Objective To explore the effect and mechanism of somatostatin and proton pump inhibitor (PPI) on Iks in pancreatic acini (RPA) by testing the change of slowly-activiting voltagedependent K+ channel current (Iks).Methods Pancreatic acini was isolated and its suspension was obtained.Iks values of different groups were recorded by an EPC10 patch clamp amplifier and the difference among the groups was compared.The groups were divided as followed:washing solution treated as control group,5 μmol/L or 20 μmol/L 293B groups,5 nmol/L secretin group,5 nmol/L secretin with somatostatin at 100 nmol/L or 10 nmol/L or 1 nmol/L groups,0.3 nmol/L 8Br-cAMP group,0.3 nmol/L 8Br-cAMP with 100 nmol/L somatostatin group,1 μmol/L acetylcholine group,1μmol/L acetylcholine with 100 nmol/L somatostatin group,5 nmol/L secretin with omeprazole at 10-5 mol/L or 10-6 mol/L or 10-7 mol/L groups,1 μmol/L acetylcholine with omeprazole at 10-5 mol/L or 10-6mol/L or 10-7 mol/L groups.Groups were compared by ANOVA,P ＜ 0.05 was considered statistically significant.Results The resting membrane voltage of rat RPA was - (40 ± 0.8) mV,and when depolarization to +10 mV,Iks of control group was (420.0±3.2) pA.After treated with 5 μmol/L or 20 μmol/L 293B,Iks of RPA decreased to (60.4±4.2)％ and (30.2±3.1)％(F=6.87,P＜0.05) of control group.Stimulated with 5 nmol/L secretin,Iks increased to (823.0±2.2) pA,and after adding somatostatin at 100 nmol/L or 10 nmol/L or 1 nmol/L,Iks decreased to (510.0±3.2) pA,(584.0±2.8) pA and (789.0±6.9) pA respectively(F=5.67,P＜0.05),after adding omeprazole at 10-5mol/L or 10-6mol/L or 107mol/L,the peak of Iks was (806.5±3.6) pA,(814.8±3.2) pA and (816.3±2.9) pA (P＞0.05).After stimulated with 1 μmol/L acetylcholine,the peak value of Iks was (966.0± 3.2) pA,and after adding omeprazole at 10-5 mol/L or 10-6 mol/L or 107mol/L,the peak of Iks was (956.3±10.3) pA,(957.5±8.6) pA and (960.0±8.4) pA (P＞0.05).Conclusion Somatostatin can inhibit Iks opening,and there is no significant inhibition of PPI on this channel.

OBJECTIVE:To observe the clinical efficacy and safety of Vitamin C tablets after grinding for local use in the treatment of oral ulcer.METHODS:Two hundved and eighty-six cases of oral ulcer in our hospital during Feb.2014-Fed.2016 were selected and divided into treatment group and control group according to random number tabel,with 143 cases in each group.Control group was given Guilin watermellon frost,2-3 times a day;treatment group was given Vitamin C tablets powder for local use,2-3 times a day.Clinical efficacy,the time of ulcer disappearance,the incidence of ADR,recurrence rate of ulcer,treatment satisfactory degree were observed in 2 groups.RESULTS:Total response rate of treatment group was 96.5 ％,which was significantly higher than 80.4％ of control group,with statistical significance (P＜0.05).The time of ulcer disappearance was (2.37 ± 0.89) d in treatment group,which was significantly shorter than (4.12 ± 1.33) d of control group;recurrence rate of ulcer was 3.5％,which was significantly lower than 8.4％ of control group;treatment satisfactory degree was 90.9％,which was significantly higher than 79.7％ of control group,with statistical significance (P＜0.05).There was no statistical significance in the incidence of ADR between 2 groups (P＞0.05).CONCLUSIONS:Vitamin C tablets after grinding for local use has obvious effect on oral ulcer,significantly shortens the time of ulcer disappearance,reduces recurrence rate,and improves treatment satisfactory degree with good safety.

The expressions of Ki67 and FHIT were detected by immunohistochemical staining in 15 cases of adrenocortical carcinoma, 42 cases with adrenocortical adenoma,6 cases of adrenocortical hyperplasia, and 10 cases of normal adrenocortical tissue. The results showed that the highest expression of Ki67 and the lowest expression of FHIT were found in adrenocortical carcinoma. There were significant differences in the Ki67 and FHIT between adrenocortical adenoma and adrenocortical carcinoma ( both P < 0. 05 ). There existed negative correlation between the expressions of Ki67 and FHIT( r=-0. 712, P<0.05 ). Ki67 over-expression and loss of FHIT expression may be involved in the occurrence and development of adrenocortical carcinoma. It is suggested that combined detections of Ki67 and FHIT may have reference significance in the differentiation of adrenocortical adenoma from adrenocortical carcinoma.

AIM: To explore the influence of herbs (9602 prescription) on brain energy metabolism in cerebral ischemia reperfusion mice.METHODS: Ischemia reperfusion in cerebral injury model was duplicated in mice. Using nuclear magnetic resonance (NMR) and high performance liquid chromatography (HPLC), the brain metabolism were measured. The influence of 9602 prescription on cerebral energy metabolism in ischemia reperfusion mice model was dynamicly observed. RESULTS: Phase Ⅰ: The spectrum of NMR showed that after 10 min of ischemia, the PCr peak dropped significantly, while the Pi peak rose significantly in both the control and the “9602” group. There was no remarkable difference between the two groups. After reperfusion the PCr peak in the control group continued dropping slowly and remained at a low level (55.50?14.94) after 10 min of reperfusion, while after reperfusion the fallen PCr peak in the “9602” group started rising till 76.72?13.37 (P0.05). Objective To explore the value of miniprobe sonography (MPS) in differentiating Crohn??s disease (CD) and ulcerative colitis (UC). Methods 46 patients with active inflammatory bowel disease (IBD) were included into the patient group (16 patients with CD and 30 cases with UC). 20 healthy cases ( underwent physical examination in outpatient department) were recruited as the control group. All cases underwent MPS and high sensitive C-reactive protein (Hs-CRP) test.The MPS measurement index include thetotalwallthickness(TWT), mucosal thickness (MT), submucosal thickness (SMT), muscularispropria thickness (MPT), and the number of enlarged colic and paracolic lymph nodes. Results TheTWT, M, SM, MP(mm)and Hs-CRP(mg/L)in CD, UC and control group are 5.84 ± 1.42, 1.48 ± 0.23, 1.88 ± 0.28, 2.31 ± 0.85, 40.58 ± 19.33, 4.74 ± 1.01, 1.79 ± 0.35, 1.41 ± 0.25, 1.32 ± 0.34, 22.41 ± 15.25,2.86 ± 0.23, 0.97 ± 0.13, 1.06 ± 0.11, 0.64 ± 0.0 and 1.70 ± 0.65. TWT, MT, SMT, MPT and Hs-CRP in UC group has significant different with those in controp group(P<0.05). The mean value of TWT, MT, SMT, MPT and Hs-CRP in CD group are higher than those in UC group. M in UC group is higher than that in CE group. The difference is significant(P<0.05). 5 cases in CD and 2 cases with UC had enlarged colic or paracolic lymph nodes. Conclusions The MPS can distinctly observe the changesof different colonic tissue layers and the surrounding tissue structures in IBD patients,which helps for distinguishing active CD from UC.
