1.Reactive protein, plasminogen activator inhibitor type-1 (PAI-1) levels, PAI-1 promoter 4G/5G polymorphism and acute myocardial infarction

Objective To investigate the relationship between CRP, plasminogen activator inhibitor type 1 (PAI-1) levels, PAI-1 gene promoter 4G/5G polymorphism and the type of acute myocardial infarction (ST elevation myocardial infarction, STEMI vs the non-ST elevation Myocardial infarction, NSTEMI). Methods One hundred seventy-six consecutive patients with AMI were included for the study, of whom 60 had STEMI and 56 had NSTEMI, and 60 adults without cardiovascular and cerebrovascular disease were selected as controls. Blood samples were obtained from patients within 6 h of AMI and the plasma PAI-1, CRP, and the gene polymorphism were measured. Results Plasma levels of PAI-1 and CRP were higher in AMI groups, compared those in the control group, and plasma levels of PAI-1 were significantly higher in patients with STEMI compared to those with NSTEMI (80.12ng/ml VS.73.01ng/ml, P <0.01), while CRP levels were not significantly different between patient with STEMI and NSTEMI (3.87±0.79 mg/ml VS.4.01±0.69mg/ml, P>0.05). PAI-1 levels presented a significant correlation with CRP levels in the NSTEMI subjects. However, PAI-1 and CRP levels could explain the lack of a significant relationship between them in control and STEMI subjects.The frequencies of 4G/4G genotype in the AMI group were higher than those in the control group and higher in patient with STEMI than in patient with NSTEMI. Plasma levels of PAI-1 in subjects with 4G/4G genotype were significantly increased as compared to those in subjects with 4G/5G and 5G/5G genotype. Conclusions Plasma PAI-1 levels were associated with different myocardial infarction type, and PAI-1 promoter 4G/5G polymorphisms and CRP may be related to plasma PAI-1 levels.
2.Construction of Phage Display cDNA Library from Adult Worms of Schistosoma japonicum

Objective To screen protective antigen genes and construct the T7 phage display library from adult worms of Schistosoma japonicum.Methods Total RNA was extracted from adult worms of S.japonicum by Trizol reagent and mRNA was isolated from the total RNA.The ds cDNA was synthesized by reverse transcription using random primer.Directional EcoRⅠ/HindⅢ linkers were ligated into the ends of ds cDNA and the ds cDNA was digested with EcoRⅠand HindⅢ,which resulted in ds cDNA with EcoRⅠand HindⅢ adhering ends.The digested ds cDNA fragments longer than 300 bp in length were fractionated and ligated into T7 Select 10-3b vector.After packaging in vitro,the T7 Select 10-3b vector was transformed into BLT5403 to construct the T7 phage display cDNA library.Plaque assay and PCR were used to evaluate the library.Seven known objective genes of S.japonicum were screened by PCR to detect the representation of the library.Result Primary library capacity was 4.98?106 pfu,and the titer of amplified library was 3.85?1011 pfu/mL.The PCR identification result of 96 clones picked at random showed that recombination rate was 93.8%,in which 95.6% inserted cDNA fragments were longer than 300 bp in length.All the seven known objective genes of S.japonicum were amplified from the library.Conclusion The T7 phage display library from adult worms of Schistosoma japonicum was constructed.
3.Screening the target genes of Schistosoma japonicum related to the naturely resistance of Microtus fortis

Objective To look for the genes of Schistosoma japonicum related to the Schistosoma-resistance of Microtus fortis.Methods The fresh sera of Microtus fortis were used to screen a T7 phage display cDNA library from worms of Schistosoma japonicum established in our lab.The positive clones were sequenced and functionally analysed through bioinformatics.Results The specific phages binding to the sera of Microtus fortis were enriched 857-fold after three rounds of biopanning,and 58 positive clones picked at random were sequenced and 10 ESTs were obtained.BLASTn results showed that 7 ESTs had 99%-100% similarity to the genes of Shistosoma japonicum reported in GenBank and 1 EST had 82% similarity to a zinc finger protein encoden gene from Pan troglodytes.The results of these ESTs function prediction indicated most of them were involved in the regulation of gene expresion of Schistosoma japonicum.Conclusions Several target genes of Schistosoma japonicum related to the Schistosoma-resistance of Microtus fortis are obtained and those would lay foundation to expatiate the native resistance mechnism of Microtus fortis to Schistosoma japonicum.
4.Monitoring the progression of renal fibrosis by T2-weighted signal intensity and diffusion weighted magnetic resonance imaging in cisplatin induced rat models.

