1.Determination of ginsenoside R_e and R_(g1) in Compound Jiangtang Oral Liquid by HPLC

AIM: To establish the determination of ginsenoside R e and R g1 in Compound Jiangtang Oral Liquid(Radix Ginseng, Fructus Schisandrae Chinensis, Radix Rehmannia, Radix Asparagi, etc.) by HPLC. METHODS: C 18 ODS was used as a stationary phase, acetonitrile-0.05% phosphoric acid (96∶400) as a mobile phase and detection wavelength at 203nm. RESULTS: The linear range of ginsenoside R e concentration was from 0.66 to 3.29?g?mL -1 and correlation coefficient was 0.9991, the linear range of ginsenoside R g1 concentration was from 1.05 to 5.27?g?mL -1 and correlation coefficient was 0.9998. The average recovery of sample was 98.2% and RSD 2.2% ( n =5), respectively. CONCLUSION: The method is simple and convenient, accurate with a good reproducibility and can be used for determination of ginsenoside R e and R g1 in Compound Jiangtang Oral Liquid.
2.Relationship between serum apelin level in patients with type 2 diabetes mellitus and microalbuminuria

Objective To explore the role of apelin in the pathogenesis of diabetic nephropathy.Methods A total of one hundred and fourteen patients with type 2 diabetes mellitus in Qingdao Municipal Hospital from July 2015 to June 2016 were divided into normal albuminuria group (56 cases in the NA group) and micro-albuminuria group (58 cases in the MA group),depending on whether the level of urinary micro-albumin in 24 h was higher than 30 mg,with 60 healthy subjects included as the controls group (NC group).Levels of serum apelin and other biochemical indexes were tested.Results (1) WC,BMI,TC,TG,LDL-C,FBG,FINS,HOMA-IR and apelin in the patients of the NA group and MA group were respectively (87.99±10.27) cm,(24.75±3.35) kg/m2,(5.27±1.28) mmol/L,(1.84±1.25) mmol/L,(3.36±0.91) mmol/L,(7.58±0.87) mmol/L,(13.29±3.57) U/L,4.50±1.41,(364.22±85.03) ng/L ,(90.10±8.97) cm,(25.47±2.82) kg/m2,(5.19±1.11) mmol/L,(2.23±1.43) mmol/L,(3.68±0.83) mmol/L,(7.89±1.11) mmol/L,(15.50±5.00) U/L,5.55±2.27,(397.42±91.29) ng/L,which were higher than those of the NC group ((83.20±5.36) cm,(22.59±2.67) kg/m2,(4.68±1.10) mmol/L,(1.37±0.58) mmol/L,(2.56±0.94) mmol/L,(5.11±0.82) mmol/L,(7.17±2.80) U/L,1.65±0.77,(309.34±68.28) ng/L,P<0.05).FINS,HOMA-IR and apelin in the MA group were significantly higher than those in the NA group (P<0.05).(2) After age,sex and BMI were taken into control,partial correlate analysis showed that serum apelin was positively correlated with LDL-C,FPG,FINS and HOMA-IR (r=0.183,0.314,0.374,0.378,P<0.05).(3) Multifactor regression analysis showed that HOMA-IR and BMI were independent related factors to serum apelin (r2=0.288,0.389,P<0.05).Conclusion Apelin levels in the MA group were significantly higher than those in the NA group,and the serum apelin was positively correlated with HOMA-IR.Thus,apelin may contribute to the pathogenesis and progress of diabetic nephropathy.
3.Studies on Th2 Type Cytokines in Patients with Neurocysticercosis

