1.Effect of somatostatin and proton pump inhibitor on the siowly-activiting voltage-dependent K+ channel in the rat pancreatic acini

Objective To explore the effect and mechanism of somatostatin and proton pump inhibitor (PPI) on Iks in pancreatic acini (RPA) by testing the change of slowly-activiting voltagedependent K+ channel current (Iks).Methods Pancreatic acini was isolated and its suspension was obtained.Iks values of different groups were recorded by an EPC10 patch clamp amplifier and the difference among the groups was compared.The groups were divided as followed:washing solution treated as control group,5 μmol/L or 20 μmol/L 293B groups,5 nmol/L secretin group,5 nmol/L secretin with somatostatin at 100 nmol/L or 10 nmol/L or 1 nmol/L groups,0.3 nmol/L 8Br-cAMP group,0.3 nmol/L 8Br-cAMP with 100 nmol/L somatostatin group,1 μmol/L acetylcholine group,1μmol/L acetylcholine with 100 nmol/L somatostatin group,5 nmol/L secretin with omeprazole at 10-5 mol/L or 10-6 mol/L or 10-7 mol/L groups,1 μmol/L acetylcholine with omeprazole at 10-5 mol/L or 10-6mol/L or 10-7 mol/L groups.Groups were compared by ANOVA,P < 0.05 was considered statistically significant.Results The resting membrane voltage of rat RPA was - (40 ± 0.8) mV,and when depolarization to +10 mV,Iks of control group was (420.0±3.2) pA.After treated with 5 μmol/L or 20 μmol/L 293B,Iks of RPA decreased to (60.4±4.2)% and (30.2±3.1)%(F=6.87,P<0.05) of control group.Stimulated with 5 nmol/L secretin,Iks increased to (823.0±2.2) pA,and after adding somatostatin at 100 nmol/L or 10 nmol/L or 1 nmol/L,Iks decreased to (510.0±3.2) pA,(584.0±2.8) pA and (789.0±6.9) pA respectively(F=5.67,P<0.05),after adding omeprazole at 10-5mol/L or 10-6mol/L or 107mol/L,the peak of Iks was (806.5±3.6) pA,(814.8±3.2) pA and (816.3±2.9) pA (P>0.05).After stimulated with 1 μmol/L acetylcholine,the peak value of Iks was (966.0± 3.2) pA,and after adding omeprazole at 10-5 mol/L or 10-6 mol/L or 107mol/L,the peak of Iks was (956.3±10.3) pA,(957.5±8.6) pA and (960.0±8.4) pA (P>0.05).Conclusion Somatostatin can inhibit Iks opening,and there is no significant inhibition of PPI on this channel.

5.Clinical Observation of Vitamine C Tablets for Local Use in the Treatment of Oral Ulcer after Grinding

OBJECTIVE:To observe the clinical efficacy and safety of Vitamin C tablets after grinding for local use in the treatment of oral ulcer.METHODS:Two hundved and eighty-six cases of oral ulcer in our hospital during Feb.2014-Fed.2016 were selected and divided into treatment group and control group according to random number tabel,with 143 cases in each group.Control group was given Guilin watermellon frost,2-3 times a day;treatment group was given Vitamin C tablets powder for local use,2-3 times a day.Clinical efficacy,the time of ulcer disappearance,the incidence of ADR,recurrence rate of ulcer,treatment satisfactory degree were observed in 2 groups.RESULTS:Total response rate of treatment group was 96.5 %,which was significantly higher than 80.4% of control group,with statistical significance (P<0.05).The time of ulcer disappearance was (2.37 ± 0.89) d in treatment group,which was significantly shorter than (4.12 ± 1.33) d of control group;recurrence rate of ulcer was 3.5%,which was significantly lower than 8.4% of control group;treatment satisfactory degree was 90.9%,which was significantly higher than 79.7% of control group,with statistical significance (P<0.05).There was no statistical significance in the incidence of ADR between 2 groups (P>0.05).CONCLUSIONS:Vitamin C tablets after grinding for local use has obvious effect on oral ulcer,significantly shortens the time of ulcer disappearance,reduces recurrence rate,and improves treatment satisfactory degree with good safety.
6.Effect of individual psychological and action factor intervention on blood pressure of resistant hypertension patients

