1.Significance of anti-cell membrane associated DNA antibodies in diagnosis of systemic lupus erythematosus lacking of specific autoantibodies

Objective To investigate the effects of anti-cell membrane associated DNA (mDNA) antibodies in the diagnosis of systemic lupus erythematosus (SLE) lacking of specific autoantibodies including anti Sm,anti ds-DNA,and anti-nucleosome antibodies.Methods Indirect immunofluorescence assay was used to measure anti-mDNA antibodies in serum of 145 SLE patients,and indirect immunofluorescence,Western-blot and ELISA were used to detect the anti-dsDNA ,anti-Sm and anti- nueleosome antibodies respectively to analysis the value of anti-mDNA antibodies on the specific autoantibodies negative patients with SLE.Results The sensitivity for anti-mDNA antibodies (69.7%) in SLE was significantly higher than anti-Sm (19.7%),anti-dsDNA ( 31.9% ) and anti-nucleosome (45.8% ).The incidences of anti-mDNA antibodies in SLE lacking of anti-dsDNA,Sm and anti-nueleosome antibodies (AnuA) were 64.3% ,70.2% and 60.3% respectively.Conclusion Anti-mDNA antibodies are serologic marker of SLE and important in diagnosis of SLE lacking of anti-dsDNA,Sm and nucleosome antibodies.
2.Expression of neural cell adhesion molecule and modification of its N-glycan in ldlD-14 cells.

Neural cell adhesion molecule (NCAM) is a glycoprotein expressing on the surface of neurons, glial cells, bone cells and natural killer cells. NCAM plays an important role in the process of cell - cell adhesion and cell migration, and is also a model protein to study polysialic acid. In this paper, NCAM gene from mouse mammary gland cells (NMuMG) was cloned into eukaryotic expression vectors pcDNA3.1(+) and transfected into mutant Chinese hamster ovary cells ldlD-14. The stable transfection over-expressing NCAM was obtained through the G418 selection and confirmed by Western blotting. Due to unique characters of ldlD-14 cells, carbohydrate chain of NCAM molecule can be easily manipulated with or without adding galactose in the serum free medium, and this modification can provide the basis for further studies on the effect of glycosylation on NCAM molecular function.

3.Relationship of sleep disorder with plasma interleukin-2 and soluble interleukine-2 receptor in patients with depression

BACKGROUND: As indicated by experiments on animal, cytokine has regulatory effects on sleep. Clinical research has found abnormal level of cytokine in patients with depression. Sleep rhythm disorder is a common symptom of depression; however, the relationship between cytokine and sleep has hardly been conducted at home and abroad.OBJECTIVE: To explore the relationship between abnormal level of cytokine and clinical characteristics of sleep disorder so as to provide theoretical basis for early intervention and functional detection of sleep disorder in patients with depression.DESIGN: A case-controlled study using the patients with depression as the subjects and normal persons as the controls.SETTING: Two wards of the Department of Psychiatry of a university hospital.PARTICIPANTS: The study was completed in the Department of Psychiatry, Second Affiliated Hospital of Xinxiang Medical College. The subjects in this study were divided into depression group and control group. The patients in depression group were hospitalized in the Second Affiliated Hospital of Xinxiang Medical College from September 2001 to December 2001. Inclusion International Classification of Diseases(10th edition) and Chinese Classifiliver function, electrocardiography and kidney function examination were quilizer or lithium salt were used in the past year; had the history of alcohol Had endocrine, heart, liver and kidney diseases and other serious somatic diseases. The 35 patients who met the above criteria were 14 males and 21females aged from 18 to 60 years with the average age of(36 ± 12) years in the depression group. In the control group there were 13 males and 17 females aged from 19 to 60 years with the average age of(34 ± 10)years.METHODS: The levels of plasma interleukin-2(IL-2) and solute interleukin-2 receptor(sIL-2R) were detected with enzyme-linked immunoadsorbent assay.disorder in patients with depression.RESULTS: The level of plasma IL-2 in the depression group(77.92± 36.85) pg/L was lower than that of the normal control group (98.98± 30. 72 ) pg/L( t = 2. 446, P < 0.05). Moreover, IL-2 level was positively correlated to depth of sleep level( r = 0. 364, P < 0. 05) . No significant difference in sIL-2R level was found between the two groups( P > 0.05).CONCLUSION: The development of sleep disorder in patients with depression may be related with the level of cytokine, and the decrease of IL-2level can be the neurobiological basis of sleep level in depression.
4.Effect and Mechanism of Exercise for Patients with Cardiovascular Diseases (review)

