OBJECTIVE:To provide reference information for the adoptation of fast drug dispensing model in hospital pharmacy of China.METHODS:The procedure of fast drug dispensing system(FDDS)was analysed and compared with tradi?tional dispensing system in advantages and disadvantages.RESULTS:FDDS could raise the speed,accuracy of drug dispensing and improve the quality of pharmaceutical service.CONCLUSION:FDDS is suitable for drug dispensing in large-scale hospi?tals.
3.Expression of transcription factor Sp3 in hepatocellular carcinoma

Objective To research the expression of Sp3 andβ‐Catenin in HCC and study the assessable factors of them for prognosis in patients with hepatocellular carcinoma .Methods Western blot and RT‐PCR methods were used to detect the expres‐sion of Sp3 andβ‐Catenin in HCC and the liver tissue beside tumor among 49 cases .We analyzed the difference of these two indexes expressed in HCC and the liver tissue beside tumor .Then we detected the correlation between these two indexes and the character of clinic pathology ,and researched the correlation between Sp3 and the prognosis of HCC .Results The high expression rate of Sp3 in HCC was higher than that of liver tissue beside tumor(P<0 .05) according to Western blot and RT‐PCR ,the same toβ‐Catenin (P<0 .05) .Expression of Sp3 andβ‐Catenin were both related with size of tumor and degree of differentiation .Positive correlation existed between these two indexes according to Western blot method(r=0 .681 ,P=0 .000) and RT‐PCR method(r=0 .641 ,P=0 .000) .The prognosis of cases with high expression of Sp3 was poorer than the low expression cases(P<0 .05) .Conclusion Sp3 plays a promoter role in occurrence of HCC ,which is correlated with the grade malignancy of HCC .Sp3 might participated in occur‐rence and development of HCC via the Wnt pathway .
4.The optimal cryopreservation liquid of the stable transfected cell line named hepatic carcinoma HepG 2 Transfected by Lintivirus

Objective To investigate the optimal cryopreservation liquid of the stable transfected cell line named hepatic carcino-ma HepG2 transfected by lentivirus .Methods Glycerol and DMSO as cryopreservation liguid ,The HepG2 transfected by lentivirus and HepG2 cells were both cryopreserved with four various proportion of cryopreservation liquid .After thawing ,the survival rate of the two cells were observed by inverted contrast microscope and the viability and proliferation were detected with MTT assay .Re-sults Using glycerol as cryoprotectant liquid ,the survival rate of the HepG2 cells transfected by lentivirus was obviously higher than other 3 groups when the proportion of the cryopreservation liquid(DMEM ∶FBS∶Glycerol)was 0∶9∶1(P=0 .001) ,where-as there was no significant difference among the groups of HepG2 cells with various proportion of cryopreservation liquid (P=0 .293) .Using DMSO as cryoprotectant liquid ,the survival rate of the cells transfected by lentivirus was 0% ,whereas the survival rate of the HepG2 cells was higher than 60% ,and there was no significant difference between groups of various proportion of cryo-preservation liquid(P=0 .487) .The MTT assay demonstrated that the higher the serum levels was ,the better proliferation capacity those cells had(P<0 .05) .Conclusion The optimal cryopreservation liquid for the hepatic carcinoma cell line HepG2 transfected by lentivirus is used glycerol as cryoprotectant solution ,and the proportion of cryopreservation solution was 90% FBS + 10%Glycerol .
5.Clinical and neuroimaging features of frontotemporal dementia with parkinsonism linked to chromosome 17

Objective To explore the clinical and neuroimaging features of a frontotemporal dementia with parkinsonism linked to chromosome 17 ( FTDP-17 ) pedigree caused by mutation of microtubule-associated protein tau ( MAPT) gene.Methods The proband and one patient from a FTDP-17 pedigree were assessed through standardized clinical evaluation , neuropsychology assessment , video-electroencephalogrom ,MRI, genetic sequencing , as well as 18 F fludeoxyglucose ( FDG) SPECT for brain metabolism and 11 C 2β-carbomethoxy-3β-( 4-fluoro ) tropane ( CFT ) PET for dopamine transporter ( DAT ) distribution, respectively.Results A FTDP pedigree with 15 patients (6 still alive) was recruited to this study.The proband and one affected patient were genotyped and confirmed as MAPT c .1788T>G mutation. Parkinsonism was the first symptom for both two patients . Personality, speech changes and dementia accompanied with brain atrophy were developed at the later stage in one patient .The 18 F FDG SPECT studies illustrated asymmetric hypometabolism of the temporal , frontal lobes and basal ganglia in two patients . Regarding to the 11 C CFT PET, one affected patient showed asymmetric decreased uptake of tracer in basal ganglia regions.Conclusions FTDP-17 can display a confusingly broad clinical phenotype , with the parkinsonism as the first symptom . Brain glucose metabolism and DAT distribution could be potential biomarkers in early diagnosis of FTDP-17.
7.Proteomics of apoptosis of multiple myeloma cells induced by proteasome inhibitor PS-341

Objective To compare the proteome difference between multiple myeloma cell line U266 cells treated and untreated with PS-341, to investigate the potential drug targets, and to provide theoretical evidence for clinical therapy of multiple myeloma. Methods Two-dimensional gel electrophoresis (2-DE) was performed to separate proteins from treated and untreated U266 cells with proteasome inhibitor PS-341. ImageMaster 2D Platinum software was used to analyze 2-DE image, and matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) was used to identify the differentially expressed proteins. The expression levels of differential protein BAG-2 in the 2 groups of U266 cells lines were detected by Western blot. Results The 2-DE reference pattern of treated and untreated U266 cells with PS-341 was established. A total of 31 differential proteins were identified by MALDI-TOF-MS, 27 of which were down-regulated after PS-341 treatment. The differential expression level of BAG-2 in the 2 groups of U266 cells was confirmed by Western blot. Conclusion Some down-regulated proteins may be the potential drug targets of proteasome inhibitor PS-341.
8.Molecular inversion probe-based single base mutations of hepatitis B virus resistance gene analysis

