1.The clinical research on the treatment of female conyloma acuminatum by CO2 laser combined with traditional Chinese medicine

Objective To evaluate the efficacy of treating female conyloma acuminatum with the CO2 laser under colposcopy guidance and traditional Chinese medicine sticking therapy.Methods Female patients with conyloma acuminatum whose diagnosis were confirmed by pathologic examination,were randomly assigned to two groups:a treatment group of 100 cases and a control group of 80 cases,with no statistical differences of the age and the duration.The treatment group was treated by CO2 laser to remove conyloma acuminatum and subclinical damage,and then by TCM wart-washing decoction sticking after operation; The control group was treated by CO2 laser to remove the wart,and then by TCM wart-washing Decotion for fumigating after operation.Results The curative rate of the treatment group and the control group was 79.00%and 60.00% respectively.There was significant difference between the two groups (x2=7.723,P<0.01).Conclusion The treatment of CO2 laser under colposcopy guidance combined with Chinese medicine sticking therapy is effective for female conyloma acuminatum.
2.Method of information system risk analysis and its application

Based on computer and network technologies,the information system has grown into an important part for normal operation of hospitals,pushing up the importance of its risk analysis and defense.This article briefly described general methods and processes of risk analysis for information system,brought forward methods of risk classification and level-defense to identify and manage risks effectively,highlighted the importance of risk analysis,after-defense evaluating and tracking.This method is called into play to identify risks exposure of the information system of Beijing Red Cross Blood Center,contributing to a risk response table,developing a sustained improvement process for risk identification,control,appraisal and tracking,with defense carried out in specific system upgrading projects.

From Aug 2014 to July 2015, researches in the field of gonadal diseases have achieved rapid advances with image technology, the technology of biochemistry, and molecular biotechnology progressing. Considerable literatures concerning the pathogenesis, gene and clinical diagnosis, and treatment of gonadal diseases have been published. We reviewed and analyzed some more instructive literatures for clinical practice, proposed some suggestions, and the work plans for the clinicians.
4.Research advances in microRNAs in regulating hepatitis C virus replication and antiviral therapy

Hepatitis C virus (HCV)infection is one of the most common causes of chronic hepatitis,liver cirrhosis,and hepatocellular car-cinoma.MicroRNAs (miRNAs),a class of small,non -coding RNA,are involved in a variety of physiological and pathological processes in human bodies.The mechanism by which miRNAs regulate HCV replication is described,and the effects of liver -specific microRNA -122 antagonists on hepatitis C antiviral therapy are discussed.Our study indicates that miRNAs play an important regulatory role in HCV ex-pression.Targeting miRNAs may be a potential therapeutic approach for treating HCV infection,but further studies are still in need.
5.Effects of a small interfering RNA targeting HPV16E7 on proliferation and apoptosis of SiHa cells and expressions of six tumor suppressor genes

Objective To evaluate effects of human papilloma virus(HPV)16E7 on expressions of six tumor suppressor genes(including MT1G, NMES1, RRAD, SFRP1, SPARC and TFPI2)in a cell line SiHa, as well as on its proliferation and apoptosis. Methods SiHa cells were divided into two groups to be transfected with a small interfering RNA targeting HPV16E7(E7SiRNA, experimental group)and an empty vehicle(negative control group) respectively, with SiHa cells receiving no treatment serving as the blank control group. After 48 hours, qPCR was performed to measure the mRNA expressions of E7 and six tumor suppressor genes, CCK?8 assay to evaluate cellular proliferative activity, and flow cytometry to assess apoptosis of SiHa cells. Results At 48 hours after the transfection, the experimental group showed significantly decreased E7 mRNA expression(0.25 ± 0.036, P<0.05), but increased mRNA expressions of the six genes(MT1G 1.403 ± 0.190, NMES1 1.720 ± 0.060, RRAD 1.390 ± 0.160, SFRP1 1.493 ± 0.120, SPARC 2.157 ± 0.144, TFPI2 2.060 ± 0.122, all P < 0.05). The proliferative activity of SiHa cells was significantly decreased(0.554 ± 0.130 vs. 1.028 ± 0.236 and 1.220 ± 0.126, both P<0.05), but the apoptosis rate was significantly increased(9.222%vs. 0.246%and 0.123%, both P<0.05)in the experimental group compared with the negative control group and blank control group. No significant differences were observed between the negative control group and blank control group in proliferative activity or apoptosis rate of SiHa cells(both P>0.05). Conclusion E7 may participate in HPV16?induced cellular malignant transformation by suppressing the mRNA expressions of 6 tumor suppressor genes, including MT1G, NMES1, RRAD, SFRP1, SPAR and TFPI2.
6.Optimization of the Extraction and Preparation Process of Total Terpene Constituents in Platycladi Seed

