2.Biological treatment for intervertebral disc degeneration.

Neck shoulder pain or lumbocrural pain caused by intervertebral disc degeneration (IDD) could seriously affect the qualities life of patients. Current treatments mainly focus on alleviating pain and the symptoms of nerve compression, which could not radically stop the process of intervertebral disc degeneration, but conversely lead to high recurrence rate. In recent years, scholars have turned to study the biological treatment for repair and rebuild the intervertebral disc by biological molecular therapy, gene therapy, cell therapy and tissue engineering to solve the problem of intervertebral disc degeneration, while most of the above methods are still in animal experiments or in vitro experiments and the clinical application is still a long way to go.

3.Relationship of TAP-1 allele gene polymorphism to recurrent condyloma acuminatum

Objective To investigate the relationship of polymorphisms at codons 333 and 637 of TAP-1 allele gene to recurrent condyloma acuminatum (CA) in a Chinese population. Methods Amplificatory refraction mutation system-PCR (ARMS-PCR) was performed to detect TAP1 polymorphic residues at codon 333 in 88 patients with recurrent CA and 81 age- and sampling date-matched controls and at codon 637 in 60 patients with recurrent CA and 60 age- and sampling date-matched controls. Results The frequencies of AA,GG and AG genotypes at codon 333 of TAP-1 gene were 86.36%, 0, 13.64%, respectively, in patients with recurrent CA, 79.01%, 0 and 20.99%, respectively, in the controls, and no significant difference was observed between the two groups (χ2 = 1.604, P > 0.05). However, a significant difference was observed in the frequencies of AA, GG and AG genotypes at codon 637 of TAP-1 gene between the patients and controls (3.33% vs 10.0%, 95.00% vs 60.00%, 1.67% vs 30.00%, χ2 = 21.551, P < 0.01 ). Conclusions The recurrence of CA may be associated with the polymorphism at codon 637, but not with that at codon 333, of TAP-1 gene.
4.The expression of P-glycoprotein in breast carcinoma

Objective To study P-glycoprotein(P-gp),the mdr-1 gene product,expression and response to pirarubicin containing regimen in recurrent and metastatic breast carcinoma.Methods P-gp expression in 40 postoperative patients with recurrent and metastatic breast carcinoma was detected by SABC immunohistochemical method.The patients were treated with combination regimen of cyclophosphamide,pirarubicin and 5-fluorouracil.The correlation between P-gp expression and chemotherapeutic response was analyzed.Results The positive rate of P-gp expression in patients with lung or liver metastasis was higher than that in skin or lymphnode metastasis(P=0.049);The overall response rate was 52.5% in 40 patients;The responserate 84.2% of the P-gp negative group was higher than that 33.3% of the P-gp positive group(P
5.Determination of ginsenoside R_e and R_(g1) in Compound Jiangtang Oral Liquid by HPLC

AIM: To establish the determination of ginsenoside R e and R g1 in Compound Jiangtang Oral Liquid(Radix Ginseng, Fructus Schisandrae Chinensis, Radix Rehmannia, Radix Asparagi, etc.) by HPLC. METHODS: C 18 ODS was used as a stationary phase, acetonitrile-0.05% phosphoric acid (96∶400) as a mobile phase and detection wavelength at 203nm. RESULTS: The linear range of ginsenoside R e concentration was from 0.66 to 3.29?g?mL -1 and correlation coefficient was 0.9991, the linear range of ginsenoside R g1 concentration was from 1.05 to 5.27?g?mL -1 and correlation coefficient was 0.9998. The average recovery of sample was 98.2% and RSD 2.2% ( n =5), respectively. CONCLUSION: The method is simple and convenient, accurate with a good reproducibility and can be used for determination of ginsenoside R e and R g1 in Compound Jiangtang Oral Liquid.
6.Discussion on the effective approaches in cultivating the graduate students' innovations from their own viewpoints