Objective To establish a molecular inversion probe ( MIP) method for detection of single base drug-resistance mutation in Hepatitis B virus ( HBV) gene.Methods The HBV wild type and YVDD mutant strain were isolated by Daping Hospital of the Third Military Medical University.The MIP was designed and applied to detect the HBV drug-resistance YVDD mutation in one case of wild type and one case of YVDD mutant HBV strain isolated previously.The results of MIP method were compared with that of sequencing to evaluate the detection accuracy.Results Thermal cycling single-base extension and connection reaction performed by Taq DNA Ligase and Ampligase DNA Ligase could ensure the specificity of the detection.The optimum probe concentration of MIP was 1 nmol/L.Through detection of the target gene with different DNA concentrations , the detection sensitivity of MIP was determined as 1 nmol/L.The results of MIP were consistent with that of sequencing method in detection of the clinical samples.Conclusion MIP is successfully used to detect single-base drug-resistance mutation in HBV gene.
9.Clinical application of dynamic neutralization system (K-Rod) in treating multisegmental lumbar degenerative disease.

OBJECTIVETo evaluate the clinical effects of dynamic neutralization system (K-Rod) in treating multisegmental lumbar degenerative disease.
METHODSFrom October 2011 to October 2013, 20 patients with multisegmental lumbar degenerative disease were treated with dynamic neutralization system (K-Rod). There were 8 males and 12 females with an average age of 45.4 years old (ranged from 31 to 65) and an average course of 3.8 years (ranged from 9 months to 6.25 years). All patients had the history of low back and legs pain. Among them, 10 cases were far lateral lumbar disc herniation, 7 cases were lumbar spinal stenosis, 3 cases were lumbar spondylolisthesis (degree I in 2 cases and degree II in 1 case). Every patient had only one responsible segment which causing the symptom would have to be rigidly fixed during operations, and the adjacent intervertebral disc of the responsible segments at least 1 segment has already obvious degenerated. All patients underwent the operation to relieve compressed nerves and reconstruct spinal stability with K-Rod system (the responsible segments were fixed with interbody fusion, and the adjacent segments were fixed with dynamic stabilization). Visual analogue scale (VAS), Japanese Orthopaedic Association Scores (JOA) and Oswestry Disability Index (ODI) were used to evaluate the clinical effects. Imaging data were used to analyze the range of motion (ROM), intervertebral disc height and intervertebral disc signal (according to modified Pfirrmann grading system) in degenerative adjacent segment.
RESULTSAll patients were followed up for more than 1 year, and preoperative symptoms obviously relieved. There were significant differences in VAS, JOA, ODI between preoperative and postoperative (postoperative at 1 week and 1 year) (P<0.05). Radiological examination showed that all responsible segments had already fused, and no looseness, displacement and breakage of internal fixations were found. Postoperative at 1 year, the ROM of adjacent segments were decreased (P<0.05). There was no significant difference in intervertebral disc height between preoperative and postoperative at 1 year (P>0.05). According to modified Pfirrmann grading system to classification for the 25 disks of adjacent segment, 8 disks (32%) got improvement, 15 disks (60%) got no change and 2 disks (8%) got aggravation at 1 year after operation.
CONCLUSIONDynamic neutralization system (K-Rod) combined with interbody fusion could obtain short-term clinical effects in the treatment of multisegmental lumbar degenerative disease.

10.Effects of moxibustion with seed-sized moxa cone at "Ganshu" (BL 18) on liver function in rats with precancerous lesion of hepatic cellular cancer.

OBJECTIVETo explore the effects of moxibustion with seed-sized moxa cone at "Ganshu" (BL 18) on liver furiction and morphology in rat with precancerous lesion of hepatic cellular cancer MCC).
METHODSA total of 60 male Wistar rats were randomly divided into a normal group (10 rats), a model group (20 rats), a 20-day treatment group (15 rats) and a 40-day treatment group (15 rats). HCC model was established by intraperitoneal injection of diethylnitrosamine (DEN). Rats in the normal group received no treatment. Rats in the model group were treated with fixation. Rats in the 20-day treatment group and 40-day treatment group were treated by moxibustion with seed-sized moxa cone at "Ganshu" (BL 18), once every other day, for 20 days and 40 days, respectively. Blood sample in each group was collected 1 d before model establishment, 20 d, 40 d and 84 d after model establishment. Chemical method was applied to test the activity of ALT (alamine aminotransferase), AST (aspartate transaminase) and GGT (glutamyl transpeptidase); at the end of model establishment, all the rats were sacrificed to observe the liver morphology changes.
RESULTSAfter the first therapeutic course, the. content of ALT and AST in the 20-day treatment group was significantly lower than that in the model group (all P<0. 05); after the second therapeutic course, the content of ALT, AST and GGT in the 40-day treatment group was insignificantly lower than that in the model group (all P>0. 05). Under light microscope, the slice of liver tissue indicated that primary tumor was induced in the model group, and the tumor cells were stained and irregular; the cytoplasm in the 20-day treatment group was even, and the tumor cells were few with several nodules alone. In the 40-day treatment group the liver morphology was normal and the staining was even; the tumor cells were few without nodules or a few. Conclusion Moxibustion with seed-sized moxa cone at "Ganshu" (BL 18) could reduce the serum content of ALT, AST and GGT in rats with HCC, which could protect the liver and: delay the DEN-induced precancerous lesion on some levels.
