2.Clinical efficacy of improved vaginal tightening surgery on vaginal relaxation

Objective To explore more effective methods in vaginal tightening surgery for vaginal relaxation treatment. Methods 132 cases of patients who suffered the decline in quality of sex life owing to vaginal relaxation, requiring improved vaginal tightening surgery. The vaginal surgery was performed to repair the muscles and fascia of the anterior and posterior walls effectively, meanwhile the perineal laceration was also repaired. According to the hammock theory of the treatment of urethral disruption, patients with urinary incontinence with tension was treated to strengthen one suture needle on muscle fascia in the middle of urethra, when repairing anterior of vagina. For patients with constipation, the posterior of vagina was repaired to strengthen 3 suture needles on levator ani during repairing the anterior and posterior wall, so that the vaginal mucosa could get maximum protection. Results 118 patients had been followed up for 2-7 years. A questionnaire survey was conducted about four elements targeted on improving sexual satisfaction, tension incontinence, constipation and vaginal discharge. The clinical results were satisfactory. Conclusions The surgical design is professional and highly effective, and the patients are satisfied with the results. It is suitable for experienced specialists to perform the procedures.
3.Expression of P53 ,Livin and PARP in epithelial ovarian cancer and its impact on chemotherapy resistance and clinical prognosis

Objective To explore the expression of P53 ,Livin and PARP in epithelial ovarian cancer and its correlation with the chemotherapy resistance and clinical prognosis.Methods Seventy-four specimens of epithelial ovarian cancer confirmed by opera-tion and pathology in the gynecology department of our hospital from January 2009 to June 2011 were selected ,followed up and di-vided into chemotherapy sensitivity group and chemotherapy resistance group according to the recurrence condition.The difference of drug resistance genes ,survival rate and influencing factors of survival time were compared between the two groups.Results The positive rates of P53 ,Livin and PARP in the chemotherapy sensitivity group were 47.1% ,56.9% and 52.9% respectively ,which in the chemotherapy resistance group were 73.9% ,95.7% and 95.7% respectively ,the sensitivity rate of each item in the chemother-apy sensitivity group was lower than that in the chemotherapy resistance group (P<0.05).The median survival time in the survival rate in the chemotherapy sensitivity group was 52.0 months ,its 3-year and 5-year survival rates were 68.63% and 41.18% respec-tively ,the medium survival time in the chemotherapy resistance group was 31.5 months ,its 3-year and 5-year survival rates were 47.83% and 26.10% respectively ,the survival rate of various stages in the chemotherapy sensitivity group was higher than that in the chemotherapyresistance group ,the difference was statistically significant (P< 0.05).The age ,pathological differentiation de-gree ,clinical staging and chemotherapy sensitivity degree were introduced into the Cox regression model ,the clinical staging and chemotherapy sensitivity degree had significant influence on the survival time (P<0.05).Conclusion The expression of P53 ,Livin and PARP is correlated with the chemotherapy resistance in the patients with epithelial ovarian cancer ,therefore ,which can be used to predict the chemotherapeutic effect.For the patients with high expression of drug resistance genes ,the personalized coping scheme may be given for improving the patient's prognosis.
4.Vascular endothelial growth factor 165 transfection promotes proliferation of adipose-derived mesenchymal stem cells

BACKGROUND:Vascular endothelial growth factor is the most important pro-angiogenic factor, which can promote revascularization and survival of fat grafts during fat transplantation. OBJECTIVE:To investigate the effect of vascular endothelial growth factor 165 transfection on the proliferation of adipose-derived mesenchymal stem cel s. METHODS:Recombinant vascular endothelial growth factor 165 mRNA fragment was transmitted into adenovirus pAdEasy-1 systems that were packaged to measure viral titer (experimental group). Empty adenovirus was also packaged as control group. Two kinds of packaged adenovirus solution were transferred into adipose-derived mesenchymal stem cel s at the best multiplicity of infection=100. Cel s with no transfection served as blank group. RT-PCR and western blot methods were used to detect the expression of vascular endothelial growth factor 165 at mRNA and protein levels;MTT method was adopted to detect cel proliferation. RESULTS AND CONCLUSION:The expressions of vascular endothelial growth factor 165 mRNA and protein were significantly higher in the experimental group than the control and blank groups (P<0.05). The division and proliferation of transfected adipose-derived mesenchymal stem cel s were increased significantly in the experimental group, which was significantly different from the control and blank groups (P<0.05). These finding indicate that vascular endothelial growth factor 165 transfection cannot only sustain the target protein expression of adipose-derived mesenchymal stem cel s, but also promote the proliferation of adipose-derived mesenchymal stem cel s remarkably.
5.Platelet-derived endothelial cell growth factor transfection of adipose-derived mesenchymal stem cells promotes vascularization of fat grafts