Objective To analyze the viewpoints of the graduate students on the effective ap-proaches in cultivating their innovations. Methods Using Eugenie Creativity Test scale and self-made questionnalre, the graduate students in a medical university were surveyed on their general informa-tion, innovation levels and viewpoints of the important links and effective approaches in cultivating the innovations. The statistical software package-SPSS 16.0 was used for questionnalres. The statistical analysis was based on chi-square test for categorical data in the levels of graduate students' innovation and variance analysis for quantitative data in graduate students' understanding of the importance of cultivating approaches. Results 1 000 questionnalres were given out and effective questionnalre 936 copies were collected. The innovations of the students were malnly medium (557/936, 59.6%) and strong (356/936, 38.0%). They thought research and practice were the most important (F=225.49, P=0.00) in the tralning links and strengthening the tutorial staff was the foremost in the cultivating ap-proaches, which was followed by the sorted tralning (F=50.36, P=0.00). Conclusion The views of the graduate students on the cultivation jobs should be pald attention to. According to this study results, the management of research and practice, tutorial staff and assorted tralning should be fully considered to advance their innovations.
7.Comparison of MRI manifestations and histopathologic findings of the elderly carotid arteries in ex vivo

ObjectiveTo evaluate the ability of high resolution magnetic resonance (MR) imaging at 3.0 tesla to depict the characterization of human carotid arterial vessel wall and detect atherosclerotic lesions ex vivo in comparison to histopathologic results. MethodsEighteen carotid arteries obtained from 9 elderly donors underwent fat-suppressed T1 - and T2-weighted MR imaging at 3.0 tesla MR system with a mouse coil. Corresponding histological sections were obtained for the comparison. Correlation between MR images and histopathologic slices was obtained by Pearson or Spearman correlation coefficient.Cohen K was computed to quantify the agreement between MRI and histopathologic findings.Results Lumen area,intima and media area measured on fat-suppressed T2-weighted images showed stronger correlation with the corresponding histopathologic slices [ MRI vs.histopathology:( 27. 53 ± 6. 77 ) mm2 vs.( 25.83 ±6. 69 ) mm2, r2 = 0. 91,P2 = 0. 024, ( 12.31 ± 3. 31 ) mm2 vs.( 12. 28 ± 3.71 ) mm2, r2 = 0. 70, P2 = 0. 020,Median 12. 29 mm2(Min 1.12 mm2, Max 33.18 mm2) vs.Median 11.62 mm2(Min 0.89 mm2, Max 32. 84 mm2 ), r2 = 0. 74, P2 = 0. 016, respectively]. The Cohen K score between the MR imaging and American Heart Association classifications was 0. 74, which corresponds to a good agreement. Conclusions 3.0 T high-resolution multi-sequence MRI can clearly show the structure of ex vivo carotid artery wall and allow quantitative assessment.Fat-suppressed T2Wimaging has a greater advantagein presenting atherosclerotic lesions.
8.Influences of escitalopram on cognitive function and cardiac function in elderly patients with chronic heart failure complicated with depression disorder

Objective To explore the short-term influences of conventional internal medical treatment combined with escitalopram on cognitive function and cardiac function in elderly patients with chronic heart failure (CHF) complicated with depression disorder.Methods A total of 97 patients with CHF complicated with depression disorder in Chongqing Ninth People's Hospital,from October 2014 to October 2015,were selected and randomly divided into the antidepressant group and control group.Both groups were undergoing conventional internal medical treatment.Additionally,patients in the antidepressant group were administrated with escitalopram,while patients in the control group were treated with placebo.The degree of depression and anxiety and cognitive function were assessed by using 24-item Hamilton depression scale (HAMD-24),14-item Hamilton anxiety scale (HAMA-14) and Montreal cognitive assessment(MoCA) scale,and the plasma level of NT-proBNP,left ventricular end-diastolic dimension (LVEDD) and left ventricular ejection fraction (LVEF) were measured before and after 6-week treatment.Results After 6-week treatment,the HAMD-24 and HAMA-14 scores and plasma level of NT-proBNP in antidepressant group were lower than those in the control group,while the attention score and LVEF were greater than those in the control group,there were statistically significant differences(P<0.05).After 6-week treatment,no statistically significant difference was found in other observed indicators between the two groups(P>0.05).Conclnsion For elderly patients with chronic heart failure complicated with depression disorder,it is indicated that escitalopram could not only relieve their anxiety and depression,but also improve their cardiac function and attention.
9.Analysis of Extra-cardiac Findings by Coronary Computed Tomography Angiography in Patients With Suspected Coronary Artery Disease