BACKGROUNDDiffusion weighted imaging (DWI), with the applying of intravoxel incoherent motion model, has showed promising results in obtaining additional information about microperfusion and tubular flow associated with morphologic changes in chronic kidney diseases. The study aims to evaluate the potential of T2-weighted signal intensity (SI) and DWI with mono- and bi-exponential models to reflect the serial changes on cisplatin (CP) induced rat renal fibrosis models.
METHODSMagnetic resonance exams were performed prior to and 2 nd day, 4 th day, 6 th day, 8 th day, 2 nd week, 3 rd week and 4 th week after CP injection at a 3.0T with an animal coil. Besides T2-weighted images (T2WI), DWI of 13 b values from 0 to 1500 s/mm 2 was acquired. Apparent diffusion coefficient (ADC), fluid fraction f, pure diffusivity D and pseudodiffusivity DFNx01 values were calculated. The regions of interest were placed on cortex (CO), outer stripe of the outer medulla (OM) and inner stripe of the outer medulla (OM), parameters were measured and compared among different time points. Five rats were scarified at each time point for pathological examination.
RESULTSOM revealed remarkable hyperintense and broadened before it became an obscure thread, while CO demonstrated moderate hyperintense and IM didn't show significant change on T2WI. On all three stripes, ADC values decreased firstly then kept increasing since the 4 th day; f values decreased on all stripes; D values had a tendency to increase with fluctuations but the changes didn't achieve statistical significance; DFNx01 values increased at the 2 nd day then tended to be steady thereafter. Pathological findings revealed tubules epitheliums swelling followed by inflammation cells infiltration, interstitial fibrosis was observed since the 2 nd week.
CONCLUSIONSAll of T2-weighted SI, ADC, and biexponential models parameters vary during fibrotic process; biexponential model is superior to monoexponential model in separating changes of microperfusion together with tubular flow from pure diffusion.

5.Association between adolescent internet addiction and suicidal behaviors

Objective To examine the association between internet addiction and suicidal behaviors in adolescences and to explore whether the association could be partly or fully accounted by depression or/and behaviors related to the impulsity. Methods A total of 3507 urban adolescent students in Hefei were administered to complete the questionnaire concerning interact addiction disorder(IAD), suicidal behaviors during the 12 months preceding the survey, behaviors related to the impulsity, Center for Epidemiologic Studies Depression Scale and early childhood adversities,respectively. Hierarchical logistic regressions were used to evaluate the associations and possible roles of depression and behaviors related to the impulsity between internet addition and suicidal behaviors after adjustment for confounding factors. Results Of the 3507 participants, 5.2% were diagnosed as IAD, 27.4% reported suicidal ideation during the 12 months preceding the survey, with another 9.5% had a plan and 2.6% had an attempt. Internet addition was associated with suicidal ideation(OR=1.79, 95%CI: 1.32-2.44), plan(OR=2.32, 95%CI: 1.57-3.42), and attempt(OR=2.08, 95%CI:1.03-4.22). After adjustment for potential confounding factors, the association between internet addition and attempt was insignificant. When depression and behaviors related to the impulsity were entered into Hierarchical regression respectively, the associations between internet addition, suicidal ideation and plan were substantially reduced. While internet addition was enter into Hierarchical regression, the associations between depression, behaviors related to the impulsity, suicidal ideation and plan were not reduced. Conclusion Adolescent intemet addition seems to be associated with suicidal ideation and plan, and the association is possibly mediated by depression and behaviors related to the impulsity.
6.Construction of a T7 phage display cDNA library from lung of Microtus fortis

Objective To construct a T7 phage display cDNA library from the lung of Microtus fortis for further screening the schistosomiasis-resistence-related genes of Microtus fortis. Methods mRNA was isolated from total RNA extracted from the lungs of Microtus fortis by TRIzol reagent, and was used to synthesize double strain cDNA by the reverse transcription. Then the double strain cDNA was given with EcoRⅠ and Hind Ⅲ adhering ends by ligation with the directional EcoRⅠ/Hind Ⅲ linkers and digestion with EcoRⅠ and Hind Ⅲ. The double strain cDNA fragments longer than 300 bp in length were fractionated by the Mini Column, and ligated into the T7 Select 10-3b vector with EcoRⅠ and Hind Ⅲ adhering ends. After packaging in vitro, the recombinant T7 Select 10-3b was transformed into BLT5403 to construct a T7 phage display cDNA library. Results The library constructed here contained 1.5?106 clones and the titer of the amplied library was 1.1?1012 pfu/ml. The PCR identification results of 100 clones picked at random showed that 91% clones were recombinant and 90% of recombinant clones contained cDNA fragments longer than 300 bp in length. Conclusion A T7 phage display cDNA library from the lung of Microtus fortis is successfully constructed.
7.Preliminary molecular analysis of bacterial composition in periapical lesions with primary endodontic infections of deciduous teeth.