Objective To research the effect of individual psychological and action factor intervention on blood pressure of resistant hypertension.Methods There are 120 resistant hypertension patients selected in the hospital,according to hospital admission number,they were graded into control group and experimental group randomly.The control group accept the a common nurse intervene,the experimental group accept the individuation nurse intervene mentality action factor,and other therapies were the same in two groups.Compare blood pressure level before and after nurse intervene and after discharge in two groups.Results Before the intervertion,the blood pressure level was no significant difference between two groups (P > 0.05 ).After the intervention and after discharge,MAP were respectively ( 108.00 ±0.8),( 106.00 ±0.7) mm Hg in experimental group,lower than in control group ( 125.33 ± 0.9),( 120.00 ± 0.8 ) mm Hg ( t =108.31,116.67 ;P <0.05) ; SBP and DBP in two groups ater the intervention and after discharge there were significant difference (P < 0.05 ).Conclusions The individuation nurse intervene mentality action factor can efficacious correct badness mentality action factor of resistant hypertension,improve control rate of high blood pressure.and help patients get well,recede rate of deformity and death in cordis and vasculardisease.
7.Effect of 9602 prescription on brain energy metabolism in cerebral ischemia reperfusion mice

AIM: To explore the influence of herbs (9602 prescription) on brain energy metabolism in cerebral ischemia reperfusion mice.METHODS: Ischemia reperfusion in cerebral injury model was duplicated in mice. Using nuclear magnetic resonance (NMR) and high performance liquid chromatography (HPLC), the brain metabolism were measured. The influence of 9602 prescription on cerebral energy metabolism in ischemia reperfusion mice model was dynamicly observed. RESULTS: Phase Ⅰ: The spectrum of NMR showed that after 10 min of ischemia, the PCr peak dropped significantly, while the Pi peak rose significantly in both the control and the “9602” group. There was no remarkable difference between the two groups. After reperfusion the PCr peak in the control group continued dropping slowly and remained at a low level (55.50?14.94) after 10 min of reperfusion, while after reperfusion the fallen PCr peak in the “9602” group started rising till 76.72?13.37 (P0.05). AIM: To investigate the effect of advanced glycosylation end products on the expression of receptor for advanced glycosylation end products in human monocyte-derived dendritic cells. METHODS: Monocytes were purified (over 98%) using anti-CD14+ microbeads. After 8 d culture in RPMI-1640 medium containing rhGM-CSF (100 ?g/L) and rhIL-4 (50 ?g/L), immature MDCs were derived, then exposed to AGE-BSA (0 or 200 mg/L) for 24 h. Expression of RAGE was semi-quantified by RT-PCR and Western blotting. At the same time, supernatants were collected. IFN-? and IL-12 were analyzed by ELISA. Objective To investigate the xenotransplantation model of fetal pig skin precursor tissue and its development after transplantaion. Methods Porcine skin precursor tissue was obtained from the embryo of gestation day 56 (E56), and made into microskin or stamp skin graft. The microskin was transplanted to the dorsal wound in BALB/c nude mice, then covered with human corpse skin. The stamp skin graft was imbedded subcutaneously into the back of nude mice, and microskin was injected subcutaneously into the auricles of nude mice. Their growth and development were observed and they were examined by HE staining at 6th and 12th week after transplantation respectively. Two-sample t test was used to analyze the size of newly grown skin tissue. Results Porcine skin precursor tissue graft in three models above survived and continued growth after transplantation, and growth ability of the dorsal wound transplantation model was significantly stronger than that of the auricle model. Epidermis and hypodermis were detected in newly grown skin tissues. Hair follicles, a few of sebaceous glands, but no sweat glands were observed in auricle model, while many sebaceous glands and sweat glands were observed in the dorsal wound model. Conclusion Transplantation of microskin to dorsal wound is the optimal model of investigating the xenotransplantation of fetal pig skin precursor tissue and its development after transplantion.

18 fungal strains including N.coenophialum,N.lolii, N.huerfanum、N.chisosum、N.aotearoae、N.sp.and 8 varieties of grass seeds belonging to Festuca arundinacea and Lolium perenne have been studied.With amplification of IS1~IS3 and F1~R1 of genomic DNA, the primers Tub-2-F~Tub-2-R from Tubulin-2 gene and F3~R3 from NC25 gene have been designed.A PCR method to detect N.coenophialum and N.lolii was established, and also a nested-PCR method to detect N.coenophialum and N.lolii in single seed was established.These PCR detection methods are strongly special and much credible and rapid-speeded.
