1.Advances of environmental DNA technology in schistosomiasis surveillance

Abstract: Schistosomiasis, an important zoonotic parasitic disease, is one of the six major tropical diseases identified by WHO, and also one of the most important parasitic diseases for prevention and control in China. After more than 70 years of efforts, the prevention and control of schistosomiasis in China has made great achievements, and the current epidemic of schistosomiasis in China has entered an extremely low epidemic state, but the distribution base of the only intermediate host of schistosomiasis, Oncomelania hupensis, is still large. For now, the techniques used to monitor schistosomiasis have shortcomings such as time-consuming, laborious and low sensitivity, which cannot meet the current needs of China. Environmental DNA (eDNA) refers to DNA that can be extracted from environmental samples (such as soil, water or air) without isolating any target organisms, which is a complex mixture of genomic DNA and its degradation products from different organisms in the same environment. eDNA technology can reflect the community or species composition information in the ecosystem through DNA extraction and detection of environmental samples. Compared with traditional biological monitoring methods, eDNA technology has the advantages of high efficiency, high sensitivity and environmental friendliness. eDNA has been successfully used for the specific detection of Schistosoma mansoni, Schistosoma haematobium and Schistosoma japonicum. This paper reviews the current detection methods of eDNA, the application and technical limitations of eDNA technology in schistosomiasis monitoring, aiming to provide scientific reference for research in the field of schistosomiasis surveillance.
2.Determination of polysaccharides content of Gentiana farreri from different producing areas based on anthrone-sulfuric acid method.

Study a method for the detemination of the content of polysaccharides in Gentiana farreri, and analysis of the content of polysaccharides from different producing areas. The results showed that using the anthrone-sulfuric acid method, simple operation, accurate result. Sample was measured at 620 nm absorbance after anthrone-sulfuric acid color, at this wavelength, solution absorption and glucose showed a good linear relationship; The linearity was in the range of 0.01-0.07 g x L(-1) (r = 0.996 7). The recovery rate was 99.41%, with RSD of 2.0%. Considering the experimental conditions, to determine the solid-liquid ratio 1:60, extracting time 50 min, concentration of ethanol 80%. The mass fraction of polysaccharides was the highest to reached 0.743% in G. farreri from Gansu Xiahe. This experiment has laid a good foundation for further study on G. farreri.

4.Association between ambient temperature and hospital emergency room visits for cardiovascular diseases:a case-crossover study

Objective To explore the association between ambient average temperature and hospital emergency room visits for cardiovascular diseases(International Classification of Diseases,Tenth Vision ICD-10:I00-I99) in Beijing,China.Methods Data was collected on daily hospital emergency room visits for cardiovascular diseases from Peking University Third Hospital,including meteorological data(daily average temperature,relative humidity,wind speed,and atmospheric pressure) from the China Meteorological Data Sharing Service System,and on air pollution from the Beijing Municipal Environmental Monitoring Center.Time-stratified case-crossover design was used to analyze data on 4 seasolls.Results After adjusting data on air pollution,1 degree(℃) increase of ambient average temperature would associate with the emergency room visits of odds ratio(Ors)as 1.282(95%CI:1.250-1.315).1.027(95%CI:1.001-1.055),0.661(95%CI:0.637-0.687),and 0.960 (95%CI:0.937-0.984) in spring,summer,autumn,and winter respectively.After controlling the influence of relative humidity,wind speed,and atmospheric pressure,1℃ increase in the ambient average temperature would be associated with the emergency room visits on Ors value as 1.423 (95%CI:1.377-1.471).1.082(95%CI:1.041-1.124),0.633(95%CI:0.607-0.660)and 0.971(95%CI:0.944-1.000) in spring,summer,auttmm,and winter respectively.Conclusion These data on outcomes suggested that the elevated level of ambient temperature would increase the hospital emergeney room visits for cardiovascular diseases in spring and summer while the elevated level of ambient temperature would decrease the hospital emergency room visits for the cardiovascular diseases in autumn and winter,suggesting that patients with cardiovascular diseases should pay attention to the climate change.
5.Detection of interleukin-8, matrix metalloproteinase-9 and interferon gamma levels in the cerebrospinal fluid of patients with tuberculous meningitis and viral meningitis