Exercise is one of the important techniques of cardiovascular rehabilitation. Exercise can reduce inflammatory response to improve endothelial function, and improve mitochondrial function to increase myocardial cell activity. For cardiovascular risk factors, exer-cise can promote the activity of lipoprotein, increase the level of high-density lipoprotein;improve the function of insulin receptor to reduce insulin resistance, reduce platelet aggregation and improve endothelial function to reduce blood pressure. For the respiratory system, aerobic exercise can improve the function of respiratory muscle, thus relieve the dyspnea. Exercise can promote the activation of immune factor and increase metabolism, to increase immune function and anti-aging. Resistance exercise can improve mitochondrial function and promote fi-ber type conversion, to improve the function of skeletal muscle system.
5. Preparation of asiaticoside sodium alginate repair patch and its wound healing effect

Objective: Asiaticoside-sodium alginate repair patch (As-SA RP) was prepared and its effect on skin wound healing was investigated. Methods: Based on the cross-linking characteristics of sodium alginate and Ca2+, As-SA RP was prepared using asiaticoside as a drug model through blending with sodium alginate. The compatibility of the blend films was investigated by scanning electron microscope and FTIR. The whole layer of skin on both sides of the back spine of New Zealand's big ear rabbit was used to establish the trauma model. The repair effect of the external application of As-SA RP on skin wounds was investigated by observing the healing status and determining the expression levels of collagen (hydroxyproline as an index) and pro-inflammatory (TNF-α, IL-1β and IL-6) factors in the injured skin tissue. Results: Asiaticoside had good compatibility with sodium alginate and can be successfully prepared As-SA RP. Compared with other groups, the red swelling, bleeding, infection, and exudate on injured skin were significantly reduced, and the wound healing rate was also relatively increased after As-SA RP treatment. Moreover, the less inflammatory cells infiltration, more new capillaries and intact wound skin structure relatively tight and orderly arrangement of collagen fibers were observed in the As-SA RP group. On the other hand, the biochemical analysis showed that As-SA RP could effectively inhibit the high expression of TNF-α, IL-1β, IL-6 in the injured skin tissue and significantly increase the contents of hydroxyproline and collagen fiber. Conclusion: As-SA RP could improve the skin absorption of asiaticoside and show the better effect of skin wound healing than the active component asiaticoside.

7.Effect of different contrast agent doses on the semi-quantitative parameters of dynamic contrast-enhanced MRI of rodent mammary tumor model

Objective To explore the effects of different doses of contrast agent (CA) on types of time-signal intensity curves (TICs) and semi-quantitative parameters after dynamic contrast-enhanced MRI (DCE-MRI) on Walker 256 murine breast tumor model.Methods A total of 12 rats burdened Walker256 breast cancer models were established and divided into 3 groups randomly, 4 in each group.Routine MR and DCE-MRI scans of the rats using Bruker Pharmascan 7T MR scanner were performed.Doses for the 3 groups were 0.2, 0.3, and 0.5 mmol/kg, respectively.MR data, TICs types and semi-quantitative parameters from each different dose group were statistically studied and compared to observe the differences.Results Tumors were enhanced significantly after injection.The types of TICs in all tumors were the Ⅲ pattern which was not influenced by CA doses.Semi-quantitative parameters of first enhancement (Efirst), maximal enhancement (Emax), washout enhancement (Ewash), and signal enhancement ratio (SER) showed statistical differences among the three dose groups (P<0.05).Semi-quantitative parameters of time to peak (Tpeak) and washout velocity (Vwash) showed no statistical differences among the three dose groups (P>0.05).Mean signal intensity of each group was highly negatively linear correlated with scan times after the peak (r=-0.972, P=0.000;r=-0.971, P=0.000;r=-0.989, P=0.000).The washout slope (slopewash) showed no statistical differences among the three groups (P>0.05).Conclusions Injection doses of CA didn't change the TIC type, Tpeak, Vwash, and Slopewash.These parameters are comparable among different medical centers and can be considered as prior parameters to monitor the efficacy of neoadjuvant chemotherapy.
8.BRAF V600E Mutation as a Predictive Factor of Anti-EGFR Monoclonal Antibodies Therapeutic Effects in Metastatic Colorectal Cancer:a Meta-analysis

