Objective To explore the effect and mechanism of somatostatin and proton pump inhibitor (PPI) on Iks in pancreatic acini (RPA) by testing the change of slowly-activiting voltagedependent K+ channel current (Iks). Methods Pancreatic acini was isolated and its suspension was obtained.Iks values of different groups were recorded by an EPC10 patch clamp amplifier and the difference among the groups was compared. The groups were divided as followed:washing solution treated as control group,5 μmol/L or 20 μmol/L 293B groups,5 nmol/L secretin group,5 nmol/L secretin with somatostatin at 100 nmol/L or 10 nmol/L or 1 nmol/L groups,0.3 nmol/L 8Br-cAMP group,0.3 nmol/L 8Br-cAMP with 100 nmol/L somatostatin group,1 μmol/L acetylcholine group,1μmol/L acetylcholine with 100 nmol/L somatostatin group,5 nmol/L secretin with omeprazole at 10-5 mol/L or 10-6 mol/L or 10-7 mol/L groups,1 μmol/L acetylcholine with omeprazole at 10-5 mol/L or 10-6mol/L or 10-7 mol/L groups.Groups were compared by ANOVA,P < 0.05 was considered statistically significant. Results The resting membrane voltage of rat RPA was - (40 ± 0.8) mV,and when depolarization to +10 mV,Iks of control group was (420.0±3.2) pA.After treated with 5 μmol/L or 20 μmol/L 293B,Iks of RPA decreased to (60.4±4.2)% and (30.2±3.1)%(F=6.87,P<0.05) of control group.Stimulated with 5 nmol/L secretin,Iks increased to (823.0±2.2) pA,and after adding somatostatin at 100 nmol/L or 10 nmol/L or 1 nmol/L,Iks decreased to (510.0±3.2) pA,(584.0±2.8) pA and (789.0±6.9) pA respectively(F=5.67,P<0.05),after adding omeprazole at 10-5mol/L or 10-6mol/L or 107mol/L,the peak of Iks was (806.5±3.6) pA,(814.8±3.2) pA and (816.3±2.9) pA (P>0.05).After stimulated with 1 μmol/L acetylcholine, the peak value of Iks was (966.0± 3.2) pA,and after adding omeprazole at 10-5 mol/L or 10-6 mol/L or 107mol/L,the peak of Iks was (956.3±10.3) pA,(957.5±8.6) pA and (960.0±8.4) pA (P>0.05).Conclusion Somatostatin can inhibit Iks opening, and there is no significant inhibition of PPI on this channel.

Objective To analyze incidence of thyroid carcinoma and to explore and conclude the principle of its diagnosis and treatment. Methods One hundred and forty six patients of thyroid carcinoma in our hospital were analyzed and discussed with literature. Results 101 cases were diagnosed as thyroid carcinoma by pathological diagnosis of fast frozen sections, the positive ratio is approximately 93.5%.totle resection of affected lobe and isthmus was performed on 35 cases, total resection of affected lobe and isthmus and majority of opposite lobe was done on 37 cases. The lymph node metastasis was found by postoperative pathological diagnosis with a rate of 32.5%.Conclusions Preoperative diagnose of thyroid carcinoma is rarely available, diagnosis of thyroid carcinoma by fast frozen sections is optimal.total resection of affected lobe and isthmus or total resection of affected lobe and isthmus and majority of opposite lobe are the main operation patterns.

Chinese Traditional and Herbal Drugs 2016;47(15):2727-2733
Objective: To screen the reference genes of Lonicera macranthoides for gene expression analysis and to study the spatio-temporal expression characteristics of LmAGL15 which was a member of Mads-Box family. Methods: In this study, 18 S rRNA, Ubiquilin, Actin and Efl-β of L. macranthoides were cloned and the stabilities of the four housekeeping genes were evaluated in different positions (leaves, stems, and buds) and different periods of bud development. In addition, the spatio-temporal expression of LmAGL15 gene was analyzed. Results: 18 S rRNA was the most suitable reference gene for spatio-temporal expression analysis in L. macranthoides; The relative expression of LmAGL15 was low in leaves and stems, and that in buds was higher. Conclusion: 18 S rRNA is the most suitable reference gene in L. macranthoides. The relative expression of LmAGL15 changes significantly in leaves, stems, and buds.

[Objective] To summarize image manifestations in intestinal tuberculosis (ITB) examined by multi-slice computed tomography enteroclysis (MSCT) and evaluate the diagnostic value of MSCTE in ITB,[Methods] The imaging findings were retrospectively analyzed in 15 cases of ITB that were confirmed by endoscopy or postoperative pathologic examination, including the location, number, shape, edge, surrounding tissue alterations of ITB and other associated changes in the peritoneum, mesentery and solid abdominal organs, and compared with endoscopy and pathology.[Results] In 15 patients, ileocecum was involved in 13 cases (87%),4 cases (27%) showed multi-segmental symmetric intestinal mural thickening,9 patients (60%) showed solid masses,1 case (7%) showed multi-segmental symmetric intestinal mural thickening and solid masses,1 case (7%) showed homogenous enhanced masses and perforation,12 cases (86%) showed enlarged lymph nodes (LNs) with rim enhancement, and 2 cases (13%) showed the comb sign of enhanced mesenteric vessels.Based on enhancement pattern of MSCTE,ITB was divided into three types:homogenous enhancement type (n=5);target sign type (n=2);caseous necrosis type (n=2).[Conclusion] The imaging features of ITB are diverse.MSCTE can clearly display the shape of intestinal mucosa, the alterations of intestinal wall and the relationship between lesion and adjacent tissues, which provides valuable information for the clinic diagnosis of ITB.

Objective To elucidate whether and how injection site diversity influences on the sodium hyaluronate (HA) distribution on cartilage surface of the knee joint. Methods From September 2014 to December 2014, 32 human cadaveric knees were sorted from small to large based on femoral condyle width (FCW) and divided into group Ⅰ (n=16, odd number, the patella medial approach) and group Ⅱ (n=16, even number, injected through the medial joint line approach).Knees of both groups were interfused with a 5% (mg: ml) methylene blue tracer. Each specimen was then simulated a 5°-45° alternating walking for 2 h in the Electro Force platform.Afterwards, all the knee cavities were cut open to examine the range of HA distribution.Based on the eight-zoning classification law for the knee cartilage surface, the coverage area of HA in each zone was scored and the characteristics of HA distribution was depicted, as well as HA zonal distribution diversity between groups were statistically analyzed. Results HA of both patella medial and medial joint line subgroups showed analogical distribution in all zones except the lateral tibial plateau at the time of FCW ≤ 7.0 cm.However, HA coverage through the patella medial approach showed significantly higher distribution scores in the patella zone and the femoral trochlear zone, but significant lower distribution scores in the antero-lateral/postero-lateral femoral condyle, the medial posterior femoral condyle zone and the lateral tibial plateau zone compared with medial joint line approach at the time of FCW > 7.0 cm.Conclusion When delivered through different injection approaches, HA showed analogical distribution characteristics in most zones of the knee cavity in population with a small bodily shape (FCW ≤ 7.0cm), but significant diverse distribution characteristics in population with a big bodily shape (FCW >7.0 cm). When FCW > 7.0cm, HA injected through the medial-patellar approach tended to be distributed mainly over the patella-femoral articulation and the anterior knee cavity, while HA injected through the joint line approach tended to be distributed mainly over the tibio-femoral articulation and the posterior knee cavity.