BACKGROUND:Platelet-derived endothelial cel growth factor (PD-ECGF) can promote revascularization in fat transplantation. OBJECTIVE: To explore the dual effects of PD-ECGF and adipose-derived mesenchymal stem cels on the survival rate of fat grafts. METHODS:(1) Adipose-derived mesenchymal stem cels were isolated from the inguinal subcutaneous fat of New Zealand white rabbits, and then cultured. Passage 3 adipose-derived mesenchymal stem cels were divided into experimental group (Lenti-PD-ECGF-EGFP transfected adipose-derived mesenchymal stem cels), control group (Lenti-EGFP transfected adipose-derived mesenchymal stem cels) and blank group (adipose-derived mesenchymal stem cels with no transfection). (2) Lenti-PD-ECGF-EGFP transfected adipose-derived mesenchymal stem cels were cultured in DMEM complete medium, and then mixed with fat tissues as group A; adipose-derived mesenchymal stem cels with no transfection were cultured in DMEM complete medium and then mixed with fat tissues as group B; DMEM complete medium with no cels served as group C. Then, the grafts in groups A, B, C were respectively injected subcutaneously into the upper left, lower left and upper right parts of the rabbits’ black. RESULTS AND CONCLUSION:(1) In the experimental group, PD-ECGF mRNA and protein expressions were significantly higher than those in the control and blank groups (P < 0.05), and cel proliferation was also the fastest. (2) Graft weight and the number of capilaries were greater in group A than groups B and C. These findings indicate that PD-ECGF transfection of adipose-derived mesenchymal stem cels not only can continuously express the PD-ECGF protein, but also can promote the proliferation of adipose-derived mesenchymal stem cels.

Aspheric intraocular lenses(IOLs)have been shown to provide better quality of vision than spherical IOLs.However,these lenses are not ideal for all patients.Surgeons may have different strategies for compensating for the patients's cornea spherical aberration,targeting a postoperative spherical aberration of zero or +0.10 ?m.But we must know that the negative spherical aberration aspheric IOLs require very good centration with respect to the visual axis of the eye.
8.Expression and significance of TRAIL and its receptor DR5 in murine myocardium with acute viral myocarditis

Objective To deterine the expression of TNF-related apoptosis inducing ligand (TRAIL) and its receptor DR5 in murine myocardium with acute viral myocarditis(VM).Methods We builted the model of VM.Eight mice of the VM group and the normal control group were sacrificed on the 7th,10th,14th,21st,28th day after inoculation CVB3 virus.The myocardial histopathological scores were counted.The terminal reansferase-mediated dUPT-biotin nick end-labeling (TUNEL) assays was used to quantified apoptosis rate.The expression of TRAIL and DR5 protein and mRNA were determined by the method of immunohistochemistry and reverse-transcription polymerase chain reaction (RT-PCR).Results The expression of TRAIL and DR5 protein and mRNA were found in myocardium of both the normal control group and the VM group.The expression of TRAIL protein of the VM group(14th) were significantly higher than which in the normal group at the same time and the VM group(7th)(F=9.17,P<0.01).The DR5 protein of the VM group(10th,14th,21st)were significantly higher than which in the normal group at the same time and the VM group(7th)(F=13.32,P<0.01).The expression of TRAIL and DR5 protein were positive correlated with the myocardial histopathological scores and the apoptosis rate.The expression of TRAIL mRNA of the VM group(10th) were significantly higher than which in the normal group at the same time and the VM group(7th)(F=10.86,P<0.01).The DR5 mRNA of the VM group(10th,14th)were significantly higher than which in the normal group at the same time and the VM group(7th)(F=22.75,P<0.01).Conclusion High characteristic expressions of protein/mRNA TRAIL and DR5 were observed in the myocardium of mice with VM.The level was positive correlationed with the account of pathology and the rate of apoptosis.The apoptosis induced by TRAIL and DR5 may participate in the pathophysiological processes of VM.
10. Optimization of extraction process for Xinshenning Tablets by Plackett- Burnman, Box-Behnken, and response surface methodology

Objective: To optimize the extraction process for Xinshenning Tablets. Methods: The extracting condition was optimized by Plackett-Burnman, Box-Behnken, and response surface methodology with the extraction rate, geniposide, and spinosin as indexes. Results: The optimal extracting condition was extracting 3 times with 7.5 fold 70% alcohol, 1.5 h for each time. Conclusion: The optimized process condition is simple and feasible. It can be used in the production.