Objective To explore the level of Th2 type cytokines including IL-4, IL-10 and IL-13, and the immunoreg-ulation of cytokines in patients with neurocysticercosis. Methods Lymphocytes in patients with neurocysticercosis were separated from the blood sample with density gradient centrifugation and the total RNA was extracted by guanidine isothiocynate method. cDNA was synthesized by reversed transcription reaction. The target gene was then amplified by PCR. The PCR products were analyzed by electrophoresis. Results Results of RT-PCR showed that cytokines mRNA in lymphocytes of peripheral blcod were detected in 27 patients with neurocysticercosis but not in the other 3 cases. Among the positive cases, mRNA of IL-4, IL-10 and IL-13 was detected in 16, 17 and 14 cases respectively and mRNA of IL-4, IL-10 and/or IL-13 was detected in all the 27 cases. In the detection of lymphocytes in peripheral blcod of 10 healthy subjects, expression of IL-4 and IL-10 was found in only one case at low level. Conclusion The study revealed that Th2 associated cytokines were expressed at high level and the humoral immunocompetence was relatively strong in patients with neurocysticercosis.

5.CT Diagnosis of Thymoma

Objective To study CT features of thymoma,so that to improve the accuracy of CT diagnosis and differential diagnosis.Methods 31 cases of thymomas proved by surgery and pathology were examined with conventional CT scans.CT findings of thymoma were analyzed.Results The lesions in 27 cases(87.1%)were located in the anterior upper and middle mediastinum.There were benign lesion in 11,including mass-cardiovascular interface(MCI) with convex type(8 cases),flatness type(1 cases) and concave type(2 cases).20 cases were malignant lesion,including MCI with cast type(18 cases) and concave type(2 cases).Irregular invasion to adjacent organs was found in 11 cases,others included pericardiac effusion(n=6),pericardial and mediastinal invasion(n=2),pleural effusion(n=4),pneumonia(n=2),lung,bone,mediastinal lymphadens metastasis(n=2) and liver,pancreas metastasis(n=1).Conclusion CT scans is of significant value in diagnosis and differential diagnosis of thymoma.
6.Experimental study on arsenic trioxide-induced apoptosis of human pan creatic cancer cell line

Objective: To study the effect of arsenic trioxide (As2O3) on human pancreatic cancer cell proliferation and apoptosis (mainly early stage) in vitro. Methods: SW1990 cells line were trea ted with As2O3 at different concentration. Cell proliferation was evaluated by MTT and apoptosis by Annexin-Ⅴ-fluostaining, electron-microscopy, flow cytometry and immunocytochemical staining of Bcl-2 and Bax. Results: As2O3 and cisplatin had the same cytotoxity on SW1990. The cytotoxic effe ct on tumor cell was produced by induction of apoptosis. Twelve hours after cult ure with 10 μg/ml As2O3, much more SW1990 cells went into apoptosis than t he control. The apoptosis rate reached 24% after 48 h with the similar concentra tion of As2O3. Immunohistochemical study revealed that the expression of Bcl -2 was decreased after treated with As2O3. Conclusion: As 2O3 can depress the proliferation of SW1990 in vitro, mainly through the i nduction of apoptosis, and it is a potential agent for pancreatic cancer chemoth erapy.
7.Cross-sectional study of the clinic behavior and therapeutic status of rheumatoid arthritis patients

Objective To investigate and analyze the behavior and therapeutic status of rheumatoid arthritis (RA) patients. Methods Out patients diagnosed with rheumatoid arthritis in our hospital from May to August 2007 were enrolled. The data including sex, age of onset, site of first hospitalization and medication status were collected and analyzed. Results In this 181 RA patients, the mean age of onset was (53±11) year-old, mean history duration was (10±8) years, the ratio of male to female was 1:4.2. The orthopedics department was the most common site of first hospital visit (32.0%, 58/181) and rheumatology department was the most common site to clarify the diagnosis (62.4% ,113/181). The diagnosis delay caused by patients themselves was (5.9±17.2) months and the delay caused by doctors was (9.0±22.0) months. More than half of the patients were not treated appropriately before they came to our hospital. Methotrexate was the most commonly used DMARDs (67.3%), followed by leflunomide (46.4%), sulfasalazine (37.5%) and hydroxyehloroquine (19.6%). Conclusion In this cohort, the proportion of patients who come to rheumatology department immediately after disease onset is low. There is delay between symptoms and final diagnosis. More than half of the patients are not treated appropriately.
8.The molecular epidemiology and infections risk factors of clinical linezolid-resistant Enterococci isolates