Objective:To investigate the optimal extraction and dripping pills preparation of total terpene constituents in platycladi seed. Methods:The extraction processes of ultrasound, reflux, microwave and homogenization were investigated by single factor tests. The effects of extraction time, temperature, solid-liquid ratio and extraction times on the extraction of total terpene constituents in platy-cladi seed were investigated by orthogonal tests. Orthogonal tests were used to investigate the effects of the ratio of polyethylene glycol 6000 to poloxamer 188, the ratio of drug to matrix, the temperature of liquid and the dropping process on the quality of dropping pills. Results:The homogeneous extraction was used, and the optimum extraction conditions were as follows: the extraction time was 10 min, the temperature was 80℃, the solid-liquid ratio was 1 :10, and extraction time was once. The best preparation process of drop-ping pills was as follows:the ratio of polyethylene glycol 6000 to poloxamer 188 was 5 :1, the ratio of drug to matrix was 1 :3, the temperature of liquid was 80℃ and the dropping distance was 10 cm. Conclusion: The method of homogeneous extraction is simple, which can break the materials and extract the active ingredients simultaneously. The preparation process of dropping pills is reasonable and feasible, which is easy for the production and application.
7.Study and biomechanical evaluation of the device of gear-distraction plate for spine reduction and fixation

8.Clinical efficacy of ulinastatin combined with naloxone in patients with cardiogenic shock after acute myocardial infarction

Objective To study clinical efficacy of ulinastatin combined with naloxone in patients with cardiogenic shock(CS) after acute myocardial infarction (AMI).Methods Eighty patients with CS after AMI were randomly divided into routine treatment group (n =19),ulinastatin group (n =20),naloxone group (n =21) and ulinastatin combined with naloxone group (n =20).The levels of serum cardiac troponin I (cTnI),brain natriuretic peptide(BNP),tumor necrosis factor-α(TNF-α) and interleukin-6 (IL-6)were measured before and a week after treatment.In the meantime,recovery time of shock,the average hospitalization days and 28-day mortality rate were recorded.Results After the treatment,the levels of serum cTnI,BNP,TNF-α and IL-6decreased in all groups(P < 0.01),and there was significant difference on the decreasing degree of cTnI,BNP,TNF-α and IL-6 in ulinastatin combined with naloxone group when compared with those in routine treatment group,ulinastatin group and naloxone group(cTnI:(1.04 ± 0.17) ng/L vs.(2.06 ± 0.15) ng/L,(1.59 ± 0.16)ng/L,(1.97 ± 0.14) ng/L; BNP:(143.21-56.94) ng/L vs.(261.07 ± 71.43) ng/L,(203.46 ± 65.73) ng/L,(252.96 ± 68.85) ng/L; TNF-α:(13.42 ± 8.93) ng/L vs.(31.21 ± 12.32) ng/L,(20.39 ± 11.08) ng/L,(28.98 ± 11.76) ng/L ; IL-6:(37.58 ± 11.14) ng/L vs.(80.46 ± 27.15) ng/L,(59.84 ± 20.72) ng/L,(76.15 ±26.45) ng/L; P < 0.01).The recovery time of shock,the average hospitalization days and 28-day mortality rate in ulinastatin combined with naloxone group were significantly lower than those in routine treatment group,ulinastatin group and naloxone group(recovery time of shock:(7.16 ± 1.52) d vs.(11.43 ± 2.40) d,(8.05 ±1.81)d,(8.74 ± 1.98)d;the average hospitalization days:(15.03 ±3.23)d vs.(22.64 ±4.18)d,(18.93 ±3.97)d,(19.21 ±3.94)d ;28-day mortality rate:(41.62% vs.61.20%,50.74%,52.31% ; P <0.01)).Conclusion The application of ulinastatin combined with naloxone can effectively inhibit the cardiac injury and inflammatory response,promote the recovery of circulation function and improve prognosis in patients with CS after AMI.

The template-independent teminal transferase activity of Taq DNA polymerase results in an overhanging dA at the 3′end of its PCR products. The pGEMX vector constructed in this experiment forms a single overhanging dT at its 3′end as the result of cleavage with Xcm I restriction enzyme. This vector is very efficient for direct cloning of PCR product obtained by using Taq DNA polymerase.Recombinant colonies can be selected by Blue/white screening. Moreover,insertion fragment can be easily released from the vector simply with either BamH I or Hind III digestion.