Objective To investigate the correlation between BRAF V600E mutation and anti-epidermal growth factor receptor (EGFR) monoclonal antibodies (MoAbs) therapeutic effects in metastatic colorectal cancer.
Methods Studies were included into meta-analysis to investigate the association between BRAF V600E mutation and clinical outcome in metastatic colorectal cancer patients treated with anti-EGFR MoAbs.
Results A total of 7 studies were included in this meta-analysis. The 7 studies included 1352 patients in total, sample sizes ranged from 67 to 493. Objective response rate (ORR), progression-free survival (PFS) and overall survival (OS) were collected from included studies and were used to assess the strength of the relation. In patients with wild-type KRAS, the pooled odds ratio for ORR of mutant BRAF over wild-type BRAF was 0.27 (95%CI=0.10-0.70). BRAF mutation predicted a deterioration in PFS and OS in wild-type KRAS patients treated with anti-EGFR MoAbs (hazard ratio=2.78, 95% CI=1.62-4.76;hazard ratio=2.54, 95%CI=1.93-3.32).
Conclusion BRAF V600E mutation is related to lack of response and worse survival in wild-type KRAS metastatic colorectal cancer patients treated with anti-EGFR MoAbs.
9.Inducible and constitutive expression of fip-fve from Flammulina velutipes in Pichia pastoris GS115.

We transformed the fip-fve gene into Pichia pastoris GS115 for inducible and constitutive expression to obtain feasible bioactvie recombinant Fip-fve. The fip-fve gene was cloned from Flammulina velutipes fruting body by PCR and ligated to pPIC9 to construct inducible expression vector pPIC9-FIP-fve, and promotor pgap was used to replace the paox1 to construct constitutive expression vector pPIC9-PGAP-FIP-fve. These two vectors were used to transform P. pastoris by PEG method. The fip-fve was expressed after histamine-absence screening and yeast colony PCR. The inducible expression level reached 158.2 mg/L at the fourth day and the constitutive expression level was 46.3 mg/L and 29.5 mg/L using glucose and glycerol, respectively. The SDS-PAGE and Western blotting both proved the correctness of rFip-fve, and the hemagglutination test indicats the rFip-fve's bioactivity.

10.A quantitative study on ultrasonographic texture analysis of injured porcine striped muscle induced by thermal and chemical factors

Objective To quantitatively analyze and compare the texture features of thermal and chemical lesions on the porcine striated muscle, in vitro extracted from high-frequency ultrasonograms using computer-assisted image analysis technique, and to investigate the application values. Methods The thermal lesion and chemical lesion were induced in vitro in porcine striated muscle by microwave ablation and anhydrous acetic acid injection, respectively. The two dimension (2D) ultrasonographic ifndings were qualitatively compared between the groups of thermal and chemical lesion models, in which eight textural features in geometric mathematics extracted from 2D ultrasonograms were quantitatively analyzed by a technique of computer-assisted image analysis named multiscale decomposition method of echo intensity of interface relfections. Results As expected, microwave ablation and anhydrous acetic acid caused signiifcant changes of several texture features extracted from ultrasonograms. There were significant differences between the normal group and microwave ablation group in grayscale mean (Mean), irregularity (IRGL) and periodicity of distribution (POD) as follows (Mean: 1.9143±0.2914 vs 1.2334±0.3357, t=-5.306, P=0.000; IRGL: 0.5577±0.0334 vs 0.5092±0.0459, t=-2.957, P=0.007; POD: 0.000 27±0.000 005 vs 0.000 29±0.000 008, t=4.782, P=0.000). There were signiifcant differences between the normal group and anhydrous acetic acid injection group in number of blobs (NOB), size of blobs (SOB) and periodicity of distribution (POD) as follows (NOB: 51.0324±13.6998 vs 31.6042±4.8315, t=4.633, P=0.000; SOB:16.4843±3.9349 vs 25.6230±2.3555, t=6.903, P=0.000;POD:0.000 26±0.000 015 vs 0.000 29±0.000 008, t=-4.459, P=0.000). For each group of injured regions, there were significant differences between the microwave ablation group and anhydrous acetic acid injection group in Mean, IRGL, NOB and SOB as follows (Mean: 1.2664±0.2688 vs 1.9143±0.2914, t=-5.661, P=0.000; IRGL: 0.5220±0.0422 vs 0.5577±0.0334, t=-2.295, P=0.032;NOB:51.0324±13.6998 vs 34.5856±2.6362, t=4.048, P=0.000;SOB:16.4843±3.9349 vs 25.3176±2.3501, t=-6.676, P=0.000). Conclusion Technique of computer-assisted image analysis named multiscale decomposition method of echo intensity of interface relfections, based on multiscale blob features extraction, was useful to differentiate ultrasonic texture features between the groups injured in our study, which established quantitative muscle ultrasound as a practical and reliable tool for the muscle injury diagnosis to distinguish the structural changes induced by different physiochemical factors.