Objective: To quantitatively study the incidental extra-cardiac ifndings (ECFs) by coronary computed tomography angiography (CCTA) in patients with suspected coronary artery disease (CAD) in order to better recognize those lesions in clinical practice.
Methods: A total of 1169 suspected CAD patients received CCTA in our hospital from 2011-06 to 2013-03 and 1030 patients were enrolled in this study. There were 589 in-patients, 441 out-patients and 549 patients≥60 years of age,481 patients < 60 years of age. 3 physicians evaluated the incidental ECFs in the full ifeld of view (FOV) in different window level and window width for lung, mediastinum, thorax and upper abdominal areas. Clinical relevance of ECFs were classiifed by corresponding scores. Score 1, the patients with severe lesion need immediate treatment, score 2, the lesion with clinical and prognostic signiifcance and score 3, the ifnding without clinical signiifcance.
Results: There were 197/1030 (19.1%) patients having 224 ECFs and 27 (2.6%) patients having 2 ECFs; 90/1030 (8.7%) patients having 106 signiifcant lesions including 3 (0.3%) of lung cancer and 8 (0.8%) of pulmonary embolism; 107 patients with 118 lesions without signiifcance. ECFs were found in 114/589 (19.4%) in-patients and in 83/441 (18.8%) out-patients, P>0.05; 76/481 (15.8%) of patients < 60 years of age and 121/549 (22.0%) of patients≥60 years,P<0.05.
Conclusion: Unexpected ECFs detection rate was 19.1% in patients undergoing CCTA without further radiation exposure by reconstruction with the full FOV setting, and 8.7% of ECFs had clinical signiifcance. Radiologists should routinely analyze the extra-cardiac organs in CCTA.
10.Construction of Recombinant Plasmid of Human RIP3 Gene and Its Effects on the Death of Breast Cancer MCF7 Cells

Objective To construct the recombinant RIP3 over-expressed plasmids and transfect them in breast cancer MCF7 cells, and identify the expression and localization of fusion protein, as well as its effect on the death way of MCF7 cells. Methods The expression levels of RIP3 mRNA in four breast cancer cell lines and normal mammary epithelial cells were detected by reverse transcription polymerase chain reaction (RT-PCR). The RIP 3 coding sequence was amplified by polymerase chain reaction and subcloned into mCherry vector to construct recombinant plasmids. The plasmids were transfected into MCF7 cells by lentivirus after DNA sequencing, then screened by basticidin (4 mg/L) for 1 week. The efficiencies of RIP3 expression were validated by Western blotting assay. The death way of mCherry-RIP3-MCF7 cells was observed under the treatment of TNF-αand Z-VAD-FMK. Results The lowest expression of RIP3 mRNA was found in MCF7 cells. The sequencing results validated the well recombinant plasmids. The expression of mCherry-RIP3 fusion pro-tein with a molecular weight of 85 ku was detected by Western blot assay. The mCherry-RIP3 expression enhanced the sensi-tivity of MCF7 cells to TNF-αand Z-VAD-FMK induced cell death. Conclusion The recombinant RIP3 over-expressed plasmids were successfully constructed, and the stable MCF7 cells with ectopic RIP3 transfection were obtained. The mCher-ry-RIP3 fusion protein was expressed in the cytoplasm and was conformed to mediate TNF-αinduced necroptosis.