Objective To investigate the diagnostic values of interleukin-8 (IL-8), matrix metalloproteinase-9 (MMP-9) and interferon gamma (INF-γ) levels in patients with tuberculous meningitis and viral meningitis by detecting the contents of these biomarkers in the cerebrospinal fluid (CSF). Methods Enzyme-linked immunosorbent assay (ELISA) was used to detect the levels of IL-8,MMP-9 and INF-γ in the CSF of patients with tuberculous meningitis (n=20),viral meningitis (n=15) and noninfectious neurologic diseases (n=20) who admitted to our hospital from August 2010 to November 2011. Results The IL-8,MMP-9 and INF-γlevels in the samples from the tuberculous meningitis patients were significantly higher than those from either viral meningitis or noninfectious neurologic diseases (P<0.05).The contents of IL-8 and MMP-9 in patients with viral meningitis were significantly higher than those in patients with noninfectious neurologic diseases (P<0.05),while that of INF-γ showed no significant difference between the 2 groups (P>0.05). Conclusion Detection of IL-8,MMP-9 and INF-γ in CSF is useful in the diagnosis of tuberculous meningitis; IL-8 and MMP-9 participate in the progress of viral meningitis; the detective value of high concentration of INF-γ in CSF is more significant in diagnosis of tuberculous meningitis than that of IL-8 and MMP-9.
7.Do rice water weevils and rice stem borers compete when sharing a host plant?

The rice water weevil (RWW) Lissorhoptrus oryzophilus Kuschel (Coleoptera: Curculionidae) is an invasive insect pest of rice Oryza sativa L. in China. Little is known about the interactions of this weevil with indigenous herbivores. In the present study, adult feeding and population density of the weevil, injury level of striped stem borer Chilo suppressalis (Walker) (Lepidoptera: Pyralidae) and pink stem borer Sesamia inferens (Walker) (Lepidoptera: Noctuidae) to rice, as well as growth status of their host plants were surveyed in a rice field located in Southeastern Zhejiang, China, in 2004 with the objective to discover interspecific interactions on the rice. At tillering stage, both adult feeding of the weevil and injury of the stem borers tended to occur on larger tillers (bearing 5 leaves) compared with small tillers (bearing 2~4 leaves), but the insects showed no evident competition with each other. At booting stage, the stem borers caused more withering/dead hearts and the weevil reached a higher density on the plants which had more productive tillers and larger root system; the number of weevils per tiller correlated negatively with the percentage of withering/dead hearts of plants in a hill. These observations indicate that interspecific interactions exist between the rice water weevil and the rice stem borers with negative relations occurring at booting or earlier developmental stages of rice.

Abstract: Schistosomiasis is a serious major parasitic disease that threatens human life and health. A better understanding
of the mechanism of host-schistosome interactions is the key to designing new prevention and control strategies. MicroRNAs
(miRNAs) are endogenous small non-coding RNA molecules, which lead to the degradation of the target messenger RNA
(mRNA) or inhibition of its translation in a sequence-specific manner. Both schistosome and its host produce miRNAs, which
can be secreted by extracellular vesicles (EVs). There is accumulating evidence that miRNAs from schistosome can be taken
up by host cells, and finely manipulate the phenotype of host cells for their survival or pathogenesis in a cross-species manner,
even inhibiting the growth and metastases of hepatoma cells. It is still unknown whether host free miRNAs can be taken up by
schistosome, but this phenomenon is highly probable. miRNA-mediated cross-species regulation has emerged as a novel
mechanism for host-schistosome interactions, and this review summarizes the advances in this regard.
9.Observation on therapeutic effect of Jingjin therapy on migraine.

OBJECTIVETo observe therapeutic effect of Jingjin therapy on migraine and search for an ideal way for non-medical treatment of migraine.
METHODSOne hundred cases of migraine were randomly divided into a observation group and a control group, 50 cases in each group. The observation group were treated with Jingjin therapy, and the control group with oral administration of Nimodipine.
RESULTSThe total effective rate was 100.0% in the observation group and 70.0% in the control group with a significant difference between the two groups (P < 0.01). The therapeutic effect for attacking times, attacking lasting time, headache index and accompanied symptoms in the observation group was better than that in the control group (P < 0.01). CONCLUSION Jingjin therapy has an obvious therapeutic effect on migraine.

10.Risk factors for capillary leak syndrome in children with tetralogy after operation.

OBJECTIVETo determine risk factors of capillary leak syndrome(CLS) in children with tetralogy after operation.
METHODSClinical data were retrospectively collected and analyzed from 32 tetralogy cases with CLS and 50 cases without CLS(control group), who received operation under cardiopulmonary bypass (CBP) in our hospital from October 2002 to December 2006. Risk factors with statistical significance were screened with univariate logistic regression analysis, independent risk factors of CLS were determined with multivariate logistic regression analysis. Postoperative outcome was compared between CLS group and control group.
RESULTLogistic analysis showed that the risk factors for CLS were age(OR=6.783), duration of CBP(OR=4.756)and MGoon index (OR=3.826). There were statistical differences in injection of colloid, time of inotropic drugs and ventilation between CLS and control groups(P<0.01). Eight CLS cases underwent peritoneal dialysis and 2 CLS cases died.
CONCLUSIONThe risk factors of CLS in children with tetralogy after CBP are age, duration of CBP and MGoon index.
