TGF-β1 , types Ⅰ and Ⅲ collagen protein were increased significantly in the Group B compared with that in the Group A, when the expression of TGF-β1 was increased in the early phase of the compression, reached the peak at the 4th week, and then decreased slowly.The expressions of collagenⅠand collagen Ⅲ were increased after compression, reached a peak at the 6th week and then maintained a relatively high level.The number of the fibrous tissues was decreased significantly and the content of types Ⅰ and Ⅲ collagen protein declined in the Group C, with statistical difference compared with the Group B (P < 0.05).ConclusionsPeripheral nerve fibrosis can be caused by chronic nerve compression.TGF-β1 plays an important role in effectively inhibiting the collagen synthesis and ameliorating the nervous fibrosis of the protein following peripheral nervous entrapment.

Orally disintegrating tablets (ODT), a kind of new solid tablet that rapidly disintegrates to work in the mouth, has became the hot form of new drug research in recent years with many advantages, such as the convenient taking, a widely applicable people, fast acting, high bioavailability, good compliance, and so on. ODT has been widely used in chemical medicines, while the application of it in traditional Chinese medicines (TCMs) is still in the stage of development The development of TCMs ODT provides a new direction for the research of Chinese medicine new dosage, accelerates the pace of connecting to the world and modernization of Chinese medicine. This dosage has a broad market prospect, and its quality control and assessment standards, taste, the disintegration time in vitro and evaluation method are the key factors that affect the industrialization, standardization of Chinese medicine ODT. Therefore, this paper reviewed the characteristics, preparation, taste masking technology and quality evaluation with new technology of ODT. Meantime, numerous application examples of ODT used in traditional Chinese medicine were described. We expect to provide the reference and utilization for the development of traditional Chinese medicine orally disinteeratine tablets.

7.Effect of TGF-β1 and CTGF on skeletal muscle fibrosis in nerve compression

ObjectiveTo investigate expression of TGF-β1,CTGF and collagen deposition in skeletal muscle during chronic entrapment of peripheral nerve. MethodsFifty rats were separated into two groups,control group and experimental group. At different time points after operation, the right gastrocnemius of 5rats from each group were collected for further analysis such as HE, Masson stain, immunohistochemical staining,RT-PCR and Western-blot. Results It was observed that axon degeneration occurred during chronic nerve entrapment,and which was in line with reports from other groups.Moreover,it had been demonstrated that after nerve entrapment,skeletal muscles may form fibrosis and degeneration consequently.Within this pathological procedure,expression of TGF-β1. CTGF and deposition of collagenⅠ changed rapidly when compared with control group.ConclusionOverall,these results indicated that these factors may be important during skeletal muscle degeneration after chronic nerve entrapment.
8.Effect of intermittent high glucose on INS-1 cell proliferation and the expression of skp2-p27

Objective To investigate the effect of intermittent high glucose on proliferation, apoptosis, and cell cycle progression of INS-1 cells, and the possible intracellular pathways activated by intermittent high glucose. Methods Cell viability was evaluated by cell counting kit, the cell cycle was determined by flow cytometry,Annexin-V/PI double-labeled cell apoptosis detection kit was used to monitor cell apoptosis. Cell cycle related protein Skp2 and p27 expressions were detected by Western blot. Results ( 1 ) Both intermittent and constant high glucose significantly inhibited the growth of INS-1 cells, and the former effect was more significant. ( 2 ) Intermittent and constant high glucose levels significantly increased apoptosis in INS-1 cells, and the former effect was more significant. (3) Intermittent and constant high glucose levels significantly inhibited the cell process, the G0/G1 cell cycle arrest also was induced by intermittent high glucose, resulting in lowered proportion of the G2/M phase and S phase of INS-1 cells. (4) Intermittent and constant high glucose significantly decreased the level of protein Skp2 and increased the level of cell cycle related protein p27. Conclusion Intermittent high glucose levels affect INS-1 cell growth and proliferation, as well as induce cell apoptosis, probably by decreasing the level of protein Skp2 and increasing the level of p27 in the cells, resulting in arrest of progression through the G1 phase to the S phase of INS1 cells, and thus impairment of cell proliferation.
9.Analysis of G6PD Screening Results in Different Group of the Southeast Dongguan

Objective To study different groups of deficiency rate of glucose-6-phosphate dehydrogenase (G6PD)and enzyme activity assay in the detection rate of female heterozygote in the Southeast Dongguan.Methods From January 2007 to April 2013,of 39 475 cases of test results were collected in Tangxia Hospital of Dongguan city,the gene frequency and the detec-tion rate of female heterozygote could be calculated through genetic equilibrium law in different group.Results The male de-ficiency rates of G6PD in different group were Adult group(A)5.03%,Neonatal Group(B)5.10% and Total group(C) 5.06%,respectively,and there were no significant difference between each groups (χ2 =0.0404,P =0.980).The detection rate of female heterozygote of A,B and C in each groups were 27.13%,14.49% and 23.87%,respectively,and the differ-ence were statistically significant between different groups (χ2 =32.26,P =0.000).Conclusion Prevalence of G6PD defi-ciency in this area was 5.06% and there were differences between the deficiency rate of G6PD in different populations.The enzyme activity assay in female heterozygote detection rate is not satisfactory,especially in group B,which is conducive to ge-netic counseling,prenatal diagnosis and birth defects,such as providing more comprehensive information.
10.Endovascular treatment for patients with spontaneous isolated superior mesenteric artery dissection: single-center experiences

Objective:To summarize the midterm to long-term outcomes and experiences of endovascular treatment (ET) of spontaneous isolated superior mesenteric artery dissection (SISMAD).Methods:The clinical data of 31 SISMAD patients from Jan 2011 to Dec 2019 treated with ET was retrospectively analyzed.Results:Successful ET was achieved in 29 patients with a technical success rate of 93.5%. A total of 36 self-expandable bare stents were planted in 28 patients and plain old balloon angioplastry (POBA) was performed in 1 patient. Abdominal pain disappeared within 24 hours in 89.3% of the patients after stenting. The rate of perioperative complication was 3.2%. There was no SMA dissection rupture bleeding, nor perioperative death occurred. The mean follow-up time was 53.5 (range, 6 to 110) months. There was no dissecting aneurysm formation, no SMA rupture and bleeding, and no stent rupture during the follow-up. The post ET 1-year, 3-year, and 5-year free from reintervention rate were 100%, 100%, and 91.7%, respectively.Conclusions:ET for SISMAD is safe and effective with satisfactory perioperative and midterm to long-term outcomes.