Objective To investigate the molecular epidemiology and infectious risk factors of linezolid-resistant Enterococci (LRE) isolates in the First Affiliated Hospital of Chongqing Medical University.Methods Thirteen LRE isolates were collected from 2011 to 2013 and confirmed by broth dilution susceptibility testing.The minimum inhibitory concentrations (MIC) of twelve antimicrobial agents were analyzed using Vitek 2 compact.The molecular epidemiology of LRE isolates was determined by multilocus sequence typing (MLST), pulsed-field gel electrophoresis (PFGE) and the Diversilab.A casecontrol study was conducted for the analysis of risk factors, and Logistic regression was performed to evaluate the independent risk factors.Results All thirteen LRE isolates showed low-level resistance to linezolid, and most of these isolates were resistant to tetracycline, erythromycin and ciprofloxacin.However, they had high sensitivity to penicillin, ampicillin and tigecycline.Sequence type 480 was predominant in the hospital, and three isolates (isolates 3, 4, and 5) from July to September 2013 were found to have the same ST, PFGE pattern and rep-PCR group, indicating the same resistance clone.Admission to intensive care unit (ICU), peripheral vascular disease, males, hypoalbuminaemia, enema and linezolid therapy were identified as significant risk factors for LRE infections.Among these factors, admission to ICU, enema and linezolid therapy were independent risk factors for the acquisition of LRE.Conclusions Thirteen LRE isolates collected in the hospital showed a multidrug-resistant phenotype, and a small-scale prevalence was detected from 2011 to 2013.Therefore, attention should be paid to monitor the LRE in the hospital to decrease the prevalence of LRE infections.

Objective Through the comparative study of the acute toxic effects of PM2.5 in nickel-contaminated area in rats to explore the possible mechanism of acute toxicity of PM2.5.Methods A nickel-contaminated area and a control area were selected,the air quality of the two areas was monitored and the concentration of nickel in the collected PM2.5 was determined. Forty two male Wistar rats(SPF Grade)were randomly divided into three groups,including one saline control group and two experimental groups,the nickel-polluted group and the non-polluted group,in each.There were three subgroups.PM2.5 were administered to rats by intratraeheal instillation at doses of 1.6 mg/kg,8.0 mg/kg and 40.0 mg/kg respectively.The rats were sacrificed 24 hours after treatment and the brochoalveolar tavage fluid(BALF)were collected,the phagocytic function of alveola macrophages of rats were tested.Results There was no significant difference between the level of environmental quality in the two areas,the concentration of Ni in PM2.5 in the nickel-polluted area was 48.75 times of that in the non-polluted control area.The phagocytic function of alveola macrophages of rats in the experimental groups decreased significantly(P
10.The biological behavior and significance of aging bone mesenchymal stem cells

Bone marrow mesenchymal stem cells (BMSCs),which can be isolated from organs and tissues,are cell populations with high degree of plasticity,similar to hematopoietic stem cells in bone marrow,and can be in vitro cultured,induced and amplified.BMSCs are increasingly used in gene therapy,cell therapy and tissue engineering.BMSCs have been currently studied in a number of autologous transplantations,while not all BMSCs are suitable for autologous transplantation.BMSCs exhibit biological aging gradually when cultured in vitro.Biological aging can be divided into age-induced senescence and long-term passage induced senescence.Aging BMSCs demonstrate changes in biological behaviour which reduces the success rate of autologous transplantation.In this review,biological aging BMSCs are discussed in order to provide help for the transplantation of BMSCs.