BACKGROUNDThe bacterial composition of periapical lesions in deciduous teeth has not been well documented. This study was designed to explore the bacterial compositions, especially the dominant bacteria in periapical lesions using 16S rRNA sequencing.
METHODSTissue samples were collected from 11 periapical lesions in deciduous teeth with primary endodontic infections. DNA was extracted from each sample and analyzed using 16S rRNA cloning and sequencing for the identification of bacteria.
RESULTSAll DNA samples were positive for 16S rRNA gene PCR. One hundred and fifty-one phylotypes from 810 clones were identified to eight phyla, and each sample contained an average of 25.9 phylotypes. In addition, 59 phylotypes were detected in more than two samples, and Fusobacterium (F.) nucleatum (8/11), Dialister (D.) invisus (8/11), Campylobacter (C.) gracilis (7/11), Escherichia (E.) coli DH1 (6/11), Aggregatibacter (A.) segnis (6/11), and Streptococcus (S.) mitis (6/11) were the most prevalent species. Furthermore, 45 as-yet-uncultivated phylotypes were also identified.
CONCLUSIONSChronic periapical lesions in deciduous teeth contained polymicrobial infections. F. nucleatum, D. invisus, C. gracilis, E. coli DH1, A. segnis, and S. mitis were the most prevalent species detected by 16S rRNA sequencing.

8.Study on knowledge, attitude and practice regarding AIDS among migrant workers in railway construction sites.

OBJECTIVETo investigate the status and influencing factors on knowledge, attitude and practice (KAP) about AIDS among the migrant workers in railway construction sites and to provide evidence for strategy development and measures on AIDS related health education and behavioral intervention.
METHODS943 migrant workers in railway construction sites were surveyed. Univariate analysis, multivariate Logistic regression and correlation analysis were used to explore their influencing factors.
RESULTSThe migrant workers in railway construction sites had poor AIDS knowledge as a whole. People with high educational level and income had higher AIDS correlative knowledge than other groups. Only 37.5% (275) of 734 migrant workers who had heard about AIDS felt pity for people with HIV/AIDS. There were 39.6%, 7.3% and 6.9% of 943 migrant workers did not object to "pre-marital sex", "prostitution" or "wenching". There were 90 (9.5%) migrant workers admitted having non-marital sex for the last three months but 22 (28.6%) admitted never using condoms,and only 8 (10.4%) had used each time. It appeared that the higher educational level and scores about AIDS knowledge one had,the higher the rate of using a condom. Both longer time interval away from their families and working time were, the lower they would usage a condom.
CONCLUSIONMigrant workers in construction sites were potential high risk population for HIV/AIDS. AIDS health education and behavioral intervention should be carried out

9.Studies on flavonoids from Blumea riparia.

OBJECTIVETo separate and identify the chemical constituents of Blumea riparia.
METHODThe compounds were separated and purified by repeated silica gel, Sephadex LH -20 column chromatographiy. The structures of these compounds isolated were identified by analysis of their spectral data, physical and chemical properties.
RESULTSix flavonoids were isolated from B. riparia. and their structures were identified as eriodictyol-7, 4'-dimethyl ether (1), eriodictyol-7, 3'-dimethyl ether (2), eriodictyol-7-methyl ether (3), quercetin-7, 3', 4'-trimethyl ether (4), tamarixetin (5), rhamnocitrin (6).
CONCLUSIONCompound 1-6 were obtained from B. riparia for the first time.

10.An Analysis of Self-reported Income Status and Related Determinants among Primary Care Providers in a County in Dali

Objective To assess the income status of primary care providers and to explore the determinants of income in a county of Dali. Methods In August 2016, the questionnaire was employed to collect the data of income status of 191 rural health workers and 217 village doctors in the county. Results Through the study, we found that the income of rural health workers in the county was 34, 000 (26, 000, 46,000) yuan with a satisfaction rate of 62.3% (95% CI 55.4%~69.2%) and no change (74.7%) was seen in the income among majorities after implementing the Zero Mark-up Policy for essential medicines. For the village doctors, the income was 20,000 (15,000, 24,000) yuan with a satisfaction rate of 40.6% (95% CI 34.0%~47.1%) and a fall of the income was found in more than half of the doctors after the implement of the policy. Conclusion Health care workers in towns are quite satisfied with their income whereas those in health stations of villages are not content, compared with the average income at the national level. We should increase government's investments on grass-root healthcare team, improve the incentive pay plans and promote the integrated management of health facilities in towns and villages.
